    Consuming transgenic goats' milk containing the antimicrobial protein lysozyme helps resolve diarrhea in young pigs.

    Childhood diarrhea is a significant problem in many developing countries and E. coli is a main causative agent of diarrhea in young children. Lysozyme is an antimicrobial protein highly expressed in human milk, but not ruminant milk, and is thought to help protect breastfeeding children against diarrheal diseases. We hypothesized that consumption of milk from transgenic goats which produce human lysozyme (hLZ-milk) in their milk would accelerate recovery from bacterial-induced diarrhea. Young pigs were used as a model for children and infected with enterotoxigenic E. coli. Once clinical signs of diarrhea developed, pigs were fed hLZ-milk or non-transgenic control goat milk three times a day for two days. Clinical observations and complete blood counts (CBC) were performed. Animals were euthanized and samples collected to assess differences in histology, cytokine expression and bacterial translocation into the mesenteric lymph node. Pigs consuming hLZ-milk recovered from clinical signs of infection faster than pigs consuming control milk, with significantly improved fecal consistency (p = 0.0190) and activity level (p = 0.0350). The CBC analysis showed circulating monocytes (p = 0.0413), neutrophils (p = 0.0219), and lymphocytes (p = 0.0222) returned faster to pre-infection proportions in hLZ-milk fed pigs, while control-fed pigs had significantly higher hematocrit (p = 0.027), indicating continuing dehydration. In the ileum, pigs fed hLZ-milk had significantly lower expression of pro-inflammatory cytokine IL-8 (p = 0.0271), longer intestinal villi (p<0.0001), deeper crypts (p = 0.0053), and a thinner lamina propria (p = 0.0004). These data demonstrate that consumption of hLZ-milk helped pigs recover from infection faster, making hLZ-milk an effective treatment of E. coli-induced diarrhea

    The Value and Feasibility of Farming Differently Than the Local Average

    The purpose of this research is to quantify the value of being different than the local average and feasibility of distinguishing particular parts of an operation from the local average. Kansas crop farms are broken down by their farm characteristics, production practices, and management performances. An ordinary least squares regression model is used to quantify the value of having different than average characteristics, practices, and management performances. The degree farms have distinguished particular parts of their operations from the average, and how consistently they distinguish their cost, yields, and prices from the average are also analyzed. Farms’ relative size, workers per acre, planting intensity, machine costs, yields, and prices are all significantly related to farm relative performance. Farms’ characteristics are generally more different from one another than their production practices and management performances, while over time farms’ relative cost management performances are more consistent than their relative yield and price management performances

    The examination of human chromosomes has been a part of the physician’s laboratory armamentarium since the correct diploid number of human chromosomes was established and a method was developed for the in vitro growth of peripheral blood leukocytes to yield metaphase chromosomes. The discovery that on ultraviolet microscopy (UV), metaphase chromosomes stained with fluorochrome dyes displayed a characteristic pattern of bright and dull bands unique for a given pair of homologous chromosomes, was a major technological breakthrough in human cytogenetics; for the first time, every chromosome in the karyotype could be unequivocally identified. Although the short storage life of fluorochrome-stained chromosomes and the costs of UV microscopy have limited the usability of fluorescence banding, the introduction of one discriminating procedure quickly led to the development of an array of similar banding techniques for conventional microscopy that yield comparable information. Some of these technical procedures depend on enzyme and/or heat denaturation of the chromosomes, resulting in the characteristic banding patterns seen by the trypsin-Giemsa method, the 5M urea method, and the acid-saline-Giemsa technique. A typical human karyotype prepared from metaphase chromosomes treated with trypsin, stained with Giemsa, and photographed with brightfield photomicrographic techniques is shown in Figure 1. Careful examination of this karyotype reveals that each chromosome in the homologous pair has an array of dark and light bands identical with those of its homolog and that each homologous pair, autosomes number 1 to number 22, has a characteristic, easily identifiable banding pattern

    Preserving Family: Themes from a Qualitative Study of Kin Caregivers

    This article presents themes from a qualitative study of 58 African American female kinship caregivers in San Francisco. Core concepts that emerged describe various paths along which children move into kin homes, and caregivers\u27 mixed emotional reactions to becoming surrogate parents. Women also discussed multiple family roles they assumed after taking in children. Responses highlight three primary reasons for becoming caregivers that center on providing for and protecting these children—particularly from the perceived threat of the public foster care system—and ultimately preserving the family unit. Paradoxically, caregivers\u27 reasons mirror the stated goals of the public foster care system, which they view as a threat to family stability. We discuss the problems of implementing practice and policy recommendations for permanency and family preservation and how to bridge the gap between the deeply held negative beliefs of African American caregivers towards the public system and begin to build trust

    Genetic Variation at Nuclear Loci Fails to Distinguish Two Morphologically Distinct Species of Aquilegia

    Aquilegia formosa and pubescens are two closely related species belonging to the columbine genus. Despite their morphological and ecological differences, previous studies have revealed a large degree of intercompatibility, as well as little sequence divergence between these two taxa [1], [2]. We compared the inter- and intraspecific patterns of variation for 9 nuclear loci, and found that the two species were practically indistinguishable at the level of DNA sequence polymorphism, indicating either very recent speciation or continued gene flow. As a comparison, we also analyzed variation at two loci across 30 other Aquilegia taxa; this revealed slightly more differentiation among taxa, which seemed best explained by geographic distance. By contrast, we found no evidence for isolation by distance on a more local geographic scale. We conclude that the extremely low levels of genetic differentiation between A. formosa and A.pubescens at neutral loci will facilitate future genome-wide scans for speciation genes
