    Átomos individuais: Um desafio para a química verde

    The growing demand for various consumer products due to the increasing world population directly impacts the environment. Recently, the study and development of new materials based on single atoms (SA) is the new paradigm for green chemistry to deal with the negative effect of raw material overexploitation. This paper explains key concepts to understand SA-based materials, their synthesis, applications, the main analytical techniques for their characterization, and their relationship with environmental chemistry as a crucial technological element for sustainable development.La creciente demanda de diversos productos de consumo debido al aumento de la población mundial impacta directamente el ambiente. En años recientes, el estudio y desarrollo de nuevos materiales basados en átomos individuales (SA) constituyen un nuevo paradigma de la eficiencia en la química verde para enfrentar los impactos negativos de la sobreexplotación de las materias primas.     Simultaneous assessment of rotavirus-specific memory B cells and serological memory after B cell depletion therapy with rituximab

    Q2Q11-12The mechanisms that contribute to the maintenance of serological memory are still unclear. Rotavirus (RV) memory B cells(mBc) are enriched in IgM+and CD27-subpopulations, which are associated with autoimmune diseases pathogenesis. Inpatients with autoimmune diseases treated with Rituximab (RTX), some autoantibodies (auto-Abs) decrease after treatment,but other auto-Abs and pathogen-specific IgG Abs remain unchanged. Thus, maintenance of autoimmune and pathogen-specific serological memory may depend on the type of antigen and/or Ab isotype evaluated. Antigen-specific mBc andantigen-specific Abs of different isotypes have not been simultaneously assessed in patients after RTX treatment. To study the relationship between mBc subpopulations and serological memory we characterized total, RV- and tetanus toxoid (TT)-specific mBc by flow cytometry in patients with autoimmune diseases before and after treatment with RTX. We also measured total, RV- and TT-Abs, and some auto-Abs by kinetic nephelometry, ELISA, and EliA tests, respectively. Minor differences were observed between the relative frequencies of RV-mBc in healthy controls and patients with autoimmune disease. After RTX treatment, naı ̈ve Bc and total, RV- and TT-specific mBc [IgM+, switched (IgA+/IgG+), IgM+only, IgD+only, and CD27-(IgA+/IgG+/IgM+)] were significantly diminished. An important decrease in total plasma IgM and minor decrease in total IgG and IgA levels were also observed. IgM rheumatoid factor, IgG anti-CCP, and IgG anti-dsDNA were significantly diminished. In contrast, RV-IgA, RV-IgG and RV-IgG1, and TT-IgG titers remained stable. In conclusion, in patients with autoimmunity, serological memory against RV and TT seem to be maintained by long-lived plasma cells, unaffected by RTX, and an important proportion of total IgM and serological memory against some auto-antigens seem to be maintained by short-lived plasma cells, dependent on mBc precursors depleted by RTX

    Análisis de la Implementación del Sistema de Gestión, Seguridad, y salud en el trabajo en el área Administrativa de la empresa Vida Plena, para el primer semestre del año 2021

    Este trabajo se enmarcó en el análisis del sistema de Gestión de la Seguridad y Salud en el Trabajo; cuyo objetivo principal es establecer propuesta estratégica de mejoramiento del SG-SST, del área administrativa de la Empresa Vida Plena, El tipo de investigación utilizado en la elaboración del presente proyecto es de estudio de tipo descriptivo cuantitativo; lo anterior se realizó a través de una metodología de corte transversal. Para este caso se analizaron e identificaron factores de riesgo existentes en el área administrativa y su grado de incidencia en el desarrollo normal de las actividades propias de las funciones de cada uno de los trabajadores. En los resultados; evidenciamos que el objeto de estudio conoce que existe un programa de SG-SST; pero se refleja que no se realizan capacitaciones, ni tampoco evalúan que porten el equipo de seguridad, motivos y factores que incidieron, en la realización de esta investigación; con el fin de definir estrategias de intervención para un control integral del mismo generados por accidentes de trabajo y exposiciones laborales. Se puede concluir que, se pueden obtener oportunidades de mejora, para la formulación de planes de acción que garanticen un mejoramiento continuo del SG-SST en Vida Plena.This work was framed in the analysis of the Occupational Health and Safety Management system; whose main objective is to establish a strategic proposal for the improvement of the SG-SST, of the administrative area of the Full Life Company. The type of research used in the preparation of this project is a quantitative descriptive study; This was done through a cross-sectional methodology. For this case, existing risk factors in the administrative area and their degree of incidence in the normal development of the activities of the functions of each of the workers were analyzed and identified. In the results; We show that the object of study knows that there is an SG-OSH program; but it is reflected that training is not carried out, nor is it evaluated that they carry the security equipment, reasons and factors that had an influence, in the realization of this investigation; in order to define intervention strategies for a comprehensive control of the same generated by accidents at work and occupational exposures. It can be concluded that opportunities for improvement can be obtained for the formulation of action plans that guarantee continuous improvement of the SG-SST in Full Life

    Lucilia eximia (Diptera: Calliphoridae), a new alternative for maggot therapy. Case series report

    RESUMEN: La terapia larval es utilizada desde los años 30 del siglo pasado para la remoción del tejido necrótico en el tratamiento de úlceras crónicas infectadas logrando con ello promover la formación de tejido granuloso para el crecimiento de piel sana; especialmente a partir de la aparición de la resistencia a los antibióticos se la reconoce como una alternativa eficaz. Se presentan los resultados de la aplicación de la terapia larval en 42 pacientes con úlceras de diferentes orígenes, utilizando, como una nueva alternativa en larviterapia, la especie Lucilia eximia. Se presenta documentación fotográfica de la aplicación de esta terapia en cuatro casosSUMMARY: Larval therapy is used in the treatment of infected chronic wounds by allowing the removal of necrotic tissue, which induces the formation of granular tissue and the growth of healthy skin. Considering the increasing prevalence of antibiotic resistance, this type of therapy may be an effective alternative in the management of infected chronic wounds. In this article we report the use of maggot therapy using the Lucilia eximia species in 42 patients with chronic skin wounds associated to different pathologies including: venous and arterial ulcers, diabetic foot, sickle cell disease, vasculopathy, elephantiasis, Berger disease, pyoderma gangrenosum (PG), traumatic wounds, erysipelas, and hospital acquired infections.     Extracellular vesicles from pristane-treated CD38-deficient mice express an antiinflammatory neutrophil protein signature, which reflects the mild lupus severity elicited in these mice

    In CD38-deficient (Cd38-/-) mice intraperitoneal injection of pristane induces a lupus-like disease, which is milder than that induced in WT mice, showing significant differences in the inflammatory and autoimmune processes triggered by pristane. Extracellular vesicles (EV) are present in all body fluids. Shed by cells, their molecular make-up reflects that of their cell of origin and/or tissue pathological situation. The aim of this study was to analyze the protein composition, protein abundance, and functional clustering of EV released by peritoneal exudate cells (PECs) in the pristane experimental lupus model, to identify predictive or diagnostic biomarkers that might discriminate the autoimmune process in lupus from inflammatory reactions and/or normal physiological processes. In this study, thanks to an extensive proteomic analysis and powerful bioinformatics software, distinct EV subtypes were identified in the peritoneal exudates of pristane-treated mice: 1) small EV enriched in the tetraspanin CD63 and CD9, which are likely of exosomal origin; 2) small EV enriched in CD47 and CD9, which are also enriched in plasma-membrane, membrane-associated proteins, with an ectosomal origin; 3) small EV enriched in keratins, ECM proteins, complement/coagulation proteins, fibrin clot formation proteins, and endopetidase inhibitor proteins. This enrichment may have an inflammation-mediated mesothelial-tomesenchymal transition origin, representing a protein corona on the surface of peritoneal exudate EV; 4) HDL-enriched lipoprotein particles. Quantitative proteomic analysis allowed us to identify an anti-inflammatory, Annexin A1- enriched pro-resolving, neutrophil protein signature, which was more prominent in EV from pristane-treated Cd38-/- mice, and quantitative differences in the protein cargo of the ECM-enriched EV from Cd38-/- vs WT mice. These differences are likely to be related with the distinct inflammatory outcome shown by Cd38-/- vs WT mice in response to pristane treatment. Our results demonstrate the power of a hypothesis-free and data-driven approach to transform the heterogeneity of the peritoneal exudate EV from pristanetreated mice in valuable information about the relative proportion of different EV in a given sample and to identify potential protein markers specific for the different small EV subtypes, in particular those proteins defining EV involved in the resolution phase of chronic inflammation.Proyecto del plan estatal, Ministerio de Ciencia e Innovacion PT13/0001/011CSIC PT17/0019/0010 PID2020-119567RB-I0

    Multicentric study of cervical cancer screening with human papillomavirus testing and assessment of triage methods in Latin America : the ESTAMPA screening study protocol

    Q1Q1Introduction Human papillomavirus (HPV) testing is replacing cytology in primary screening. Its limited specificity demands using a second (triage) test to better identify women at high-risk of cervical disease. Cytology represents the immediate triage but its low sensitivity might hamper HPV testing sensitivity, particularly in low-income and middle-income countries (LMICs), where cytology performance has been suboptimal. The ESTAMPA (EStudio multicéntrico de TAMizaje y triaje de cáncer de cuello uterino con pruebas del virus del PApiloma humano; Spanish acronym) study will: (1) evaluate the performance of different triage techniques to detect cervical precancer and (2) inform on how to implement HPV-based screening programmes in LMIC. Methods and analysis Women aged 30–64 years are screened with HPV testing and Pap across 12 study centres in Latin America. Screened positives have colposcopy with biopsy and treatment of lesions. Women with no evident disease are recalled 18 months later for another HPV test; those HPV-positive undergo colposcopy with biopsy and treatment as needed. Biological specimens are collected in different visits for triage testing, which is not used for clinical management. The study outcome is histological high-grade squamous intraepithelial or worse lesions (HSIL+) under the lower anogenital squamous terminology. About 50 000 women will be screened and 500 HSIL+ cases detected (at initial and 18 months screening). Performance measures (sensitivity, specificity and predictive values) of triage techniques to detect HSIL+ will be estimated and compared with adjustment by age and study centre. Ethics and dissemination The study protocol has been approved by the Ethics Committee of the International Agency for Research on Cancer (IARC), of the Pan American Health Organisation (PAHO) and by those in each participating centre. A Data and Safety Monitoring Board (DSMB) has been established to monitor progress of the study, assure participant safety, advice on scientific conduct and analysis and suggest protocol improvements. Study findings will be published in peer-reviewed journals and presented at scientific meetings. Trial registration number NCT01881659Revista Internacional - Indexad

    Validation of <i>N</i>-myristoyltransferase as Potential Chemotherapeutic Target in Mammal-Dwelling Stages of <i>Trypanosoma cruzi</i>

    BACKGROUND:Trypanosoma cruzi causes Chagas disease, an endemic and debilitating illness in Latin America. Lately, owing to extensive population movements, this neglected tropical disease has become a global health concern. The two clinically available drugs for the chemotherapy of Chagas disease have rather high toxicity and limited efficacy in the chronic phase of the disease, and may induce parasite resistance. The development of new anti-T. cruzi agents is therefore imperative. The enzyme N-myristoyltransferase (NMT) has recently been biochemically characterized, shown to be essential in Leishmania major, Trypanosoma brucei, and T. cruzi¸ and proposed as promising chemotherapeutic target in these trypanosomatids. METHODOLOGY/PRINCIPAL FINDINGS:Here, using high-content imaging we assayed eight known trypanosomatid NMT inhibitors, against mammal-dwelling intracellular amastigote and trypomastigote stages and demonstrated that three of them (compounds 1, 5, and 8) have potent anti-proliferative effect at submicromolar concentrations against T. cruzi, with very low toxicity against human epithelial cells. Moreover, metabolic labeling using myristic acid, azide showed a considerable decrease in the myristoylation of proteins in parasites treated with NMT inhibitors, providing evidence of the on-target activity of the inhibitors. CONCLUSIONS/SIGNIFICANCE:Taken together, our data point out to the potential use of NMT inhibitors as anti-T. cruzi chemotherapy
