    Evaluation of lung recovery after static administration of three different perfluorocarbons in pigs.

    International audienceBackground: The respiratory properties of perfluorocarbons (PFC) have been widely studied for liquid ventilation inhumans and animals. Several PFC were tested but their tolerance may depend on the species. Here, the effects of asingle administration of liquid PFC into pig lungs were assessed and compared. Three different PFC having distinctevaporative and spreading coefficient properties were evaluated (Perfluorooctyl bromide [PFOB], perfluorodecalin[PFD] and perfluoro-N-octane [PFOC]).Methods: Pigs were anesthetized and submitted to mechanical ventilation. They randomly received an intra-trachealadministration of 15 ml/kg of either PFOB, PFD or PFOC with 12 h of mechanical ventilation before awakening andweaning from ventilation. A Control group was submitted to mechanical ventilation with no PFC administration. Allanimals were followed during 4 days after the initial PFC administration to investigate gas exchanges and clinicalrecovery. They were ultimately euthanized for histological analyses and assessment of PFC residual concentrationswithin the lungs using dual nuclei fluorine and hydrogen Magnetic Resonance Imaging (MRI). Sixteen animals wereincluded (4/group).Results: In the PFD group, animals tended to be hypoxemic after awakening. In PFOB and PFOC groups, blood gaseswere not significantly different from the Control group after awakening. The poor tolerance of PFD was likely related toa large amount of residual PFC, as observed using MRI in all lung samples (≈10% of lung volume). This percentage waslower in the PFOB group (≈1%) but remained significantly greater than in the Control group. In the PFOC group, thepercentage of residual PFC was not significantly different from that of the Control group (≈0.1%). Histologically, themost striking feature was an alveolar infiltration with foam macrophages, especially in the groups treated by PFD orPFOB.Conclusions: Of the three tested perfluorocarbons, PFOC offered the best tolerance in terms of lung function, gasexchanges and residuum in the lung. PFOC was rapidly cleared from the lungs and virtually disappeared after 4 dayswhereas PFOB persisted at significant levels and led to foam macrophage infiltration.     The original problem of supervised classification considers the task of automatically assigning objects to their respective classes on the basis of numerical measurements derived from these objects. Classifiers are the tools that implement the actual functional mapping from these measurements---also called features or inputs---to the so-called class label---or output. The fields of pattern recognition and machine learning study ways of constructing such classifiers. The main idea behind supervised methods is that of learning from examples: given a number of example input-output relations, to what extent can the general mapping be learned that takes any new and unseen feature vector to its correct class? This chapter provides a basic introduction to the underlying ideas of how to come to a supervised classification problem. In addition, it provides an overview of some specific classification techniques, delves into the issues of object representation and classifier evaluation, and (very) briefly covers some variations on the basic supervised classification task that may also be of interest to the practitioner

    Attraction Basins as Gauges of Robustness against Boundary Conditions in Biological Complex Systems

    One fundamental concept in the context of biological systems on which researches have flourished in the past decade is that of the apparent robustness of these systems, i.e., their ability to resist to perturbations or constraints induced by external or boundary elements such as electromagnetic fields acting on neural networks, micro-RNAs acting on genetic networks and even hormone flows acting both on neural and genetic networks. Recent studies have shown the importance of addressing the question of the environmental robustness of biological networks such as neural and genetic networks. In some cases, external regulatory elements can be given a relevant formal representation by assimilating them to or modeling them by boundary conditions. This article presents a generic mathematical approach to understand the influence of boundary elements on the dynamics of regulation networks, considering their attraction basins as gauges of their robustness. The application of this method on a real genetic regulation network will point out a mathematical explanation of a biological phenomenon which has only been observed experimentally until now, namely the necessity of the presence of gibberellin for the flower of the plant Arabidopsis thaliana to develop normally

    Patients with an Open Abdomen in Asian, American and European Continents: A Comparative Analysis from the International Register of Open Abdomen (IROA)

    Background: International register of open abdomen (IROA) enrolls patients from several centers in American, European, and Asiatic continent. The aim of our study is to compare the characteristics, management and clinical outcome of adult patients treated with OA in the three continents. Material and methods: A prospective analysis of adult patients enrolled in the international register of open abdomen (IROA). Trial registration: NCT02382770. Results: 1183 patients were enrolled from American, European and Asiatic Continent. Median age was 63 years (IQR 49–74) and was higher in the European continent (65 years, p < 0.001); 57% were male. The main indication for OA was peritonitis (50.6%) followed by trauma (15.4%) and vascular emergency (13.5%) with differences among the continents (p < 0.001). Commercial NPWT was preferred in America and Europe (77.4% and 52.3% of cases) while Barker vacuum pack (48.2%) was the preferred temporary abdominal closure technique in Asia (p < 0.001). Definitive abdominal closure was achieved in 82.3% of cases in America (fascial closure in 90.2% of cases) and in 56.4% of cases in Asia (p < 0.001). Prosthesis were mostly used in Europe (17.3%, p < 0.001). The overall entero-atmospheric fistula rate 2.5%. Median open abdomen duration was 4 days (IQR 2–7). The overall intensive care unit and hospital length-of-stay were, respectively, 8 and 11 days (no differences between continents). The overall morbidity and mortality rates for America, Europe, and Asia were, respectively, 75.8%, 75.3%, 91.8% (p = 0.001) and 31.9%, 51.6%, 56.9% (p < 0.001). Conclusion: There is no uniformity in OA management in the different continents. Heterogeneous adherence to international guidelines application is evident. Different temporary abdominal closure techniques in relation to indications led to different outcomes across the continents. Adherence to guidelines, combined with more consistent data, will ultimately allow to improving knowledge and outcome

    Flavivirus envelope proteins have been shown to play a major role in virus assembly. These proteins are anchored into cellular and viral membranes by their C-terminal domain. These domains are composed of two hydrophobic stretches separated by a short hydrophilic segment containing at least one charged residue. We investigated the role of the transmembrane domains of prM and E in the envelope formation of the flavivirus yellow fever virus (YFV). Alanine scanning insertion mutagenesis has been used to examine the role of the transmembrane domains of prM and E in YFV subviral particle formation. Most of the insertions had a dramatic effect on the release of YFV subviral particles. Some of these mutations were introduced into the viral genome. The ability of these mutant viruses to produce infectious particles was severely reduced. The alanine insertions did not affect prM-E heterodimerization. In addition, replacement of the charged residues present in the middle of the transmembrane domains had no effect on subviral particle release. Taken together, these data indicate that the transmembrane domains of prM and E play a crucial role in the biogenesis of YFV envelope. In addition, these data indicate some differences between the transmembrane domains of the hepaciviruses and the flaviviruses

    Improving fast pyrolysis of lignin using three additives with different modes of action

    Lignin holds the potential to obtain key monoaromatic compounds upon its depolymerization. Depolymerization of woody biomass by pyrolysis is well established but often unsuccessful for lignin due to a combination of its melting, agglomeration, and modest yields towards aromatics. Therefore, several lignin additives have been put forth to overcome one or more of these hurdles. Although some seem promising, a direct comparison is obscured by differences in applied technical lignin types, reactor configurations/scales, and product analyses. Moreover, the effects of additives have either been evaluated mostly on an analytical scale or their mode of action is not entirely understood. This work involves the addition of clays, calcium hydroxide and sodium formate to lignin, each having a different (putative) mode of action, in a well-defined and comparable manner. Organosolv lignin and lignin with additives were analysed by TGA/DSC and py-GC/MS. Pyrolysis was performed in a lab-scale reactor (350 g feeding). The pyrolysis liquids were characterised through elemental analysis, GCxGC-FID, GCxGC-HR-ToF-MS, GPC, and HSQC NMR analyses. All additives overcame melting issues and led to increased liquid yields but the most promising were attapulgite and calcium hydroxide. Lignin with attapulgite resulted in a heavy phase with the highest carbon yield (25.7%) and a substantial monomer yield (18.9%, mostly alkylphenols). Lignin with calcium hydroxide resulted in a heavy phase with the highest monomer yield (23.8%, mostly alkylphenols) at a substantial carbon yield (15.1%).
