Overexpression of mitochondrial if1 prevents metastatic disease of colorectal cancer by enhancing anoikis and tumor infiltration of NK cells

Increasing evidences show that the ATPase Inhibitory Factor 1 (IF1), the physiological inhibitor of the ATP synthase, is overexpressed in a large number of carcinomas contributing to metabolic reprogramming and cancer progression. Herein, we show that in contrast to the findings in other carcinomas, the overexpression of IF1 in a cohort of colorectal carcinomas (CRC) predicts less chances of disease recurrence, IF1 being an independent predictor of survival. Bioinformatic and gene expression analyses of the transcriptome of colon cancer cells with differential expression of IF1 indicate that cells overexpressing IF1 display a less aggressive behavior than IF1 silenced (shIF1) cells. Proteomic and functional in vitro migration and invasion assays confirmed the higher tumorigenic potential of shIF1 cells. Moreover, shIF1 cells have increased in vivo metastatic potential. The higher metastatic potential of shIF1 cells relies on increased cFLIP-mediated resistance to undergo anoikis after cell detachment. Furthermore, tumor spheroids of shIF1 cells have an increased ability to escape from immune surveillance by NK cells. Altogether, the results reveal that the overexpression of IF1 acts as a tumor suppressor in CRC with an important anti-metastatic role, thus supporting IF1 as a potential therapeutic target in CRCThis research was funded by grants from Ministerio de Ciencia, Innovación y Universidades (SAF2013-41945-R, SAF2016-75916-R and SAF2016-75452-R), CIBERER-ISCIII (CB06/07/0017) and Fundación Ramón Areces, Spai

‘Se hace camino al andar’: El legado de la poesía pedestre del Romanticismo inglés en Miguel de Unamuno y Antonio Machado

    Get PDF
En este trabajo sostenemos que el caminar tiene un efecto poético similar en la obra de Miguel de Unamuno y Antonio Machado, que unen a la lectura de la poesía pedestre de Wordsworth y Coleridge su propia experiencia del campo españolWalking in Nature was for the English Romantics an intense poetic experience, which originated some of their best poems, based on the search of a total immersion in Nature and of the gaining of the poet’s self-knowledge. The poetry of W. Wordsworth and S. T. Coleridge epitomises this epistemological journey in the natural world, in which the limits of the poet’s identity are blurred, being that identity reconstructed afterwards in its intimate communion with Nature. A similar poetic effect, we contend, is produced in two major early twentieth-century Spanish poets, namely Miguel de Unamuno and Antonio Machado, both by means of their reading of Wordsworth’s and Coleridge’s pedestrian poems, and by their own exposure to the Spanish countrysid

Hypothalamic and mesencephalic regions involved in the control of laryngeal activity and subglottic pressure in spontaneously breathing anaesthetized rats

Changes of laryngeal caliber allow changes in airflow which are necessary for the vibration of the vocal folds and emission of voice. It is known that stimulation of the Periaqueductal Gray matter (PAG) and nucleus retroambiguus (nRA) produces vocalization, and lesions in PAG cause mutism in animals and humans. The nRA is the perfect target to turn passive into active expiration modifying the activity of laryngeal motoneurons located in the nucleus ambiguous (Paton and Nolan, 2000). We have shown that rostral and ventral pontine structures are involved in changes of laryngeal caliber (Lara et al., 2002). It has been also demonstrated a high expression of FOXP2 protein (transcription factor related to vocalization) at mesencephalic and pontine regions (PAG, Parabrachial complex and A5 Region) involved in cardiorespiratory control. The aim of this study was to characterize the relations between hypothalamic and mesencephalic regions involved in cardiorespiratory control and their possible role in modulating laryngeal activity. Experimental studies were carried out with non-inbred male rats (n=7), SPF, Sprague-Dawley (250-300 g) housed under standard conditions. Animals were anesthetized with sodium pentobarbitone (60 mg/kg i.p., initial dose, supplemented 2 mg/kg, i.v., as necessary). A double tracheal cannulation to develop the classical technique of the “glottis isolated in situ” and for the recording of respiratory airflow was carried out. DMH-PeF, dlPAG, and CnF stimulations evoked a significant decrease of laryngeal resistance (subglottal pressure) (p<0.01) accompanied with an inspiratory facilitatory response consisted of an increase in respiratory rate, together with a pressor and tachycardic response. The results of our study contribute with new data on the role of the hypothalamic-mesencephalic neuronal circuits in the control mechanisms of subglottic pressure and laryngeal activity.Universidad de Málaga. Upregulating Positive Affectivity in the Transdiagnostic Treatment of Emotional Disorders: A Randomized Pilot Study

    Get PDF
Transdiagnostic cognitive-behavioral therapy for emotional disorders (ED) has proven to be effective. However, current transdiagnostic treatment protocols address only the regulation of negative affectivity, and they do not include treatment components to more directly target the regulation of positive affectivity. In this study, we propose to evaluate the preliminary efficacy and acceptability of a transdiagnostic treatment protocol for ED that includes, as an innovative feature, a specific treatment component to directly upregulate positive affectivity based on positive psychology interventions. A total of 24 participants were randomized to either a transdiagnostic treatment protocol (n = 12) or a transdiagnostic treatment protocol with an additional component designed to regulate positive affectivity (n = 12). Participants completed measures of anxiety, depression, positive and negative affectivity, and quality of life, as well as treatment acceptability at pre- and posttreatment and at the 3-month follow-up. Both interventions led to improvements in all measures at posttreatment, and these outcomes were maintained at the 3-month follow-up, with large effect sizes for all measures. The effect sizes for positive affect were larger in the condition that included the component to upregulate positive affectivity. Attrition rate was low, and both treatment protocols were well accepted by participants. The results obtained in this study indicate the feasibility of testing the treatment protocol in a larger, randomized, controlled trial, and they suggest the potential of including treatment components for directly upregulating positive affectivity in future research on transdiagnostic treatment protocols for ED

Phytochemical Assessment of Native Ecuadorian Peppers (Capsicum spp.) and Correlation Analysis to Fruit Phenomics

[Abstract] In this work, the impact of pepper (Capsicum spp.) fruits morphology on their composition for health-promoting compounds was investigated. For that purpose, pepper accessions from Ecuador, one of the hotspots in Capsicum’s origin, were analyzed for ascorbic acid, polyphenols, capsaicinoids, and prevention of cholesterol oxidation. Plant and fruit phenomics were assessed with conventional descriptors and Tomato Analyzer digital traits. Significant differences among accessions and species revealed a large diversity within the collection. The Capsicum frutescens group displayed the highest levels of capsaicinoids, whereas the polyphenols shortly varied among the five domesticated species. Capsicum pubescens exhibited the lowest content of ascorbic acid. The conventional descriptors describing the magnitude of plants and fruits, as well as digital attributes under the categories of size, shape index, and latitudinal section, mostly explained the variance among Capsicum groups. Correlation test revealed that phytochemical components were negatively correlated with the morphometric fruit attributes, suggesting that huge fruits contained lower amounts of nutraceutical compounds. Multivariate analysis showed that parameters related to fruit size, shape, and nutraceutical composition primarily contribute to the arrangement of pepper accessions. Such results suggested that those traits have been subjected to higher selection pressures imposed by humans.Instituto Nacional de Investigación y Tecnología Agraria y Alimentaria; RTA2015-00042-C02-02Xunta de Galicia; ED431C 2018/5
