    MicroRNAs from saliva of anopheline mosquitoes mimic human endogenous miRNAs and may contribute to vector-host-pathogen interactions

    During blood feeding haematophagous arthropods inject into their hosts a cocktail of salivary proteins whose main role is to counteract host haemostasis, inflammation and immunity. However, animal body fluids are known to also carry miRNAs. To get insights into saliva and salivary gland miRNA repertoires of the African malaria vector Anopheles coluzzii we used small RNA-Seq and identified 214 miRNAs, including tissue-enriched, sex-biased and putative novel anopheline miRNAs. Noteworthy, miRNAs were asymmetrically distributed between saliva and salivary glands, suggesting that selected miRNAs may be preferentially directed toward mosquito saliva. The evolutionary conservation of a subset of saliva miRNAs in Anopheles and Aedes mosquitoes, and in the tick Ixodes ricinus, supports the idea of a non-random occurrence pointing to their possible physiological role in blood feeding by arthropods. Strikingly, eleven of the most abundant An. coluzzi saliva miRNAs mimicked human miRNAs. Prediction analysis and search for experimentally validated targets indicated that miRNAs from An. coluzzii saliva may act on host mRNAs involved in immune and inflammatory responses. Overall, this study raises the intriguing hypothesis that miRNAs injected into vertebrates with vector saliva may contribute to host manipulation with possible implication for vector-host interaction and pathogen transmission

    Inventory of Vietri-Maiori landslides induced by the storm of October 1954 (southern Italy)

    On 25 October 1954, a storm hit the area surrounding the villages of Vietri sul Mare and Maiori of the Amalfi Coast (southern Italy) causing more than 300 deaths and severe damage to infrastructures and agriculture. This event has been among the most catastrophic historically documented in Campania Region. On this basis, and considering the lack of an existing complete characterization of the event in terms of triggered slope processes, we used multiple sets of stereoscopic aerial photos and a LiDAR-derived high-resolution topography to produce an event landslide inventory map. Our map provides an overview of the landslide distribution and extent in the area that mainly suffered the effect of the storm and is the basis for a morphometric characterization of landslide source areas that we present in this paper as simplified statistical analysis. In addition, we compared the rainfall distribution with the spatial density of source areas

    VR for Cultural Heritage. A VR-WEB-BIM for the future maintenance of Milan’s Cathedral.

    The work presented here is the final step of a multidisciplinary research project conducted on the Milan Cathedral for eight years (2008–2015). Three main topics, consequentially related, will be here addressed: (i) the survey of the structure, meant to update the old drawings; (ii) the construction of an accurate and detailed 3D model to be used to produce measurements at a 1:20–1:50 representation scale; (iii) the development of a Building Information System (BIM) to collect all the data relating to the restoration projects, as well as all information relating to past, current and future maintenance activities of the cathedral. The result of this research project is a complex and accurate digital 3D model of the main spire of the cathedral and of other parts of the building. This model can be visualized, navigated and used by the Veneranda Fabbrica technicians as an info-data catalogue, thanks to a common web browser connected with the remote BIM System Server and the modelling software where ad hoc I/O plugins are implemented. The last step of this long project was to take advantage of the nascent potential of immersive visualization techniques and to transpose the BIM system in a VR environment, thus obtaining two main results. The first was a high-appeal visualization system that allows a virtual visit of the Main Spire of the cathedral, the building’s highest part that has been closed to visitors since the beginning of the XX century. The second was the possibility to use this technology to virtually explore the cathedral from a technical point of view: by using an immersive visualization technology, operators can improve their understanding of the structure and obtain real-time information about the state of conservation, including current and past maintenance activities, in a sort of “augmented reality system in a virtual environment”

    Mitochondrial Fission Factor (MFF) inhibits mitochondrial metabolism and reduces breast cancer stem cell (CSC) activity

    Elevated mitochondrial biogenesis and metabolism represent key features of breast cancer stem cells (CSCs), whose propagation is conducive to disease onset and progression. Therefore, interfering with mitochondria biology and function may be regarded as a useful approach to eradicate CSCs. Here, we used the breast cancer cell line MCF7 as a model system to interrogate how mitochondrial fission contributes to the development of mitochondrial dysfunction toward the inhibition of metabolic flux and stemness. We generated an isogenic MCF7 cell line transduced with Mitochondrial Fission Factor (MCF7-MFF), which is primarily involved in mitochondrial fission. We evaluated the biochemical, molecular and functional properties of MCF7-MFF cells, as compared to control MCF7 cells transduced with the empty vector (MCF7-Control). We observed that MFF over-expression reduces both mitochondrial mass and activity, as evaluated using the mitochondrial probes MitroTracker Red and MitoTracker Orange, respectively. The analysis of metabolic flux using the Seahorse XFe96 revealed the inhibition of OXPHOS and glycolysis in MCF7-MFF cells, suggesting that increased mitochondrial fission may impair the biochemical properties of these organelles. Notably, CSCs activity, assessed by 3D-tumorsphere assays, was reduced in MCF7-MFF cells. A similar trend was observed for the activity of ALDH, a well-established marker of stemness. We conclude that enhanced mitochondrial fission may compromise CSCs propagation, through the impairment of mitochondrial function, possibly leading to a quiescent cell phenotype. Unbiased proteomic analysis revealed that proteins involved in mitochondrial dysfunction, oxidative stress-response, fatty acid metabolism and hypoxia signaling are among the most highly up-regulated in MCF7-MFF cells. Of note, integrated analysis of top regulatory networks obtained from unbiased proteomics in MCF7-MFF cells predicts that this cell phenotype activates signaling systems and effectors involved in the inhibition of cell survival and adhesion, together with the activation of specific breast cancer cell death programs. Overall, our study shows that unbalanced and abnormal activation of mitochondrial fission may drive the impairment of mitochondrial metabolic function, leading to inhibition of CSC propagation, and the activation of quiescence programs. Exploiting the potential of mitochondria to control pivotal events in tumor biology may, therefore, represent a useful tool to prevent disease progression

    Learning mutational graphs of individual tumour evolution from single-cell and multi-region sequencing data

    Background. A large number of algorithms is being developed to reconstruct evolutionary models of individual tumours from genome sequencing data. Most methods can analyze multiple samples collected either through bulk multi-region sequencing experiments or the sequencing of individual cancer cells. However, rarely the same method can support both data types. Results. We introduce TRaIT, a computational framework to infer mutational graphs that model the accumulation of multiple types of somatic alterations driving tumour evolution. Compared to other tools, TRaIT supports multi-region and single-cell sequencing data within the same statistical framework, and delivers expressive models that capture many complex evolutionary phenomena. TRaIT improves accuracy, robustness to data-specific errors and computational complexity compared to competing methods. Conclusions. We show that the application of TRaIT to single-cell and multi-region cancer datasets can produce accurate and reliable models of single-tumour evolution, quantify the extent of intra-tumour heterogeneity and generate new testable experimental hypotheses
