    ‘And yet I’m an adult now’. The influence of parental criticism on women’s body satisfaction/dissatisfaction during emerging adulthood

    Body dissatisfaction is widely recognized as an important public health concern mainly for women because of its increasing incidence worldwide. The aim of this exploratory study was to test the predictive role of family influence on body satisfaction (BS) among young Italian women, during a decade conceptualized as \u2018emerging adulthood\u2019. Instruments designed to assess BS, appearance-focused parental comments, maternal and paternal criticism were administered to 688 women aged between 18 and 28 years (M age = 23.40; SD = 2.33). Two regression analyses were used with life satisfaction levels, parental comments, and maternal/paternal criticism as independent variables to explain variance in BS for younger (18\u201323 years) and older (24\u201328 years) women. The data reveal an interesting difference in the influence of parental criticism on BS depending on the age cohort: maternal criticism negatively predicts BS in younger women, while paternal criticism is a risk factor for BS in older women

    The New World challenge : Performance trends in wine production in major wine-exporting countries in the 2000s and their implications for the Australian wine industry

University of Adelaide Press, Adelaide publication of an index of revealed comparative advantage suggests that the Australian wine industry had come under increased competition from other "New World" producers in the first decade of this century. We examine this influence by comparing the transformation of wine grapes into wine volume and value in the 11 largest wine-exporting countries during the years, 2000-2009. Our focus is on the challenge issued by other New World producers from the Southern Hemisphere to Australian producers, and the continuing challenge to Old World global supremacy by New World producers and its response. Four performance measures are used this study. Two key trends are evident. First, all countries migrated to higher price points, albeit with differing degrees of success: slightly declining productivity in transforming wine grapes into wine output was over whelmed by price/quality effects, leading to substantial gains in transforming wine grapes in to wine value. Second, New World producers plus Portugal and Spain were much more successful in achieving gains in their export value proposition than they were in extracting value in their domestic markets. Results show that Australian wine producers had lost some of their competitive advantage during the 2000s as their pre- existing strategy dominated by the export of high-volume wines by large companies at low to medium price points, and their reliance on a reputation for reliable good quality for the price point was beginning to fail in the face of competition from both New World and Old World producers. Acknowledgement of this outcome has led to a good deal of introspection, and recognition of the need to promote the wine regions of Australia, based on higher-quality wines,and to select and promote quality indicators

    Innovative strategy for polyhydroxyalkanoates recovery from mixed microbial cultures. Effects of aqueous phase and solvent extraction on polymer properties

    This work aimed to investigate various methods of polyhydroxyalkanoates (PHA) extraction and to optimize the recovery in the view of reducing solvents’ use and waste. The extraction tests were applied on PHA-rich biomass collected at the end of the accumulation step, conducted both at pilot and lab scale. PHA-rich biomass obtained from the lab scale process was produced from synthetic feedstock (mixture of acetic and propionic acids), while fermented organic waste was used for PHA production at pilot scale. In the extraction tests, different solutions of NaOH, also in combination with a surfactant (sodium dodecyl sulfate, SDS), were used to recover the polymer from the non-polymeric cellular material (NPCM). Different times (3, 6 and 24 h) and concentrations (0.05 – 1 M) were tested, evaluating also the effect of adding SDS (0.1 % w/v). As a benchmark, solvent extraction with chloroform and oxidation with NaClO were also conducted. Finally, extracted samples were characterized through several techniques: DSC, TGA, GC-FID, capillary viscosimetry. Alkaline treatment at short times and low concentrations resulted to be more efficient in terms of purity (85 – 99 % w/w PHA) and recovery (higher than 80 % w/w), than higher concentration. On the other hand, the presence of SDS had dramatic effects on the recovery (lower than 50 % w/w) and also on the molecular weight, which was two folds lower than that obtained from alkaline extraction. Overall, extraction with aqueous phase reagents had no effects on thermal properties, which resulted to be in the range of those reported in literature

    Polyhydroxyalkanoates production by mixed microbial cultures in sequencing batch reactors operated under different feeding conditions

    The production of polyhydroxyalkanoates (PHA) by mixed microbial cultures (MMC) requires a multistage process, whereby the microbial selection of PHA-storing microorganisms plays a key role on the overall performance. A strategy to favor the microbial selection consists in the alternance of excess (feast phase) and absence (famine phase) of the external carbon source. In this work, three runs of a lab-scale Sequencing Batch Reactor (SBR) operated under different working conditions for the establishment of the feast and famine (F/F) regime were analyzed. A fixed organic loading rate of 4.25 gCOD (Chemical Oxygen Demand)/L d, and a fixed cycle length of 12 h were applied to the SBR. The F/F regime consisted of fully aerobic dynamic (ADF) or aerobic/anoxic (AE/ANOX) conditions. Results showed an intracellular PHA content as high as 40 ± 2 (%, w/w) when ADF conditions were applied with the organic feeding solution made of acetate (85 % on COD basis) and propionic (15%) acids. The hydroxyvalerate content in the stored polymer increased (from 25 ± 1 to 41 ± 3, % w/w) by increasing the propionic fraction (up to 35%) in the feeding solution. The AE/ANOX condition resulted in a lower PHA-storing ability which warrants further investigations

    Simplified reactor design for mixed culture-based electrofermentation toward butyric acid production

    Mixed microbial culture (MMC) electrofermentation (EF) represents a promising tool to drive metabolic pathways toward the production of a specific compound. Here, the MMC-EF process has been exploited to obtain butyric acid in simplified membrane-less reactors operated by applying a difference of potential between two low-cost graphite electrodes. Ten values of voltage difference, from -0.60 V to -1.5 V, have been tested and compared with the experiment under open circuit potential (OCP). In all the tested conditions, an enhancement in the production rate of butyric acid (from a synthetic mixture of glucose, acetate, and ethanol) was observed, ranging from 1.3- to 2.7-fold relative to the OCP. Smaller enhancements in the production rate resulted in higher values of the calculated specific energy consumption. However, at all applied voltages, a low flow of current was detected in the one-chamber reactors, accounting for an average value of approximately -100 µA. These results hold a substantial potential with respect to the scalability of the electrofermentation technology, since they pinpoint the possibility to control MMC-based bioprocesses by simply inserting polarized electrodes into traditional fermenters

    SerpinB3 as hepatic marker of post-resective shear stress

    Post-resective liver failure is a frequent complication of liver surgery and it is due to portal hyperperfusion of the remnant liver and to arterial vasoconstriction, as buffer response of the hepatic artery. In this context, splenectomy allows a reduction of portal flow and increases the survival chance in preclinical models. SerpinB3 is over-expressed in the liver in oxidative stress conditions, as a mechanism of cell defense to provide survival by apoptosis inhibition and cell proliferation. In this study, the expression of SerpinB3 was assessed as predictor of liver damage in in vivo models of major hepatic resection with or without splenectomy. Wistar male rats were divided into 4 groups: group A received 30% hepatic resection, group B > 60% resection, group C > 60% resection with splenectomy and group D sham-operated. Before and after surgery liver function tests, echo Doppler ultrasound and gene expression were assessed. Transaminase values and ammonium were significantly higher in groups that underwent major hepatic resection. Echo Doppler ultrasound showed the highest portal flow and resistance of the hepatic artery in the group with > 60% hepatectomy without splenectomy, while the association of splenectomy determined no increase in portal flow and hepatic artery resistance. Only the group of rats without splenectomy showed higher shear-stress conditions, reflected by higher levels of HO-1, Nox1 and of Serpinb3, the latter associated with an increase of IL-6. In conclusion, splenectomy controls inflammation and oxidative damage, preventing the expression of Serpinb3. Therefore, SerpinB3 can be considered as a marker of post-resective shear stress

    Hypoxia up-regulates SERPINB3 through HIF-2\u3b1 in human liver cancer cells.

    SERPINB3 is a cysteine-proteases inhibitor up-regulated in a significant number of cirrhotic patients carrying hepatocellular carcinoma (HCC) and recently proposed as a prognostic marker for HCC early recurrence. SERPINB3 has been reported to stimulate proliferation, inhibit apoptosis and, similar to what reported for hypoxia, to trigger epithelial-to-mesenchymal transition (EMT) and increased invasiveness in liver cancer cells. This study has investigated whether SERPINB3 expression is regulated by hypoxia-related mechanisms in liver cancer cells. Exposure of HepG2 and Huh7 cells to hypoxia up-regulated SERPINB3 transcription, protein synthesis and release in the extracellular medium. Hypoxia-dependent SERPINB3 up-regulation was selective (no change detected for SERPINB4) and operated through hypoxia inducible factor (HIF)-2\u3b1 (not HIF-1\u3b1) binding to SERPINB3 promoter, as confirmed by chromatin immuno-precipitation assay and silencing experiments employing specific siRNAs. HIF-2\u3b1-mediated SERPINB3 up-regulation under hypoxic conditions required intracellular generation of ROS. Immuno-histochemistry (IHC) and transcript analysis, performed in human HCC specimens, revealed co-localization of the two proteins in liver cancer cells and the existence of a positive correlation between HIF-2\u3b1 and SERPINB3 transcript levels, respectively. Hypoxia, through HIF-2\u3b1-dependent and redox-sensitive mechanisms, up-regulates the transcription, synthesis and release of SERPINB3, a molecule with a high oncogenic potential

    The role of very low calorie ketogenic diet in sympathetic activation through cortisol secretion in male obese population

    Adipose tissue is considered an endocrine organ, and its excess compromises the immune response and metabolism of hormones and nutrients. Furthermore, the accumulation of visceral fat helps to increase the synthesis of cortisol. The hypothalamus-pituitary-adrenal (HPA) axis is a neuroendocrine system involved in maintaining homeostasis in humans under physiological conditions and stress, and cortisol is the main hormone of the HPA axis. It is known that a stress-induced diet and cortisol reactivity to acute stress factors may be related to dietary behavior. In obesity, to reduce visceral adipose tissue, caloric restriction is a valid strategy. In light of this fact, the aim of this study was to assess the effects of a commercial dietary ketosis program for weight loss on the sympathetic nervous system and HPA axis, through evaluation of salivary cortisol and GSR levels. Thirty obese subjects were recruited and assessed before and after 8 weeks of Very Low Calorie Ketogenic Diet (VLCKD) intervention to evaluate body composition and biochemical parameters. Salivary cortisol levels and GSR significantly decreased after dietary treatment; in addition, body composition and biochemical features were ameliorated. The VLCKD had a short-term positive effect on the SNS and HPA axes regulating salivary cortisol levels. Finally, the effects of the VLCKD on the SNS and HPA axis may lead to more individualized treatment strategies that integrate obesity and stress and support the usefulness of such therapeutic interventions in promoting the reduction of the individual disease burden
