    Detection of the toxicity of a candidate compound at an early stage of drug development is an emerging area of interest. It is difficult to determine all of the effects of metabolism of a compound using traditional approaches such as histopathology and serum biochemistry. The goal of a metabolomics approach is to determine all metabolites in a living system, with the potential to detect and identify biomarkers involved in toxicity onset.     Electron Abundance in Protostellar Cores

    The determination of the fractional electron abundance, Xe, in protostellar cores relies on observations of molecules, such as DCO+, H13CO+ and CO, and on chemical models to interpret their abundance. Studies of protostellar cores have revealed significant variations of Xe from core to core within a range 10^-8<Xe<10^-6. The physical origin of these large variations in Xe is not well understood, unless unlikely variations in the cosmic ray ionization rate or ad hoc values of metal depletion are assumed. In this work we explore other potential causes of these variations in Xe, including core age, extinction and density. We compute numerically the intensity of the radiation field within a density distribution generated by supersonic turbulence. Taking into account the lines of sight in all directions, the effective visual extinction in dense regions is found to be always much lower than the extinction derived from the column density along a fixed line of sight. Dense cores with volume and column densities comparable to observed protostellar cores have relatively low mass-averaged visual extinction, 2mag <= A_V <= 5mag, such that photo-ionization can sometimes be as important as cosmic ray ionization. Chemical models, including gas-grain chemistry and time dependent gas depletion and desorption, are computed for values of visual extinction in the range 2mag <= A_V <= 6mag, and for a hydrogen gas density of 10^4cm^-3$, typical of protostellar cores. The models presented here can reproduce some of the observed variations of ion abundance from core to core as the combined effect of visual extinction and age variations.     大学生に必要な能力として高く要求されているものに,コミュニケーション力がある。現在,この力を養成するためのアクティブ・ラーニングを中心とした授業を行っている。この中でコミュニケーション力が養成されているかを検証するため,コミュニケーション力の測定を授業初回と最終回に実施した。この測定では,「小グループ」,「集会」,「会話」,「スピーチ」の場面についての測定を行った。これに加えて,学生の授業の振り返りを検討し,授業をとおして効果的にコミュニケーション力が養成されているのかについて,検証した。この結果,コミュニケーションについてのそれぞれの場面での感じ方は,個人差があることが明らかになった。また,コミュニケーションに関する苦手意識があっても,コミュニケーション行動はできていることが,明らかになった。Communication skills are considered to be very important for university students. The measurement of communication skills was conducted at the first and the last lessons of a communication skills enhancement course using an active learning approach, in order to assess its effectiveness. The measurement included four aspects, namely, small group activity, meeting, conversation, and speech. In addition, the students\u27 reflection sheets were analyzed to determine whether their communication abilities were improved through the course. The results show that perceptions on communication in each of the above-mentioned aspects vary between individuals. It is also suggested that communicative actions can be taken, even when challenged by negative attitudes towards communication

    γ-Tocopherol Accelerated Sodium Excretion in a Dose-Dependent Manner in Rats with a High Sodium Intake

    We have previously reported that γ-tocopherol (γ-Toc) displays a natriuretic potency in rats fed a NaCl diet and administered 20 mg γ-Toc. In this study, we investigated whether γ-Toc has natriuretic potency at a dose lower or higher than 20 mg in rats given a NaCl diet. Male rats were fed a control diet or a NaCl diet and administered either placebo or 10, 20 or 40 mg of γ-Toc. The rat urine was collected for 24 hours (divided into 6 hour periods) and the 2,7,8-trimethyl-2-(2'-carboxyethyl)-6-hydroxychroman (γ-CEHC) level, the sodium excretion content, and the urine volume were determined. The 24-hour γ-CEHC and sodium levels in the urine of the NaCl groups given 20 mg or 40 mg γ-Toc were significantly higher than those in the placebo group. The peak levels of urine sodium and γ-CEHC in the NaCl group given 40 mg γ-Toc appeared at 0–6 h, which was a more rapid increase than that seen in the group given 20 mg γ-Toc. The 24-hour urine volumes of the NaCl groups given 10 and 20 mg γ-Toc were significantly higher than the urine volume of the placebo group. Our findings suggested that γ-Toc increased sodium excretion in a dose-dependent manner in rats fed a NaCl diet. Moreover, a high dose of γ-Toc may accelerate its metabolism and cause an increase in the rate of sodium excretion

    Development and applicability of Hospital Survey on Patient Safety Culture (HSOPS) in Japan

    <p>Abstract</p> <p>Background</p> <p>Patient safety culture at healthcare organizations plays an important role in guaranteeing, improving and promoting overall patient safety. Although several conceptual frameworks have been proposed in the past, no standard measurement tool has yet been developed for Japan.</p> <p>Methods</p> <p>In order to examine possibilities to introduce the Hospital Survey on Patient Safety Culture (HSOPS) in Japan, the authors of this study translated the HSOPS into Japanese, and evaluated its factor structure, internal consistency, and construct validity. Healthcare workers (n = 6,395) from 13 acute care general hospitals in Japan participated in this survey.</p> <p>Results</p> <p>Confirmatory factor analysis indicated that the Japanese HSOPS' 12-factor model was selected as the most pertinent, and showed a sufficiently high standard partial regression coefficient. The internal reliability of the subscale scores was 0.46-0.88. The construct validity of each safety culture sub-dimension was confirmed by polychoric correlation, and by an ordered probit analysis.</p> <p>Conclusions</p> <p>The results of the present study indicate that the factor structures of the Japanese and the American HSOPS are almost identical, and that the Japanese HSOPS has acceptable levels of internal reliability and construct validity. This shows that the HSOPS can be introduced in Japan.</p

    A noncoding RNA gene on chromosome 10p15.3 may function upstream of hTERT

    <p>Abstract</p> <p>Background</p> <p>We attempted to clone candidate genes on 10p14–15 which may regulate hTERT expression, through exon trapping using 3 BAC clones covering the region. After obtaining 20 exons, we examined the function of RGM249 (RGM: RNA gene for miRNAs) we cloned from primary cultured human hepatocytes and hepatoma cell lines. We confirmed approximately 20 bp products digested by Dicer, and investigated the function of this cloned gene and its involvement in hTERT expression by transfecting the hepatoma cell lines with full-length dsRNA, gene-specific designed siRNA, and shRNA-generating plasmid.</p> <p>Results</p> <p>RGM249 showed cancer-dominant intense expression similar to hTERT in cancer cell lines, whereas very weak expression was evident in human primary hepatocytes without telomerase activity. This gene was predicted to be a noncoding precursor RNA gene. Interestingly, RGM249 dsRNA, siRNA, and shRNA inhibited more than 80% of hTERT mRNA expression. In contrast, primary cultured cells overexpressing the gene showed no significant change in hTERT mRNA expression; the overexpression of the gene strongly suppressed hTERT mRNA in poorly differentiated cells.</p> <p>Conclusion</p> <p>These findings indicate that RGM249 might be a microRNA precursor gene involved in the differentiation and function upstream of hTERT.</p
