    Nutrients and Chlorophyll at the Shelf Break Off the Southeastern United States During the Genesis of Atlantic Lows Experiment: Winter 1986

    The outer shelf and upper slope off Charleston, South Carolina, were the site of oceanographic and meteorological measurements during the winter of 1986. The purpose of the study was to test ideas about front formation, heat transport, and stratification during cold air outbreaks. An ancillary part of the study was the observation of nutrients and chlorophyll concentrations. The observations extended across the shelf and sometimes crossed the Gulf Stream front. The results show slightly elevated nitrate concentrations in outer shelf waters (1 - 2 μM NO3) with chlorophyll concentrations in the 1 - 1.8 μg L-1 range. When effects of Gulf Stream frontal eddies are eliminated, the estimated wintertime wind-driven transport of nutrients from the deep ocean to the shelf is comparable to that observed during spring and summer. Thus significant onshore nutrient transport can occur throughout the year in this region

    Raman excitation spectroscopy of carbon nanotubes: effects of pressure medium and pressure

    Raman excitation and emission spectra for the radial breathing mode (RBM) are reported, together with a preliminary analysis. From the position of the peaks on the two-dimensional plot of excitation resonance energy against Raman shift, the chiral indices (m, n) for each peak are identified. Peaks shift from their positions in air when different pressure media are added - water, hexane, sulphuric acid - and when the nanotubes are unbundled in water with surfactant and sonication. The shift is about 2 - 3 cm-1 in RBM frequency, but unexpectedly large in resonance energy, being spread over up to 100meV for a given peak. This contrasts with the effect of pressure. The shift of the peaks of semiconducting nanotubes in water under pressure is orthogonal to the shift from air to water. This permits the separation of the effects of the pressure medium and the pressure, and will enable the true pressure coefficients of the RBM and the other Raman peaks for each (m, n) to be established unambiguously.Comment: 6 pages, 3 Figures, Proceedings of EHPRG 2011 (Paris

    A bi-directional relationship between obesity and health-related quality of life : evidence from the longitudinal AusDiab study

    Objective: To assess the prospective relationship between obesity and health-related quality of life, including a novel assessment of the impact of health-related quality of life on weight gain.Design and setting: Longitudinal, national, population-based Australian Diabetes, Obesity and Lifestyle (AusDiab) study, with surveys conducted in 1999/2000 and 2004/2005.Participants: A total of 5985 men and women aged 25 years at study entry.Main outcome measure(s): At both time points, height, weight and waist circumference were measured and self-report data on health-related quality of life from the SF-36 questionnaire were obtained. Cross-sectional and bi-directional, prospective associations between obesity categories and health-related quality of life were assessed.Results: Higher body mass index (BMI) at baseline was associated with deterioration in health-related quality of life over 5 years for seven of the eight health-related quality of life domains in women (all P0.01, with the exception of mental health, P&gt;0.05), and six out of eight in men (all P&lt;0.05, with the exception of role-emotional, P=0.055, and mental health, P&gt;0.05). Each of the quality-of-life domains related to mental health as well as the mental component summary were inversely associated with BMI change (all P&lt;0.0001 for women and P0.01 for men), with the exception of vitality, which was significant in women only (P=0.008). For the physical domains, change in BMI was inversely associated with baseline general health in women only (P=0.023).Conclusions: Obesity was associated with a deterioration in health-related quality of life (including both physical and mental health domains) in this cohort of Australian adults followed over 5 years. Health-related quality of life was also a predictor of weight gain over 5 years, indicating a bi-directional association between obesity and health-related quality of life. The identification of those with poor health-related quality of life may be important in assessing the risk of future weight gain, and a focus on health-related quality of life may be beneficial in weight management strategies.<br /

    Engineering enzymes with non-canonical active site functionality

    The combination of computational enzyme design and laboratory evolution provides an attractive platform for the creation of protein catalysts with new function. To date, designed mechanisms have relied upon Nature’s alphabet of 20 genetically encoded amino acids, which greatly restricts the range of functionality which can be installed into enzyme active sites. Here, we have exploited engineered components of the cellular translation machinery to create a protein catalyst which operates via a non-canonical catalytic nucleophile. We have subsequently shown that powerful laboratory evolution protocols can be readily adapted to allow optimization of enzymes containing non-canonical active site functionality. Crystal structures obtained along the evolutionary trajectory highlight the origins of improved activity. Thus our approach merges beneficial features of organo- and biocatalysis, by combining the intrinsic reactivities and greater versatility of small molecule catalysts with the rate enhancements, reaction selectivities and evolvability of proteins.     Objectively Measured Physical Activity and the Subsequent Risk of Incident Dysglycemia: The Australian Diabetes, Obesity and Lifestyle Study (AusDiab)

    OBJECTIVE - To investigate pedometer-measured physical activity (PA) in 2000 and change in PA over 5 years with subsequent risk of dysglycemia by 2005. RESEARCH DESIGN AND METHODS - This prospective cohort study in Tasmania, Australia, analyzed 458 adults with normal glucose tolerance and a mean (SD) age of 49.7 (12.1) years in 2000. Variables assessed in 2000 and 2005 included PA, by pedometer and questionnaire, nutrient intake, and other lifestyle factors. Incident dysglycemia was defined as the development of impaired fasting glucose or impaired glucose tolerance revealed by oral glucose tolerance testing in 2005, without type 2 diabetes. RESULTS - Incident dysglycemia developed in 26 participants during the 5-year period. Higher daily steps in 2000 were independently associated with a lower 5-year risk of incident dysglycemia (adjusted odds ratio [AOR] 0.87 [95% CI 0.77-0.97] per 1,000-step increment). Higher daily steps in 2005, after controlling for baseline steps in 2000 (thus reflecting change in steps over 5 years), were not associated with incident dysglycemia (AOR 1.02 [0.92-1.14]). Higher daily steps in 2000 were also associated with lower fasting blood glucose, but not 2-h plasma glucose by 2005. Further adjustment for BMI or waist circumference did not remove these associations. CONCLUSIONS - Among community-dwelling adults, a higher rate of daily steps is associated with a reduced risk of incident dysglycemia. This effect appears to be not fully mediated through reduced adiposity

    Chemical Pressure and Physical Pressure in BaFe_2(As_{1-x}P_{x})_2

    Measurements of the superconducting transition temperature, T_c, under hydrostatic pressure via bulk AC susceptibility were carried out on several concentrations of phosphorous substitution in BaFe_2(As_{1-x}P_x)_2. The pressure dependence of unsubstituted BaFe_2As_2, phosphorous concentration dependence of BaFe_2(As_{1-x}P_x)_2, as well as the pressure dependence of BaFe_2(As_{1-x}P_x)_2 all point towards an identical maximum T_c of 31 K. This demonstrates that phosphorous substitution and physical pressure result in similar superconducting phase diagrams, and that phosphorous substitution does not induce substantial impurity scattering.Comment: 5 pages, 4 figures, to be published in Journal of the Physical Society of Japa

    Measurement of bitumen viscosity in the room-temperature drop experiment: student education, public outreach and modern science in one

    Slow flow of the viscous liquid is a thought-provoking experiment that challenges students, academics and public to think about some fundamental questions in modern science. In the Queensland demonstration, the world-longest running experiment earning the Ig Nobel prize, one drop of pitch takes about 10 years to fall, leading to problems of actually observing the drops. Here, we describe our recent demonstration of slowly-flowing bitumen where appreciable flow is observed on the time scale of months. The experiment is free from dissipative heating effects and has the potential to improve the accuracy of measurement. Bitumen viscosity was calculated by undergraduate students during the summer project. The worldwide access to the running experiment is provided by webcams uploading the images to a dedicated website, enhancing student education experience and promotion of science. This demonstration serves as an attractive student education exercise and stimulates the discussion of fundamental concepts and hotly debated ideas in modern physics research: difference between solids and liquids, the nature of liquid-glass transition, emergence of long time scales in a physical process, and the conflict between human intuition and physical reality

    A review of the surgical conversion rate and independent management of spinal extended scope practitioners in a secondary care setting

    INTRODUCTION Spinal orthopaedic triage aims to reduce unnecessary referrals to surgical consultants, thereby reducing waiting times to be seen by a surgeon and to surgical intervention. This paper presents an evaluation of a spinal orthopaedic triage service in the third largest spinal unit in the UK. METHODS A retrospective service evaluation spanning 2012 to 2014 was undertaken by members of the extended scope practitioner (ESP) team to evaluate the ESPs’ ability to manage patient care independently and triage surgical referrals appropriately. Data collected included rates of independent management, referral rates for surgical consideration and conversion to surgery. Patient satisfaction rates were evaluated retrospectively from questionnaires given to 5% of discharged patients. RESULTS A total of 2,651 patients were seen. The vast majority (92%) of all referrals seen by ESPs were managed independently. Only 8% required either a discussion with a surgeon to confirm management or for surgical review. Of the latter, 81% were considered to be suitable surgical referrals. A 99% satisfaction rate was reported by discharged patients. CONCLUSIONS ESP services in a specialist spinal service are effective in managing spinal conditions conservatively and identifying surgical candidates appropriately. Further research is needed to confirm ESPs’ diagnostic accuracy, patient outcomes and cost effectiveness
