Arc Stability Characterization of Double Coated Electrodes for Hardfacing
The aim of this work is to establish the influence of the relative position of the alloy charge C-Cr-Mn in the structure of the coating of rutile electrodes for hardfacing, on the operational behavior (arc stability). For this, three variants of electrodes with similar chemical composition are elaborated in the metallic core and the coating, differentiating only in the relative position that occupies the alloy charge (C-Cr-Mn) in the structure of the coating: internal, external and homogeneous. For the development of the research, a completely random design is used. The operative characterization of the electrodes is performed in terms of the arc stability. In conclusion, it can be seen that the position of the alloy load influences the operation behavior (stability of the arc). The variant of internal alloy charge in the coating presents the better arc stability performance
La Agenda 2030 y las prácticas de responsabilidad social corporativa en las federaciones deportivas colombianas (The 2030 Agenda and corporate social responsibility practices in Colombian sports federations)
The United Nations Sustainable Development Goals (SDGs) represent the framework for sports organisations to develop sustainable and socially responsible agendas. This study provides an insight into the perception of federated sports managers on the SDGs whose feasibility is most relevant within the Colombian sport context, and the most appropriate Corporate Social Responsibility (CSR) practices for their implementation. For this purpose, an analytical survey of 51 questions was applied to managers of national sports federations (n = 36/61). Subsequently, an analysis of the possible interconnection between the results obtained and the financial availabilities of the sports federations is carried out. Within the framework of the sustainability paradigm, social and economic actions are more important than those related to environmental preservation, and there is also a weak relationship between the managers' perceptions of the SDGs and CSR and the financial resources of the federations. Given the predominance of studies on CSR focused on professional sports and the Anglo-Saxon and European world, this study provides the vision of a sector that is little addressed in the literature, such as the sports federations of a Latin American country. Additionally, a quantitative methodology is presented as a contribution to research historically dominated by qualitative approaches in both Sport for Development and Peace (SDD) and CSR

Regeneración de plantas via embriogénesis somática en Phaseolus acutifolius A. Gray
Plant regeneration protocols are a requirement to develop plants transformation systems. The capacity of Phaseolus acutifolius A. Gray to regenerate plants from embryogenic callus formation was investigated. Two explants were used to form calli on a culture medium containing thidiazuron and indole-3-acetic acid. The embryonic axes showed better capacity than cotyledons to form embryos. Solar light was the most favourable to develop the in vitro plants. The 32% of somatic embryos with complete germination was achieved. These results indicate that somatic embryos formation and their germination in Phaseolus acutifolius A. Gray depended on the explant, culture medium and illumination conditions. In vitro-germinated plantlets were established in the greenhouse. Lead Optimization of 3,5-Disubstituted-7-Azaindoles for the Treatment of Human African Trypanosomiasis.
Neglected tropical diseases such as human African trypanosomiasis (HAT) are prevalent primarily in tropical climates and among populations living in poverty. Historically, the lack of economic incentive to develop new treatments for these diseases has meant that existing therapeutics have serious shortcomings in terms of safety, efficacy, and administration, and better therapeutics are needed. We now report a series of 3,5-disubstituted-7-azaindoles identified as growth inhibitors of Trypanosoma brucei, the parasite that causes HAT, through a high-throughput screen. We describe the hit-to-lead optimization of this series and the development and preclinical investigation of 29d, a potent antitrypanosomal compound with promising pharmacokinetic (PK) parameters. This compound was ultimately not progressed beyond in vivo PK studies due to its inability to penetrate the blood-brain barrier (BBB), critical for stage 2 HAT treatments. The authors acknowledge funding from the National Institute of Allergy and Infectious Diseases (M.P.P. and M.N., R01AI114685; M.P.P., 1R21AI127594, R01AI124046; C.R.C., R21AI126296; https://www.niaid.nih.gov/), the Spanish Ministerio de Economí a, Industria y Competitividad (M.N., SAF2015-71444-P; D.G.-P., SAF2016-79957-R; http://www.mineco.gob.es), Subdireccion General de Redes ́ y Centros de Investigacion Cooperativa (RICET, https://www.ricet.es/) (M.N., RD16/0027/0019; D.G.P., RD16/ 0027/0014), and RTI2018-097210-B-I00 (MINCIU-FEDER) to F.G. New Electrocardiographic Algorithm for the Diagnosis of Acute Myocardial Infarction in Patients With Left Bundle Branch Block
Background Current electrocardiographic algorithms lack sensitivity to diagnose acute myocardial infarction (AMI) in the presence of left bundle branch block. Methods and Results A multicenter retrospective cohort study including consecutive patients with suspected AMI and left bundle branch block, referred for primary percutaneous coronary intervention between 2009 and 2018. Pre-2015 patients formed the derivation cohort (n=163, 61 with AMI); patients between 2015 and 2018 formed the validation cohort (n=107, 40 with AMI). A control group of patients without suspected AMI was also studied (n=214). Different electrocardiographic criteria were tested. A total of 484 patients were studied. A new electrocardiographic algorithm (BARCELONA algorithm) was derived and validated. The algorithm is positive in the presence of ST deviation ≥1 mm (0.1 mV) concordant with QRS polarity, in any lead, or ST deviation ≥1 mm (0.1 mV) discordant with the QRS, in leads with max (R|S) voltage (the voltage of the largest deflection of the QRS, ie, R or S wave) ≤6 mm (0.6 mV). In both the derivation and the validation cohort, the BARCELONA algorithm achieved the highest sensitivity (93%-95%), negative predictive value (96%-97%), efficiency (91%-94%) and area under the receiver operating characteristic curve (0.92-0.93), significantly higher than previous electrocardiographic rules (P<0.01); the specificity was good in both groups (89%-94%) as well as the control group (90%). Conclusions In patients with left bundle branch block referred for primary percutaneous coronary intervention, the BARCELONA algorithm was specific and highly sensitive for the diagnosis of AMI, leading to a diagnostic accuracy comparable to that obtained by ECG in patients without left bundle branch block
Activation-induced cytidine deaminase targets SUV4-20-mediated histone H4K20 trimethylation to class-switch recombination sites
Activation-induced cytidine deaminase (AID) triggers antibody diversification in B cells by catalysing deamination and subsequently mutating immunoglobulin (Ig) genes. Association of AID with RNA Pol II and occurrence of epigenetic changes during Ig gene diversification suggest participation of AID in epigenetic regulation. AID is mutated in hyper-IgM type 2 (HIGM2) syndrome. Here, we investigated the potential role of AID in the acquisition of epigenetic changes. We discovered that AID binding to the IgH locus promotes an increase in H4K20me3. In 293F cells, we demonstrate interaction between co-transfected AID and the three SUV4-20 histone H4K20 methyltransferases, and that SUV4-20H1.2, bound to the IgH switch (S) mu site, is replaced by SUV4-20H2 upon AID binding. Analysis of HIGM2 mutants shows that the AID truncated form W68X is impaired to interact with SUV4-20H1.2 and SUV4-20H2 and is unable to bind and target H4K20me3 to the Smu site. We finally show in mouse primary B cells undergoing class-switch recombination (CSR) that AID deficiency associates with decreased H4K20me3 levels at the Smu site. Our results provide a novel link between SUV4-20 enzymes and CSR and offer a new aspect of the interplay between AID and histone modifications in setting the epigenetic status of CSR sites
