3D tuned porous carbon monolith as catalysts in the wet peroxide oxidation of paracetamol
In recent years, many pharmaceuticals have been identified at trace levels worldwide in the aquatic
environment [1]. Municipal wastewater treatment plants (WWTPs) are considered the main sources of
these pollutants as they are not generally prepared to deal with such complex substances and thus,
they are usually ineffective in their removal [1]. Despite the low concentration of drugs contained in
those effluents, the presence of pharmaceuticals, even in trace concentrations, affects the quality of
water and constitutes a risk of toxicity for the ecosystems and living organisms [1-2]. Consequently,
new regulation for micropollutants discharge and monitoring has been issued in Europe (Directive
2013/39/EU). Paracetamol (PCM) deserves particular attention, since it has recently been discovered
as a potential pollutant of waters, largely accumulated in the aquatic environment [3]. This work deals
with the treatment of PCM, used as a model pharmaceutical contaminant of emerging concern, by
catalytic wet peroxide oxidation using carbon-based monoliths (Fig. 1a) as catalysts. Monoliths were
prepared by stereolithographic 3D printing of a photoresin, which was later converted into porous
carbon by oxidation in air (300 °C, 6 h) and subsequent pyrolysis in N2 (900 °C, 15 min) as described
elsewhere [4]. The materials revealed catalytic activity in the CWPO of PCM allowing to reach PCM

3D-printed activated carbon for post-combustion CO2 capture
The applicability of 3D-printed activated carbons for their use to CO2 capture in post-combustion streams and the
influence of activation conditions on CO2 uptake and CO2 to N2 selectivity were studied. For two monoliths with
the same open cellular foam geometry but low and high burnoff during activation, a series of fixed-bed breakthrough
adsorption experiments under typical post-combustion conditions, in a wide range of temperature (313
and 373 K), and partial pressure of CO2 up to 120 kPa were carried out. It is shown that the higher burnoff during
activation of the 3D printed carbon enhances the adsorption capacity of CO2 and N2 due to the increased specific
surface area with sorption uptakes that can reach 3.17 mol/kg at 313 K and 120 kPa. Nevertheless, the lower
burnoff time on monolith 1 leads to higher selectivity of CO2 over N2, up to 18 against 10 on monolith 2,
considering a binary interaction to a mixture of CO2/N2 (15/85 vol%) at 313 K. The single and multicomponent
adsorption equilibrium is conveniently described through the dual-site Langmuir isotherm model, while the
breakthrough curves simulated using a dynamic fixed-bed adsorption linear driving force model. Working capacities
for the 3D printed carbon with lower burnoff time lead to the best results, varying of 0.15â1.1 mol/kg for
the regeneration temperature 300â390 K. Finally, consecutive adsorption-desorption experiments show excellent
stability and regenerability for both 3D printed activated carbon monoliths and the whole study underpins the

9\. AnlageIm Rahmen dieser Arbeit wurde eine MeĂapparatur zur zeitkorrelierten
EinzelphotonenzĂ€hlung fĂŒr den Einsatz an Synchrotronstrahlungsquellen
entwickelt und zum Einsatz gebracht. Mit dem neuen MeĂaufbau wurden
Experimente zur zeitaufgelösten Fluoreszenzanisotropie von an den
Membranproteinen Bacteriorhodopsin und Rinderrhodopsin gebundenen
Fluoreszenzmarkern (Labeln) durchgefĂŒhrt. Die sulfhydrylreaktiven
Fluoreszenzmarker wurden ortsspezifisch an verschiedene Positionen innerhalb
der die Transmembranhelices verbindenden Schleifenkonformationen (Loops)
gebunden, um die Dynamik und die sterische BeschrÀnkung dieser flexiblen
Bereiche zu studieren. Es sollte ein VerstĂ€ndnis fĂŒr das Bewegungsverhalten
der Fluoreszenzmarker unter der Beteiligung der flexiblen Polypeptidketten
entwickelt werden.
Die empirische Anpassung der gemessenen zeitaufgelösten
Fluoreszenzanisotropiekurven erfolgte mit einer Summe von ein bis drei
Exponentialfunktionen. Die Kurzzeitkomponente von einigen Hundert Pikosekunden
wurde der Eigenbewegung der Fluorophore zugeordnet. Die mittlere
Zeitkomponente von einigen Nanosekunden wurde als Anteil der Loops
interpretiert. Die Langzeitkomponente von einigen zehn Nanosekunden bei den
Mizellen beruhte auf der Depolarisation durch die Gesamtrotationsdiffusion.
Neben der Rotationsdiffusion trat bei einer Reihe von markierten Proteinproben
zusÀtzlich strahlungsloser Energietransfer auf. Der strahlungslose Transfer
der Energie vom Fluoreszenzmarker (Donator) zum Retinal-Chromophor (Akzeptor)
fĂŒhrte zu keinen signifikanten VerĂ€nderungen des Anisotropieverlaufs. Der
Energie-Homotransfer der Donatoren untereinander fĂŒhrte dagegen zu einer
verstĂ€rkten Depolarisation. Mit Hilfe des Energietransfers konnten fĂŒr
verschiedene Positionen Abstandsbereiche zwischen den Transferpartnern
berechnet werden.
Gleichlichtexperimente zur PolaritÀt der Labelumgebung sowie
Quenchingexperimente mit Jodionen wiesen auf eine freie ZugÀnglichkeit und ein
wĂ€Ăriges Umgebungsmilieu der gebundenen Fluoreszenzmarker hin.
Ein wesentliches Resultat dieser Arbeit war der direkte Nachweis von
KonformationsÀnderungen an der zytoplasmatischen OberflÀche des Rhodopsins mit
Hilfe der Methode der zeitaufgelösten Fluoreszenzanisotropie. Die Experimente
zum Anisotropieverhalten im Grundzustand und im aktiven Intermediatszustand
MII fĂŒhrten zu der Beobachtung einer Erhöhung des Ordnungsparameters durch die
Photoaktivierung. Dieser Effekt an den Positionen C316 und C140 wurde als eine
Erhöhung der sterischen BewegungsbeschrÀnkung der Label-Loop-Konstrukte im
aktiven MII-Zustand interpretiert.A new optical set up was developed for the time-correlated single photon
counting using synchrotron radiation for excitation. With this new set up
time-resolved fluorescence anisotropy experiments were executed. The
sulfhydryl reactive fluorescence markers were specifically bound at different
positions within the loops, connecting the helices of the membrane proteins
bacteriorhodopsin and bovine rhodopsin., in order to study the dynamics and
the sterical restriction of these flexible polypeptide chains.
For the empirical analysis of the measured time-resolved fluorescence
anisotropy curves a sum of one to three exponential functions was used. The
short time component of some hundred picoseconds was assigned to the
independent movement of the fluorophores. The middle time component of some
nanoseconds was caused by the loops. The long-term component of some ten
nanoseconds was based on the depolarization by total rotation diffusion of the
micelle systems.
Apart from rotation diffusion additionally the process of radiationless energy
transfer occurred at the marked protein samples. The radiationless transfer of
the energy of the fluorescence marker (donor) to the retinal chromophor
(acceptor) led to no significant modifications of the anisotropy process. The
energy homotransfer of the donors among themselves led against it to an
intensified depolarization. With the help of the energy transfer distance
ranges between the transfer partners could be calculated for different
positions.
Experiments to test the polarity of the label environment as well as
fluorescence quenching with iodine ions referred to a free accessibility and
an aqueous environment of the bound fluorescence markers.
A substantial result of this work was the direct proof of conformational
changes at the cytoplasmic surface of the rhodopsin by the method of the
fluorescence anisotropy. The experiments to the anisotropy behaviour in the
initial state and in the active intermediate state MII led to the observation
of an increase of the order parameter by the photo activation. This effect at
the positions C316 and C140 was interpreted as an increase of the sterical
restriction of the label-loop-construction in the active state
Der Einfluss von microRNAs auf die Expression von VEGF-Rezeptor 2 im sensorischen und motorischen Teil des Nervensystems
VEGFR-2 (Vascular Endothelial Growth Factor Receptor 2) zeigte in der Vergangenheit nicht nur Effekte auf die Angiogenese, sondern auch auf die Neurogenese und Neuroprotektion. Dazu wurde in vorherigen Arbeiten eine Regulation des VEGFR-2 nachgewiesen. Die molekularen Mechanismen dieser Regulation sind bisher noch unklar. In dieser Forschungsarbeit sollte eine erste Idee zur Expressionsregulation von VEGFR-2 entwickelt werden. Im Fokus standen hierbei miRNAs (microRNAs). Im ersten Teil wurde eine altersabhaÌngige Expression von VEGFR-2 im sensorischen und motorischen Nervensystem nachgewiesen. Im zweiten Teil wurde eine altersabhaÌngige Expression der miRNAs in beiden Systemen nachgewiesen. Im dritten Teil der Dissertationsarbeit wurde ein positiver Einfluss von miR-130a-3p auf die Expression von VEGFR-2 mRNA im sensorischen Nervensystem gezeigt. miR-129-5p hatte einen hemmenden Einfluss auf die Expression von VEGFR-2 mRNA im motorischen Nervensystem
