    Trophic ecology of piscivorous Arctic charr (Salvelinus alpinus (L.)) in subarctic lakes with contrasting food-web structures

    The trophic ecology of piscivorous Arctic charr (Salvelinus alpinus (L.); charr) in the food webs of large subarctic lakes is not well understood. We assessed charr diets, parasites, growth, maturity, and stable isotope ratios in Fennoscandian subarctic lakes dominated by monomorphic or polymorphic whitefish (Coregonus lavaretus (L.)) populations. Charr density was low in all lakes, except in profundal habitats. Charr shifted to piscivory at small size (16-25 cm total length) and consumed a range of prey-fish sizes (2-25 cm). Cannibalism was observed in a few individuals from one monomorphic whitefish lake. Charr matured at 37-51 cm (5-8 years old), grew to 52-74 cm maximum observed length and 47-83 cm asymptotic length. Charr increased total area of convex hull and core stable isotopic diversity area of the fish community by 51-98% and 44-51% in monomorphic whitefish lakes, but only 8-11% and 7-10% in polymorphic whitefish lakes. The difference was due to increasing food-chain length in monomorphic whitefish lakes, whereas reliance on littoral carbon did not change. Charr were the top piscivores in monomorphic whitefish lakes, but played a less important role in polymorphic whitefish lakes, which contained a more diverse predator fauna.Peer reviewe

    Transport Modelling of a pyro-convection event in Alaska

    International audienceSummer 2004 saw severe forest fires in Alaska and the Yukon Territory that were mostly triggered by lightning strikes. The area burned (>2.7×106 ha) in the year 2004 was the highest on record to date in Alaska. Pollutant emissions from the fires lead to violation of federal standards for air quality in Fairbanks. This paper studies deep convection events that occurred in the burning regions at the end of June 2004. The convection was likely enhanced by the strong forest fire activity (so-called pyro-convection) and penetrated into the lower stratosphere, up to about 3 km above the tropopause. Emissions from the fires did not only perturb the UT/LS locally, but also regionally. POAM data at the approximate location of Edmonton (53.5° N, 113.5° W) show that the UT/LS aerosol extinction was enhanced by a factor of 4 relative to unperturbed conditions. Simulations with the particle dispersion model FLEXPART with the deep convective transport scheme turned on showed transport of forest fire emissions into the stratosphere, in qualitatively good agreement with the enhancements seen in the POAM data. A corresponding simulation with the deep convection scheme turned off did not result in such deep vertical transport. Lidar measurements at Wisconsin on 30 June also show the presence of substantial aerosol loading in the UT/LS, up to about 13 km. In fact, the FLEXPART results suggest that this aerosol plume originated from the Yukon Territory on 25 June

    Liver secretin receptor predicts portoenterostomy outcomes and liver injury in biliary atresia

    Biliary atresia (BA) is a chronic neonatal cholangiopathy characterized by fibroinflammatory bile duct damage. Reliable biomarkers for predicting native liver survival (NLS) following portoenterostomy (PE) surgery are lacking. Herein we explore the utility of 22 preidentified profibrotic molecules closely connected to ductular reaction (DR) and prevailing after successful PE (SPE), in predicting PE outcomes and liver injury. We used qPCR and immunohistochemistry in a BA cohort including liver samples obtained at PE (n = 53) and during postoperative follow-up after SPE (n = 25). Of the 13 genes over-expressed in relation to cholestatic age-matched controls at PE, only secretin receptor (SCTR) expression predicted cumulative 5-year NLS and clearance of jaundice. Patients in the highest SCTR expression tertile showed 34–55% lower NLS than other groups at 1–5 years after PE (P = 0.006–0.04 for each year). SCTR expression was also significantly lower [42 (24–63) vs 75 (39–107) fold, P = 0.015] among those who normalized their serum bilirubin after PE. Liver SCTR expression localized in cholangiocytes and correlated positively with liver fibrosis, DR, and transcriptional markers of fibrosis (ACTA2) and cholangiocytes (KRT7, KRT19) both at PE and after SPE.     From clear lakes to murky waters - tracing the functional response of high-latitude lake communities to concurrent 'greening' and 'browning'

    Climate change and the intensification of land use practices are causing widespread eutrophication of subarctic lakes. The implications of this rapid change for lake ecosystem function remain poorly understood. To assess how freshwater communities respond to such profound changes in their habitat and resource availability, we conducted a space-for-time analysis of food-web structure in 30 lakes situated across a temperature-productivity gradient equivalent to the predicted future climate of subarctic Europe (temperature +3 degrees C, precipitation +30% and nutrient +45 mu g L-1 total phosphorus). Along this gradient, we observed an increase in the assimilation of pelagic-derived carbon from 25 to 75% throughout primary, secondary and tertiary consumers. This shift was overwhelmingly driven by the consumption of pelagic detritus by benthic primary consumers and was not accompanied by increased pelagic foraging by higher trophic level consumers. Our data also revealed a convergence of the carbon isotope ratios of pelagic and benthic food web endmembers in the warmest, most productive lakes indicating that the incorporation of terrestrial derived carbon into aquatic food webs increases as land use intensifies. These results, reflecting changes along a gradient characteristic of the predicted future environment throughout the subarctic, indicate that climate and land use driven eutrophication and browning are radically altering the function and fuelling of aquatic food webs in this biome.Peer reviewe

    We have measured the temperature dependence of the time of flight of helium excimer molecules He2* in superfluid 4He and find that the molecules behave ballistically below 100mK and exhibit Brownian motion above 200 mK.     Increased sedentariness has been linked to the growing prevalence of obesity in children, but some longitudinal studies suggest that sedentariness may be a consequence rather than a cause of increased adiposity. We used Mendelian randomization to examine the causal relations between body mass index (BMI) and objectively assessed sedentary time and physical activity in 3-8 year-old children from one Finnish and two Danish cohorts [NTOTAL=679]. A genetic risk score (GRS) comprised of 15 independent genetic variants associated with childhood BMI was used as the instrumental variable to test causal effects of BMI on sedentary time, total physical activity, and moderate-to-vigorous physical activity (MVPA). In fixed effects meta-analyses, the GRS was associated with 0.05 SD/allele increase in sedentary time (P=0.019), but there was no significant association with total physical activity (beta=0.011 SD/allele, P=0.58) or MVPA (beta=0.001 SD/allele, P=0.96), adjusting for age, sex, monitor wear-time and first three genome-wide principal components. In two-stage least squares regression analyses, each genetically instrumented one unit increase in BMI z-score increased sedentary time by 0.47 SD (P=0.072). Childhood BMI may have a causal influence on sedentary time but not on total physical activity or MVPA in young children. Our results provide important insights into the regulation of movement behaviour in childhood

    Ultrafast relaxation of photoexcited superfluid He nanodroplets

    The relaxation of photoexcited nanosystems is a fundamental process of light-matter interaction. Depending on the couplings of the internal degrees of freedom, relaxation can be ultrafast, converting electronic energy in a few fs, or slow, if the energy is trapped in a metastable state that decouples from its environment. Here, we study helium nanodroplets excited resonantly by femtosecond extreme-ultraviolet (XUV) pulses from a seeded free- electron laser. Despite their superfluid nature, we find that helium nanodroplets in the lowest electronically excited states undergo ultrafast relaxation. By comparing experimental pho- toelectron spectra with time-dependent density functional theory simulations, we unravel the full relaxation pathway: Following an ultrafast interband transition, a void nanometer-sized bubble forms around the localized excitation (He ) within 1 ps. Subsequently, the bubble collapses and releases metastable He at the droplet surface. This study highlights the high level of detail achievable in probing the photodynamics of nanosystems using tunable XUV pulses

    A rare regulatory variant in the MEF2D gene affects gene regulation and splicing and is associated with a SLE sub-phenotype in Swedish cohorts

Systemic lupus erythematosus (SLE) is an autoimmune disorder with heterogeneous clinical presentation and complex etiology involving the interplay between genetic, epigenetic, environmental and hormonal factors. Many common SNPs identified by genome wide-association studies (GWAS) explain only a small part of the disease heritability suggesting the contribution from rare genetic variants, undetectable in GWAS, and complex epistatic interactions. Using targeted re-sequencing of coding and conserved regulatory regions within and around 215 candidate genes selected on the basis of their known role in autoimmunity and genes associated with canine immune-mediated diseases, we identified a rare regulatory variant rs200395694:G > T located in intron 4 of the MEF2D gene encoding the myocyte-specific enhancer factor 2D transcription factor and associated with SLE in Swedish cohorts (504 SLE patients and 839 healthy controls, p = 0.014, CI = 1.1–10). Fisher’s exact test revealed an association between the genetic variant and a triad of disease manifestations including Raynaud, anti-U1-ribonucleoprotein (anti-RNP), and anti-Smith (anti-Sm) antibodies (p = 0.00037) among the patients. The DNA-binding activity of the allele was further studied by EMSA, reporter assays, and minigenes. The region has properties of an active cell-specific enhancer, differentially affected by the alleles of rs200395694:G > T. In addition, the risk allele exerts an inhibitory effect on the splicing of the alternative tissue-specific isoform, and thus may modify the target gene set regulated by this isoform. These findings emphasize the potential of dissecting traits of complex diseases and correlating them with rare risk alleles with strong biological effects
