    Composição proximal em filés, rendimento de carcaça e de cortes de jundiá (rhamdia quelen) alimentados com rações contendo diferentes óleos vegetais.

    Este trabalho teve por objetivos avaliar a influência de óleos vegetais no desenvolvimento corporal de jundiá (Rhamdia quelen, Heptapteridae) e sua relação com o rendimento de carcaça, partes comestíveis e composição química do filé. Testaram-se seis rações com 32% de proteína bruta, sendo avaliados óleos de arroz, canola ou soja com 5 e 10% de inclusão durante 90 dias. Foram utilizados 180 peixes (peso inicial=71±0,81g) distribuídos ao acaso em 18 caixas de 280L (10 peixes/caixa) em um sistema de recirculação de água, com temperatura controlada (25,9±0,9ºC). Não houve diferenças para os parâmetros produtivos entre os tratamentos testados ao final do experimento (P<0,05). Os peixes alimentados com maiores níveis de óleo nas dietas depositaram maior porcentagem de gordura no filé. Conclui-se que os óleos de canola, arroz e soja utilizados como alternativas em dietas na recria de jundiá proporcionam bom rendimento de carcaça e de partes comestíveis. E, as rações contendo óleo de canola possibilitam menor deposição de gordura no filé de juvenis de jundiá em nível de 5%

    O objetivo do trabalho foi avaliar o crescimento, a composição e o perfil lipídico corporal de jundiás criados em diferentes densidades de estocagem (DE: 4,2; 6,4; 8,6 e 10,8kg m-3) em sistema com recirculação de água. Durante 120 dias, 420 peixes (peso inicial=182,95±2,96g) foram alimentados uma vez ao dia, até a saciedade aparente, com dieta peletizada formulada. Observou-se que, quanto maior a DE utilizada, menor é o peso individual dos peixes, entretanto a biomassa por tanque aumentou (P<0,05). Melhor conversão alimentar (1,69) e maior quantidade de gordura intraperitoneal (3,92) foram observadas nos peixes submetidos à densidade 4,2kg m-3. A proteína corporal foi afetada pelas DEs. Ocorreu diminuição dos ácidos graxos saturados e aumento dos insaturados nos peixes de acordo com o aumento das DEs. Conclui-se que a densidade de estocagem influência no peso final e na produtividade por volume do jundiá

    Transcriptional activation of pericentromeric satellite repeats and disruption of centromeric clustering upon proteasome inhibition

    Heterochromatinisation of pericentromeres, which in mice consist of arrays of major satellite repeats, are important for centromere formation and maintenance of genome stability. The dysregulation of this process has been linked to genomic stress and various cancers. Here we show in mice that the proteasome binds to major satellite repeats and proteasome inhibition by MG132 results in their transcriptional de-repression; this de-repression is independent of cell-cycle perturbation. The transcriptional activation of major satellite repeats upon proteasome inhibition is accompanied by delocalisation of heterochromatin protein 1 alpha (HP1α) from chromocentres, without detectable change in the levels of histone H3K9me3, H3K4me3, H3K36me3 and H3 acetylation on the major satellite repeats. Moreover, inhibition of the proteasome was found to increase the number of chromocentres per cell, reflecting destabilisation of the chromocentre structures. Our findings suggest that the proteasome plays a role in maintaining heterochromatin integrity of pericentromeres

    This study was carried out to obtain more information about the physicochemical properties, composition, and antioxidant activity of pitanga fruits (Eugenia uniflora L.), particularly fruits from the State of Rio Grande do Sul, Brazil. Pitanga with different flesh colors (purple, red, and orange) from tree selections cultivated at Embrapa Clima Temperado (RS-Brazil) were analyzed. Only slight differences were observed in the quality parameters and in the proximate and fatty acid compositions among the fruits studied. The extracts from purple-fleshed pitanga had the highest total phenolic and anthocyanin contents along with the highest antioxidant capacity. The antioxidant capacity (DPPH and FRAP assays) of methanolic pitanga extracts was highly correlated with the total phenolic content, but in ethanolic extracts, the anthocyanin content was correlated only with the FRAP antioxidant capacity. Orange fleshed pitanga had higher β-cryptoxanthin and β-carotene levels than those of the red fruit, which had higher lycopene content. The results indicate that the purple-fleshed pitanga, cultivated in Rio Grande do Sul, is a rich source of phenolic compounds and has high antioxidant capacity.     Negli ultimi 20 anni il miglioramento dellassistenza neonatale ha determinato un progressivo aumento dei bambini che sopravvivono in presenza di gravi malformazioni o patologie congenite. Questi bambini richiedono una riabilitazione prolungata, talora multidisciplinare e complessa. Purtroppo, unorganizzazione adeguata alla riabilitazione della disfagia, dei disturbi della comunicazione e della respirazione non è sempre disponibile, non è sempre coordinata in equipe multidisciplinari che operino sia negli ospedali che sul territorio e non è facile mantenere tutte le figure professionali coinvolte al passo con le sempre più rapide innovazioni. Scopo del presente lavoro è presentare un aggiornamento su alcuni aspetti tuttora controversi della riabilitazione in età pediatrica

    Optimization and validation of an enzymatic method to quantify glucose in potato tubers

    The processing industry requires potato tubers with low levels of reduced sugars, glucose and fructose, which is responsible for the whiteness of the chip color. The objective of this research was to optimize and validate a simple, inexpensive, and precise enzymatic method to quantify glucose in potato tubers. This method will be useful in breeding programs to select clones for processing purposes with low levels of glucose. The validation procedure followed the recommendations described in INMETRO document DOQ-CGCRE-008 in 2003. The method was linear between 1.25 to 40µg of glucose in the sample. The quantification limit was 0.319mg g-1 of potato fresh weight. The linear working interval of the method started at the quantification limit and ended at 1.32mg g-1 of potato fresh weight. The average recovery in the extraction was 99.0%. The proposed method was used to determine glucose level in samples of Macaca tubers, which ranged from 1.4 to 2.9mg g-1 potato fresh weight

    Cross-generational trans fat intake exacerbates UV radiation-induced damage in rat skin

    AbstractWe evaluated the influence of dietary fats on ultraviolet radiation (UVR)-induced oxidative damage in skin of rats. Animals from two consecutive generations born of dams supplemented with fats during pregnancy and breastfeeding were maintained in the same supplementation: soybean-oil (SO, rich in n-6 FA, control group), fish-oil (FO, rich in n-3 FA) or hydrogenated-vegetable-fat (HVF, rich in TFA). At 90days of age, half the animals from the 2nd generation were exposed to UVR (0.25J/cm2) 3×/week for 12weeks. The FO group presented higher incorporation of n-3 FA in dorsal skin, while the HVF group incorporated TFA. Biochemical changes per se were observed in skin of the HVF group: greater generation of reactive oxygen species (ROS), lower mitochondrial integrity and increased Na+K+-ATPase activity. UVR exposure increased skin wrinkles scores and ROS generation and decreased mitochondrial integrity and reduced-glutathione levels in the HVF group. In FO, UVR exposure was associated with smaller skin thickness and reduced levels of protein–carbonyl, together with increased catalase activity and preserved Na+K+-ATPase function. In conclusion, while FO may be protective, trans fat may be harmful to skin health by making it more vulnerable to UVR injury and thus more prone to develop photoaging and skin cancer

    O objetivo deste trabalho foi avaliar a influência de óleos vegetais no desenvolvimento corporal e composição do filé de jundiás. Foram testadas seis dietas (32%PB), sendo avaliados óleos de arroz, canola ou soja com 5 ou 10% de inclusão, durante 90 dias. Os peixes (180, peso inicial=71,0±0,8g) foram distribuídos ao acaso em 18 caixas (280L) de um sistema de recirculação de água, com temperatura controlada. Para avaliação de desempenho, estimaram-se variáveis como peso, comprimentos totais, padrão, altura caudal, sobrevivência, taxa de crescimento específico, fator de condição, consumo diário, conversão alimentar aparente e rendimento de partes comestíveis. Além disso, foram avaliadas a composição centesimal e taxas de deposição de proteína e gordura nos filés. Não houve diferenças entre os óleos e os níveis sobre o crescimento dos peixes; entretanto, os jundiás alimentados com 10% de óleo depositaram mais gordura no filé. Conclui-se que os óleos de canola, arroz e soja proporcionam bom crescimento e rendimento de partes comestíveis. As dietas contendo óleo de canola, com 5% de inclusão, possibilitam menor deposição de gordura no filé
