    An alkaliphilic cyclodextrin glycosyltransferase from a new Bacillus agaradhaerens WN-I strain isolated from an Egyptian soda lake: Purification and properties

    Alkaliphilic bacteria were isolated from soil and water samples obtained from Egyptian soda lakes in the Wadi Natrun area. Screening for cyclodextrin glycosyltransferase- producing alkaliphilic bacteria resulted in the isolation of 15 positive strains. Strain WN-I was selected as the best producer of CGTase. 16S rDNA sequence analysis and DNA-DNA hybridization identified the isolate as Bacillus agaradhaerens. The enzyme was purified to homogeneity up to 21 fold by starch adsorption and anion exchange chromatography with a yield of 26.40%. The pure enzyme was a monomer with an estimated molecular weight of 85 kDa. The enzyme was stable, at 25°C, over a pH range of 5.0 to 11, with a maximum activity at pH 9.0. The enzyme activity exhibited an optimum temperature of 55°C and was stable at 40°C for at least 1 h. Thermal stability was improved in the presence of maltodextrin, starch or CaCl2. The enzyme was slightly stimulated by CaCl2, KC1 and BaCl2 but was completely inhibited in the presence of FeCl2 and strongly inhibited by ZnCl2 and CoCl2 and to a lower extent by CuCl2, EDTA, 2-mercaptoethanol, and dithiothritol. The enzyme produced mainly β-CD (71.20% of the total cyclodextrin amount).     Hospital pharmacy practice in Saudi Arabia: Prescribing and transcribing in the Riyadh region

    AbstractPurposeThe purpose of this survey is to outline pharmacy services in hospitals on a regional level in the Kingdom of Saudi Arabia.MethodsA modified-American Society of Health-System Pharmacists (ASHP) survey questionnaire as pertinent to Saudi Arabia was used to conduct a national survey. After discussing with the pharmacy directors of 48 hospitals in the Riyadh region over the phone on the survey’s purpose, the questionnaires were personally delivered and collected upon completion. The hospital lists were drawn from the Ministry of Health hospital database.ResultsTwenty-nine hospitals participated in the survey giving a response rate of 60.4%. Approximately 60% of the hospitals which participated in the survey required prior approval for the use of non-formulary medications. About 83.3% of hospitals reviewed compliance with clinical practice guidelines and 72.7% hospitals reported that pharmacists are also actively involved in these activities. Pharmacists in more than 95% of hospitals provided consultations on drug information. A staff pharmacist routinely answering questions was the most frequently cited (74.1%) method by which objective drug information was provided to prescribers. Electronic drug information resources were available in 77.7% of hospitals, although internet use is not widely available to hospital pharmacists, with only 58.6% of hospitals providing pharmacist access to the internet. About, 34.5% of hospitals had computerized prescriber order entry (CPOE) systems with clinical decision-support systems (CDSSs) and 51.9% of the hospitals had electronic medical record (EMR) system.ConclusionHospital pharmacists are increasingly using electronic technologies to improve prescribing and transcribing of medications in Saudi Arabia

    Early Diagnosis of Brain Tumour MRI Images Using Hybrid Techniques between Deep and Machine Learning.

    Cancer is considered one of the most aggressive and destructive diseases that shortens the average lives of patients. Misdiagnosed brain tumours lead to false medical intervention, which reduces patients' chance of survival. Accurate early medical diagnoses of brain tumour are an essential point for starting treatment plans that improve the survival of patients with brain tumours. Computer-aided diagnostic systems have provided consecutive successes for helping medical doctors make accurate diagnoses and have conducted positive strides in the field of deep and machine learning. Deep convolutional layers extract strong distinguishing features from the regions of interest compared with those extracted using traditional methods. In this study, different experiments are performed for brain tumour diagnosis by combining deep learning and traditional machine learning techniques. AlexNet and ResNet-18 are used with the support vector machine (SVM) algorithm for brain tumour classification and diagnosis. Brain tumour magnetic resonance imaging (MRI) images are enhanced using the average filter technique. Then, deep learning techniques are applied to extract robust and important deep features via deep convolutional layers. The process of combining deep and machine learning techniques starts, where features are extracted using deep learning techniques, namely, AlexNet and ResNet-18. These features are then classified using SoftMax and SVM. The MRI dataset contains 3,060 images divided into four classes, which are three tumours and one normal. All systems have achieved superior results. Specifically, the AlexNet+SVM hybrid technique exhibits the best performance, with 95.10% accuracy, 95.25% sensitivity, and 98.50% specificity

    In vitro growth inhibition of bloodstream forms of Trypanosoma brucei and Trypanosoma congolense by iron chelators

    African trypanosomes exert significant morbidity and mortality in man and livestock. Only a few drugs are available for the treatment of trypanosome infections and therefore, the development of new anti-trypanosomal agents is required. Previously it has been shown that bloodstream-form trypanosomes are sensitive to the iron chelator deferoxamine. In this study the effect of 13 iron chelators on the growth of Trypanosoma brucei, T. congolense and human HL-60 cells was tested in vitro. With the exception of 2 compounds, all chelators exhibited anti-trypanosomal activities, with 50% inhibitory concentration (IC(50)) values ranging between 2.1 – 220 μM. However, the iron chelators also displayed cytotoxicity towards human HL-60 cells and therefore, only less favourable selectivity indices compared to commercially available drugs. Interfering with iron metabolism may be a new strategy in the treatment of trypanosome infections. More specifically, lipophilic iron-chelating agents may serve as lead compounds for novel anti-trypanosomal drug development
