Dairying and Employment in the Amuri: 1983 to 2002
The dairy industry is currently experiencing a staff shortage, as are many other industries. It has experienced rapid expansion, and the situation is made worse by the poor image of the industry. This expansion has often occurred in areas with little prior dairying. The Amuri region, North Canterbury, is one such area. The conversion of farms to dairying began in I983, following the commissioning of the 17000-hectare Waiau Plains Irrigation Scheme in 1980. There are now 49 herds in the area. Some of the initial dairy farming experiences were very bad, giving all dairying in the region a poor reputation. The Amuri region is 'geographically isolated basin', and situated approximately 90 minutes from Christchurch, with limited social opportunities for the farm staff This resulted in extreme difficulties attracting and retaining good staff in the area. In response to this situation, the dairy farmers in the area formed the Amuri Dairy Employers Group, in March 2000. This group established a constitution including: Mandatory member employer training; Agreeing to an independent annual audit of member employment practices; and Agreeing to a Code of Practice for employment standards. I have undertaken a two-year investigation of the effects of the Amuri Dairy Employers Group, on dairy farming employment and the wider social effects in the Amuri area. The multiplicity of value in the front-end of projects: The case of London transportation infrastructure
There is growing interest in the ways that value is understood in the context of projects and within project-based settings. Recent studies emphasise the multiplicity of project value in various project settings as perceived by different project actors. Drawing on previous work on project value and project front-end, this study expands on the idea of multiplicity of project value in the early project definition phase. To this end, the study draws from empirical data on infrastructure projects provision, including semi-structured interviews with a set of highly experienced and senior level informants with extensive knowledge and familiarity of infrastructure project planning and front-end decision making. The study is bounded with a focus on London, UK as an example of a complex, highly established global city with a great reliance on its infrastructure and a well-established projects ecology. Through inductive qualitative data analysis the study explores the role of infrastructure projects as solutions to policy problems, the multiple and complex nature of value in project definition and identifies three value levels, which are instrumental for project definition: local value, sector value and user value. The multi-level value framework in the project front-end extends and complements early decision making in planning and setting up of infrastructure projects
Does sentinel lymph node biopsy have a role in node-positive head and neck squamous carcinoma?
OBJECTIVES/HYPOTHESIS: The objective of the study was to determine whether sentinel lymph node biopsy (SLNB) can be used to reduce clinical overstaging of cervical nodes in head and neck squamous cell carcinoma (SCC) in a developing world setting. STUDY DESIGN: Sentinel and echelon lymph nodes were identified by means of a combination of lymphoscintigraphy, gamma probe and blue dye identification. They were analysed histologically and their pathological status was compared with the rest of the neck dissection specimen to determine diagnostic accuracy in patients with T1-4 N0-3 SCC of the oral cavity or oropharynx undergoing primary surgical resection and neck dissection. RESULTS: Thirty-three patients were included in the study, 13 in the node-negative (N0) and 20 in the node-positive (N+) group. In the clinically N0 group the sensitivity of SLNB was 100% and the negative predictive value (NPV) 100%. In the clinically N+ group the sensitivity was 71% and the NPV 60% for staging the nodal status of the neck. CONCLUSIONS; The accuracy of SLNB in the clinically N+ neck is too low for SLNB to be a means of avoiding comprehensive neck dissection. LEVEL OF EVIDENCE: 2B
Chronic otorrhoea: Spectrum of microorganisms and antibiotic sensitivity in a South African cohort
BACKGROUND: Chronic otorrhoea is difficult to treat, with treatment in South Africa (SA) being protocol driven and generally initiated at the primary healthcare level. There is a lack of local studies that focus on the bacteriology and antimicrobial sensitivities of chronic otorrhoea, which underpins the management advice offered. AIMS: To determine the microbiological profile and antimicrobial susceptibility of patients with chronic otorrhoea and the validity of the Department of Health's (DoH) current guideline. METHODS: We conducted a prospective study at Groote Schuur Hospital from 2005 to 2009. We included patients with chronic otorrhoea classified as either otitis media or otitis externa, according to our definitions. Pus swabs were taken, from which microorganisms were cultured and tested for antimicrobial susceptibility. RESULTS: Of 79 patients with otorrhoea, 50 had otitis media, 21 had otitis externa and the condition was not determined in 8 patients. The most common organism isolated with otitis media was Proteus mirabilis (18/50; 36%) and with otitis externa, Pseudomonas aeruginosa (7/21; 33%). Otorrhoea had a different microbial spectrum compared with international reports, with methicillin-resistant Staphylococcus aureus infection in a single patient. The organisms isolated were susceptible mainly to fluoroquinolones (96%) and aminoglycosides (81%). CONCLUSION: Amoxicillin is a poor choice of antibiotic due to its low sensitivity, which calls into question the current DoH guideline for otorrhoea. Antimicrobial treatment protocols should be based on local data and be revisited from time to time. This study suggests that, should first-line treatment fail, an antibiotic with Gram-negative cover, e.g. a topical fluoroquinolone, should be considered
Commensurate period Charge Density Modulations throughout the Pseudogap Regime
Theories based upon strong real space (r-space) electron electron
interactions have long predicted that unidirectional charge density modulations
(CDM) with four unit cell (4) periodicity should occur in the hole doped
cuprate Mott insulator (MI). Experimentally, however, increasing the hole
density p is reported to cause the conventionally defined wavevector of
the CDM to evolve continuously as if driven primarily by momentum space
(k-space) effects. Here we introduce phase resolved electronic structure
visualization for determination of the cuprate CDM wavevector. Remarkably, this
new technique reveals a virtually doping independent locking of the local CDM
wavevector at throughout the underdoped phase diagram of the
canonical cuprate . These observations have significant
fundamental consequences because they are orthogonal to a k-space (Fermi
surface) based picture of the cuprate CDM but are consistent with strong
coupling r-space based theories. Our findings imply that it is the latter that
provide the intrinsic organizational principle for the cuprate CDM state
Deletion Mutations in an Australian Series of HNPCC Patients
Hereditary non polyposis colorectal cancer (HNPCC) is characterized by the presence of early onset colorectal cancer and other epithelial malignancies. The genetic basis of HNPCC is a deficiency in DNA mismatch repair, which manifests itself as DNA microsatellite instability in tumours. There are four genes involved in DNA mismatch repair that have been linked to HNPCC; these include hMSH2, hMLH1, hMSH6 and hPMS2. Of these four genes hMLH1 and hMSH2 account for the majority of families diagnosed with the disease. Notwithstanding, up to 40 percent of families do not appear to harbour a change in either hMSH2 or hMLH1 that can be detected using standard screening procedures such as direct DNA sequencing or a variety of methods all based on a heteroduplex analysis
Machine Learning in Electronic Quantum Matter Imaging Experiments
Essentials of the scientific discovery process have remained largely
unchanged for centuries: systematic human observation of natural phenomena is
used to form hypotheses that, when validated through experimentation, are
generalized into established scientific theory. Today, however, we face major
challenges because automated instrumentation and large-scale data acquisition
are generating data sets of such volume and complexity as to defy human
analysis. Radically different scientific approaches are needed, with machine
learning (ML) showing great promise, not least for materials science research.
Hence, given recent advances in ML analysis of synthetic data representing
electronic quantum matter (EQM), the next challenge is for ML to engage
equivalently with experimental data. For example, atomic-scale visualization of
EQM yields arrays of complex electronic structure images, that frequently elude
effective analyses. Here we report development and training of an array of
artificial neural networks (ANN) designed to recognize different types of
hypothesized order hidden in EQM image-arrays. These ANNs are used to analyze
an experimentally-derived EQM image archive from carrier-doped cuprate Mott
insulators. Throughout these noisy and complex data, the ANNs discover the
existence of a lattice-commensurate, four-unit-cell periodic,
translational-symmetry-breaking EQM state. Further, the ANNs find these
phenomena to be unidirectional, revealing a coincident nematic EQM state.
Strong-coupling theories of electronic liquid crystals are congruent with all

Assessment of potential anti-cancer stem cell activity of marine algal compounds using an in vitro mammosphere assay
Background: The cancer stem cell (CSC) theory proposes that tumours arise from and are sustained by a subpopulation of cells with both cancer and stem cell properties. One of the key hallmarks of CSCs is the ability to grow anchorage-independently under serum-free culture conditions resulting in the formation of tumourspheres. It has further been reported that these cells are resistant to traditional chemotherapeutic agents.
Methods: In this study, the tumoursphere assay was validated in MCF-7 cells and used to screen novel marine algal compounds for potential anti-cancer stem cell (CSC) activity in vitro.
Results: MCF-7 breast cancer cells were observed to generate tumourspheres or mammospheres after 3-5 days growth in anchorage-independent conditions and an apparent enrichment in potential CSCs was observed by an increase in the proportion of CD44high/CD24low marker-bearing cells and Oct4 expression compared to those in the bulk population grown in regular adherent conditions. Using this assay, a set of algal metabolites was screened for the ability to inhibit mammosphere development as a measure of potential anti-CSC activity. We report that the polyhalogenated monoterpene stereoisomers RU017 and RU018 isolated from the red alga Plocamium cornutum, both of which displayed no cytotoxicity against either adherent MCF-7 breast cancer or MCF-12A non-transformed breast epithelial cells, were able to prevent MCF-7 mammosphere formation in vitro. On the other hand, neither the brown algal carotenoid fucoxanthin nor the chemotherapeutic paclitaxel, both of which were toxic to adherent MCF-7 and MCF-12A cells, were able to inhibit mammosphere formation. In fact, pre-treatment with paclitaxel appeared to enhance mammosphere formation and development, a finding which is consistent with the reported resistance of CSCs to traditional chemotherapeutic agents.
Conclusion: Due to the proposed clinical significance of CSC in terms of tumour initiation and metastasis, the identification of agents able to inhibit this subpopulation has clinical significance
