    DESIGN, DEVELOPMENT AND CHARACTERIZATION OF PACLITAXEL LOADED SOLID LIPID NANOPARTICLES AS A COLLOIDAL DRUG CARRIER

    Objective: This study was aimed to design and characterize Paclitaxel-loaded Solid Lipid Nanoparticles (SLNs) to achieve site specificity,reduce toxicity and sustained release pattern. Methods: Paclitaxel-loaded solid lipid nanoparticles were fabricated by microemulsion followed by probe sonication technique using stearic acid as lipid and stabilized of the mixture of surfactants. In this study, 32 full factorial design was employed for optimizing the concentration of lipid as stearic acid and surfactant (soya lecithin) for the nanoparticles. The optimization was done by studying the dependent variable of particle size and % entrapment efficiency. Results: The results showed that the paclitaxel-loaded solid lipid nanoparticles prepared with the concentration of 33.31 % stearic acid and 500 mg of soya lecithin were optimum characteristic than other formulations. They showed the average particles size 149±4.10 nm and PDI 250±2.04. The zeta potential, % EE and % drug loading capacity was found to be respectively-29.7, 93.38±1.90 and 0.81±0.01. The optimized batch of Paclitaxel SLNs exhibited spherical shape with smooth surface analyzed by Transmission Electron Microscopy. In vitro study showed sustained release profile and was found to follow Higuchi Kinetics Equation. Conclusion: The SLNs of paclitaxel m et al. l the requirements of a colloidal drug delivery system. They had a particle size in nanosize; their size distribution was narrow and all the particles were in a spherical shape

    Colorimetric method for simultaneous estimation of amlodipine besylate from plasma

    Aim: The present work was to develop the method of analysis which can estimate drug in combined form without prior separation. Materials and method: By using UV spectroscopy colorimetric method was used for determination of Amlodipine besylate (AML) from plasma. Result and conclusion: This method is based on the formation of green colour in reaction between AML and 0.4 % Ferric chloride (FC) and 0.2 % Potassium ferricyanide (PF).The absorbance was measured at 775 nm. Result of tablet analysis showed % S.D. values in the range of 098.22 to 100,63%. Standard deviation value for tablet analysis by using methanol ranging from 98.01 to 101,13 % which proves the ability of the method to remain unaffected by small but deliberate change in reaction conditions and this method is used for estimation of AML from biological samples.Objetivo. El objetivo del presente estudio era desarrollar un método de análisis que permitiera estimar la cantidad de fármaco en forma combinada sin separación previa. Material y Método. Se utilizó espectroscopía colorimétrica UV para la determinación de Amlodipino Besilato (AML) plasmático Resultados. El presente método está basado en la formación de color verde en la reacción entre Amlodipino Besilato (AML) y cloruro férrico 0,4% y ferrocianuro potásico 0,2%. La medida de la absorbancia se realizó a 775nm. El resultado del análisis de los comprimidos mostró unos valores de DE comprendidos entre 098,22 y 100,63%. El valor de la DE utilizando metanol oscilan entre 98,01 y 101,13% lo que demuestra la capacidad del método de permanecer inalterado por pequeños pero intencionados cambios en las condiciones de la reacción, este método es usado para la estimación de Amlodipino Besilato (AML) en muestras biológicas

    Design and Development of SPM for Face Milling & Two Hole Boring of a CR-22 Block

    Abstract: Special purpose machine are widely used for special kind of operations, which are not economical on conventional machines. It is design for getting higher accuracy at desired condition. Face milling in industrial component is followed by a milling process. In the initial core bore is develop on the component with the help of required tool material. A Special Purpose Machine introduced here can be operated manually or automatically, as per the condition or a mechatronic based system can be developed that will work with different speed. A SPM can be developing with two separate units which perform milling & boring operation sequentially. SPM introduced here is automatically start operation by sensing work piece on table; it helps in start & end of cycle. Milling Machine performs first milling operation of face of 105 x 135mm size and then boring machine for ø41.30H 7 × 20mm deep cylinder block. In this the job is machined on two machines and two fixtures are used and operation is performed, hence it takes 10 min. to completion of one job. In this method Bore to face perpendicularity not maintained, so accuracy is low. The production rate is very low due to time required for two times loading and unloading of job, operational accuracy required. The manpower is required is more & skilled. In order to successfully come up with these problems, company decided to develop Special Purpose Machine. It will save production time and increase the production rate

    LIQUISOLID COMPACTS: A PROMISING APPROACH FOR SOLUBILITY ENHANCEMENT

    At present 40% of the drugs within the development pipelines, and approximately 60 % of the drugs coming directly from synthesis area unit poorly soluble.  Solubility is one of the important parameter to obtain desired concentration of drug in systemic circulation. Liquisolid compacts technique is a new and promising approach to overcome this consequence and that can change the dissolution rate of water insoluble drugs and increase the bioavailability of the drugs. This technique is an efficient method for formulating water insoluble and water soluble drugs. This technique relies upon the admixture of drug loaded solutions with applicable carrier and coating materials. Liquisolid system is characterized by flow behaviour, wettability, powder bed hydrophilicity, saturation solubility, drug content, differential scanning calorimetry, Fourier transform infra-red spectroscopy, powder X-ray diffraction, scanning electron microscopy, in-vitro release and in-vivo evaluation.     Impact of smartphone on mental health among medical undergraduates: A cross-sectional study

    BACKGROUND: Smartphones have become an indispensable part of almost everyone’s life. India has now become the leading and second-largest Smartphone market in the world. It has been noted that the purpose of Smartphone usage has exceptionally changed over the past few years, especially among students, professionals, and the common man. Smartphones have now become essential and the need of the hour, and medical students are no exception. Smartphones can make “smart students smarter”! There are now 10,275 unique applications labeled under the “Medical” and “Healthcare and Fitness” categories. The common medical applications used are MedCalc, Drug Infusion, Flashcards, Encyclopedia, Merck Manual, Medscape, PubMed, Epocrates, MedlinePlus, Lab test applications, Medical Dictionary, Eponyms etc. Despite the advantages and needs of smartphones, they have proven to be a source of potential hazard to human health, not only physical but also mental, social, and emotional well-being. There is consistent evidence for co-morbidity (such as obesity, heart diseases, neck and back pain, etc.) between excessive smartphone use and other psychiatric disorders, such as depression, anxiety, obsessive and compulsive disorder (OCD), and attention deficit hyperactivity disorder (ADHD) similar to internet addiction. The significant association of this addiction with poorer sleep quality and higher perceived stress has been a cause for concern. Hence, further investigation to explore the association between smartphone addiction and mental health, this study was undertaken. MATERIALS AND METHODS: Our study was undertaken in Dr VMGMC, Solapur, from June to August 2022, after obtaining approval from the ethical committee, approval number 172/22. Total voluntary participation for the study was 600 (from first to final year), and accordingly convenient sample size was taken. RESULTS: We found that out of the total participants, 42% of the participants had an average screen time of 4–6 h daily. A very small percentage of participants (4%) spent less than two hours in front of a screen. Alarmingly, 65% of the participants had an average screen time of more than 4 hours, which puts them at risk for the negative health impacts of prolonged screen time. Around 12% of them had symptoms of mild stress, 10.3% for mild anxiety, and 15.6% for mild depression. 10.6% had symptoms of moderate stress, 23.3% for moderate anxiety, and 16% for moderate depression. A small proportion of undergraduates, that is, 5%, 16%, and 11.6%, had symptoms of severe and extremely severe stress, anxiety, and depression, respectively. CONCLUSION: The study participants did not feel a lot of stress, anxiety, or depression symptoms when smartphones were used judiciously and mostly for non-social purposes (such as studying, listening to music, or watching videos). This investigation led us to the conclusion that there are some positive effects of smartphones on mental health. However, those who spent an excessive amount of time on their smartphones for social contact, with an average screen time of 5 h, showed signs of mild to moderate sadness, moderate anxiety, and tension, demonstrating that social media had a negative impact on the mental health of medical undergraduates. Therefore, efforts should be made to inform medical students about how using a smartphone is harming their mental health

    Brevibacterium spp. are aerobic, nonbranched, asporogenous, gram-positive, rod-shaped bacteria which may exhibit a rod-coccus cycle when cells get older and can be found in various environments. ​Several Brevibacterium species have industrial importance and are capable of biotransformation of various contaminants. Here we describe the draft genome sequence of Brevibacterium epidermidis EZ-K02 isolated from nitrocellulose-contaminated wastewater environments. The genome comprises 3,885,924 bp, with a G + C content of 64.2%. This whole genome shotgun project has been deposited at DDBJ/ENA/GenBank under the accession PDHL00000000.
