AbstractObjectiveTo investigate capacity of Hyptis suaveolens (H. suaveolens) methanol extract as an antioxidant to protect against carbon tetrachloride (CCl4)-induced oxidative stress, hepatotoxicity in Albino Wistar rats and cytoprotective effect of hydrogen peroxide (H2O2) induced cell death in HepG2 cell line.MethodsTwo different doses of methanol extract of H. suaveolens were evaluated for the hepatoprotective activity against carbon tetrachloride (CCl4) induced hepatotoxicity in rats. Animals in Group I: served as control, group II: H. suaveolens (100 mL/kg b.w), group III: H. suaveolens (50 mL/kg b.w) + CCl4 (1 mg/kg), group IV: H. suaveolens (100 mL/kg b.w) + CCl4 (1 mL/kg) and group V: CCl4 (1 mL/kg). Histopathologic changes of liver were also evaluated. Cytotoxicity was also determined by 3, (4,5-dimethylthiazol-2-yl)2,5-diphenyl-tetrazolium bromide (MTT) assay.ResultsOral sigle dose treatment of CCl4 produced a marked elevation in the serum levels of aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP) and Lactate dehydrogenase (LDH). Histopathological analysis of the liver of CCl4-induced rats revealed marked liver cell necrosis with inflammatory collections that were conformed to increase in the levels of SOD, GSH, GST, GR and LPO. Treatment with H2O2 significantly induced death of HepG2 cell. Pretreatment with H. suaveolens methanol extract inhibited or attenuated H2O2 induced cytotoxicity.ConclusionsThis study shows that H. suaveolens methanol extract can be proposed to protect the liver against CCl4-induced oxidative damage in rats and protect the cells against H2O2-induced oxidative damage in HepG2 cells. The hepatoprotective and cytoprotective effects might be correlated with its antioxidant and free radical scavenger effects

Analisis Karakteristik Pekarangan dalam Mendukung Penganekaragaman Pangan Keluarga di Kabupaten Bogor
Pekarangan is a type of traditional Indonesian home gardens that utilize the land around the house with the status and clear boundaries. Pekarangan in rural areas have high agrobiodiversity, good agroecosystem and should be optimized as an area to meet the needs of daily life, especially to support the diversification of food consumption of the household. Therefore, the purpose of this study is to analyze the ecology characteristics of pekarangan to support food consumption diversification of the household. The study was conducted in Bogor Regency, which located in altitudes at 165 – 460 m height with high level of urbanization, from December 2013 to June 2014. Diversity of food in pekarangan, specially the crops and livestocks, is the focus of this research. The results of the study showed that the most common crops in Bogor Regency are vegetables, fruits, and spices plants. Analysis of plant diversity index showed that the pekarangan in Bogor Regency has a diversity in the mid category (1.95) with the dominance of seasonal crops. Diversification of food in pekarangan can be done by optimizing the existing area, utilization of all zoning for the cultivation of diverse functions crops and livestock. For diversification of food consumption, the owner should use different types of food, as well as considering the crop calendar to a wide variety of food that can be consumed on a daily basis. Therefore, it's necessary to educate the housewives about the potency of diversification crops in pekarangan for consumption, strong motivation and assistance from the government, as well as optimizing the role of KWT group to optimize the role of pekarangan for the diversification of food

Biomarker discovery and redundancy reduction towards classification using a multi-factorial MALDI-TOF MS T2DM mouse model dataset
Diabetes like many diseases and biological processes is not mono-causal. On the one hand multifactorial studies with complex experimental design are required for its comprehensive analysis. On the other hand, the data from these studies often include a substantial amount of redundancy such as proteins that are typically represented by a multitude of peptides. Coping simultaneously with both complexities (experimental and technological) makes data analysis a challenge for Bioinformatics
New Vesicovaginal Fistula Formation in a Rabbit Model
Objective: To create an animal model of vesicovaginal fistula as a modality to develop therapy. Material and Methods: This study was an preliminary experimental study to create new fistula vesicovaginal formation in a rabbit model. Six female New Zealand white rabbits of Stem Cell Research and Development Center of Universitas Airlangga were used in this study and were divided into 3 groups in which there were 2 rabbits in each group. Vesicovaginal fistula formation was performed through laparotomy surgery. After the abdomen is opened, the Nasogastric tube is inserted vaginally into the anterior vaginal wall. The incision is made on the anterior vaginal wall and the posterior wall of the bladder. The Nasogastric tube that comes out of the vagina is inserted into the bladder incision to maintain the formation of vesicovaginal fistula. Both mucosal walls are sutured and tested to determine if there is any leak. Evaluation the model of vesicovaginal fistula was carried out 3 weeks postoperatively according the group. Result: Four out of six rabbits have formed vesicovaginal fistulas, which are from groups two and three. There are no complications during the treatment period until termination Conclusion: Rabbits have the potential to become experimental animals for the vesicovaginal fistula model. The best time to release a nasogastric tube to maintain a fistula is 2 weeks
Adaptation to acetaminophen exposure elicits major changes in expression and distribution of the hepatic proteome.
Acetaminophen overdose is the leading cause of acute liver failure. One dose of 10-15 g causes severe liver damage in humans, whereas repeated exposure to acetaminophen in humans and animal models results in autoprotection. Insight of this process is limited to select proteins implicated in acetaminophen toxicity and cellular defence. Here we investigate hepatic adaptation to acetaminophen toxicity from a whole proteome perspective, using quantitative mass spectrometry. In a rat model, we show the response to acetaminophen involves the expression of 30% of all proteins detected in the liver. Genetic ablation of a master regulator of cellular defence, NFE2L2, has little effect, suggesting redundancy in the regulation of adaptation. We show that adaptation to acetaminophen has a spatial component, involving a shift in regionalisation of CYP2E1, which may prevent toxicity thresholds being reached. These data reveal unexpected complexity and dynamic behaviour in the biological response to drug-induced liver injury
