    An electrostatic elliptical mirror for neutral polar molecules

    Focusing optics for neutral molecules finds application in shaping and steering molecular beams. Here we present an electrostatic elliptical mirror for polar molecules consisting of an array of microstructured gold electrodes deposited on a glass substrate. Alternating positive and negative voltages applied to the electrodes create a repulsive potential for molecules in low-field-seeking states. The equipotential lines are parallel to the substrate surface, which is bent in an elliptical shape. The mirror is characterized by focusing a beam of metastable CO molecules and the results are compared to the outcome of trajectory simulations.Comment: 5 pages, 4 figure

    The use of storm fragments and biodegradable replanting methods allows for a low-impact habitat restoration method of seagrass meadows, in the eastern Aegean Sea

    Seagrasses are important marine ecosystems but are vulnerable to physical damage from anthropogenic activities such as anchoring and trawling. Replanting damaged areas can represent a viable restoration strategy, yet current methods rely on the removal of plants from existing meadows and in some cases the use of non-sustainable planting materials. In this paper, we present evidence of a sustainable replanting strategy. Storm fragments of the endemic Mediterranean seagrass, neptune grass Posidonia oceanica were collected from the shore and shallow water, both the plagiotropic and orthotropic (horizontal and vertical) growth forms were then replanted using one of two biodegradable materials, coconut fibre pots or bamboo stakes, to secure them to the seafloor. Establishment of plagiotropic fragments were increased by bamboo anchorage (x̅ = 89% SE ± 0%) compared to orthotropic storm fragments (x̅ = 66.5% SE ± 6.5%). By contrast a coconut fibre method resulted in greater establishment of orthotropic fragments (x̅ = 79% SE ± 7%) compared to plagiotropic (x̅ = 51% SE ± 11%). Fragments showed some blade growth, but little shoot growth after 15 months. The fragment shoot and blade growth did not differ between the plagiotropic or orthotropic fragments replanted by bamboo stakes or coconut fibre pot. Our results suggest that the use of storm fragments and biodegradable anchoring materials constitutes a viable, non-destructive replanting technique in seagrass restoration. Furthermore success can be increased by selecting a growth-form appropriate planting method

    We report here the genome sequence of a clinical isolate of <i>Staphylococcus aureus</i> from an orthopedic infection. Phenotypically diverse <i>Staphylococcus aureus</i> strains are associated with orthopedic infections and subsequent implant failure, and some are highly resistant to antibiotics. This genome sequence will support further analyses of strains causing orthopedic infections

    Lessons Learned in Applying the U.S. EPA Proposed Cancer Guidelines to Specific Compounds

    An expert panel was convened to evaluate the U.S. Environmental Protection Agency’s “Proposed Guidelines for Carcinogen Risk Assessment” through their application to data sets for chloroform (CHCl3) and dichloroacetic acid (DCA). The panel also commented on perceived strengths and limitations encountered in applying the guidelines to these specific compounds. This latter aspect of the panel’s activities is the focus of this perspective. The panel was very enthusiastic about the evolution of these proposed guidelines, which represent a major step forward from earlier EPA guidance on cancer-risk assessment. These new guidelines provide the latitude to consider diverse scientific data and allow considerable flexibility in dose-response assessments, depending on the chemical’s mode of action. They serve as a very useful template for incorporating state-of-the-art science into carcinogen risk assessments. In addition, the new guidelines promote harmonization of methodologies for cancer- and noncancer-risk assessments. While new guidance on the qualitative decisions ensuing from the determination of mode of action is relatively straightforward, the description of the quantitative implementation of various risk-assessment options requires additional development. Specific areas needing clarification include: (1) the decision criteria for judging the adequacy of the weight of evidence for any particular mode of action; (2) the role of mode of action in guiding development of toxicokinetic, biologically based or case-specific models; (3) the manner in which mode of action and other technical considerations provide guidance on margin-of-exposure calculations; (4) the relative roles of the risk manager versus the risk assessor in evaluating the margin of exposure; and (5 ) the influence of mode of action in harmonizing cancer and noncancer risk assessment methodologies. These points are elaborated as recommendations for improvements to any revisions. In general, the incorporation of examples of quantitative assessments for specific chemicals would strengthen the guidelines. Clearly, any revisions should retain the emphasis present in these draft guidelines on flexibility in the use of scientific information with individual compounds, while simultaneously improving the description of the processes by which these mode-of-action data are organized and interpreted

    Transfected CD59 protects mesangial cells from injury induced by antibody and complement. CD59 is a complement regulatory protein on the glomerular cells that inhibits C5b-9 assembly and insertion. We employed an overexpression strategy to determine the functional significance of CD59 in mesangial cells. We made a CD59 expression vector tagged with FLAG utilizing site-directed mutagenesis and PCR, which allows transfected CD59 to be distinguished from the constitutively expressed protein. In stable clones, overexpressed CD59 was clearly detected immunocytochemically both by anti-FLAG and anti-CD59 antibody in a granular pattern. The overexpression of CD59 was also confirmed by Western blotting. To determine if overexpression of CD59 by mesangial cells protected these cells from C5b-9 attack, we performed complement-mediated cell lysis assays. CD59-transfected mesangial cells demonstrated marked resistance to complement-mediated cell lysis which was reversed in the presence of antibody to CD59. We also investigated the role of CD59 in protecting cells from the effects of membrane insertion of sublytic quantities of C5b-9. Overexpressed CD59 suppressed production of superoxide, one of the inflammatory mediators induced by sublytic C5b-9 attack. These results demonstrate directly that transfected CD59 functions as a potent protector of mesangial cells against both lytic and sublytic attack by C5b-9. CD59 may be an important regulator of complement-mediated disease in the glomerular mesangium

    Epidemiology, prehospital care and outcomes of patients arriving by ambulance with dyspnoea: An observational study

    Background: This study aimed to determine epidemiology and outcome for patients presenting to emergency departments (ED) with shortness of breath who were transported by ambulance. Methods: This was a planned sub-study of a prospective, interrupted time series cohort study conducted at three time points in 2014 and which included consecutive adult patients presenting to the ED with dyspnoea as a main symptom. For this sub-study, additional inclusion criteria were presentation to an ED in Australia or New Zealand and transport by ambulance. The primary outcomes of interest are the epidemiology and outcome of these patients. Analysis was by descriptive statistics and comparisons of proportions. Results: One thousand seven patients met inclusion criteria. Median age was 74 years (IQR 61-68) and 46.1 % were male. There was a high rate of co-morbidity and chronic medication use. The most common ED diagnoses were lower respiratory tract infection (including pneumonia, 22.7 %), cardiac failure (20.5%) and exacerbation of chronic obstructive pulmonary disease (19.7 %). ED disposition was hospital admission (including ICU) for 76.4 %, ICU admission for 5.6 % and death in ED in 0.9 %. Overall in-hospital mortality among admitted patients was 6.5 %. Discussion: Patients transported by ambulance with shortness of breath make up a significant proportion of ambulance caseload and have high comorbidity and high hospital admission rate. In this study, >60 % were accounted for by patients with heart failure, lower respiratory tract infection or COPD, but there were a wide range of diagnoses. This has implications for service planning, models of care and paramedic training. Conclusion: This study shows that patients transported to hospital by ambulance with shortness of breath are a complex and seriously ill group with a broad range of diagnoses. Understanding the characteristics of these patients, the range of diagnoses and their outcome can help inform training and planning of services

    AA-Amyloidosis Can Be Transferred by Peripheral Blood Monocytes

    Spongiform encephalopathies have been reported to be transmitted by blood transfusion even prior to the clinical onset. Experimental AA-amyloidosis shows similarities with prion disease and amyloid-containing organ-extracts can prime a recipient for the disease. In this systemic form of amyloidosis N-terminal fragments of the acute-phase reactant apolipoprotein serum amyloid A are the main amyloid protein. Initial amyloid deposits appear in the perifollicular region of the spleen, followed by deposits in the liver. We used the established murine model and induced AA-amyloidosis in NMRI mice by intravenous injections of purified amyloid fibrils (‘amyloid enhancing factor’) combined with inflammatory challenge (silver nitrate subcutaneously). Blood plasma and peripheral blood monocytes were isolated, sonicated and re-injected into new recipients followed by an inflammatory challenge during a three week period. When the animals were sacrificed presence of amyloid was analyzed in spleen sections after Congo red staining. Our result shows that some of the peripheral blood monocytes, isolated from animals with detectable amyloid, contained amyloid-seed that primed for AA-amyloid. The seeding material seems to have been phagocytosed by the cells since the AA-precursor (SAA1) was found not be expressed by the monocytes. Plasma recovered from mice with AA amyloidosis lacked seeding capacity. Amyloid enhancing activity can reside in monocytes recovered from mice with AA-amyloidosis and in a prion-like way trigger amyloid formation in conjunction with an inflammatory disorder. Human AA-amyloidosis resembles the murine form and every individual is expected to be exposed to conditions that initiate production of the acute-phase reactant. The monocyte-transfer mechanism should be eligible for the human disease and we point out blood transfusion as a putative route for transfer of amyloidosis

    An Assessment of the Effectiveness of High Definition Cameras as Remote Monitoring Tools for Dolphin Ecology Studies.

    Research involving marine mammals often requires costly field programs. This paper assessed whether the benefits of using cameras outweighs the implications of having personnel performing marine mammal detection in the field. The efficacy of video and still cameras to detect Indo-Pacific bottlenose dolphins (Tursiops aduncus) in the Fremantle Harbour (Western Australia) was evaluated, with consideration on how environmental conditions affect detectability. The cameras were set on a tower in the Fremantle Port channel and videos were perused at 1.75 times the normal speed. Images from the cameras were used to estimate position of dolphins at the water’s surface. Dolphin detections ranged from 5.6 m to 463.3 m for the video camera, and from 10.8 m to 347.8 m for the still camera. Detection range showed to be satisfactory when compared to distances at which dolphins would be detected by field observers. The relative effect of environmental conditions on detectability was considered by fitting a Generalised Estimation Equations (GEEs) model with Beaufort, level of glare and their interactions as predictors and a temporal auto-correlation structure. The best fit model indicated level of glare had an effect, with more intense periods of glare corresponding to lower occurrences of observed dolphins. However this effect was not large (-0.264) and the parameter estimate was associated with a large standard error (0.113).The limited field of view was the main restraint in that cameras can be only applied to detections of animals observed rather than counts of individuals. However, the use of cameras was effective for long term monitoring of occurrence of dolphins, outweighing the costs and reducing the health and safety risks to field personal. This study showed that cameras could be effectively implemented onshore for research such as studying changes in habitat use in response to development and construction activities
