    Bioactive Materials Subjected to Erosion/Abrasion and Their Influence on Dental Tissues

    Objective: The objective of this study was to evaluate the effect of erosion or erosion-abrasion on bioactive materials and adjacent enamel/dentin areas. Methods and Materials: Enamel and dentin blocks (4×4×2 mm) were embedded side by side in acrylic resin, and a standardized cavity (1.2×4×1.5 mm) was prepared between them. Preparations were restored with the following materials: composite resin (Filtek Z350, control); experimental composite containing di-calcium phosphate dihydrate particles (DCPD); Giomer (Beautifil II), high viscosity glass ionomer cement (GIC, Fuji IX); and a resin-modified GIC (Fuji II LC). The specimens were submitted to two cycling models (n=10): erosion or erosion-abrasion. The challenges consisted of five-minute immersion in 0.3% citric acid solution, followed by 60-minute exposure to artificial saliva. Toothbrushing was carried out twice daily, 30 minutes after the first and last exposures to acid. Dental and material surface loss (SL, in μm) were determined by optical profilometry. Data were analyzed with Kruskal-Wallis and Dunn tests (α=0.05). Results: Under erosion, for enamel, only the GIC groups presented lower SL values than Z350 (p0.05). For material, the GICs had significantly higher SL values than those of Z350 (p<0.001 for Fuji IX and p=0.002 for Fuji II LC). Under erosion-abrasion, the enamel SL value was significantly lower around Fuji II LC compared with the other materials (p<0.05). No significant differences were observed among groups for dentin SL (p=0.063). The GICs and Giomer showed higher SL values than Z350 (p<0.001 for the GICs and p=0.041 for Giomer). Conclusion: Both GIC-based materials were susceptible to erosive wear; however, they promoted the lowest erosive loss of adjacent enamel. Against erosion-abrasion, only Fuji II LC was able to reduce enamel loss. For dentin, none of the materials exhibited a significant protective effect

    River damming affects seedling communities of a floodplain forest in the Central Amazon

    The flood pulse of black water rivers in the Amazon basin determines the composition of species along the flood gradient in igapó forests. The Balbina dam, built on the Uatumã River, has altered the flood pulse and caused changes in the floristic composition of adult trees throughout the downstream area. There is a lack of studies on how communities of seedlings in igapó forests respond to changes in the flood pulse. This study investigates the response of seedling communities in the igapó forest downstream the Balbina dam and compares it with two pristine areas. The areas were sampled with transects of 1x25 m within 36 plots (25x25 m) along the flood gradient. Richness and dominance were calculated by simple regression and ordination analyses. The pristine areas had the same pattern of richness, dominance and genera distribution along the flood gradient. However, the affected Uatumã area formed different groups of genera by NMDS analysis, which divided them along the flood gradient with significantly increased dominance of three genera. The insertion of the Balbina dam resulted a loss of lateral and longitudinal connectivity for the Uatumã River, and the alteration to seedling communities may alter the future landscape of downstream igapó forests

    Characterization of monoolein-based lipoplexes using fluorescence spectroscopy

    Lipoplexes are commonly used as delivery systems in vitro and in vivo, the role of a neutral lipid as helper being of extreme importance in these systems. Cationic liposomes composed of dioctadecyldimethylammonium bromide (DODAB) with monoolein (MO) as a helper, at different molar ratios (1:2; 1:1 and 1:0.5) were prepared, and subsequently titrated to DNA. The structural and physicochemical properties of the lipid/DNA complexes were assessed by Ethidium Bromide (EtBr) exclusion, 90º Static Light Scattering (90º SLS) assays and Fluorescence Resonance Energy Transfer (FRET). In EtBr exclusion assays, the steady-state fluorescence spectra of EtBr were decomposed into the sum of two lognormal emissions, emanating from two different environments – H2O and DNA, and the effect of charge ratio (+/-) was observed. 90º SLS assays gave an important contribution, detecting size variations in systems with different MO fractions on the lipoplexes. In FRET assays, 2-(3-(diphenylhexatrienyl)propanoyl)-1-hexadecanoyl-sn-glycero-3-phosphocholine (DPH-HPC) was used as donor and EtBr as acceptor. The DNA component previously calculated by EtBr exclusion, was used to determine the energy transfer efficiency, as an indirect measurement of the lipoplexes structural and physicochemical properties. Our results demonstrate that the inclusion of monoolein in the cationic liposomes formulation significantly modifies the rate of DNA complexation, being DODAB:MO (1:1) the system with higher DNA condensation efficiency.Fundação para a Ciência e a Tecnologia (FCT

    Cooperative localisation using image sensors in a dynamic traffic scenario

    Localisation is one of the key elements in navigation. Especially due to the development in automated driving, precise and reliable localisation becomes essential. In this paper, we report on different cooperation approaches in visual localisation with two vehicles driving in a convoy formation. Each vehicle is equipped with a multi-sensor platform consisting of front-facing stereo cameras and a global navigation satellite system (GNSS) receiver. In the first approach, the GNSS signals are used as excentric observations for the projection centres of the cameras in a bundle adjustment, whereas the second approach uses markers on the front vehicle as dynamic ground control points (GCPs). As the platforms are moving and data acquisition is not synchronised, we use time dependent platform poses. These time dependent poses are represented by trajectories consisting of multiple 6 Degree of Freedom (DoF) anchor points between which linear interpolation takes place. In order to investigate the developed approach experimentally, in particular the potential of dynamic GCPs, we captured data using two platforms driving on a public road at normal speed. As a baseline, we determine the localisation parameters of one platform using only data of that platform. We then compute a solution based on image and GNSS data from both platforms. In a third scenario, the front platform is used as a dynamic GCP which can be related to the trailing platform by markers observed in the images acquired by the latter. We show that both cooperative approaches lead to significant improvements in the precision of the poses of the anchor points after bundle adjustment compared to the baseline. The improvement achieved due to the inclusion of dynamic GCPs is somewhat smaller than the one due to relating the platforms by tie points. Finally, we show that for an individual vehicle, the use of dynamic GCPs can compensate for the lack of GNSS data

    Multiethnic genome-wide meta-analysis of ectopic fat depots identifies loci associated with adipocyte development and differentiation

    Variation in body fat distribution contributes to the metabolic sequelae of obesity. The genetic determinants of body fat distribution are poorly understood. The goal of this study was to gain new insights into the underlying genetics of body fat distribution by conducting sample-size weighted fixed-effects genome-wide association meta-analyses in up to 9,594 women and 8,738 men for six ectopic fat traits in European, African, Hispanic, and Chinese ancestry populations, with and without sex stratification. In total, 7 new loci were identified in association with ectopic fat traits (ATXN1, UBE2E2, EBF1, RREB1, GSDMB, GRAMD3 and ENSA; PATXN1 and UBE2E2 in primary mouse adipose progenitor cells impaired adipocyte differentiation, suggesting a physiological role for ATXN1 and UBE2E2 in adipogenesis. Future studies are necessary to further explore the mechanisms by which these genes impact adipocyte biology and how their perturbations contribute to systemic metabolic disease

    Evaluating different water-land-boundary approximations to improve sar-derived digital elevation models

    The coastline of the German Wadden Sea is constantly subjected to the tides and the tidal-induced environmental changes like erosion and accumulation of sediments need to be monitored constantly. This task requires digital elevation models (DEMs), which are derived from remote sensing data. To model those DEMs, a separation of data collected over landmasses and water bodies is required. In the GeoWAM project the potential of airborne SAR-data (F-SAR) is investigated for monitoring purposes in the Wadden Sea. As part of the project, this paper focuses on the suitability of F-SAR data regarding the derivation of water-land-boundaries (WLBs). Therefore, water-land-boundaries based on independent data sets are compared and evaluated. Analyzed data sets include data collected via F-SAR, airborne laserscanning (ALS), on site GNSS measured WLB points and sea-level data from two acoustic gauges. The algorithms were tested on a study site on Spiekeroog island. Our results show, that the accuracies of the derived WLBs mostly depend on the on-site topography and sediments. The spatial deviation between the reference data and the approximated WLBs is mostly less than 2 m horizontally and 0.15 m vertically. Identified challenges to overcome are mostly related to processing of F-SAR data in areas with highly water saturated sediments. Our results suggest, that F-SAR data in tidal flats is not necessarily dependent on further supplementing surveys, as one of the main advantages of the F-SAR data is the potential to derive DEMs and WLBs from the same data set

    Dioctadecyldimethylammonium:monoolein nanocarriers for efficient in vitro gene silencing

    This study describes a novel liposomal formulation for siRNA delivery, based on the mixture of the neutral lipid monoolein (MO) and cationic lipids of the dioctadecyldimethylammonium (DODA) family. The cationic lipids dioctadecyldimethylammonium bromide (DODAB) and chloride (DODAC) were compared in order to identify which one will most efficiently induce gene silencing. MO has a fluidizing effect on DODAC and DODAB liposomes, although it was more homogeneously distributed in DODAC bilayers. All MO-based liposomal formulations were able to efficiently encapsulate siRNA. Stable lipoplexes of small size (100-160 nm) with a positive surface charge (>+45 mV) were formed. A more uniform MO incorporation in DODAC:MO may explain an increase of the fusogenic potential of these liposomes. The siRNA-lipoplexes were readily internalized by human nonsmall cell lung carcinoma (H1299) cells, in an energy dependent process. DODAB:MO nanocarriers showed a higher internalization efficiency in comparison to DODAC:MO lipoplexes, and were also more efficient in promoting gene silencing. MO had a similar gene silencing ability as the commonly used helper lipid 1,2-dioleyl-3-phosphatidylethanolamine (DOPE), but with much lower cytotoxicity.     HDAC9 is implicated in atherosclerotic aortic calcification and affects vascular smooth muscle cell phenotype.

    Aortic calcification is an important independent predictor of future cardiovascular events. We performed a genome-wide association meta-analysis to determine SNPs associated with the extent of abdominal aortic calcification (n = 9,417) or descending thoracic aortic calcification (n = 8,422). Two genetic loci, HDAC9 and RAP1GAP, were associated with abdominal aortic calcification at a genome-wide level (P &lt; 5.0 × 10-8). No SNPs were associated with thoracic aortic calcification at the genome-wide threshold. Increased expression of HDAC9 in human aortic smooth muscle cells promoted calcification and reduced contractility, while inhibition of HDAC9 in human aortic smooth muscle cells inhibited calcification and enhanced cell contractility. In matrix Gla protein-deficient mice, a model of human vascular calcification, mice lacking HDAC9 had a 40% reduction in aortic calcification and improved survival. This translational genomic study identifies the first genetic risk locus associated with calcification of the abdominal aorta and describes a previously unknown role for HDAC9 in the development of vascular calcification

    Onsager-Machlup theory for nonequilibrium steady states and fluctuation theorems

    A generalization of the Onsager-Machlup theory from equilibrium to nonequilibrium steady states and its connection with recent fluctuation theorems are discussed for a dragged particle restricted by a harmonic potential in a heat reservoir. Using a functional integral approach, the probability functional for a path is expressed in terms of a Lagrangian function from which an entropy production rate and dissipation functions are introduced, and nonequilibrium thermodynamic relations like the energy conservation law and the second law of thermodynamics are derived. Using this Lagrangian function we establish two nonequilibrium detailed balance relations, which not only lead to a fluctuation theorem for work but also to one related to energy loss by friction. In addition, we carried out the functional integrals for heat explicitly, leading to the extended fluctuation theorem for heat. We also present a simple argument for this extended fluctuation theorem in the long time limit.Comment: 20 pages, 2 figure
