    Genome-wide identification, characterization and expression profiling of the ubiquitin-proteasome genes in biomphalaria glabrata

    Biomphalaria glabrata is the major species used for the study of schistosomiasis-related parasite-host relationships, and understanding its gene regulation may aid in this endeavor. The ubiquitin-proteasome system (UPS) performs post-translational regulation in order to maintain cellular protein homeostasis and is related to several mechanisms, including immune responses. The aims of this work were to identify and characterize the putative genes and proteins involved in UPS using bioinformatic tools and also their expression on different tissues of B. glabrata. The putative genes and proteins of UPS in B. glabrata were predicted using BLASTp and as queries reference proteins from model organism. We characterized these putative proteins using PFAM and CDD software describing the conserved domains and active sites. The phylogenetic analysis was performed using ClustalX2 and MEGA5.2. Expression evaluation was performed from 12 snail tissues using RPKM. 119 sequences involved in the UPS in B. glabrata were identified, which 86 have been related to the ubiquitination pathway and 33 to proteasome. In addition, the conserved domains found were associated with the ubiquitin family, UQ_con, HECT, U-box and proteasome. The main active sites were lysine and cysteine residues. Lysines are responsible and the starting point for the formation of polyubiquitin chains, while the cysteine residues of the enzymes are responsible for binding to ubiquitin. The phylogenetic analysis showed an organized distribution between the organisms and the clades of the sequences, corresponding to the tree of life of the animals, for all groups of sequences analyzed. The ubiquitin sequence was the only one with a high expression profile found in all libraries, inferring its wide range of performance. Our results show the presence, conservation and expression profile of the UPS in this mollusk, providing a basis and new knowledge for other studies involving this system.     We investigate the domain of coupling constants which achieve binding for a 3-body system, while none of the 2-body subsystems is bound. We derive some general properties of the shape of the domain, and rigorous upper bounds on its size, using a Hall--Post decomposition of the Hamiltonian.     Jatropha curcasand Ricinus communisdisplay contrasting photosynthetic mechanisms in response to environmental conditions

    Higher plants display different adaptive strategies in photosynthesis to cope with abiotic stress. In this study, photosynthetic mechanisms and water relationships displayed byJatropha curcasL. (physic nuts) andRicinus communisL. (castor bean), in response to variations in environmental conditions, were assessed.R. communis showed higher CO2 assimilation, stomatal and mesophyll conductance thanJ. curcas as light intensity and intercellular CO2 pressure increased. On the other hand,R. communis was less effective in stomatal control in response to adverse environmental factors such as high temperature, water deficit and vapor pressure deficit, indicating lower water use efficiency. Conversely,J. curcas exhibited higher photosynthetic efficiency (gas exchange and photochemistry) and water use efficiency under these adverse environmental conditions.R. communisdisplayed higher potential photosynthesis, but exhibited a lowerin vivo Rubisco carboxylation rate (Vcmax) and maximum electron transport rate (Jmax). During the course of a typical day, in a semiarid environment, with high irradiation, high temperature and high vapor pressure deficit, but exposed to well-watered conditions, the two studied species presented similar photosynthesis. Losing potential photosynthesis, but maintaining favorable water status and increasing non-photochemical quenching to avoid photoinhibition, are important acclimation mechanisms developed byJ. curcas to cope with dry and hot conditions. We suggest thatJ. curcas is more tolerant to hot and dry environments thanR. communis but the latter species displays higher photosynthetic efficiency under well-watered and non-stressful conditions

    Longevidade de sementes de camu-camu submetidas a diferentes ambientes e formas de conservação

    O camu-camu (Myrciaria dubia (H.B.K.) McVaugh) produz fruto com grande potencial para extração de ácido ascórbico, que apresenta relevante importância econômica e social, com inúmeras aplicações industriais. O objetivo deste trabalho foi avaliar o efeito de diferentes formas de conservação e ambientes de armazenamento, sobre a manutenção da viabilidade das sementes. O delineamento experimental inteiramente casualizado, utilizando parcelas subsubdivididas, onde os fatores foram: ambiente de armazenamento (ao ar livre, em água, em câmara a 5 e a 10 ºC), forma de conservação (sementes com polpa - CP, sementes sem polpa - SP e sementes lavadas e tratadas - LT) e período de armazenamento (0; 2; 4 e 6 meses). O critério de avaliação adotado foi a protrusão da raiz primária. As sementes armazenadas em água, nas formas SP e LT, apresentaram germinação maior que 90%. As sementes nas formas CP e LT, armazenadas a 5 e a 10 ºC, tiveram germinação acima de 89%. O armazenamento em água e ao ar livre não afetou a germinação, podendo as sementes serem armazenadas durante o período de seis e quatro meses, respectivamente. As sementes CP, armazenadas em água, tiveram menor tempo médio para germinação. O índice de velocidade de germinação aumentou com o período de armazenamento e foi maior nas sementes armazenadas na água em todos os períodos. As sementes com polpa apresentaram menor índice de velocidade de germinação em todo o período de armazenamento avaliado

    EFFECT OF CHARCOAL -ENRICHED SUBSTRATE ON SEEDLINGS OF RHIZOBIUM-INOCULATED LEGUME TREES

    ABSTRACT Native legume trees are planted in agroforestry systems for their hardiness and symbiosis with soil bacteria of the genus rhizobium, efficient in N2 fixation. The enrichment of the substrate composition with fine charcoal for seedling production of these trees is interesting for increasing soil porosity, water retention and the proliferation of beneficial microorganisms. Experiments were carried out to analyze the effect of substrate enrichment with charcoal on the quality of Clitoria fairchildiana, Enterolobium schomburgkii and Inga edulis seedlings. The treatments consisted of a 3:2:0.5 (v:v) mixture of clay soil, sand and bovine manure and a 3:2 (v:v) mixture of clay soil and sand combined with charcoal rates of 0, 10, 19 and 29%. After mixing the components, substrate samples were collected and chemically analyzed. The experiment was arranged in a completely randomized design with 5 treatments and 10 replications. The seedlings were inoculated with homologues rhizobia and growth controlled monthly. The plants were collected to determine the number of nodules and dry biomass of roots, shoots and nodules. Seedling growth was similar on substrates containing charcoal or manure, except for E. schomburkii, which increased by more than 100% on the charcoal-containing substrates. The number and dry biomass of nodules in the charcoal-containing substrates was up to 100% and 300% higher than in the manure-containing treatment, respectively. The results indicated that the substitution of manure by charcoal favors the seedling quality of the studied species
