    Effect of argon plasma abutment activation on soft tissue healing: RCT with histological assessment.

    OBJECTIVE To assess the peri-implant soft tissue profiles between argon plasma treatment (PT) and non-treated (NPT) healing abutments by comparing clinical and histological parameters 2 months following abutment placement. MATERIALS AND METHODS Thirty participants were randomly assigned to argon-plasma treatment abutments group (PT) or non-treated abutments (NPT) group. Two months after healing abutment placement, soft peri-implant tissues and abutment were harvested, and histological and clinical parameters including plaque index, bleeding on probing, and keratinized mucosa diameter (KM) were assessed. Specialized stainings (hematoxylin-eosin and picrocirious red) coupled with immunohistochemistry (vimentin, collagen, and CK10) were performed to assess soft tissue inflammation and healing, and the collagen content keratinization. In addition to standard statistical methods, machine learning algorithms were applied for advanced soft tissue profiling between the test and control groups. RESULTS PT group showed lower plaque accumulation and inflammation grade (6.71% vs. 13.25%, respectively; p-value 0.02), and more advanced connective tissue healing and integration compared to NPT (31.77% vs. 23.3%, respectively; p = 0.009). In the control group, more expressed keratinization was found compared to the PT group, showing significantly higher CK10 (>47.5%). No differences in KM were found between the groups. SIGNIFICANCE PT seems to be a promising protocol for guided peri-implant soft tissue morphogenesis reducing plaque accumulation and inflammation, and stimulating collagen and soft tissue but without effects on epithelial tissues and keratinization

    Facial scanning technologies in the era of digital workflow: A systematic review and network meta-analysis.

    PURPOSE The aim of this network meta-analysis is to evaluate the accuracy of various face-scanning technologies in the market, with respect to the different dimensions of space (x, y, and z axes). Furthermore, attention will be paid to the type of technologies currently used and to the best practices for high-quality scan acquisition. MATERIAL AND METHODS The review was conducted following the PRISMA guidelines and its updates. A thorough search was performed using the digital databases MEDLINE, PubMed, EMBASE, and the Cochrane Central Register of Controlled Trials by entering research lines or various combinations of free words. The main keywords used during the search process were "photogrammetry", "laser scanner", "optical scanner", "3D, and "face". RESULTS None of the included technologies significantly deviated from direct anthropometry. The obtained mean differences in the distances between the considered landmarks range from 1.10 to -1.74 mm. CONCLUSION Limiting the movements of the patient and scanner allows for more accurate facial scans with all the technologies involved. Active technologies such as laser scanners (LS), structured light (SL), and infrared structured light (ISL) have accuracy comparable to that of static stereophotogrammetry while being more cost-effective and less time-consuming

    Antitumor effects of table grape extracts

    Grape (Vitis vinifera L.) is a fruit rich in polyphenols, bioactive compounds able to prevent cancer, reduce tumorigenesis, and influence critical cancer-related pathways. This research shows the main results obtained in our previous works: 1) the characterization of the polyphenolic content and antioxidant activity of two table grape skin extracts (GSEs), Autumn Royal, and Egnatia; 2) the GSEs effects on Caco2 colon cancer cell; 3) the effects of GSEs on the lipid composition and the fluidity of the cell membrane. These in vitro studies suggested that Autumn Royal and Egnatia contain high levels of polyphenols, possess antiproliferative activity on the Caco2 human colon carcinoma cell line and inhibit cell migration by acting on membrane fatty acids composition. Moreover, these results highlighted that the new grape variety Egnatia is an exciting source of phenolic compounds that could interest the food and pharmaceutical industries

    Gene expression signature induced by grape intake in healthy subjects reveals wide-spread beneficial effects on peripheral blood mononuclear cells

    Abstract Using a transcriptomic approach, we performed a pilot study in healthy subjects to evaluate the changes in gene expression induced by grape consumption. Blood from twenty subjects was collected at baseline (T0), after 21 days of grape-rich diet (T1) and after one-month washout (T2). Gene expression profiling of peripheral blood mononuclear cells from six subjects identified 930 differentially expressed transcripts. Gene functional analysis revealed changes (at T1 and/or T2) suggestive of antithrombotic and anti-inflammatory effects, confirming and extending previous finding on the same subjects. Moreover, we observed several other favourable changes in the transcription of genes involved in crucial processes such as immune response, DNA and protein repair, autophagy and mitochondrial biogenesis. Finally, we detected significant changes in many long non-coding RNAs genes, whose regulatory functions are being increasingly appreciated. Altogether, our data suggest that a grape diet may exert its beneficial effects by targeting different strategic pathways

    Evaluation of Anti-Candida Activity of Vitis vinifera L. Seed Extracts Obtained from Wine and Table Cultivars

    For the first time, grape seed extracts (GSEs), obtained from wine and table cultivars of Vitis vinifera L., cultured in experimental fields of Lazio and Puglia regions of Italy and grown in different agronomic conditions, have been tested on 43 Candida species strains.We demonstrated a significant correlation between the content of the flavan-3-ols in GSEs extracts, with a polymerization degree 654, and anti-Candida activity.Moreover, we demonstrated thatGSEs, obtained from plants cultured with reduced irrigation, showed a content of polymeric flavan-3-ols >250mg/gwith geometric mean MICvalues between 5.7 and 20.2 mg/L against Candida albicans reference strains. GSE, showing 573mg/g of polymeric flavan-3-ols, has been tested in an experimental murine model of vaginal candidiasis by using noninvasive in vivo imaging technique. The results pointed out a significant inhibition of Candida albicans load 5 days after challenge.These findings indicate that GSEs with high content of polymeric flavan-3-ols can be used in mucosal infection as vaginal candidiasis

    Antiproliferative and Apoptotic Effects Triggered by Grape Seed Extract (GSE) versus Epigallocatechin and Procyanidins on Colon Cancer Cell Lines

    Grape seed extract has been proven to exert anticancer effects on different tumors. These effects are mainly ascribed to catechin and procyanidin content. Analytical studies demonstrated that grape seed extract composition is complex and it is likely other components could exert biological activities. Using cell count and flow cytometry assays, we evaluated the cytostatic and apoptotic effects produced by three different grape seed extracts from Italia, Palieri and Red Globe cultivars, on Caco2 and HCT-8 colon cancer cells. These effects were compared to those induced by epigallocatechin and procyanidins, alone or in association, on the same cell lines. All the extracts induced growth inhibition and apoptosis in Caco2 and HCT-8 cells, along the intrinsic apoptotic pathway. On both cell lines, growth inhibition induced by Italia and Palieri grape seed extracts was significantly higher than that it has been recorded with epigallocatechin, procyanidins and their association. In Caco2 cells, the extract from Red Globe cultivar was less effective in inducing growth inhibition than procyanidins alone and in association with epigallocatechin, whereas, in HCT-8 cells, only the association of epigallocatechin and procyanidins triggers a significant proliferation decrease. On both cell lines, apoptosis induced by Italia, Palieri and Red Globe grape seed extracts was considerably higher than has been recorded with epigallocatechin, procyanidins and their association. These data support the hypothesis by which other compounds, present in the grape seed extracts, are likely to enhance the anticancer effects

    Analysis of high-identity segmental duplications in the grapevine genome

    <p>Abstract</p> <p>Background</p> <p>Segmental duplications (SDs) are blocks of genomic sequence of 1-200 kb that map to different loci in a genome and share a sequence identity > 90%. SDs show at the sequence level the same characteristics as other regions of the human genome: they contain both high-copy repeats and gene sequences. SDs play an important role in genome plasticity by creating new genes and modeling genome structure. Although data is plentiful for mammals, not much was known about the representation of SDs in plant genomes. In this regard, we performed a genome-wide analysis of high-identity SDs on the sequenced grapevine (<it>Vitis vinifera</it>) genome (PN40024).</p> <p>Results</p> <p>We demonstrate that recent SDs (> 94% identity and >= 10 kb in size) are a relevant component of the grapevine genome (85 Mb, 17% of the genome sequence). We detected mitochondrial and plastid DNA and genes (10% of gene annotation) in segmentally duplicated regions of the nuclear genome. In particular, the nine highest copy number genes have a copy in either or both organelle genomes. Further we showed that several duplicated genes take part in the biosynthesis of compounds involved in plant response to environmental stress.</p> <p>Conclusions</p> <p>These data show the great influence of SDs and organelle DNA transfers in modeling the <it>Vitis vinifera </it>nuclear DNA structure as well as the impact of SDs in contributing to the adaptive capacity of grapevine and the nutritional content of grape products through genome variation. This study represents a step forward in the full characterization of duplicated genes important for grapevine cultural needs and human health.</p
