Identifying components required for OMP biogenesis as novel targets for antiinfective drugs
The emergence of multiresistant Gram-negative bacteria requires new therapies for combating bacterial infections. Targeting the biogenesis of virulence factors could be an alternative strategy instead of killing bacteria with antibiotics. The outer membrane (OM) of Gram-negative bacteria acts as a physical barrier. At the same time it facilitates the exchange of molecules and harbors a multitude of proteins associated with virulence. In order to insert proteins into the OM, an essential oligomeric membrane-associated protein complex, the ss-barrel assembly machinery (BAM) is required. Being essential for the biogenesis of outer membrane proteins (OMPs) the BAM and also periplasmic chaperones may serve as attractive targets to develop novel antiinfective agents. Herein, we aimed to elucidate which proteins belonging to the OMP biogenesis machinery have the most important function in granting bacterial fitness, OM barrier function, facilitating biogenesis of dedicated virulence factors and determination of overall virulence. To this end we used the enteropathogen Yersinia enterocolitica as a model system. We individually knocked out all non-essential components of the BAM (BamB, C and E) as well as the periplasmic chaperones DegP, SurA and Skp. In summary, we found that the most profound phenotypes were produced by the loss of BamB or SurA with both knockouts resulting in significant attenuation or even avirulence of Ye in a mouse infection model. Thus, we assume that both BamB and SurA are promising targets for the development of new antiinfective drugs in the future.Peer reviewe
Deep sequencing uncovers numerous small RNAs on all four replicons of the plant pathogen Agrobacterium tumefaciens
Agrobacterium species are capable of interkingdom gene transfer between bacteria and plants. The genome of Agrobacterium tumefaciens consists of a circular and a linear chromosome, the At-plasmid and the Ti-plasmid, which harbors bacterial virulence genes required for tumor formation in plants. Little is known about promoter sequences and the small RNA (sRNA) repertoire of this and other α-proteobacteria. We used a differential RNA sequencing (dRNA-seq) approach to map transcriptional start sites of 388 annotated genes and operons. In addition, a total number of 228 sRNAs was revealed from all four Agrobacterium replicons. Twenty-two of these were confirmed by independent RNA gel blot analysis and several sRNAs were differentially expressed in response to growth media, growth phase, temperature or pH. One sRNA from the Ti-plasmid was massively induced under virulence conditions. The presence of 76 cis-antisense sRNAs, two of them on the reverse strand of virulence genes, suggests considerable antisense transcription in Agrobacterium. The information gained from this study provides a valuable reservoir for an in-depth understanding of sRNA-mediated regulation of the complex physiology and infection process of Agrobacterium
Evaluierung und Optimierung biologischer Verfahren zur Regulierung des Pflaumenwicklers (Cydia funebrana) und der Monilia-Krankheit im ökologischen Steinobstanbau
Im Verbundforschungsprojekt FKZ 06OE198, FKZ 06OE057 und FKZ 06OE348 wurden Möglichkeiten der Regulierung des Pflaumenwicklers (Cydia funebrana) an Zwetschgen in Labor-, Halbfreiland und Freilandversuchen in Geisenheim, Darmstadt, Klein-Altendorf und Weinsberg untersucht. Labor- und Freilandversuche zur Regulierung der Monilia-Krankheit wurden in Geisenheim, Weinsberg und Dresden durchgefĂŒhrt. Sauerkirschsorten-Versuche sind in Weinsberg und Dresden aufgepflanzt.
Umfangreiche Freilandversuche zur Verwirrung des Pflaumenwicklers mit âIsomate OFM Rossoâ in Baden-WĂŒrttemberg und Rheinland-Pfalz bestĂ€tigten mit Wirkungsgraden von bis zu 70 % die Wirksamkeit dieser Methode bei ausreichend groĂen FlĂ€chen und niedrigem Befallsniveau. FĂŒr den kleinparzellierten Anbau und bei höherem Befallsdruck wurden entomopathogene Nematoden im Freiland gegen ĂŒberwinternde Larven mit heterogenen Wirkungsgraden erprobt.
Es gelang nicht Pflaumenwickler ganzjĂ€hrig unter Laborbedingen zu zĂŒchten, jedoch wurden mit Tieren aus Freilandsammlungen bzw. deren Eiern nach der Ăberwinterung Versuche zur Infektion der neonaten Larven mit dem Apfelwicklergranulosevirus, Eiparasitierungsversuche mit Trichogramma cacoeciae, zu Applikationen mit NeemAzal-T/S und zum Einsatz von insektenpathogen Pilzen gegen ĂŒberwinternde Larven durchgefĂŒhrt. Bei weiterfĂŒhrenden Versuchen im Freiland wurden mit dem Apfelwicklergranulosevirus V15 sehr heterogene Ergebnisse erzielt. Der Einsatz von Trichogramma in Form von Trichokarten ergab Wirkungsgrade von bis zu 47 %. Eine bessere Verteilung der Trichogramma in der Anlage sollte mit einer spritzfĂ€higen Formulierung erreicht werden, dafĂŒr liegen jedoch erst einjĂ€hrige Ergebnisse vor. FĂŒr die BekĂ€mpfung der ĂŒberwinternden Pflaumenwickler mit insektenpathogenen Pilzen stellte sich Isaria fumosorosea, als am geeignetsten heraus. Jedoch gestaltete sich bisher ein Nachweis der Wirksamkeit des mit dem Pilz infizierten Mulches im Freiland sehr schwierig. Die Pflaumenwicklerlarve sucht sich zur Ăberwinterung kleinste Ritzen am Stamm aus. Mit Wellpappe, die um den Stamm gewickelt wurde, konnte den Larven ein attraktives Ăberwinterungsquartier angeboten werden. Durch Entfernen und Verbrennen der Pappringe im Winter kann der Befallsdruck im Folgejahr minimiert werden.
FĂŒr die Monilia-Regulierung an Zwetschgen konnte im Labor unter einer Vielzahl an getesteten PrĂ€paraten lediglich Serenade mit einer antagonistischen Wirkung ĂŒberzeugen. Es hat jedoch keine Zulassung als Pflanzenschutzmittel im Steinobst. Versuche im Freiland ergaben auĂerdem, dass das konsequente Eliminieren von möglichen Infektionsquellen durch das Entfernen von Monilia-Nestern vor der Ernte und von Fruchtmumien nach der Ernte letztlich bessere Effekte erzielte, als Behandlungen mit mittelgut wirksamen PflanzenstĂ€rkungsmitteln.
FĂŒr die Behandlung der Monilia-SpitzendĂŒrre an Sauerkirschen wurden verschiedene PrĂ€parate im Freiland ĂŒberprĂŒft. Die Handelsprodukte BoniProtectÂź forte und BoniProtectÂź reduzierten tendenziell die BlĂŒteninfektionen. Da fĂŒr SPU 2720 und Cuprozin bisher nur aus einem Jahr Versuchsergebnisse vorliegen, ist keine abschlieĂende Beurteilung möglich. Jedoch lĂ€sst sich bereits durch den konsequenten RĂŒckschnitt befallener Triebe der Befallsdruck erheblich reduzieren.
An beiden Standorten erscheinen die Sauerkirschsorten âMorinaâ, âJadeâ, âCoralinâ und âSafirâ als geeignet fĂŒr den ökologischen Anbau, in Weinsberg zusĂ€tzlich âTopasâ und âUngarische Traubigeâ. Allerdings war der Monilia-Druck nur in wenigen Jahren hoch. Bis jetzt wurden keine Infektionen mit Gloeosporium sp., einer weiteren bedrohlichen Krankheit im ökologischen Sauerkirschanbau, beobachtet
Chartis Measurement of Collateral Ventilation:Conscious Sedation versus General Anesthesia
BACKGROUND: Absence of interlobar collateral ventilation using the Chartis measurement is the key predictor for successful endobronchial valve treatment in severe emphysema. Chartis was originally validated in spontaneous breathing patients under conscious sedation (CS); however, this can be challenging due to cough, mucus secretion, mucosal swelling, and bronchoconstriction. Performing Chartis under general anesthesia (GA) avoids these problems and may result in an easier procedure with a higher success rate. However, using Chartis under GA with positive pressure ventilation has not been validated. OBJECTIVES: In this study we investigated the impact of anesthesia technique, CS versus GA, on the feasibility and outcomes of Chartis measurement. METHODS: We retrospectively analyzed all Chartis measurements performed at our hospital from October 2010 until December 2017. RESULTS: We analyzed 250 emphysema patients (median forced expiratory volume in 1 s 26%, range 12-52% predicted). In 121 patients (48%) the measurement was performed using CS, in 124 (50%) using GA, and in 5 (2%) both anesthesia techniques were used. In total, 746 Chartis readings were analyzed (432 CS, 277 GA, and 37 combination). Testing under CS took significantly longer than GA (median 19 min [range 5-65] vs. 11 min [3-35], p < 0.001) and required more measurements (3 [1-13] vs. 2 [1-6], p < 0.001). There was no significant difference in target lobe volume reduction after treatment (-1,123 mL [-3,604 to 332] in CS vs. -1,251 mL [-3,333 to -1] in GA, p = 0.35). CONCLUSIONS: In conclusion, Chartis measurement under CS took significantly longer and required more measurements than under GA, without a difference in treatment outcome. We recommend a prospective trial comparing both techniques within the same patients to validate this approach
Collateral Ventilation Measurement Using Chartis Procedural Sedation vs General Anesthesia:procedural sedation versus general anesthesia
BACKGROUND: Absence of interlobar collateral ventilation is key to successful endobronchial valve treatment in patients with severe emphysema and can be functionally assessed by using the Chartis measurement. This system has been validated during spontaneous breathing, undergoing procedural sedation (PS), but can also be performed under general anesthesia. Performing the Chartis measurement under PS is often challenging because of coughing, mucus secretion, and difficulties in maintaining an adequate level of sedation. The objective of this study was to investigate whether there is a difference in Chartis measurement outcomes between PS and general anesthesia.
METHODS: In this prospective study, patients underwent Chartis measurements under both PS and general anesthesia. Study outcomes were Chartis measurement duration, number of measurements, feasibility, and success rate.
RESULTS: The study included 30 patients with severe emphysema (mean age, 62 years; median FEV1, 29% of predicted). Chartis measurement duration was significantly longer under PS than under general anesthesia (mean, 20.3 +/- 4.2 min vs 15.1 +/- 4.4 min; P < .001). There was no difference in the number (median [range]) of measurements performed (2 [1-3] for PS vs 1 [1-3] for general anesthesia; P= 1.00). Chartis measurement was more feasible during general anesthesia (median sum of all feasibility scores, 12 [range, 6-26] for PS vs 7 [5-13] for general anesthesia; P < .001). There was no statistical difference in success rate: 77% of PS cases vs 97% of general anesthesia cases (P = .07).
CONCLUSIONS: This study found that Chartis measurement under general anesthesia is faster and more feasible to perform compared with performance with PS, without affecting measurement outcomes
Comparative grading scales, statistical analyses, climber descriptors and ability grouping: International Rock Climbing Research Association position statement
The research base for rock climbing has expanded substantially in the past 3 decades as worldwide interest in the sport has grown. An important trigger for the increasing research attention has been the transition of the sport to a competitive as well as recreational activity and the potential inclusion of sport climbing in the Olympic schedule. The International Rock Climbing Research Association (IRCRA) was formed in 2011 to bring together climbers, coaches and researchers to share knowledge and promote collaboration. This position statement was developed during and after the 2nd IRCRA Congress which was held in Pontresina, in September 2014. The aim of the position statement is to bring greater uniformity to the descriptive and statistical methods used in reporting rock climbing research findings. To date there is a wide variation in the information provided by researchers regarding the climbersâ characteristics and also in the approaches employed to convert from climbing grading scales to a numeric scale suitable for statistical analysis. Our paper presents details of recommended standards of reporting that should be used for reporting climber characteristics and provides a universal scale for the conversion of climbing grades to a number system for statistical analysis
Utilization of livers donated after circulatory death for transplantation - An international comparison.
BACKGROUND AND AIM
Liver graft utilization rates are a hot topic due to the worldwide organ shortage and an increasing number of transplant candidates on waiting lists. Liver perfusion techniques have been introduced in several countries, and may help to increase the organ supply, as they potentially allow the assessment of livers before use.
METHODS
Liver offers were counted from donation after circulatory death (DCD) donors (Maastricht-type-III) arising during the past decade in eight countries, including Belgium, France, Italy, the Netherlands, Spain, Switzerland, UK, and US. Initial DCD-type-III liver offers were correlated with accepted, recovered and implanted livers.
RESULTS
A total number of 34`269 DCD livers were offered, resulting in 9`780 liver transplants (28.5%). The discard rates were highest in UK and US, ranging between 70 and 80%. In contrast, much lower DCD liver discard rates, e.g., between 30-40%, were found in Belgium, France, Italy, Spain and Switzerland. In addition, large differences were recognized in the use of various machine perfusion techniques, and in terms of risk factors in the cohorts of implanted livers. For example, the median donor age and functional donor warm ischemia were highest in Italy, e.g., >40minutes, followed by Switzerland, France, and the Netherlands. Importantly, such varying risk profiles of accepted DCD livers between countries did not translate into large differences in five-year graft survival rates, which ranged between 60-82% in this analysis.
CONCLUSIONS
We highlight a significant number of discarded and consequently unused DCD liver offers. Countries with more routine use of in- and ex-situ machine perfusion strategies showed better DCD utilization rates without compromised outcome.
IMPACT AND IMPLICATIONS
A significant number of Maastricht type III DCD livers are discarded across Europe and North America today. The overall utilization rate among eight Western countries is 28.5%, but varies significantly between 18.9% and 74.2%. For example, the median DCD III liver utilization in five countries, e.g., Belgium, France, Italy, Switzerland, and Spain is 65%, in contrast to 24% in the Netherlands, UK and US. Despite this, and despite different rules and strategies for organ acceptance and preservation, the one and five-year graft survival remains currently relatively comparable among all participating countries. Factors which impact on DCD liver acceptance rates include the national pre-selections of donors, before the offer is made, as well as cutoffs for key risk factors, including donor age and donor warm ischemia time. In addition, a highly varying experience with modern machine perfusion technology is noticed. In situ and ex situ liver perfusion concepts, and assessment tools for type III DCD livers before transplantation may be one key part for the observed differences in better DCD III utilization
Pan-cancer Alterations of the MYC Oncogene and Its Proximal Network across the Cancer Genome Atlas
Although theMYConcogene has been implicated incancer, a systematic assessment of alterations ofMYC, related transcription factors, and co-regulatoryproteins, forming the proximal MYC network (PMN),across human cancers is lacking. Using computa-tional approaches, we define genomic and proteo-mic features associated with MYC and the PMNacross the 33 cancers of The Cancer Genome Atlas.Pan-cancer, 28% of all samples had at least one ofthe MYC paralogs amplified. In contrast, the MYCantagonists MGA and MNT were the most frequentlymutated or deleted members, proposing a roleas tumor suppressors.MYCalterations were mutu-ally exclusive withPIK3CA,PTEN,APC,orBRAFalterations, suggesting that MYC is a distinct onco-genic driver. Expression analysis revealed MYC-associated pathways in tumor subtypes, such asimmune response and growth factor signaling; chro-matin, translation, and DNA replication/repair wereconserved pan-cancer. This analysis reveals insightsinto MYC biology and is a reference for biomarkersand therapeutics for cancers with alterations ofMYC or the PMN

The association between frailty and MRI features of cerebral small vessel disease
Abstract: Frailty is a common syndrome in older individuals that is associated with poor cognitive outcome. The underlying brain correlates of frailty are unclear. The aim of this study was to investigate the association between frailty and MRI features of cerebral small vessel disease in a group of non-demented older individuals. We included 170 participants who were classified as frail (n = 30), pre-frail (n = 85) or non-frail (n = 55). The association of frailty and white matter hyperintensity volume and shape features, lacunar infarcts and cerebral perfusion was investigated by regression analyses adjusted for age and sex. Frail and pre-frail participants were older, more often female and showed higher white matter hyperintensity volume (0.69 [95%-CI 0.08 to 1.31], p = 0.03 respectively 0.43 [95%-CI: 0.04 to 0.82], p = 0.03) compared to non-frail participants. Frail participants showed a non-significant trend, and pre-frail participants showed a more complex shape of white matter hyperintensities (concavity index: 0.04 [95%-CI: 0.03 to 0.08], p = 0.03; fractal dimensions: 0.07 [95%-CI: 0.00 to 0.15], p = 0.05) compared to non-frail participants. No between group differences were found in gray matter perfusion or in the presence of lacunar infarcts. In conclusion, increased white matter hyperintensity volume and a more complex white matter hyperintensity shape may be structural brain correlates of the frailty phenotype
