Real-time Alarm Monitoring System for Detecting Driver Fatigue in Wireless Areas
The purpose of this paper was to develop a real-time alarm monitoring system that can detect the fatigue driving state through wireless communication. The drivers’ electroencephalogram (EEG) signals were recorded from occipital electrodes. Seven EEG rhythms with different frequency bands as gamma, hbeta, beta, sigma, alpha, theta and delta waves were extracted. They were simultaneously assessed using relative operating characteristic (ROC) curves and grey relational analysis to select one as the fatigue feature. The research results showed that the performance of theta wave was the best one. Therefore, theta wave was used as fatigue feature in the following alarm device. The real-time alarm monitoring system based on the result has been developed, once the threshold was settled by using the data of the first ten minutes driving period. The developed system can detect driver fatigue and give alarm to indicate the onset of fatigue automatically
Enhance Reasoning for Large Language Models in the Game Werewolf
This paper presents an innovative framework that integrates Large Language
Models (LLMs) with an external Thinker module to enhance the reasoning
capabilities of LLM-based agents. Unlike augmenting LLMs with prompt
engineering, Thinker directly harnesses knowledge from databases and employs
various optimization techniques. The framework forms a reasoning hierarchy
where LLMs handle intuitive System-1 tasks such as natural language processing,
while the Thinker focuses on cognitive System-2 tasks that require complex
logical analysis and domain-specific knowledge. Our framework is presented
using a 9-player Werewolf game that demands dual-system reasoning. We introduce
a communication protocol between LLMs and the Thinker, and train the Thinker
using data from 18800 human sessions and reinforcement learning. Experiments
demonstrate the framework's effectiveness in deductive reasoning, speech
generation, and online game evaluation. Additionally, we fine-tune a 6B LLM to
surpass GPT4 when integrated with the Thinker. This paper also contributes the
largest dataset for social deduction games to date
Towards Real-World Visual Tracking with Temporal Contexts
Visual tracking has made significant improvements in the past few decades.
Most existing state-of-the-art trackers 1) merely aim for performance in ideal
conditions while overlooking the real-world conditions; 2) adopt the
tracking-by-detection paradigm, neglecting rich temporal contexts; 3) only
integrate the temporal information into the template, where temporal contexts
among consecutive frames are far from being fully utilized. To handle those
problems, we propose a two-level framework (TCTrack) that can exploit temporal
contexts efficiently. Based on it, we propose a stronger version for real-world
visual tracking, i.e., TCTrack++. It boils down to two levels: features and
similarity maps. Specifically, for feature extraction, we propose an
attention-based temporally adaptive convolution to enhance the spatial features
using temporal information, which is achieved by dynamically calibrating the
convolution weights. For similarity map refinement, we introduce an adaptive
temporal transformer to encode the temporal knowledge efficiently and decode it
for the accurate refinement of the similarity map. To further improve the
performance, we additionally introduce a curriculum learning strategy. Also, we
adopt online evaluation to measure performance in real-world conditions.
Exhaustive experiments on 8 wellknown benchmarks demonstrate the superiority of
TCTrack++. Real-world tests directly verify that TCTrack++ can be readily used

Epigenetic hypomethylation and upregulation of GD3s in triple negative breast cancer.
Background: Breast cancer remains a major health problem in the world. Triple-negative breast cancer (TNBC) is an aggressive subtype with very poor prognosis. Up to now, the mechanism behind TNBC\u27s activity is still unclear and no candidate drug target has been identified. Thus, it is of critical importance to elucidate the pathways in TNBC and identify the relevant biomarkers. Recent studies showed that ganglioside D3 synthase (GD3s) played a very important role in development of cancers. However, the physiological functions and associated pathways of GD3s in TNBC are still unclear.

miR-29c plays a suppressive role in breast cancer by targeting the TIMP3/STAT1/FOXO1 pathway.
Background: miR-29c has been associated with the progression of many cancers. However, the function and mechanism of miR-29c have not been well investigated in breast cancers.
Methods: Real-time quantitative PCR was used to assess expression of miR-29c and DNMT3B mRNA. Western blot and immunochemistry were used to examine the expression of DNA methyltransferase 3B (DNMT3B) protein in breast cancer cells and tissues. The functional roles of miR-29c in breast cancer cells such as proliferation, migration, invasion, colony formation, and 3D growth were evaluated using MTT, transwell chambers, soft agar, and 3D Matrigel culture, respectively. In addition, the luciferase reporter assay was used to check if miR-29c binds the 3\u27UTR of DNMT3B. The effects of miR-29c on the DNMT3B/TIMP3/STAT1/FOXO1 pathway were also examined using Western blot and methyl-specific qPCR. The specific inhibitor of STAT1, fludarabine, was used to further check the mechanism of miR-29c function in breast cancer cells. Studies on cell functions were carried out in DNMT3B siRNA cell lines.
Results: The expression of miR-29c was decreased with the progression of breast cancers and was closely associated with an overall survival rate of patients. Overexpression of miR-29c inhibited the proliferation, migration, invasion, colony formation, and growth in 3D Matrigel while knockdown of miR-29c promoted these processes in breast cancer cells. In addition, miR-29c was found to bind 3\u27UTR of DNMT3B and inhibits the expression of DNMT3B, which was elevated in breast cancers. Moreover, the protein level of TIMP3 was reduced whereas methylation of TIMP3 was increased in miR-29c knockdown cells compared to control. On the contrary, the protein level of TIMP3 was increased whereas methylation of TIMP3 was reduced in miR-29c-overexpressing cells compared to control. Knockdown of DNMT3B reduced the proliferation, migration, and invasion of breast cancer cell lines. Finally, our results showed that miR-29c exerted its function in breast cancers by regulating the TIMP3/STAT1/FOXO1 pathway.
Conclusion: The results suggest that miR-29c plays a significant role in suppressing the progression of breast cancers and that miR-29c may be used as a biomarker of breast cancers
Hippo dictates signaling for cellular homeostasis and immune defense in Crassostrea hongkongensis hemocytes
IntroductionThe Hippo signaling pathway is an evolutionarily conserved signaling cascade that plays a crucial role in regulating cell proliferation, differentiation, and apoptosis. It has been shown to be a key regulator of cell fate and cellular homeostasis in various immune processes. Despite its well-established functions in vertebrate immunity, its roles in marine invertebrate immunity remain poorly understood. Therefore, our present work provides fresh mechanistic insights into how the Hippo pathway orchestrates hemocytic functions in Crassostrea hongkongensis, with implications for studies on its major forms and modifications in animal evolution.MethodThe complete set of Hippo pathway genes, including SAV1, MOB1, LATS, YAP/TAZ, TEAD, and MST, were identified from the C. hongkongensis genome. Quantitative PCR assays were conducted to examine the mRNA expression levels of these genes in different tissues and the levels of these genes in hemocytes before and after bacterial challenges. The study also examined the crosstalk between the Hippo pathway and other immune pathways, such as the AP-1 and p53-dependent p21 signaling cascades. RNA interference was used to knock down MST and TEAD, and MST is a core orchestrator of non-canonical Hippo signaling, to investigate its impact on phagocytosis and bacterial clearance in hemocytes.ResultThe results demonstrated that members of the Hippo pathway were highly expressed in hemocytes, with their expression levels significantly increasing following bacterial challenges. Crosstalk between the Hippo pathway and other immune pathways triggered hemocytic apoptosis, which functioned similarly to the canonical Mst-Lats-Yap signaling pathway in Drosophila and mammals. Knocking down MST resulted in increased phagocytosis and boosted the efficiency of bacterial clearance in hemocytes, presumably due to mobilized antioxidant transcription by Nrf for maintaining immune homeostasis.DiscussionThis study provides novel insights into the regulatory mechanisms underlying the Hippo pathway in immune responses of C. hongkongensis hemocytes. The study highlights the importance of the Hippo pathway in maintaining immune homeostasis and orchestrating hemocytic functions in oysters. Moreover, this study demonstrates the divergence of the Hippo pathway's roles in marine invertebrate immunity from mammalian observations, indicating the need for further comparative studies across species. These findings have significant implications for future research aimed at elucidating the evolutionary trajectory and functional diversity of the Hippo signaling pathway in animal evolution
Temporal and structural patterns of hepatitis B virus integrations in hepatocellular carcinoma
Chronic infection of hepatitis B virus (HBV) is the major cause of hepatocellular carcinoma (HCC). Notably, 90% of HBV-positive HCC cases exhibit detectable HBV integrations, hinting at the potential early entanglement of these viral integrations in tumorigenesis and their subsequent oncogenic implications. Nevertheless, the precise chronology of integration events during HCC tumorigenesis, alongside their sequential structural patterns, has remained elusive thus far. In this study, we applied whole-genome sequencing to multiple biopsies extracted from six HBV-positive HCC cases. Through this approach, we identified point mutations and viral integrations, offering a blueprint for the intricate tumor phylogeny of these samples. The emergent narrative paints a rich tapestry of diverse evolutionary trajectories characterizing the analyzed tumors. We uncovered oncogenic integration events in some samples that appear to happen before and during the initiation stage of tumor development based on their locations in reconstituted trajectories. Furthermore, we conducted additional long-read sequencing of selected samples and unveiled integration-bridged chromosome rearrangements and tandem repeats of the HBV sequence within integrations. In summary, this study revealed premalignant oncogenic and sequential complex integrations and highlighted the contributions of HBV integrations to HCC development and genome instability
Hand-drawn sketch and vector map matching based on topological features
In the process of addressing, when people use words to express indistinctly, they often draw simple sketches to assist expression, which helps people to form a simple spatial scene in the brain and correspond to the actual scene one by one, and finally locate and find the target address. How to establish an one-to-one mapping relationship between the spatial objects in the hand-drawn sketch and in the vector map is the key to the realization of map addressing and location, and this process is also the process of map matching. This paper aims to address difficult problems associated with the features of hand-drawn sketches and vector map matching in order to improve the use of all potential matching points designed for application in hand-drawn sketches and spatial relation matrix structures of vector maps. To accomplish this, we use the N-queen problem solving process and improve the tabu search algorithm. In the matching process under the constraint of a single spatial relationship, and the hierarchical matching process under the constraint of multiple spatial relations, this study verifies the quality of the spatial relationship and the feasibility and effectiveness of the matching method of hand-drawn sketches and vector maps using the improved tabu search algorithm
Optimization of Traced Neuron Skeleton Using Lasso-Based Model
Reconstruction of neuronal morphology from images involves mainly the extraction of neuronal skeleton points. It is an indispensable step in the quantitative analysis of neurons. Due to the complex morphology of neurons, many widely used tracing methods have difficulties in accurately acquiring skeleton points near branch points or in structures with tortuosity. Here, we propose two models to solve these problems. One is based on an L1-norm minimization model, which can better identify tortuous structure, namely, a local structure with large curvature skeleton points; the other detects an optimized branch point by considering the combination patterns of all neurites that link to this point. We combined these two models to achieve optimized skeleton detection for a neuron. We validate our models in various datasets including MOST and BigNeuron. In addition, we demonstrate that our method can optimize the traced skeletons from large-scale images. These characteristics of our approach indicate that it can reduce manual editing of traced skeletons and help to accelerate the accurate reconstruction of neuronal morphology
