    This chapter presents a conceptual analysis of the current trends for research paradigmatic perspectives used in doctoral social justice research approaches. The chapter offers a concise resource for doctoral scholars and their research supervisors to establish and illustrate a relevant paradigmatic perspective aligned with the research method and design choice to view the dissertation research problem in doctoral social justice research. Paradigmatic perspectives from feminist, critical theoretical perspectives, and grounded theory are also included as examples of specific approaches

    R1507, a fully human monoclonal antibody targeting IGF-1R, is effective alone and in combination with rapamycin in inhibiting growth of osteosarcoma xenografts

    Background The combination of rapamycin and R1507, a fully human IgG1 monoclonal antibody targeting the IGF-1 receptor (IGF-1R), in osteosarcoma xenograft tumors in vivo is evaluated in this report. Procedure Six osteosarcoma xenograft tumor models were evaluated for growth inhibition after monotherapy with R1507, rapamycin, and the combination of both drugs. Phosphorylation of proteins involved in IGF-1R signaling is evaluated at various time points by immunoblotting. Results IGF-1R was expressed in five of the six human osteosarcoma tumor lines. Objective responses to R1507 were seen in four of the six tumor lines (OS1, OS2, OS9, and OS17) including one complete response in OS1. IGF-1R protein levels did not predict degree of response to R1507 in the sensitive tumors. However, in one of the two R1507-resistant tumors (OS33), there was a minimal expression of IGF-1R. An increase in AKT phosphorylation was observed in all the osteosarcoma tumors treated with rapamycin. However, phosphorylation of AKT was inhibited when rapamycin was used in combination with R1507. In three of the xenograft tumor lines, there was an improvement in response when R1507 was used in combination with rapamycin. Conclusions IGF-1R inhibition by R1507 induced tumor growth delays and improvement in event-free survival in four of six osteosarcoma xenograft tumor lines. R1507 negates increased signaling through AKT in response to mammalian target of rapamycin inhibition, suggesting that the combination is worthy of further evaluation in patients. As R1507 and other IGF-1R inhibitors advance in clinical trials, it will be important to understand biomarkers of response and pathways of resistance.     Understanding the Behaviour of Silver as a Low Friction Coating in Aerospace Fasteners

    Nuts and bolts used in aero-engines are manufactured from heat-resistant super-alloys. When used in a like on like couple, these materials have a high coefficient of friction, and frequently seizure occurs. In order to prevent this, a silver coating is applied to the nut threads, providing a low friction boundary at the interface. Additionally, a radial crimp is applied to the nut, in order to provide a self-locking feature preventing vibration self-loosening. In this study, the coefficient of friction of the thread contact will be investigated both during initial joint assembly, and after thermal ageing. Additionally, a finite element model will be employed to investigate the contact mechanics as a consequence of the crimp. The low coefficient of friction observed during initial assembly was found to be a consequence of shear flow of the silver coating, with an approximate doubling of this value once the coating aged. Areas of silver removal were found to be coincident with areas of high contact pressure in the joint, attributable to the crimp feature

    The general 2-dimensional fermion system with repulsive interactions (typified by the Hubbard Model) is bosonized, taking into account the finite on-shell forward scattering phase shift derived in earlier papers.     Topological string in harmonic space and correlation functions in S3S^3 stringy cosmology

    We develop the harmonic space method for conifold and use it to study local complex deformations of TS3T^{\ast}S^{3} preserving manifestly SL(2,C)SL(2,C) isometry. We derive the perturbative manifestly SL(2,C)SL(2,C) invariant partition function Ztop\mathcal{Z}_{top} of topological string B model on locally deformed conifold. Generic nn momentum and winding modes of 2D c=1c=1 non critical theory are described by highest % \upsilon_{(n,0)} and lowest components υ(0,n)\upsilon_{(0,n)} of SL(2,C)SL(2,C) spin s=n2s=\frac{n}{2} multiplets (nk,k))% (\upsilon _{(n-k,k)}) , 0kn0\leq k\leq n and are shown to be naturally captured by harmonic monomials. Isodoublets (n=1n=1) describe uncoupled units of momentum and winding modes and are exactly realized as the SL(2,C)SL(2,C) harmonic variables Uα+U_{\alpha}^{+} and VαV_{\alpha}^{-}. We also derive a dictionary giving the passage from Laurent (Fourier) analysis on TS1T^{\ast}S^{1} (S1S^{1}) to the harmonic method on TS3T^{\ast}S^{3} (S3S^{3}). The manifestly SU(2,C)SU(2,C) covariant correlation functions of the S3S^{3} quantum cosmology model of Gukov-Saraikin-Vafa are also studied.Comment: 91 page

    A systems approach to evaluate One Health initiatives

    Challenges calling for integrated approaches to health, such as the One Health (OH) approach, typically arise from the intertwined spheres of humans, animals, and ecosystems constituting their environment. Initiatives addressing such wicked problems commonly consist of complex structures and dynamics. As a result of the EU COST Action (TD 1404) “Network for Evaluation of One Health” (NEOH), we propose an evaluation framework anchored in systems theory to address the intrinsic complexity of OH initiatives and regard them as subsystems of the context within which they operate. Typically, they intend to influence a system with a view to improve human, animal, and environmental health. The NEOH evaluation framework consists of four overarching elements, namely: (1) the definition of the initiative and its context, (2) the description of the theory of change with an assessment of expected and unexpected outcomes, (3) the process evaluation of operational and supporting infrastructures (the “OH-ness”), and (4) an assessment of the association(s) between the process evaluation and the outcomes produced. It relies on a mixed methods approach by combining a descriptive and qualitative assessment with a semi-quantitative scoring for the evaluation of the degree and structural balance of “OH-ness” (summarised in an OH-index and OH-ratio, respectively) and conventional metrics for different outcomes in a multi-criteria-decision-analysis. Here, we focus on the methodology for Elements (1) and (3) including ready-to-use Microsoft Excel spreadsheets for the assessment of the “OH-ness”. We also provide an overview of Element (2), and refer to the NEOH handbook for further details, also regarding Element (4) (http://neoh.onehealthglobal.net). The presented approach helps researchers, practitioners, and evaluators to conceptualise and conduct evaluations of integrated approaches to health and facilitates comparison and learning across different OH activities thereby facilitating decisions on resource allocation. The application of the framework has been described in eight case studies in the same Frontiers research topic and provides first data on OH-index and OH-ratio, which is an important step towards their validation and the creation of a dataset for future benchmarking, and to demonstrate under which circumstances OH initiatives provide added value compared to disciplinary or conventional health initiatives

    Kinetoplastids:related protozoan pathogens, different diseases

    Kinetoplastids are a group of flagellated protozoans that include the species Trypanosoma and Leishmania, which are human pathogens with devastating health and economic effects. The sequencing of the genomes of some of these species has highlighted their genetic relatedness and underlined differences in the diseases that they cause. As we discuss in this Review, steady progress using a combination of molecular, genetic, immunologic, and clinical approaches has substantially increased understanding of these pathogens and important aspects of the diseases that they cause. Consequently, the paths for developing additional measures to control these “neglected diseases” are becoming increasingly clear, and we believe that the opportunities for developing the drugs, diagnostics, vaccines, and other tools necessary to expand the armamentarium to combat these diseases have never been better

    Regulation of Apolipoprotein A-1 and Apolipoprotein B100 Genes by Thymoquinone Rich Fraction and Thymoquinone in HEPG2 Cells.

    Thymoquinone (TQ) rich fraction (TQRF) extracted from Nigella sativa seeds using a supercritical fluid extraction technique was prepared. The regulatory effects of TQRF at 80 µg/mL and commercial TQ at 2 µg/mL on apolipoprotein B100 (Apo B100) and apolipoprotein A-1 (Apo A-1) genes in the presence or absence of 25-hydroxycholesterol (25OH), were investigated in human HepG2 cell line using quantitative real-time polymerase chain reaction. Incubating HepG2 cells in 10% human lipoprotein deficient serum (HLPDS) for 24 h in the presence of 2 µg/mL 25OH showed a significant increase in Apo B100 mRNA expression level by twofold compared to the control cells; on the other hand, no significant change in Apo A-1 mRNA level was observed. When cells were incubated with HLPDS in the absence of 25OH and treated with TQRF and TQ, the mRNA level of Apo B100 was down-regulated by 70 and 49%, respectively, in TQRF and TQ treated cells compared to untreated cells. Apo A-1 gene was up-regulated by four- and twofold in TQRF and TQ treated cells, respectively, compared to that observed in untreated cells. The present study clearly shows that TQRF and TQ are effective in regulating Apo A-1 and Apo B100 genes that influence cholesterol metabolism in HepG2 cells

    Educating the public health workforce: Issues and challenges

    Background: In public health, as well as other health education contexts, there is increasing recognition of the transformation in public health practice and the necessity for educational providers to keep pace. Traditionally, public health education has been at the postgraduate level; however, over the past decade an upsurge in the growth of undergraduate public health degrees has taken place. Discussion: This article explores the impact of these changes on the traditional sphere of Master of Public Health programs, the range of competencies required at undergraduate and postgraduate levels, and the relevance of these changes to the public health workforce. It raises questions about the complexity of educational issues facing tertiary institutions and discusses the implications of these issues on undergraduate and postgraduate programs in public health. Conclusion: The planning and provisioning of education in public health must differentiate between the requirements of undergraduate and postgraduate students – while also addressing the changing needs of the health workforce. Within Australia, although significant research has been undertaken regarding the competencies required by postgraduate public health students, the approach is still somewhat piecemeal, and does not address undergraduate public health. This paper argues for a consistent approach to competencies that describe and differentiate entry-level and advanced practice
