Oral chondroitin sulfate and prebiotics for the treatment of canine Inflammatory Bowel Disease: a randomized, controlled clinical trial
BACKGROUND
Canine inflammatory bowel disease (IBD) is a chronic enteropathy of unknown etiology, although microbiome dysbiosis, genetic susceptibility, and dietary and/or environmental factors are hypothesized to be involved in its pathogenesis. Since some of the current therapies are associated with severe side effects, novel therapeutic modalities are needed. A new oral supplement for long-term management of canine IBD containing chondroitin sulfate (CS) and prebiotics (resistant starch, ÎČ-glucans and mannaoligosaccharides) was developed to target intestinal inflammation and oxidative stress, and restore normobiosis, without exhibiting any side effects. This double-blinded, randomized, placebo-controlled trial in dogs with IBD aims to evaluate the effects of 180Â days administration of this supplement together with a hydrolyzed diet on clinical signs, intestinal histology, gut microbiota, and serum biomarkers of inflammation and oxidative stress.
RESULTS
Twenty-seven client-owned biopsy-confirmed IBD dogs were included in the study, switched to the same hydrolyzed diet and classified into one of two groups: supplement and placebo. Initially, there were no significant differences between groups (pâ>â0.05) for any of the studied parameters. Final data analysis (supplement: nâ=â9; placebo: nâ=â10) showed a significant decrease in canine IBD activity index (CIBDAI) score in both groups after treatment (pâ<â0.001). After treatment, a significant decrease (1.53-fold; pâ<â0.01) in histologic score was seen only in the supplement group. When groups were compared, the supplement group showed significantly higher serum cholesterol (pâ<â0.05) and paraoxonase-1 (PON1) levels after 60Â days of treatment (pâ<â0.01), and the placebo group showed significantly reduced serum total antioxidant capacity (TAC) levels after 120Â days (pâ<â0.05). No significant differences were found between groups at any time point for CIBDAI, WSAVA histologic score and fecal microbiota evaluated by PCR-restriction fragment length polymorphism (PCR-RFLP). No side effects were reported in any group.
CONCLUSIONS
The combined administration of the supplement with hydrolyzed diet over 180Â days was safe and induced improvements in selected serum biomarkers, possibly suggesting a reduction in disease activity. This study was likely underpowered, therefore larger studies are warranted in order to demonstrate a supplemental effect to dietary treatment of this supplement on intestinal histology and CIBDAI

âNo, no Maama! Say âShaatir ya Ouledee Shaatirâ!â Childrenâs agency in language use and socialisation
Aims and objectives: This paper investigates how children in multilingual and transnational families mobilise their multiple and developing linguistic repertoires creatively to assert their agency in language use and socialisation, and why these acts of agency are conducive to successful maintenance of the so-called âhomeâ, âcommunityâ or âminorityâ language.
Methodology: Close, qualitative analysis of mealtime multiparty conversations is carried out to examine childrenâs agency in language use and socialisation.
Data and analysis: Twelve hours of mealtime conversations within one Arabic and English speaking multilingual family in the UK were recorded over a period of eight months. The excerpts selected for analysis in this paper illustrate how agency is enacted in interaction.
Findings: The data analyses of the familyâs language practices reveal both their flexible language policy and the importance the family attaches to Arabic. The children in this family are fully aware of the language preferences of their parents and are capable of manipulating that knowledge and asserting their agency through their linguistic choices to achieve their interactional goals.
Originality: This paper explores how Arabic is maintained as a minority language by second and third generations of Arabic-speaking immigrants in the UK through close analysis of conversations.
Significance: The findings contribute to the current discussions of family language policy and maintenance by demonstrating childrenâs agentive and creative roles in language use and socialisation. Three factors are identified as the reason for the successful language learning, use and maintenance of Arabic: firstly, a family language policy that has a positive multilingual outlook; secondly, family relationship dynamics that connect and bond family members; and thirdly, the childrenâs highly developed ability to understand their parentsâ language preferences
Shifting Attention From Theory to Practice in Philosophy of Biology
Traditional approaches in philosophy of biology focus attention on biological concepts, explanations, and theories, on evidential support and inter-theoretical relations. Newer approaches shift attention from concepts to conceptual practices, from theories to practices of theorizing, and from theoretical reduction to reductive retooling. In this article, I describe the shift from theory-focused to practice-centered philosophy of science and explain how it is leading philosophers to abandon fundamentalist assumptions associated with traditional approaches in philosophy of science and to embrace scientific pluralism. This article comes in three parts, each illustrating the shift from theory-focused to practice-centered epistemology. The first illustration shows how shifting philosophical attention to conceptual practice reveals how molecular biologists succeed in identifying coherent causal strands within systems of bewildering complexity. The second illustration suggests that analyzing how a multiplicity of alternative models function in practice provides an illuminating approach for understanding the nature of theoretical knowledge in evolutionary biology. The third illustration demonstrates how framing reductionism in terms of the reductive retooling of practice offers an informative perspective for understanding why putting DNA at the center of biological research has been incredibly productive throughout much of biology. Each illustration begins by describing how traditional theory-focused philosophical approaches are laden with fundamentalist assumptions and then proceeds to show that shifting attention to practice undermines these assumptions and motivates a philosophy of scientific pluralism

Precision medicine in cats:novel niemann-pick type C1 diagnosed by whole-genome sequencing
State-of-the-art health care includes genome sequencing of the patient to identify genetic variants that contribute to either the cause of their malady or variants that can be targeted to improve treatment. The goal was to introduce state-of-the-art health care to cats using genomics and a precision medicine approach. To test the feasibility of a precision medicine approach in domestic cats, a single cat that presented to the University of Missouri, Veterinary Health Center with an undiagnosed neurologic disease was whole-genome sequenced. The DNA variants from the cat were compared to the DNA variant database produced by the 99 Lives Cat Genome Sequencing Consortium. Approximately 25Ă genomic coverage was produced for the cat. A predicted p.H441P missense mutation was identified in NPC1, the gene causing Niemann-Pick type C1 on cat chromosome D3.47456793 caused by an adenine-to-cytosine transversion, c.1322A>C. The cat was homozygous for the variant. The variant was not identified in any other 73 domestic and 9 wild felids in the sequence database or 190 additionally genotyped cats of various breeds. The successful effort suggested precision medicine is feasible for cats and other undiagnosed cats may benefit from a genomic analysis approach. The 99 Lives DNA variant database was sufficient but would benefit from additional cat sequences. Other cats with the mutation may be identified and could be introduced as a new biomedical model for NPC1. A genetic test could eliminate the disease variant from the population
