    The Relationship between Drug Consumption and Dating App Use: Results from an Italian Survey

    To date, the literature regarding the relationship between drug consumption and dating app use is still very scant and inconclusive. The present study was thus aimed at investigating the association between drug consumption and dating app use in the general population. A total of 1278 Italian respondents completed an online ad hoc questionnaire assessing drug consumption (cannabis versus other illicit drugs), dating app use, the primary motive for installing dating apps, and demographics. Multiple logistic regression analyses were run to investigate the role of demographics and dating app use on drug consumption. Being single predicted cannabis use. Using dating apps accounted for higher odds of cannabis use; however, people who intensely used the apps were less likely to consume marijuana. Conversely, dating app use was not associated with the consumption of other drugs. This study suggests the presence of common underlying factors between dating app use and cannabis use, and it highlights the mediating role of the intensity of app use. Conversely, the study suggests that dating app use and the use of other drugs are quite independent behaviors

    The Association between Dating Apps and Alcohol Consumption in an Italian Sample of Active Users, Former Users, and Non-Users

    To date, the relationship between alcohol use and dating app use has been investigated mostly in conjunction with sexual activities and in homosexual men. For this reason, the aim of this study was to explore the association between dating app use and alcohol consumption among the general population. A cross-sectional study was conducted including app users, non-users, and former users: 1278 respondents completed an online ad hoc questionnaire assessing dating app use, motivations for installing dating apps, alcohol use, and demographics. Multiple logistic regression analysis was run to investigate the association between dating app use, demographic features, and alcohol consumption. Whereas educational level, age, and gender significantly contributed to the regular consumption of alcohol, dating app use did not account for a significant amount of variance between regular and not regular drinkers. However, people who installed and used dating apps with the motivation of searching for sexual partners were more likely to be regular drinkers. Among the active users, heavy app users were less likely to drink regularly. The study indicates that underlying factors (sexual aspects, motives for using the apps) and the intensity of using the apps may mediate the relationship between dating app use and alcohol use

    Impaired release of Vitamin D in dysfunctional adipose tissue: New cues on Vitamin D supplementation in obesity

    Context: Vitamin D accumulates in adipose tissue (AT) and vitamin D deficiency is frequent in obesity. Objective: We hypothesize that trafficking of vitamin D is altered in dysfunctional AT. Design, Patients, Settings: 54 normal-weight and 67 obese males were recruited in a prospective study and randomly assigned to supplementation with 50 \ub5g/week 25-hydroxyvitamin-D3 (25(OH)D) or 150 \ub5g/week vitamin D3 for 1 year, raising dosage by 50% if vitamin D-sufficiency (serum 25(OH)D>50 nomol/l), was not achieved at 6 months; 97 subjects completed the study. Methods: Vitamin D3 (D3) and 25(OH)D were quantified by HPLC-MS in control and insulin-resistant (IR) 3T3-L1 cells and subcutaneous AT (SAT) from lean and obese subjects, incubated with or without adrenaline; expression of 25-hydroxylase (CYP27A1), 1\u3b1-hydroxylase (CYP27B1) and vitamin D receptor (VDR) were analysed by real-time PCR. Results: In IR adipocytes the uptake of D3 and 25(OH)D was higher, but after adrenaline stimulation, the decrement in D3 and 25(OH)D was stronger in control cells, which also showed increased expression of CYP27A1 and CYP27B1 and higher levels of 25(OH)D. In SAT from obese subjects, the adrenaline-induced release of D3 and 25(OH)D was blunted; in both IR cells and obese SAT, protein expression of \u3b22-adrenergic receptor was reduced. Supplementation with 25-hydroxyvitamin-D3 was more effective in achieving vitamin D sufficiency in obese, but not in normal weight subjects. Conclusion: Dysfunctional AT shows a reduced catecholamine-induced release of D3 and 25(OH)D, and altered activity of vitamin D-metabolizing enzymes, for these reasons supplementation with 25-hydroxyvitamin-D3 is more effective in obese individuals

    Heat Sensing Receptor TRPV1 Is a Mediator of Thermotaxis in Human Spermatozoa

    The molecular bases of sperm thermotaxis, the temperature-oriented cell motility, are currently under investigation. Thermal perception relies on a subclass of the transient receptor potential [TRP] channels, whose member TRPV1 is acknowledged as the heat sensing receptor. Here we investigated the involvement of TRPV1 in human sperm thermotaxis. We obtained semen samples from 16 normozoospermic subjects attending an infertility survey programme, testis biopsies from 6 patients with testicular germ cell cancer and testis fine needle aspirates from 6 patients with obstructive azoospermia undergoing assisted reproductive technologies. Expression of TRPV1 mRNA was assessed by RT-PCR. Protein expression of TRPV1 was determined by western blot, flow cytometry and immunofluorescence. Sperm motility was assessed by Sperm Class Analyser. Acrosome reaction, apoptosis and intracellular-Ca2+ content were assessed by flow cytometry. We found that TRPV1 mRNA and protein were highly expressed in the testis, in both Sertoli cells and germ-line cells. Moreover, compared to no-gradient controls at 31°C or 37°C (Ctrl 31°C and Ctrl 37°C respectively), sperm migration towards a temperature gradient of 31-37°C (T gradient) in non-capacitated conditions selected a higher number of cells (14,9 ± 4,2×106 cells T gradient vs 5,1± 0,3×106 cells Ctrl 31°C and 5,71±0,74×106 cells Ctrl 37°C; P = 0,039). Capacitation amplified the migrating capability towards the T gradient. Sperms migrated towards the T gradient showed enriched levels of both TRPV1 protein and mRNA. In addition, sperm cells were able to migrate toward a gradient of capsaicin, a specific agonist of TRPV1, whilst capsazepine, a specific agonist of TRPV1, blocked this effect. Finally, capsazepine severely blunted migration towards T gradient without abolishing. These results suggest that TRPV1 may represent a facilitating mediator of sperm thermotaxis

    Phthalates and heavy metals as endocrine disruptors in food: A study on pre-packed coffee products

    Phthalate plasticizers and heavy metals are widely recognized to be pollutants that interfere with key developmental processes such as masculinization. We investigated the release of phthalates and heavy metals in coffee brewed from coffee packed in single-serve coffee containers made from different types of materials: metal, biodegradable and plastics. We detected with GC\u2013MS small amounts phthalates, below the tolerated daily risks levels, in all the coffees prepared from the different types of capsules. Specifically, Di (2-ethyl-hexyl)-phthalate and DiBP: Diisobuthyl-pthalate were ubiquitously present despite the high variability among the samples (respective range 0.16\u20131.87 \u3bcg/mL and 0.01\u20130.36 \u3bcg/mL). Whereas, diethyl-phthalate (range 0.20\u20130.26 \u3bcg/mL) and di-n-buthyl-phthalate (range 0.02\u20130.14 \u3bcg/mL) were detected respectively in one and three out of the four types of capsule tested. In contrast, we detected by atomic mass spectrometry on mineralized samples heavy metals lead (Pb) and nickel (Ni), in all coffee tested. PB levels (respective range 0.32\u2013211.57 \u3bcg/dose) accounted for 42\u201379%, whereas Ni levels (respective range 166.25\u20131950.26 \u3bcg/dose) accounted for >100% of the tolerable daily intake. These results add to the already present concerns related to the multiple pathways of human exposure and the ubiquitous presence of these pollutants in consumer products and their long-term effect on human health

    Heterogeneous and rapid ice loss over the Patagonian Ice Fields revealed by CryoSat-2 swath radar altimetry

    The Northern and Southern Patagonian Ice Fields (NPI and SPI) in South America are the largest bodies of ice in the Southern hemisphere outside of Antarctica and the largest contributors to eustatic sea level rise (SLR) in the world, per unit area. Here we exploit swath processed CryoSat-2 interferometric data to produce maps of surface elevation change at sub-kilometer spatial resolution over the Ice Fields for six glaciological years between April 2011 and March 2017. Mass balance is calculated independently for nine sub-regions, including six individual glaciers larger than 300 km². Overall, between 2011 and 2017 the Patagonian Ice Fields have lost mass at a combined rate of 21.29 ± 1.98 Gt a−¹, contributing 0.059 ± 0.005 mm a−¹ to SLR. We observe widespread thinning on the Ice Fields, particularly north of 49° S. However the pattern of surface elevation change is highly heterogeneous, partly reflecting the importance of dynamic processes on the Ice Fields. The Jorge Montt glacier (SPI), whose tidewater terminus is approaching floatation, retreated ~2.5 km during our study period and lost mass at the rate of 2.20 ± 0.38 Gt a−¹ (4.64 ± 0.80 mwe a−¹). In contrast with the general pattern of retreat and mass loss, Pio XI, the largest glacier in South America, is advancing and gaining mass at 0.67 ± 0.29 Gt a−¹ rate
