Leishmania metacaspase: an arginine-specific peptidase.
The purpose of this chapter is to give insights into metacaspase of Leishmania protozoan parasites as arginine-specific cysteine peptidase. The physiological role of metacaspase in Leishmania is still a matter of debate, whereas its peptidase enzymatic activity has been well characterized. Among the different possible expression systems, metacaspase-deficient yeast cells (Δyca1) have been instrumental in studying the activity of Leishmania major metacaspase (LmjMCA). Here, we describe techniques for purification of LmjMCA and its activity measurement, providing a platform for further identification of LmjMCA substrates
Entropy coding for training deep belief networks with imbalanced and unlabeled data
Session 1aSCb - Speech Communication: Speech Processing Potpourri (Poster Session): no. 1aSCb1Training deep belief networks (DBNs) is normally done with large data sets. In this work, the goal is to predict traces of the surface of the tongue in ultrasoundimages of the mouth during speech. Performance on this task can be dramatically enhanced by pre-training a DBN jointly on human-supplied traces and ultrasoundimages, then training a modified version of the network to predict traces from ultrasound only. However, hand-tracing the entire dataset of ultrasoundimages is extremely labor intensive. Moreover, the dataset is highly imbalanced since many images are extremely similar. This work presents a bootstrapping method which takes advantage of this imbalance, iteratively selecting a small subset of images to be hand-traced, then (re)training the DBN, making use of an entropy-based diversity measure for the initial selection. With this approach, a three-fold reduction in human time required to trace an entire dataset with human-level accuracy was achieved.published_or_final_versio
Optimizing single-photon-source heralding efficiency at 1550 nm using periodically poled lithium niobate
We explore the feasibility of using high conversion-efficiency
periodically-poled crystals to produce photon pairs for photon-counting
detector calibrations at 1550 nm. The goal is the development of an appropriate
parametric down-conversion (PDC) source at telecom wavelengths meeting the
requirements of high-efficiency pair production and collection in single
spectral and spatial modes (single-mode fibers). We propose a protocol to
optimize the photon collection, noise levels and the uncertainty evaluation.
This study ties together the results of our efforts to model the single-mode
heralding efficiency of a two-photon PDC source and to estimate the heralding
uncertainty of such a source.Comment: 14 pages, 2 tables and 3 figures, final version accepted by

Characterizing heralded single-photon sources with imperfect measurement devices
Any characterization of a single-photon source is not complete without
specifying its second-order degree of coherence, i.e., its function.
An accurate measurement of such coherence functions commonly requires
high-precision single-photon detectors, in whose absence, only time-averaged
measurements are possible. It is not clear, however, how the resulting
time-averaged quantities can be used to properly characterize the source. In
this paper, we investigate this issue for a heralded source of single photons
that relies on continuous-wave parametric down-conversion. By accounting for
major shortcomings of the source and the detectors--i.e., the multiple-photon
emissions of the source, the time resolution of photodetectors, and our chosen
width of coincidence window--our theory enables us to infer the true source
properties from imperfect measurements. Our theoretical results are
corroborated by an experimental demonstration using a PPKTP crystal pumped by a
blue laser, that results in a single-photon generation rate about 1.2 millions
per second per milliwatt of pump power. This work takes an important step
toward the standardization of such heralded single-photon sources.Comment: 18 pages, 9 figures; corrected Eq. (11) and the description follows

High coherence photon pair source for quantum communication
This paper reports a novel single mode source of narrow-band entangled photon
pairs at telecom wavelengths under continuous wave excitation, based on
parametric down conversion. For only 7 mW of pump power it has a created
spectral radiance of 0.08 pairs per coherence length and a bandwidth of 10 pm
(1.2 GHz). The effectively emitted spectral brightness reaches 3.9*10^5 pairs
/(s pm). Furthermore, when combined with low jitter single photon detectors,
such sources allow for the implementation of quantum communication protocols
without any active synchronization or path length stabilization. A HOM-Dip with
photons from two autonomous CW sources has been realized demonstrating the
setup's stability and performance.Comment: 12 pages, 4 figure
Security bound of two-bases quantum key-distribution protocols using qudits
We investigate the security bounds of quantum cryptographic protocols using
-level systems. In particular, we focus on schemes that use two mutually
unbiased bases, thus extending the BB84 quantum key distribution scheme to
higher dimensions. Under the assumption of general coherent attacks, we derive
an analytic expression for the ultimate upper security bound of such quantum
cryptography schemes. This bound is well below the predictions of optimal
cloning machines. The possibility of extraction of a secret key beyond
entanglement distillation is discussed. In the case of qutrits we argue that
any eavesdropping strategy is equivalent to a symmetric one. For higher
dimensions such an equivalence is generally no longer valid.Comment: 12 pages, 2 figures, to appear in Phys. Rev.
Experimental realization of a low-noise heralded single photon source
We present a heralded single-photon source with a much lower level of
unwanted background photons in the output channel by using the herald photon to
control a shutter in the heralded channel. The shutter is implemented using a
simple field programable gate array controlled optical switch.Comment: 4 pages, 5 figure
Dramatic Acceleration of the Hopf Cyclization on Gold(111): From Enediynes to Peri-Fused Diindenochrysene Graphene Nanoribbons.
Hopf et al. reported the high-temperature 6π-electrocyclization of cis-hexa-1,3-diene-5-yne to benzene in 1969. Subsequent studies using this cyclization have been limited by its very high reaction barrier. Here, we show that the reaction barrier for two model systems, (E)-1,3,4,6-tetraphenyl-3-hexene-1,5-diyne (1a) and (E)-3,4-bis(4-iodophenyl)-1,6-diphenyl-3-hexene-1,5-diyne (1b), is decreased by nearly half on a Au(111) surface. We have used scanning tunneling microscopy (STM) and noncontact atomic force microscopy (nc-AFM) to monitor the Hopf cyclization of enediynes 1a,b on Au(111). Enediyne 1a undergoes two sequential, quantitative Hopf cyclizations, first to naphthalene derivative 2, and finally to chrysene 3. Density functional theory (DFT) calculations reveal that a gold atom from the Au(111) surface is involved in all steps of this reaction and that it is crucial to lowering the reaction barrier. Our findings have important implications for the synthesis of novel graphene nanoribbons. Ullmann-like coupling of enediyne 1b at 20 °C on Au(111), followed by a series of Hopf cyclizations and aromatization reactions at higher temperatures, produces nanoribbons 12 and 13. These results show for the first time that graphene nanoribbons can be synthesized on a Au(111) surface using the Hopf cyclization mechanism
A bank of unscented Kalman filters for multimodal human perception with mobile service robots
A new generation of mobile service robots could be ready soon to operate in human environments if they can robustly estimate position and identity of surrounding people. Researchers in this field face a number of challenging problems, among which sensor uncertainties and real-time constraints.
In this paper, we propose a novel and efficient solution for simultaneous tracking and recognition of people within the observation range of a mobile robot. Multisensor techniques for legs and face detection are fused in a robust probabilistic framework to height, clothes and face recognition algorithms. The system is based on an efficient bank of Unscented Kalman Filters that keeps a multi-hypothesis estimate of the person being tracked, including the case where the latter is unknown to the robot.
Several experiments with real mobile robots are presented to validate the proposed approach. They show that our solutions can improve the robot's perception and recognition of humans, providing a useful contribution for the future application of service robotics
