    Absence of dystrophin in mice reduces NO-dependent vascular function and vascular density: total recovery after a treatment with the aminoglycoside gentamicin.

    International audienceOBJECTIVE: Mutations in the dystrophin gene causing Duchenne's muscular dystrophy (DMD) lead to premature stop codons. In mice lacking dystrophin (mdx mice), a model for DMD, these mutations can be suppressed by aminoglycosides such as gentamicin. Dystrophin plays a role in flow (shear stress)-mediated endothelium-dependent dilation (FMD) in arteries. We investigated the effect of gentamicin on vascular contractile and dilatory functions, vascular structure, and density in mdx mice. METHODS AND RESULTS: Isolated mice carotid and mesenteric resistance arteries were mounted in arteriographs allowing continuous diameter measurements. Mdx mice showed lower nitric oxide (NO)-dependent FMD and endothelial NO synthase (eNOS) expression as well as decreased vascular density in gracilis and cardiac muscles compared with control mice. Treatment with gentamycin restored these parameters. In contrast, smooth muscle-dependent contractions as well as endothelium-dependent or -independent dilation were not affected by dystrophin deficiency or by gentamicin treatment. CONCLUSIONS: Dystrophin deficiency induces a selective defect in flow-dependent mechanotransduction, thus attenuating FMD and eNOS expression, and may contribute to low arteriolar density. These findings open important perspectives regarding the mechanism involved in the pathophysiology of genetic diseases related to premature stop codons such as DMD

    Defining Mediterranean and Black Sea biogeochemical Subprovinces and Synthetic ocean indicators using mesoscale oceanographic features

    The Mediterranean and Black Seas are semi-enclosed basins characterized by high environmental variability and growing anthropogenic pressure, which has led to increasing need for a bioregionalization of the oceanic environment at local and regional scales that can be used as a geographical reference with managerial applications. We aim to develop synthetic indices of the key oceanographic dynamics of each region to quantify baselines from which to assess variability and change. To do this, we compile a data set of 101 months (2002-2010) of a variety of both “classical” (i.e., sea surface temperature, surface chlorophyll a, and bathymetry) and “mesoscale” (i.e., eddy kinetic energy, finite-size Lyapunov exponents, and surface frontal gradients) ocean features that we use to characterize the surface ocean variability. We employ a k-means clustering algorithm to objectively define biogeochemical regions based on classical features, and for the first time, on mesoscale features and a combination of both classical and mesoscale features. Principal components analysis is then performed on the oceanographic variables to define integrative indices to monitor at monthly resolutions the environmental changes within each resultant region. Using both the classical and mesoscale features, we find five biogeochemical regions for the Mediterranean and Black Seas. Interestingly, the use of mesoscale variables contributes highly in the delineation of the open ocean. The first axis of the principal component analysis is explained primarily by classical ocean features and the second axis is explained by mesoscale features. Biogeochemical regions identified by the present study can be useful within the European management framework as an objective geographical framework of the Mediterranean and Black Seas, and the integrative environmental indices developed here can be used to monitor variability and long-term change.JRC.H.1-Water Resource

    Regulation of the mTOR signaling pathway: from laboratory bench to bedside and back again

    Recent publications have moved us significantly closer to a complete understanding of the mammalian target of rapamycin (mTOR) signaling pathway, which plays a central role in the control of growth and metabolism and is dysregulated in a broad spectrum of human diseases, including cancer, tuberous sclerosis, diabetes, and cardiovascular and neurodegenerative diseases. Rapamycin-related mTOR inhibitors have shown clinical efficacy in several of these diseases, and novel inhibitors currently in development will be valuable tools for further dissections of the mTOR signaling network in human health and disease

    STRENGTHENING REGIONAL COOPERATION IN FISHERIES DATA COLLECTION

    fishPi was a research project with the aim of “Strengthening regional cooperation in the area of fisheries data collection”. The project brought together over 40 experts from 13 scientific institutes in 12 countries (10 member states (MS)) and two internationally recognised survey design experts. It was funded by EU MARE grant MARE/2014/19, with a 14 month timeline commencing in April 2015. This project has trialled the way sampling designs would be developed in a regional setting and showed that collaboration and consultation is required at face to face meetings through regional groups that focus on a particular group of fisheries. The project was the first step in this process and one of the main outcomes is the framework to take the process forward; developing data formats, data sharing agreements and easily accessible software for data sharing, checking and analysis, and for the simulation testing of sampling designs.European Unio

    Report of Working Group on Widely Distributed Stocks (WGWIDE).

    As a consequence of the impact of the COVID pandemic on international travel which prevented the traditional meeting from taking place, the Working Group on Widely Distributed Stocks (WGWIDE) met online via WebEx hosted by ICES. Prior to the 2020 meeting, the generic ToRs for species and regional working groups were re-prioritised by ACOM to allow the WG to focus primarily on those ToRs most applicable to the provision of advice. WGWIDE reports on the status and considerations for management of Northeast Atlantic mackerel, blue whiting, Western and North Sea horse mackerel, Northeast Atlantic boarfish, Norwegian springspawning herring, striped red mullet (Subareas 6, 8 and Divisions 7.a-c, e-k and 9.a), and red gurnard (Subareas 3, 4, 5, 6, 7, and 8) stocks. Northeast Atlantic (NEA) Mackerel. This stock is highly migratory and widely distributed throughout the Northeast Atlantic with significant fisheries is most ICES subareas. A diverse range of fleets from smaller artisanal, handline vessels to large (100m+) factory freezer vessels and modern RSW trawlers and purse seiners take part in what is one of the most valuable European fisheries. The assessment conducted in 2020 is an update assessment, based on the configuration agreed during the most recent inter-benchmark exercise in 2019 and incorporates the most recent data available from sampling of the commercial catch in 2019, the final 2019 egg survey SSB estimate, an updated recruitment index and tagging time series along with 2020 survey data from the IESSNS swept area survey. Advice is given based on stock reference points which were updated during a management strategy evaluation carried out in 2020. Following a strong increase from 2007 to 2014, SSB has been declining although it remains well above MSY Btrigger. Fishing mortality has been below FMSY since 2016. There have been a number of large year classes since 2001 with above average recruitment over much of the most recent decade. Blue Whiting. This pelagic gadoid is widely distributed in the eastern part of the North Atlantic. The 2020 update assessment followed the protocol from the most recent inter-benchmark in 2016 and used preliminary catch data from 2020. Due to the cancellation of the 2020 acoustic survey, this data was not available. The effect on the assessment was minimal and limited to increases in uncertainty of the terminal year estimates. The SSB continues to decrease from the most recent maximum in 2017 mainly due to below average recruitment since 2017, although it remains above MSY Btrigger. Fishing mortality has been above FMSY since 2014. Norwegian Spring Spawning Herring. This is one of the largest herring stocks in the world. It is highly migratory, spawning along the Norwegian coast and feeding throughout much of the Norwegian Sea. The 2020 assessment is based on an implementation of the XSAM assessment model introduced at the benchmark in 2016. This years’ assessment indicates that the stock is continuing to decline from the peak in 2008 of 7Mt to just above MSY Btrigger due to successive years of average or below average recruitment. Catch advice for 2021 is given on the basis of the agreed management plan and represents a substantial increase over the 2020 advice due to an upward revision in the estimate of the 2016 year-class which is considered to be the most significant year-class since 2004

    Differences in foraging strategy and maternal behaviour between two sympatric fur seal species at the Crozet Islands

    Marine top-predators such as marine mammals forage in a heterogeneous environment according to their energetic requirements and to the variation in environmental characteristics. In this study, the behaviour of breeding females in 2 sympatric fur seal species, Antarctic fur seal Arctocephalus gazella and Subantarctic fur seal A. tropicalis, was investigated in relation to foraging effort. Foraging effort was hypothesised to be greater in Antarctic fur seal than in Subantarctic fur seal due to their shorter lactation period. Using satellite telemetry, time-depth recorders and satellite images of sea-surface temperature and chlorophyll a concentration, the foraging grounds, the at-sea activity budgets and the environmental features were determined for both species breeding on the Crozet Archipelago. Foraging cycle duration was similar for the 2 species, and the seals exhibited similar at-sea activity budgets. Only the proportion of time spent at sea was higher in Antarctic fur seals. Separate foraging areas were identified for the 2 species. Antarctic fur seal distribution was related to bathymetric features, while we did not find any direct relationship between chlorophyll a concentration and seal foraging areas. Our results suggest that Antarctic fur seals tend to respond to the higher needs of their pups by having a higher foraging efficiency and concentrating their foraging activity in the most productive areas

    Protocole d'actualisation des fiches d'identification de la maturité sexuelle des poissons

    Ce protocole a été réalisé dans le cadre du projet de standardisation des méthodes de lecture visuelle de la maturité sexuelle des poissons d'intérêts commerciaux (SIH IFREMER). Au cours de ce projet de 2021 à 2023, différents outils d’aide à l’identification de la maturité visuelle des poissons ont été mis en place : un répertoire de données de photographie de gonades de poissons marins (Photo_MATURITY), un protocole de photographie des gonades de poissons, des fiches d’identification de la maturité sexuelle des poissons d'intérêts commerciaux (INDEX.knit (lm-anna.github.io) ainsi que les outils de génération automatique des fiches d’identification (GitHub : MaturityScaleTools). Ces outils standardisent et apportent un soutien à la lecture visuelle de la maturité sexuelle des espèces présentes en Manche, en Mer du Nord, au niveau de la façade maritime française de la mer Méditerranée et de l’océan Atlantique, mais également présentes au niveau du littoral Guadeloupéen et Martiniquais. Ce protocole a été mis en place afin de pérenniser ces travaux. Il regroupe l’ensemble des méthodes de mise à jour des différents outils mis en place au cours de ce projet
