Effects of Two Species of VA Mycorrhizal Fungi on Drought Tolerance of Winter Wheat
Roots and soils from western Nebraska fields of native and planted grasslands, and winter wheat of varied fallow-wheat cultivation duration, were evaluated for vesicular-arbuscular (VA) mycorrhizal root infection and spore numbers and types. Increased cultivation decreased percentage mycorrhizal infection in wheat and reduced spore numbers of Glomus fasciculatus, the dominant VA mycorrhizal fungus in these soils. Spore numbers of other VA mycorrhizal fungi did not change significantly with cultivation although mean numbers of G. mosseae increased with continued wheat production. Water relations and growth were determined for greenhouse-grown non-mycorrhizal, G. fasciculatus-infected, and G. mosseae-infected wheat in wet and dry soils. Stomatal conductances were higher in mycorrhizal than in non-mycorrhizal plants in both wet and dry treatments. Stomatal closure in mycorrhizal plants occurred at lower leaf water potentials (ψ1) and after greater desiccation than in non-mycorrhizal plants, but some leaves of G. masseae-infected plants showed no stomatal response to drought and continued to transpire at ψ1 as low as -4◦1 MPa. Leaf osmotic adjustment was greater for G. fasciculatus-infected plants. Non-mycorrhizal and G. fasciculatus-infected plants had equal dry wts in both wet and dry conditions. Infection by G. fasciculatus appeared to increase wheat drought tolerance while infection by G. mosseae did not
Josephson Coupling and Fiske Dynamics in Ferromagnetic Tunnel Junctions
We report on the fabrication of Nb/AlO_x/Pd_{0.82}Ni_{0.18}/Nb
superconductor/insulator/ferromagnetic metal/superconductor (SIFS) Josephson
junctions with high critical current densities, large normal resistance times
area products, high quality factors, and very good spatial uniformity. For
these junctions a transition from 0- to \pi-coupling is observed for a
thickness d_F ~ 6 nm of the ferromagnetic Pd_{0.82}Ni_{0.18} interlayer. The
magnetic field dependence of the \pi-coupled junctions demonstrates good
spatial homogeneity of the tunneling barrier and ferromagnetic interlayer.
Magnetic characterization shows that the Pd_{0.82}Ni_{0.18} has an out-of-plane
anisotropy and large saturation magnetization, indicating negligible dead
layers at the interfaces. A careful analysis of Fiske modes provides
information on the junction quality factor and the relevant damping mechanisms
up to about 400 GHz. Whereas losses due to quasiparticle tunneling dominate at
low frequencies, the damping is dominated by the finite surface resistance of
the junction electrodes at high frequencies. High quality factors of up to 30
around 200 GHz have been achieved. Our analysis shows that the fabricated
junctions are promising for applications in superconducting quantum circuits or
quantum tunneling experiments.Comment: 15 pages, 9 figure
Soil biochemistry and microbial activity in vineyards under conventional and organic management at Northeast Brazil.

Based on the fact that organic fertilization can improve soil quality, we compared the effects of conventional and organic soil management on microbial activity and mycorrhization of seedless grape crops. We measured glomerospores number, most probable number (MPN) of propagules, richness of arbuscular mycorrhizal fungi (AMF) species, AMF root colonization, EE-BRSP production, carbon microbial biomass (C-MB), microbial respiration, fluorescein diacetate hydrolytic activity (FDA) and metabolic coefficient (qCO2). The organic management led to an increase in all variables with the exception of EE-BRSP and qCO2. Mycorrhizal colonization increased from 4.7% in conventional crops to 15.9% in organic crops. Spore number ranged from 4.1 to 12.4 per 50 g-1 soil in both management systems. The most probable number of AMF propagules increased from 79 cm-3 soil in the conventional system to 110 cm-3 soil in the organic system. Microbial carbon, CO2 emission, and FDA activity were increased by 100 to 200% in the organic crop. Thirteen species of AMF were identified, the majority in the organic cultivation system. Acaulospora excavata, Entrophospora infrequens, Glomus sp.3 and Scutellospora sp. were found only in the organically managed crop. S. gregaria was found only in the conventional crop. Targeted reconstruction of T cell receptor sequence from single cell RNA-seq links CDR3 length to T cell differentiation state
Abstract The T cell compartment must contain diversity in both T cell receptor (TCR) repertoire and cell state to provide effective immunity against pathogens. However, it remains unclear how differences in the TCR contribute to heterogeneity in T cell state. Single cell RNA-sequencing (scRNA-seq) can allow simultaneous measurement of TCR sequence and global transcriptional profile from single cells. However, current methods for TCR inference from scRNA-seq are limited in their sensitivity and require long sequencing reads, thus increasing the cost and decreasing the number of cells that can be feasibly analyzed. Here we present TRAPeS, a publicly available tool that can efficiently extract TCR sequence information from short-read scRNA-seq libraries. We apply it to investigate heterogeneity in the CD8+ T cell response in humans and mice, and show that it is accurate and more sensitive than existing approaches. Coupling TRAPeS with transcriptome analysis of CD8+ T cells specific for a single epitope from Yellow Fever Virus (YFV), we show that the recently described ‘naive-like’ memory population have significantly longer CDR3 regions and greater divergence from germline sequence than do effector-memory phenotype cells. This suggests that TCR usage is associated with the differentiation state of the CD8+ T cell response to YFV
