    Potential phenotyping methodologies to assess inter- and intravarietal variability and to select grapevine genotypes tolerant to abiotic stress

    ReviewPlant phenotyping is an emerging science that combines multiple methodologies and protocols to measure plant traits (e.g., growth, morphology, architecture, function, and composition) at multiple scales of organization. Manual phenotyping remains as a major bottleneck to the advance of plant and crop breeding. Such constraint fostered the development of high throughput plant phenotyping (HTPP), which is largely based on imaging approaches and automatized data retrieval and processing. Field phenotyping still poses major challenges and the progress of HTPP for field conditions can be relevant to support selection and breeding of grapevine. The aim of this review is to discuss potential and current methods to improve field phenotyping of grapevine to support characterization of inter- and intravarietal diversity. Vitis vinifera has a large genetic diversity that needs characterization, and the availability of methods to support selection of plant material (polyclonal or clonal) able to withstand abiotic stress is paramount. Besides being time consuming, complex and expensive, field experiments are also affected by heterogeneous and uncontrolled climate and soil conditions, mostly due to the large areas of the trials and to the high number of traits to be observed in a number of individuals ranging from hundreds to thousands. Therefore, adequate field experimental design and data gathering methodologies are crucial to obtain reliable data. Some of the major challenges posed to grapevine selection programs for tolerance to water and heat stress are described herein.     Chemical and physical properties of some hazelnut varieties grown in Portugal

    Hazelnuts are one of the most appreciated nuts worldwide due to their unique organoleptic and nutritional characteristics. The present work intended to analyse several physical and chemical properties of different hazelnut varieties grown in Portugal, namely Tonda de Giffoni, Grada de Viseu, Segorbe, Longa de Espanha, Butler, Gunslebert, and Negreta. In general, the results revealed statistically significant differences between the varieties under study. The Gunslebert had more elongated hazelnuts and with heavier shelled fruits, while the kernels of the Grada de Viseu revealed to be heavier. Grada de Viseu was harder in the shell, Gunslebert had a harder core, and Segorbe was more resistant to fracture. Fat was the more representative component for all varieties and in some cases the values of moisture and water activity were over the recommended amount ( 0.62). Tonda de Giffoni was the variety with the highest induction time, indicating the highest oxidation stability. Moreover, discriminant analysis revealed that the variables more important to distinguish the varieties were protein ( = 0.007) and water activity ( = 0.010).     Levantamentos geofísicos no setor português da Faixa Piritosa Ibérica: perspetiva global da sua importância em prospeção de sulfuretos maciços e interpretação geológica

    ABSTRACT: The application of geophysical techniques to massive sulphide deposits exploration has proven to be a success in the Iberian Pyrite Belt (IPB) metallogenetic province, both in Portugal and in Spain. Several hidden deposits were discovered through the interpretation of geological models, supported by geophysical data collected in land and airborne surveys, as well as applied using down hole techniques in selected drill holes. This paper presents an overview of the methodologies applied in Portugal in the South Portuguese Zone (SPZ) geological domain, where the IPB is included, mostly by the LNEG and mining companies, with emphasis on the interpretation of the regional gravity, magnetometry and radiometry surveys, published in 1/400 000 scale by the EXPLORA/Alentejo2020 Project. In this regional mapping program, the large volume of data collected by LNEG and mining companies since the 1960’s was compiled and processed. The integrated interpretation and correlation between these methods will enable a better planning of new IPB exploration investments. IPB Volcano-Sedimentary Complex (VSC, Famennian to Late Visean age) lineaments are well defined in the gravity and magnetic fields along the IPB western region (e.g. Alcoutim/Neves Corvo/Montinho/Sesmarias/Lousal/Caveira). Aeromagnetic data also reflect deep and large anomalies related with the SPZ basement. A rock density database is presented and used to interpret the gravity data. Radiometric map (Natural Radioactivity), based mostly in airborne radiometric surveys, show contrasts between rocks in Paleozoic, Mesozoic and Cenozoic formations and correlations with geological and hydrothermal mapping are highlighted. The use of geophysical techniques is particularly critical in areas where VSC is concealed by younger sediments (e.g. Baixo Alentejo Flysch Group and/or Cenozoic formations), allowing the selection of target areas. Case studies are discussed based on electromagnetic, seismic and magnetotelluric methods and on the high mineral potential of defined targets. Multilayered data correlation (geophysics including petrophysical data, geochemistry and stratigraphic high-resolution models) is a key prospecting methodology and is essential for the accurate modelling of ore horizons and geological structures. Considering the IPB potential for VMS deposits, including the possible discovery of new giant (> 200 Mt) deposits, an increase of investment in new geophysical methods can be predicted. LNEG development of R&D projects such as EXPLORA/Alentejo2020 and SmartExplorationTM/H2020 also contribute to a favourable setting in the province, both in near mining and green fields scenarios. Thus, new discoveries are expected, like others made in the recent times in the IPB Portuguese sector, such as Lagoa Salgada (SFM, 1992, SFM; Redcorp/EDM, 2011; Redcorp/Ascendant/EDM, 2017), Semblana (Somincor-AGC-Lundin Mining, 2010), Monte Branco (Somincor Lundin Mining, 2012) and Sesmarias (Maepa/Avrupa, 2014). The importance of these data goes beyond the mineral exploration purpose. Since some of them use rock and mineral physical properties that can be related with environmental contamination (e.g. radiometry and magnetics) they can also be used in geochemical background studies, acid mine drainage and metal dispersion. Therefore, they can also promote a deeper knowledge in other areas of research related with environment preservation.RESUMO: A aplicação de técnicas de prospeção geofísica na pesquisa de jazigos de sulfuretos maciços na Faixa Piritosa Ibérica (FPI) tem-se revelado um sucesso em Portugal e em Espanha. Vários jazigos ocultos foram descobertos a partir da interpretação de modelos geológicos, com o apoio de dados geofísicos recolhidos em levantamentos terrestres, aerotransportados ou em profundidade, em sondagens (diagrafias). Neste artigo, apresenta-se uma análise das metodologias aplicadas principalmente pelo LNEG e empresas mineiras em Portugal, na Zona Sul Portuguesa (ZSP), onde se insere a FPI. Destaca-se a interpretação dos levantamentos regionais, esc. 1/400 000, de gravimetria, magnetometria e radiometria (EXPLORA/Alentejo2020). Nestes programas, foi feito o processamento de um volume de informação significativo, a partir de dados obtidos desde a década de 1960. Neste contexto indicam-se diversos alinhamentos de gravimetria e de aeromagnetometria relativos a unidades geológicas com elevado potencial mineiro, como o Complexo Vulcano-Sedimentar (CVS, idade Famenniano a Viseano sup.), sobressaindo o eixo Alcoutim/Neves Corvo/Montinho/Sesmarias/Lousal/Caveira. Os dados de aeromagnetismo refletem também anomalias com origens mais profundas e relacionadas com a estrutura basal da ZSP. A base de dados de densidades de rocha é utilizada na interpretação gravimétrica da FPI. O mapa radiométrico (Radioatividade Natural), baseado fundamentalmente em radiometria aeroportada (contagens totais) mostra o contraste à superfície de litologias das formações paleozoicas, mesozoicas e cenozoicas, sendo salientado, no primeiro caso, a sua correlação com a cartografia de alterações hidrotermais. Em áreas onde o CVS se encontra subjacente sob sedimentos mais recentes (ex. Grupo Flysch Baixo Alentejo e formações cenozoicas), o uso de técnicas geofísicas é fundamental para a seleção de alvos. São também descritos exemplos sobre a aplicação de métodos eletromagnéticos, sísmicos e magnetotelúricos na identificação e caracterização de estruturas geológicas. A análise integrada de diferentes métodos geofísicos, complementados com informação geológica e geoquímica, constitui a melhor metodologia de prospeção, sendo essencial para a pesquisa de horizontes de mineralização. Considerando a FPI como uma das principais províncias metalogenéticas do mundo, é expectável que venham a ocorrer mais investimentos e assim serem identificados novos jazigos, inclusive com dimensão superior a 200 Mt. Através de projetos ID como o EXPLORA/Alentejo2020 e SmartExplorationTM/H2020 o LNEG contribui para um melhor conhecimento na FPI em cenários de prospeção near mining e green fields. Nesta perspetiva, são esperadas novas descobertas de jazigos à semelhança do que tem acontecido em Portugal, como por exemplo Lagoa Salgada (SFM-1992, Redcorp/EDM 2011, Redcorp/Ascendant/EDM-2017), Semblana (Somincor-AGC Lundin Mining, 2010), Monte Branco (Somincor-Lundin Mining, 2012) e Sesmarias (Maepa/Avrupa, 2014).     Response of mycorrhizal Touriga Nacional variety grapevines to high temperatures measured by calorespirometry and near-infrared spectroscopy

    Heat stress negatively affects several physiological and biochemical processes in grapevine plants. In this work, two new methods, calorespirometry, which has been used to determine temperature adaptation in plants, and near-infrared (NIR) spectroscopy, which has been used to determine several grapevine-related traits and to discriminate among varieties, were tested to evaluate grapevine response to high temperatures. ‘Touriga Nacional’ variety grapevines, inoculated or not with Rhizoglomus irregulare or Funneliformis mosseae, were used in this study. Calorespirometric parameters and NIR spectra, as well as other parameters commonly used to assess heat injury in plants, were measured before and after high temperature exposure. Growth rate and substrate carbon conversion effciency, calculated from calorespirometric measurements, and stomatal conductance, were the most sensitive parameters for discriminating among high temperature responses of control and inoculated grapevines. The results revealed that, although this vine variety can adapt its physiology to temperatures up to 40 ºC, inoculation with R. irregulare could additionally help to sustain its growth, especially after heat shocks. Therefore, the combination of calorespirometry together with gas exchange measurements is a promising strategy for screening grapevine heat tolerance under controlled conditions and has high potential to be implemented in initial phases of plant breeding programsinfo:eu-repo/semantics/publishedVersio

    Implications for Quality and Wine Production

    UID/FIS/04559/2020Nowadays, there is a growing concern about micronutrient deficits in food products, with agronomic biofortification being considered a mitigation strategy. In this context, as Zn is essential for growth and maintenance of human health, a workflow for the biofortification of grapes from the Vitis vinifera variety Fernão Pires, which contains this nutrient, was carried out considering the soil properties of the vineyard. Additionally, Zn accumulation in the tissues of the grapes and the implications for some quality parameters and on winemaking were assessed. Vines were sprayed three times with ZnO and ZnSO4 at concentrations of 150, 450, and 900 g ha−1 during the production cycle. Physiological data were obtained through chlorophyll a fluorescence data, to access the potential symptoms of toxicity. At harvest, treated grapes revealed significant increases of Zn concentration relative to the control, being more pronounced for ZnO and ZnSO4 in the skin and seeds, respectively. After winemaking, an increase was also found regarding the control (i.e., 1.59-fold with ZnSO4-450 g ha−1). The contents of the sugars and fatty acids, as well as the colorimetric analyses, were also assessed, but significant variations were not found among treatments.     Distinct epidemiology and resistance mechanisms affecting ceftolozane/tazobactam in Pseudomonas aeruginosa isolates recovered from ICU patients in Spain and Portugal depicted by WGS

    STEP and SUPERIOR study groups.[Objectives] To analyse the epidemiology, the resistome and the virulome of ceftolozane/tazobactam-susceptible or -resistant Pseudomonas aeruginosa clinical isolates recovered from surveillance studies in Portugal (STEP, 2017–18) and Spain (SUPERIOR, 2016–17).[Methods] P. aeruginosa isolates were recovered from intra-abdominal, urinary tract and lower respiratory tract infections in ICU patients admitted to 11 Portuguese and 8 Spanish hospitals. MICs were determined (ISO-standard broth microdilution, EUCAST 2020 breakpoints). A subset of 28 ceftolozane/tazobactam-resistant P. aeruginosa isolates were analysed and compared with 28 ceftolozane/tazobactam-susceptible P. aeruginosa strains by WGS.[Results] Clonal complex (CC) 235 (27%) and CC175 (18%) were the most frequent, followed by CC244 (13%), CC348 (9%), CC253 (5%) and CC309 (5%). Inter-hospital clonal dissemination was observed, limited to a geographical region (CC235, CC244, CC348 and CC253 in Portugal and CC175 and CC309 in Spain). Carbapenemases were detected in 25 isolates (45%): GES-13 (13/25); VIM type (10/25) [VIM-2 (4/10), VIM-20 (3/10), VIM-1 (2/10) and VIM-36 (1/10)]; and KPC-3 (2/25). GES-13-CC235 (13/15) and VIM type-CC175 (5/10) associations were observed. Interestingly, KPC-3 and VIM-36 producers showed ceftolozane/tazobactam-susceptible phenotypes. However, ceftolozane/tazobactam resistance was significantly associated with GES-13 and VIM-type carbapenemase production. Six non-carbapenemase producers also displayed ceftolozane/tazobactam resistance, three of them showing known ceftolozane/tazobactam resistance-associated mutations in the PBP3 gene, ftsI (R504C and F533L). Overall, an extensive virulome was identified in all P. aeruginosa isolates, particularly in carbapenemase-producing strains.[Conclusions] GES-13-CC235 and VIM type-CC175 were the most frequent MDR/XDR P. aeruginosa clones causing infections in Portuguese and Spanish ICU patients, respectively. Ceftolozane/tazobactam resistance was mainly due to carbapenemase production, although mutations in PBP-encoding genes may additionally be involved.The study was funded by MSD Portugal (protocol VP6918) and MSD Spain (protocol MSD-CEF-2016-01).     Evaluation of alternative preservation treatments (water heat treatment, ultrasounds, thermosonication and UV-C radiation) to improve safety and quality of whole tomato

    Previously optimised postharvest treatments were compared to conventional chlorinated water treatment in terms of their effects on the overall quality of tomato (‘Zinac’) during storage at 10 °C. The treatments in question were water heat treatment (WHT = 40 °C, 30 min), ultrasounds (US = 45 kHz, 80 %, 30 min), thermosonication (TS =40 °C, 30 min, 45 kHz, 80 %) and ultraviolet irradiation (UV-C: 0.97 kJ m−2). The quality factors evaluated were colour, texture, sensorial analysis, mass loss, antioxidant capacity, total phenolic content, peroxidase and pectin methylesterase enzymatic activities, and microbial load reduction. The results demonstrate that all treatments tested preserve tomato quality to some extent during storage at 10 °C. WHT, TS and UV-C proved to be more efficient on minimising colour and texture changes with the additional advantage of microbial load reduction, leading to a shelf life extension when compared to control trials. However, at the end of storage, with exception of WHT samples, the antioxidant activity and phenolic content of treated samples was lower than for control samples. Moreover, sensorial results were well correlated with instrumental colour experimental data.     Mortality from gastrointestinal congenital anomalies at 264 hospitals in 74 low-income, middle-income, and high-income countries: a multicentre, international, prospective cohort study

    Summary Background Congenital anomalies are the fifth leading cause of mortality in children younger than 5 years globally. Many gastrointestinal congenital anomalies are fatal without timely access to neonatal surgical care, but few studies have been done on these conditions in low-income and middle-income countries (LMICs). We compared outcomes of the seven most common gastrointestinal congenital anomalies in low-income, middle-income, and high-income countries globally, and identified factors associated with mortality. Methods We did a multicentre, international prospective cohort study of patients younger than 16 years, presenting to hospital for the first time with oesophageal atresia, congenital diaphragmatic hernia, intestinal atresia, gastroschisis, exomphalos, anorectal malformation, and Hirschsprung’s disease. Recruitment was of consecutive patients for a minimum of 1 month between October, 2018, and April, 2019. We collected data on patient demographics, clinical status, interventions, and outcomes using the REDCap platform. Patients were followed up for 30 days after primary intervention, or 30 days after admission if they did not receive an intervention. The primary outcome was all-cause, in-hospital mortality for all conditions combined and each condition individually, stratified by country income status. We did a complete case analysis. Findings We included 3849 patients with 3975 study conditions (560 with oesophageal atresia, 448 with congenital diaphragmatic hernia, 681 with intestinal atresia, 453 with gastroschisis, 325 with exomphalos, 991 with anorectal malformation, and 517 with Hirschsprung’s disease) from 264 hospitals (89 in high-income countries, 166 in middleincome countries, and nine in low-income countries) in 74 countries. Of the 3849 patients, 2231 (58·0%) were male. Median gestational age at birth was 38 weeks (IQR 36–39) and median bodyweight at presentation was 2·8 kg (2·3–3·3). Mortality among all patients was 37 (39·8%) of 93 in low-income countries, 583 (20·4%) of 2860 in middle-income countries, and 50 (5·6%) of 896 in high-income countries (p<0·0001 between all country income groups). Gastroschisis had the greatest difference in mortality between country income strata (nine [90·0%] of ten in lowincome countries, 97 [31·9%] of 304 in middle-income countries, and two [1·4%] of 139 in high-income countries; p≤0·0001 between all country income groups). Factors significantly associated with higher mortality for all patients combined included country income status (low-income vs high-income countries, risk ratio 2·78 [95% CI 1·88–4·11], p<0·0001; middle-income vs high-income countries, 2·11 [1·59–2·79], p<0·0001), sepsis at presentation (1·20 [1·04–1·40], p=0·016), higher American Society of Anesthesiologists (ASA) score at primary intervention (ASA 4–5 vs ASA 1–2, 1·82 [1·40–2·35], p<0·0001; ASA 3 vs ASA 1–2, 1·58, [1·30–1·92], p<0·0001]), surgical safety checklist not used (1·39 [1·02–1·90], p=0·035), and ventilation or parenteral nutrition unavailable when needed (ventilation 1·96, [1·41–2·71], p=0·0001; parenteral nutrition 1·35, [1·05–1·74], p=0·018). Administration of parenteral nutrition (0·61, [0·47–0·79], p=0·0002) and use of a peripherally inserted central catheter (0·65 [0·50–0·86], p=0·0024) or percutaneous central line (0·69 [0·48–1·00], p=0·049) were associated with lower mortality. Interpretation Unacceptable differences in mortality exist for gastrointestinal congenital anomalies between lowincome, middle-income, and high-income countries. Improving access to quality neonatal surgical care in LMICs will be vital to achieve Sustainable Development Goal 3.2 of ending preventable deaths in neonates and children younger than 5 years by 2030

    Thermoelectric Properties of Nickel and Selenium Co-Doped Tetrahedrite

    As the search continues for novel, cheaper, more sustainable, and environmentally friendly thermoelectric materials in order to expand the range of applications of thermoelectric devices, the tetrahedrite mineral (Cu12Sb4S13) stands out as a potential candidate due to its high abundance, low toxicity, and good thermoelectric performance. Unfortunately, as most current thermoelectric materials achieve zTs above 1.0, ternary tetrahedrite is not a suitable alternative. Still, improvement of its thermoelectric performance has been achieved to zTs &asymp; 1 via isovalent doping and composition tuning, but most studies were limited to a single doping element. This project explores the effects of simultaneous doping with nickel and selenium in the thermoelectric properties of tetrahedrite. Simulated properties for different stoichiometric contents of these dopants, as well as the measured thermoelectric properties of the correspondent materials, are reported. One of the samples, Cu11.5Ni0.5Sb4S12.5Se0.5, stands out with a high power factor = 1279.99 &micro;W/m&middot;K2 at 300 K. After estimating the thermal conductivity, a zT = 0.325 at 300 K was obtained for this composition, which is the highest for tetrahedrites for this temperature. However, analysis of the weighted mobility shows the presence of detrimental factors, such as grain boundaries, disorder, or ionized impurity scattering, pointing to the possibility of further improvements
