Interaction between atypical microorganisms and E. coli in catheter-associated urinary tract biofilms
Most biofilms involved in catheter-associated urinary tract infections (CAUTIs) are polymicrobial, with disease causing
(eg Escherichia coli) and atypical microorganisms (eg Delftia tsuruhatensis) frequently inhabiting the same catheter.
Nevertheless, there is a lack of knowledge about the role of atypical microorganisms. Here, single and dual-species
biofilms consisting of E. coli and atypical bacteria (D. tsuruhatensis and Achromobacter xylosoxidans), were evaluated.
All species were good biofilm producers (Log 5.84–7.25 CFU cm−2 at 192 h) in artificial urine. The ability of atypical
species to form a biofilm appeared to be hampered by the presence of E. coli. Additionally, when E. coli was added to a
pre-formed biofilm of the atypical species, it seemed to take advantage of the first colonizers to accelerate adhesion, even
when added at lower concentrations. The results suggest a greater ability of E. coli to form biofilms in conditions
mimicking the CAUTIs, whatever the pre-existing microbiota and the inoculum concentration.This work was supported by the Portuguese Science Foundation (FCT), DNA mimics Research Project [Ref. Spectral quantification of nonlinear behaviour of the nearshore seabed and correlations with potential forcings at Duck, N.C., U.S.A
Local bathymetric quasi-periodic patterns of oscillation are identified from
monthly profile surveys taken at two shore-perpendicular transects at the USACE
field research facility in Duck, North Carolina, USA, spanning 24.5 years and
covering the swash and surf zones. The chosen transects are the two furthest
(north and south) from the pier located at the study site. Research at Duck has
traditionally focused on one or more of these transects as the effects of the
pier are least at these locations. The patterns are identified using singular
spectrum analysis (SSA). Possible correlations with potential forcing
mechanisms are discussed by 1) doing an SSA with same parameter settings to
independently identify the quasi-periodic cycles embedded within three
potentially linked sequences: monthly wave heights (MWH), monthly mean water
levels (MWL) and the large scale atmospheric index known as the North Atlantic
Oscillation (NAO) and 2) comparing the patterns within MWH, MWL and NAO to the
local bathymetric patterns. The results agree well with previous patterns
identified using wavelets and confirm the highly nonstationary behaviour of
beach levels at Duck; the discussion of potential correlations with
hydrodynamic and atmospheric phenomena is a new contribution. The study is then
extended to all measured bathymetric profiles, covering an area of 1100m
(alongshore) by 440m (cross-shore), to 1) analyse linear correlations between
the bathymetry and the potential forcings using multivariate empirical
orthogonal functions (MEOF) and linear correlation analysis and 2) identify
which collective quasi-periodic bathymetric patterns are correlated with those
within MWH, MWL or NAO, based on a (nonlinear) multichannel singular spectrum

RT-qPCR reveals opsin gene upregulation associated with age and sex in guppies (Poecilia reticulata) - a species with color-based sexual selection and 11 visual-opsin genes
<p>Abstract</p> <p>Background</p> <p>PCR-based surveys have shown that guppies (<it>Poecilia reticulata</it>) have an unusually large visual-opsin gene repertoire. This has led to speculation that opsin duplication and divergence has enhanced the evolution of elaborate male coloration because it improves spectral sensitivity and/or discrimination in females. However, this conjecture on evolutionary connections between opsin repertoire, vision, mate choice, and male coloration was generated with little data on gene expression. Here, we used RT-qPCR to survey visual-opsin gene expression in the eyes of males, females, and juveniles in order to further understand color-based sexual selection from the perspective of the visual system.</p> <p>Results</p> <p>Juvenile and adult (male and female) guppies express 10 visual opsins at varying levels in the eye. Two opsin genes in juveniles, <it>SWS2B </it>and <it>RH2-2</it>, accounted for >85% of all visual-opsin transcripts in the eye, excluding <it>RH1</it>. This relative abundance (RA) value dropped to about 65% in adults, as <it>LWS-A180 </it>expression increased from approximately 3% to 20% RA. The juvenile-to-female transition also showed <it>LWS-S180 </it>upregulation from about 1.5% to 7% RA. Finally, we found that expression in guppies' <it>SWS2-LWS </it>gene cluster is negatively correlated with distance from a candidate locus control region (LCR).</p> <p>Conclusions</p> <p>Selective pressures influencing visual-opsin gene expression appear to differ among age and sex. <it>LWS </it>upregulation in females is implicated in augmenting spectral discrimination of male coloration and courtship displays. In males, enhanced discrimination of carotenoid-rich food and possibly rival males are strong candidate selective pressures driving <it>LWS </it>upregulation. These developmental changes in expression suggest that adults possess better wavelength discrimination than juveniles. Opsin expression within the <it>SWS2-LWS </it>gene cluster appears to be regulated, in part, by a common LCR. Finally, by comparing our RT-qPCR data to MSP data, we were able to propose the first opsin-to-λ<sub>max </sub>assignments for all photoreceptor types in the cone mosaic.</p
Trauma- und Gewaltfolgen – psychische Auswirkungen
This paper addresses consequences of exposure to violence and trauma. Traumata are defined as events with an extraordinary threat or catastrophic extent. Beside Posttraumatic Stress Disorder (PTSD), affected people may develop Complex PTSD, Prolonged Grief Disorder or Adjustment Disorder as direct consequences of exposure with extreme stress. Indirect trauma-related disorders are amongst others Major Depression, Substance Dependency and Personality Disorders. These disorders develop often comorbid to PTSD. The likelihood to develop a PTSD at one point during the life course is 1-4 % in Germany. A PTSD is diagnosed if for any length of time the traumatic situation is re-experienced (e.g. in pictures or nightmares), potential triggers are avoided, emotional reactivity is numb and a permanent hyperarousal is experienced. The intensity, the duration and the frequency of traumatic experiences as well as the lack of social support after the trauma are important risk factors for the development of a PTSD. In the last two decades successful treatment approaches for PTSD have been developed. The main focus of all evidence-based treatment approaches is the exposure in sensu of the traumatic experiences. Behavioral therapy approaches have shown to be most effective in the treatment of PTSD. A better understanding of the consequences of exposure to violence and trauma may help us to identify people at risk for developing trauma-related disorders already at an early stage
