Designing an effective evaluation model for the South African Department of Agriculture
Governments are under increasing pressure to deliver results. Therefore, it is important to evaluate the effectiveness, efficiency and relevance of the public service in implementing policies and programmes for social betterment. Without such evaluations, it is difficult to ensure that evidence is integrated into policy and used in practice due to lack of generalizability and learning. This paper focuses on (1) the knowledge that is relevant to understand evaluation influence, (2) the possible conceptual frameworks that enable understanding of the evaluation implementation process, (3) possible models of the process of organizational evaluation, and (4) the main ways of intervening to increase influence. Meta-evaluations in government and government institutions: A case study example from the Australian Centre for International Agricultural Research
In this paper we draw on impact assessment work of the Australian Centre for International Agricultural Research (ACIAR) to present an example of meta-evaluation – an evaluation of evaluations – in an agricultural research, development and extension setting. We explore quality issues relating to evaluation studies in the context of government institutions. Program evaluation standards (PES) are divided into categories of utility, feasibility, propriety and accuracy to provide a framework for the meta-evaluation. The PES are presented as a universal measure of evaluation study quality. The intent of using them here is to judge the adequacy of PES as a universal quality measure or meta-evaluation base and to extract useful insights from ACIAR program evaluation activities when developing a meta-evaluation model for the Limpopo Department of Agriculture (LDA). Our meta-evaluation is undertaken of 63 impact assessment reports. First, the literature guiding the conduct of a meta-evaluation is reviewed. Second, an assessment (the meta-evaluation) of the evaluation studies is carried out for 19 sampled reports from a population of relevant reports fitting the dimension of the analysis, and results are presented and discussed. Also, lessons learned are presented, using the framework provided by the meta-evaluation criteria. Third, taking into account the lessons learned, implications are drawn for a proposed systematic meta-evaluation of the LDA. Finally, we conclude that all the PES cannot be equally emphasized in a meta-evaluation model. At ACIAR, 70% of the standards were at least partially addressed. Therefore, we succeeded in using the PES in judging the ACIAR evaluation quality. Regulation of desmocollin transcription in mouse preimplantation embryos
The molecular mechanisms regulating the biogenesis of the first desmosomes to form during mouse embryogenesis have been studied. A sensitive modification of a reverse transcriptase-cDNA amplification procedure has been used to detect transcripts of the desmosomal adhesive cadherin, desmocollin. Sequencing of cDNA amplification products confirmed that two splice variants, a and b, of the DSC2 gene are transcribed coordinately. Transcripts were identified in unfertilized eggs and cumulus cells and in cleavage stages up to the early 8-cell stage, were never detected in compact 8-cell embryos, but were evident again either from the 16-cell morula or very early blastocyst (approx 32-cells) stages onwards. These two phases of transcript detection indicate DSC2 is encoded by maternal and embryonic genomes. Previously, we have shown that desmocollin protein synthesis is undetectable in eggs and cleavage stages but initiates at the early blastocyst stage when desmocollin localises at, and appears to regulate assembly of, nascent desmosomes that form in the trophectoderm but not in the inner cell mass (Fleming, T. P., Garrod, D. R. and Elsmore, A. J. (1991), Development 112, 527–539). Maternal DSC2 mRNA is therefore not translated and presumably is inherited by blastomeres before complete degradation. Our results suggest, however, that initiation of embryonic DSC2 transcription regulates desmocollin protein expression and thereby desmosome formation. Moreover, data from blastocyst single cell analyses suggest that embryonic DSC2 transcription is specific to the trophectoderm lineage. Inhibition of E-cadherin-mediated cell-cell adhesion did not influence the timing of DSC2 embryonic transcription and protein expression. However, isolation and culture of inner cell masses induced an increase in the amount of DSC2 mRNA and protein detected. Taken together, these results suggest that the presence of a contact-free cell surface activates DSC2 transcription in the mouse early embryo. <br/
Towards an understanding of neuroscience for science educators
Advances in neuroscience have brought new insights to the development of cognitive functions. These data are of considerable interest to educators concerned with how students learn. This review documents some of the recent findings in neuroscience, which is richer in describing cognitive functions than affective aspects of learning. A brief overview is presented here of the techniques used to generate data from imaging and how these findings have the possibility to inform educators. There are implications for considering the impact of neuroscience at all levels of education – from the classroom teacher and practitioner to policy. This relatively new cross-disciplinary area of research implies a need for educators and scientists to engage with each other. What questions are emerging through such dialogues between educators and scientists are likely to shed light on, for example, reward, motivation, working memory, learning difficulties, bilingualism and child development. Prevalence of Frailty in European Emergency Departments (FEED): an international flash mob study
Introduction
Current emergency care systems are not optimized to respond to multiple and complex problems associated with frailty. Services may require reconfiguration to effectively deliver comprehensive frailty care, yet its prevalence and variation are poorly understood. This study primarily determined the prevalence of frailty among older people attending emergency care.
Methods
This cross-sectional study used a flash mob approach to collect observational European emergency care data over a 24-h period (04 July 2023). Sites were identified through the European Task Force for Geriatric Emergency Medicine collaboration and social media. Data were collected for all individuals aged 65 + who attended emergency care, and for all adults aged 18 + at a subset of sites. Variables included demographics, Clinical Frailty Scale (CFS), vital signs, and disposition. European and national frailty prevalence was determined with proportions with each CFS level and with dichotomized CFS 5 + (mild or more severe frailty).
Results
Sixty-two sites in fourteen European countries recruited five thousand seven hundred eighty-five individuals. 40% of 3479 older people had at least mild frailty, with countries ranging from 26 to 51%. They had median age 77 (IQR, 13) years and 53% were female. Across 22 sites observing all adult attenders, older people living with frailty comprised 14%.
Conclusion
40% of older people using European emergency care had CFS 5 + . Frailty prevalence varied widely among European care systems. These differences likely reflected entrance selection and provide windows of opportunity for system configuration and workforce planning
Open data from the third observing run of LIGO, Virgo, KAGRA, and GEO
The global network of gravitational-wave observatories now includes five detectors, namely LIGO Hanford, LIGO Livingston, Virgo, KAGRA, and GEO 600. These detectors collected data during their third observing run, O3, composed of three phases: O3a starting in 2019 April and lasting six months, O3b starting in 2019 November and lasting five months, and O3GK starting in 2020 April and lasting two weeks. In this paper we describe these data and various other science products that can be freely accessed through the Gravitational Wave Open Science Center at https://gwosc.org. The main data set, consisting of the gravitational-wave strain time series that contains the astrophysical signals, is released together with supporting data useful for their analysis and documentation, tutorials, as well as analysis software packages
Search for eccentric black hole coalescences during the third observing run of LIGO and Virgo
Despite the growing number of binary black hole coalescences confidently observed through gravitational waves so far, the astrophysical origin of these binaries remains uncertain. Orbital eccentricity is one of the clearest tracers of binary formation channels. Identifying binary eccentricity, however, remains challenging due to the limited availability of gravitational waveforms that include the effects of eccentricity. Here, we present observational results for a waveform-independent search sensitive to eccentric black hole coalescences, covering the third observing run (O3) of the LIGO and Virgo detectors. We identified no new high-significance candidates beyond those that have already been identified with searches focusing on quasi-circular binaries. We determine the sensitivity of our search to high-mass (total source-frame mass M > 70 M⊙) binaries covering eccentricities up to 0.3 at 15 Hz emitted gravitational-wave frequency, and use this to compare model predictions to search results. Assuming all detections are indeed quasi-circular, for our fiducial population model, we place a conservative upper limit for the merger rate density of high-mass binaries with eccentricities 0 < e ≤ 0.3 at 16.9 Gpc−3 yr−1 at the 90% confidence level
Contribution of JAM-1 to epithelial differentiation and tight-junction biogenesis in the mouse preimplantation embryo
We have investigated the contribution of the tight junction (TJ) transmembrane protein junction-adhesion-molecule 1 (JAM-1) to trophectoderm epithelial differentiation in the mouse embryo. JAM-1-encoding mRNA is expressed early from the embryonic genome and is detectable as protein from the eight-cell stage. Immunofluorescence confocal analysis of staged embryos and synchronized cell clusters revealed JAM-1 recruitment to cell contact sites occurred predominantly during the first hour after division to the eight-cell stage, earlier than any other TJ protein analysed to date in this model and before E-cadherin adhesion and cell polarization. During embryo compaction later in the fourth cell cycle, JAM-1 localized transiently yet precisely to the apical microvillous pole, where protein kinase C (PKC) and PKC are also found, indicating a role in cell surface reorganization and polarization. Subsequently, in morulae and blastocysts, JAM-1 is distributed ubiquitously at cell contact sites within the embryo but is concentrated within the trophectoderm apicolateral junctional complex, a pattern resembling that of E-cadherin and nectin-2. However, treatment of embryos with anti-JAM-1-neutralizing antibodies indicated that JAM-1 did not contribute to global embryo compaction and adhesion but rather regulated the timing of blastocoel cavity formation dependent upon establishment of the trophectoderm TJ paracellular seal
Influence of maternal diet on the developmental potential of the preimplantation embryo.
The cleavage-stage embryo is sensitive to environmental conditions that can change developmental potential, leading to altered fetal and postnatal phenotype. We have developed mouse and rat models in vivo to investigate the effect of a mild form of maternal protein undernutrition exclusively during egg cleavage on long-term developmental potential. Maternal low-protein diet (LPD) during embryonic development leads to significant alteration in fetal and postnatal growth, a sustained increase in systolic blood pressure during adult life, and a changed pattern of anxiety-related behavior compared with controls fed a normal-protein diet during the embryonic period. This long-term patterning of phenotype includes gender-specific components. The sensitivity of embryos in vivo is paralleled by a similar adverse programming of phenotype in response to in vitro culture conditions, demonstrated in several species. Given the health care implications of the data arising from these models, elucidation of underlying mechanisms contributing to environment-induced changes in embryo potential is essential. Studies to date indicate that complex interactions are mediated through altered cell signaling, homeostatic regulation, proliferation, and epigenetics. In vivo programming via maternal LPD may initiate through abnormal cross-talk between maternal and embryonic cells involving amino acid and growth factor signaling, which may lead to altered setting of metabolism and biosynthesis within the embryo. Our data further indicate that in response to poor maternal periconceptional nutrition, the embryo can induce adaptations in nutrient supply during later development via the visceral yolk sac to protect fetal growth. However, such adaptations can be inappropriate and contribute to adverse fetal and postnatal programming when maternal nutrient levels exceed those predicted. Epigenetic changes involving DNA methylation in control regions of growth- and metabolism-regulating genes, including imprinted genes, have been identified in response to embryo in vitro culture conditions and sustained maternal LPD during gestation, representing additional mechanisms contributing to changes in developmental potentia
