    Field Experience as the Centerpiece of an Integrated Model for STEM Teacher Preparation

    The purpose of this study was to provide a descriptive account of one pathway for preparing high-quality STEM (science, technology, engineering, and mathematics) teachers for work in high-need urban schools. In this account, we discuss the supports that STEM majors need in learning how to think about the content that they know well, through an educational perspective that focuses on teaching and learning. We also describe the approach that we use that integrates content knowledge, pedagogical content knowledge, and three extensive teaching co-op experiences to facilitate the transition from successful STEM undergraduate students to effective teachers of STEM content. We suggest that by using the teaching co-op experiences to both filter and reflect on content and pedagogical content knowledge, the STEM undergraduates develop a particularly strong foundation of knowledge for teaching

    An Integrated Model for STEM Teacher Preparation: The Value of a Teaching Cooperative Educational Experience

    The purpose of this article is to evaluate an intensive, integrated model for teacher preparation, specifically, a preservice STEM teacher education model which incorporates science or mathematics content with pedagogical content knowledge in an early, intensive classroom immersion program based entirely in a local school. STEM preservice teachers participated in a cooperative teaching experience which placed them at the school site for their university course work and field placements, thus ensuring a more seamless connection between theory and practice. The findings from this comparative study of the STEM preservice students in the teaching co-op and STEM preservice teachers in a traditional preparation model indicate that the STEM preservice teachers in the teaching cooperative model were more confident about their teaching skills, more comfortable with their content knowledge, and prepared to work effectively with high-needs students

    Optimized Seamless Integration of Biomolecular Data

    Today, scientific data is inevitably digitized, stored in a wide variety of heterogeneous formats, and is accessible over the Internet. Scientists need to access an integrated view of multiple remote or local heterogeneous data sources. They then integrate the results of complex queries and apply further analysis and visualization to support the task of scientific discovery. Building such a digital library for scientific discovery requires accessing and manipulating data extracted from flat files or databases, documents retrieved from the Web, as well as data that is locally materialized in warehouses or is generated by software. We consider several tasks to provide optimized and seamless integration of biomolecular data. Challenges to be addressed include capturing and representing source capabilities; developing a methodology to acquire and represent semantic knowledge and metadata about source contents, overlap in source contents, and access costs; and decision support to select sources and capabilities using cost based and semantic knowledge, and generating low cost query evaluation plans.     Mast Cells and Gastrointestinal Dysmotility in the Cystic Fibrosis Mouse

    BACKGROUND: Cystic fibrosis (CF) has many effects on the gastrointestinal tract and a common problem in this disease is poor nutrition. In the CF mouse there is an innate immune response with a large influx of mast cells into the muscularis externa of the small intestine and gastrointestinal dysmotility. The aim of this study was to evaluate the potential role of mast cells in gastrointestinal dysmotility using the CF mouse (Cftr(tm1UNC), Cftr knockout). METHODOLOGY: Wild type (WT) and CF mice were treated for 3 weeks with mast cell stabilizing drugs (ketotifen, cromolyn, doxantrazole) or were treated acutely with a mast cell activator (compound 48/80). Gastrointestinal transit was measured using gavage of a fluorescent tracer. RESULTS: In CF mice gastric emptying at 20 min post-gavage did not differ from WT, but was significantly less than in WT at 90 min post-gavage. Gastric emptying was significantly increased in WT mice by doxantrazole, but none of the mast cell stabilizers had any significant effect on gastric emptying in CF mice. Mast cell activation significantly enhanced gastric emptying in WT mice but not in CF mice. Small intestinal transit was significantly less in CF mice as compared to WT. Of the mast cell stabilizers, only doxantrazole significantly affected small intestinal transit in WT mice and none had any effect in CF mice. Mast cell activation resulted in a small but significant increase in small intestinal transit in CF mice but not WT mice. CONCLUSIONS: The results indicate that mast cells are not involved in gastrointestinal dysmotility but their activation can stimulate small intestinal transit in cystic fibrosis

    The Somatic Genomic Landscape of Glioblastoma

    We describe the landscape of somatic genomic alterations based on multi-dimensional and comprehensive characterization of more than 500 glioblastoma tumors (GBMs). We identify several novel mutated genes as well as complex rearrangements of signature receptors including EGFR and PDGFRA. TERT promoter mutations are shown to correlate with elevated mRNA expression, supporting a role in telomerase reactivation. Correlative analyses confirm that the survival advantage of the proneural subtype is conferred by the G-CIMP phenotype, and MGMT DNA methylation may be a predictive biomarker for treatment response only in classical subtype GBM. Integrative analysis of genomic and proteomic profiles challenges the notion of therapeutic inhibition of a pathway as an alternative to inhibition of the target itself. These data will facilitate the discovery of therapeutic and diagnostic target candidates, the validation of research and clinical observations and the generation of unanticipated hypotheses that can advance our molecular understanding of this lethal cancer
