    A replicated, small plot study on watermelon [Citrullus lanatus (Thunberg) Matsumura and Nakai] in 1997, 1999, and 2000 revealed that production management intensity affected yields and profitability of watermelon, in Oklahoma. Management intensity was based on a combination of cultural practices and levels of use of production methods. Low intensity management (LM) consisted of use of soil fertilization and weed control. High intensity management (HM) included the same weed control and fertilization as LM but also included use of plastic mulch, drip irrigation, insect pest control, and plant disease control. Cost and return analyses were based on the range of actual prices during the cropping season and the range of yields during the three years. Yields from the seedless triploid genotype 'Gem Dandy' consistently resulted in greater positive net revenue under HM than the diploid open pollinated 'Allsweet' or the hybrid diploid 'Sangria'. Under LM, yields from the seedless triploid also resulted in greater net revenues when conditions were favorable or lost less money than the open pollinated 'Allsweet' or the hybrid diploid 'Sangria' when conditions were unfavorable.Crop Production/Industries,

    KSwAGS: A Swift X-ray and UV Survey of the Kepler Field. I

    We introduce the first phase of the Kepler-Swift Active Galaxies and Stars survey (KSwAGS), a simultaneous X-ray and UV survey of ~6 square degrees of the Kepler field using the Swift XRT and UVOT. We detect 93 unique X-ray sources with S/N>3 with the XRT, of which 60 have observed UV counterparts. We use the Kepler Input Catalog (KIC) to obtain the optical counterparts of these sources, and construct the X-ray to optical flux ratio as a first approximation of the classification of the source. The survey produces a mixture of stellar sources, extragalactic sources, and sources which we are not able to classify with certainty. We have obtained optical spectra for thirty of these targets, and are conducting an ongoing observing campaign to fully identify the sample. For sources classified as stellar or AGN with certainty, we construct SEDs using the 2MASS, UBV and GALEX data supplied for their optical counterparts by the KIC, and show that the SEDs differ qualitatively between the source types, and so can offer a method of classification in absence of a spectrum. Future papers in this series will analyze the timing properties of the stars and AGN in our sample separately. Our survey provides the first X-ray and UV data for a number of known variable stellar sources, as well as a large number of new X-ray detections in this well-studied portion of the sky.     In vivo imaging of pyrrole-imidazole polyamides with positron emission tomography

    The biodistribution profiles in mice of two pyrrole-imidazole polyamides were determined by PET. Pyrrole-imidazole polyamides are a class of small molecules that can be programmed to bind a broad repertoire of DNA sequences, disrupt transcription factor-DNA interfaces, and modulate gene expression pathways in cell culture experiments. The 18F-radiolabeled polyamides were prepared by oxime ligation between 4-[18F]-fluorobenzaldehyde and a hydroxylamine moiety at the polyamide C terminus. Small animal PET imaging of radiolabeled polyamides administered to mice revealed distinct differences in the biodistribution of a 5-ring ÎČ-linked polyamide versus an 8-ring hairpin, which exhibited better overall bioavailability. In vivo imaging of pyrrole-imidazole polyamides by PET is a minimum first step toward the translation of polyamide-based gene regulation from cell culture to small animal studies

    Reprocessing of X-rays in AGN. I. Plane parallel geometry -- test of pressure equilibrium

    We present a model of the vertical stratification and the spectra of an irradiated medium under the assumption of constant pressure. Such a solution has properties intermediate between constant density models and hydrostatic equilibrium models, and it may represent a flattened configuration of gas clumps accreting onto the central black hole. Such a medium develops a hot skin, thicker than hydrostatic models, but thinner than constant density models, under comparable irradiation. The range of theoretical values of the alpha_ox index is comparable to those from hydrostatic models and both are close to the observed values for Seyfert galaxies but lower than in quasars. The amount of X-ray Compton reflection is consistent with the observed range. The characteristic property of the model is a frequently multicomponent iron K alpha line.Comment: accepted for publication in Astronomy and Astrophysic

    Improved nuclear localization of DNA-binding polyamides

    Regulation of endogenous genes by DNA-binding polyamides requires effective nuclear localization. Previous work employing confocal microscopy to study uptake of fluorophore-labeled polyamides has demonstrated the difficulty of predicting a priori the nuclear uptake of a given polyamide. The data suggest that dye identity influences uptake sufficiently such that a dye-conjugate cannot be used as a proxy for unlabeled analogs. Polyamides capable of nuclear localization unaided by fluorescent dyes are desirable due to size and other limitations of fluorophores. Recently, a polyamide-fluorescein conjugate targeted to the hypoxia response element (HRE) was found to inhibit vascular endothelial growth factor (VEGF) expression in cultured HeLa cells. The current study uses inhibition of VEGF expression as a biological read-out for effective nuclear localization of HRE-targeted polyamides. We synthesized a focused library of non-fluorescent, HRE-targeted polyamides in which the C-terminus ‘tail’ has been systematically varied. Members of this library bind the HRE with affinities comparable or superior to that of the fluorescein-labeled analog. Although most library members demonstrate modest or no biological activity, two non-fluorescent polyamides are reported with activity rivaling that of the previously reported fluorescein-labeled polyamide. We also show evidence that promoter occupancy by HIF-1, the transcription factor that binds the HRE, is inhibited by HRE-targeted polyamides
