    Study on Reservoir Characteristics of Ta112 in Songliao Basin

    Ta34 in Songliao basin is buried 1,300 metres below the band, the exploration difficulty is big and high quality recognition is difficult. According to its complex lithology characteristics, poor porosity and strong heterogeneity, with the application of comprehensive conventional coring, thin-section analysis, scanning electron microscopy and conventional curve data for analysis of reservoir characteristics of T112, lithology and reservoir criterion are established, more over the fracture strata section of the hill is identified. The reservoirs are mainly composed of carbonate rocks and quartzite, the accurate lithology identification provides geological basis for the next step of well deployment

    Optimization of Millet Axial Flow Threshing and Separation Device Based on Discrete Element Method

    The difficulties of threshing and separation of millet have not been solved yet which has restricted the development of the millet industry because of the special biological structure and lack of professional agricultural machinery. In order to improve the quality of millet harvest and meet the market demand for millet, in this paper, according to the branching structure of millet, the millet earhead model was established by Discrete Element Method. Using virtual models of millet and device, the simulation tests were carried out whose results have shown that the threshing effect of the rasp-bar threshing element is better than that of the teeth threshing element. Then the rotor structure was optimized into a combined type of the rasp-bar and the teeth. A three-factor five-level quadratic orthogonal rotation combination test was carried out whose results have shown that the combined rotor can meet the requirements of millet harvest

    Multifunctional gold nanostar conjugates for tumor imaging and combined photothermal and chemo-therapy

    Uniform gold nanostars (Au NS) were conjugated with cyclic RGD (cRGD) and near infrared (NIR) fluorescence probe (MPA) or anti-cancer drug (DOX) to obtain multi-functional nanoconstructs, Au-cRGD-MPA and Au-cRGD-DOX respectively. The NIR contrast agent Au-cRGD-MPA was shown to have low cytotoxicity. Using tumor cells and tumor bearing mice, these imaging nanoparticles demonstrated favorable tumor-targeting capability mediated by RGD peptide binding to its over-expressed receptor on the tumor cells. The multi-therapeutic analogue, Au-cRGD-DOX, integrates targeting tumor, chemotherapy and photo-thermotherapy into a single system. The synergistic effect of photo-thermal therapy and chemotherapy was demonstrated in different tumor cell lines and in vivo using S180 tumor-bearing mouse models. The viability of MDA-MB-231 cells was only 40 % after incubation with Au-cRGD-DOX and irradiation with NIR light. Both tail vein and intratumoral injections showed Au-cRGD-DOX treated mice exhibiting the slowest tumor increase. These results indicate that the multifunctional nanoconstruct is a promising combined therapeutic agent for tumor-targeting treatment, with the potential to enhance the anti-cancer treatment outcomes

    A proteome map of primary cultured rat Schwann cells

    <p>Abstract</p> <p>Background</p> <p>Schwann cells (SCs) are the principal glial cells of the peripheral nervous system with a wide range of biological functions. SCs play a key role in peripheral nerve regeneration and are involved in several hereditary peripheral neuropathies. The objective of this study was to gain new insight into the whole protein composition of SCs.</p> <p>Results</p> <p>Two-dimensional liquid chromatography coupled with tandem mass spectrometry (2D LC-MS/MS) was performed to identify the protein expressions in primary cultured SCs of rats. We identified a total of 1,232 proteins, which were categorized into 20 functional classes. We also used quantitative real time RT-PCR and Western blot analysis to validate some of proteomics-identified proteins.</p> <p>Conclusion</p> <p>We showed for the first time the proteome map of SCs. Our data could serve as a reference library to provide basic information for understanding SC biology.</p

    Gastric Adenocarcinoma Predictive Long Intergenic Non-Coding RNA Promotes Tumor Occurrence and Progression in Non-Small Cell Lung Cancer via Regulation of the miR-661/eEF2K Signaling Pathway

    Background/Aims: Long non-coding RNAs (lncRNAs) play vital roles in carcinogenesis as oncogenes or tumor suppressor genes. This study explored the biological function of lncRNA gastric adenocarcinoma predictive long intergenic non-coding RNA (GAPLINC) in human non-small cell lung cancer (NSCLC). Methods: GAPLINC expression in NSCLC specimens and cell lines was detected by qRT-PCR and Western blot. The effect of GAPLINC on cell proliferation was investigated using CCK8-assay, colony formation assay, and xenograft model. The effects of GAPLINC on apoptosis and cell cycle were determined using flow cytometry. The mechanism of GAPLINC involved in NSCLC was explored using Western blot, luciferase reporter assay, and RNA fluorescence in situ hybridization. Results: We found that GAPLINC expression was up-regulated in NSCLC tissues and cell lines. Overexpression of GAPLINC was associated with poor prognosis in patients with NSCLC. Silencing of GAPLINC significantly inhibited cell proliferation, promoted apoptosis, and induced cell cycle arrest in the G0/G1 phase. Results from xenograft transplantation showed that GAPLINC silencing inhibited the tumor growth in vivo. Interestingly, GAPLINC silencing decreased the expression of eukaryotic elongation factor-2 kinase (eEF2K) protein both in vivo and in vitro. Bioinformatic analysis and luciferase reporter confirmed that miR-661 targeted GAPLINC and eEF2K 3’-UTR and was negatively correlated with the expression of GAPLINC and eEF2K. Conclusion: Our findings indicate that GAPLINC promotes NSCLC tumorigenesis by regulating miR-661/eEF2K cascade and provide new insights for the pathogenesis underlying NSCLC and potential targets for therapeutic strategy
