Analysis of expression of LGALS3BP gene in thyroid tissues and peripheral blood lymphocytes in patients with papillary thyroid cancer
Introduction: Galectin-3 (Gal-3), coding by LGALS3BP gene,
is a protein of the lectin family that has been associated with
neoplastic processes and seems to play an important role in
a variety of cell biological processes. In the thyroid gland,
the high expression of this protein has been described in
differentiated carcinomas, especially in papillary thyroid
cancer (PTC).
Aim: Analysis of Gal-3 protein expression in PTC and nodular
goiters; investigation of Gal-3 mRNA expression in
peripheral blood lymphocytes (PBL) in patients with PTC
and nodular goiter; analysis of correlation between Gal-3
protein expression in PTC and Gal-3 mRNA in PBL of the
same patient.
Material and methods: Gal-3 protein was evaluated by
immunohistochemisty in benign (27 multinodular goiters)
and malignant (30 papillary carcinomas) thyroid tissues and
galectin-3 mRNA expression by real-time PCR in peripheral
blood lymphocytes (PBL) from 90 patients with multinodular
goiter (n = 27), papillary carcinoma (n = 30) and
healthy controls (n = 33).
Results: In PTC we observed increased expression of Gal-3
protein (all 30 cases) in cytoplasm, nucleus and cell membranes of cancer cells. 23 of 27 benign thyroid nodular goiters
were negative for Gal-3 expression. In all examined
blood samples we observed higher LGALS3BP gene expression
than GAPDH (house keeping gene) with no difference
between both groups, without relation to the Gal-3 expression
in PTC.
Conclusions: There is no difference in Gal-3 expression in
peripheral blood lymphocytes in patients with papillary
thyroid cancer in relation to nodular goiter.Wstęp: Choroba nowotworowa wywołuje miejscowe i ogólnoustrojowe

Skomplikowane, trudne do śledzenia mechanizmy mobilizacji
komórek układu immunologicznego oraz wytwarzanie
swoistych klonów zwalczających komórki z obcym antygenem
to jedno z głównych zadań tego układu. Limfocyty
T i B odgrywają znaczącą rolę w procesie obrony
przeciwnowotworowej. Galektyna-3 (Gal-3), kodowana
przez gen LGALS3BP, jest białkiem należącym do grupy
lektyn, związanym z procesami nowotworzenia oraz odgrywającym
ważną rolę w wielu czynnościach życiowych
komórki. Jej zwiększoną ekspresję obserwuje się w wysoko
zróżnicowanych rakach tarczycy, a szczególnie w raku brodawkowatym
tarczycy (PTC, papillary thyroid cancer). Wykazano,
iż Gal-3 występuje w komórkach układu immunologicznego
aktywowanych antygenami nowotworu. Jak do
tej pory, nie przeprowadzono badań dotyczących analizy
ekspresji Gal-3 mRNA w limfocytach krwi obwodowej
u pacjentów z PTC.
Cel pracy: Analiza ekspresji białka Gal-3 w PTC oraz wolu
guzowatym, sprawdzenie, czy w limfocytach krwi obwodowej
pobranych od pacjentów z wolem guzowatym i PTC
występuje ekspresja mRNA Gal-3, jeżeli tak, czy występuje
jakakolwiek zależność pomiędzy ekspresją białka Gal-3
w tarczycy i mRNA Gal-3 w limfocytach krwi obwodowej
u tego samego pacjenta.
Materiał i metody: Przeprowadzono immunohistochemiczną
analizę bloczków parafinowych na obecność białka
Gal-3 w 27 zmianach łagodnych o charakterze wola guzkowego
obojętnego i 30 zmianach złośliwych (30 PTC). W drugiej
części pracy przeprowadzono badanie PCR w czasie
rzeczywistym dla mRNA Gal-3 w limfocytach krwi obwodowej
u 90 osób: 27 z wolem guzowatym obojętnym, 30 z PTC i 33 bez zmian w zakresie gruczołu tarczowego
(grupa kontrolna).
Wyniki: We wszystkich 30 próbkach z PTC stwierdzono
wysoką lub podwyższoną ekspresję białka Gal-3. W 23 na
27 analizowanych łagodnych zmian tarczycy - wola guzowate
- nie wykazano dodatniej reakcji z przeciwciałem
anty Gal-3. W drugiej części pracy u 30 pacjentów z PTC
i 27 z wolem guzkowym we wszystkich przypadkach obserwowano
w limfocytach krwi obwodowej zwiększoną
ekspresję genu LGALS3BP w stosunku do genu referencyjnego
GAPDH (gen metabolizmu podstawowego). Nie stwierdzono
zależności pomiędzy ekspresją genu LGALS3BP na
poziomie białka w komórkach PTC a jego ekspresją na poziomie
mRNA w limfocytach krwi obwodowej u tych samych
pacjentów. Wartości względnej ekspresji genu LGALS3BP
w odniesieniu do genu referencyjnego u chorych z rakiem
brodawkowatym tarczycy oraz u chorych z wolem guzowatym
nie różniły się między sobą w sposób istotny.
Wnioski: W limfocytach krwi obwodowej nie ma różnic
w ilości mRNA Gal-3 u chorych z PTC i zwykłym wolem
Polimorfizmy typu SNP w obrębie genu LGALS3BP u chorych z łagodnymi i złośliwymi nowotworami tarczycy
Introduction: The aim of the study was estimation of occurrence
of SNP (single nucleotide polymorphism) sites in
LGALS3BP gene in patients with benign and malignant thyroid
tumors and analysis of correlation between their frequency
and the histological type of thyroid lesions.
Material and methods: The studied group consisted
of 58 patients, 24 with papillary thyroid carcinomas, 19 with
nodular goiters and 15 with follicular adenomas. Control
group included 180 healthy volunteers. Four different SNP
polymorphisms were analysed in PCR products using the
SNaPshot test, on genetic analyser ABI Prism 310; these can
be found in NCBI database under rs numbers: 1803938
(a/g), 11024 (g/c), 1801463 (a/c) and 1131516 (c/t).
Results: In all patients analysis of SNP sites revealed: lack
of polymorphism in a/g rs 18039380; polymorphism identical
to database in g/c rs 11024 and polymorphism different
from database in a/g rs 1801463 and c/g rs 131516. There were no significant differences between patients with thyroid
lesions and group of healthy controls.
Conclusion: Single nucleotide polymorphism (SNP) of
LGALS3BP gene (found in NCBI) database are not characteristic
for papillary thyroid cancer, follicular adenomas and
nodular goiter.Wstęp: Badanie polimorfizmu genetycznego jest w ostatnich
latach coraz częściej wykorzystywane w ocenie predyspozycji
dziedzicznej do chorób nowotworowych i metabolicznych.
Gen LGALS3BP kodujący galektynę-3 (Gal-3),
białko ulegające nadekspresji w komórkach raka brodawkowatego
tarczycy (PTC, papillary thyroid carcinoma), posiada
w swym obrębie kilka miejsc polimorficznych typu SNP
(single nucleotide polymorphism). Poznanie ewentualnych zależności
pomiędzy zmianami patologicznymi w gruczole
tarczowym a polimorfizmem omawianego genu może mieć
znaczenie w badaniu patogenezy raka tarczycy.
Cel pracy: Ocena występowania wybranych miejsc polimorficznych
typu SNP genu LGALS3BP u pacjentów z łagodnymi
i złośliwymi guzami tarczycy oraz analiza korelacji pomiędzy
częstością występowania danej formy allelicznej
genu a typem histologicznym zmiany w obrębie gruczołu
tarczowego.
Materiał i metody: Badana grupa obejmowała 58 osób,
w tym 24 chorych z rakiem brodawkowatym, 19 chorych
z wolem guzowatym i 15 chorych z gruczolakiem pęcherzykowym.
Grupę kontrolną stanowiło, w zależności od
konkretnie badanego polimorfizmu, 130-180 osób zdrowych.
DNA izolowano z leukocytów krwi obwodowej
i z guzów tarczycy pacjentów oraz z krwi grupy kontrolnej
za pomocą zestawu "Sherlock" do izolacji DNA. Po dokonaniu
oceny ilościowej i jakościowej, DNA posłużył jako
matryca do amplifikacji in vitro wybranego fragmentu genu
za pomocą reakcji łańcuchowej polimerazy (PCR, polymerase
chain reaction). Produkty PCR analizowano w kierunku polimorfizmu
typu SNP techniką "SNaPshot", wykorzystując analizator genetyczny ABI Prism 310. Do porównania otrzymanych
sekwencji użyto programu SeqEval (Applied Biosystems).
Oceniono 4 miejsca polimorficzne typu SNP genu
LGALS3BP znalezione w bazie danych NCBI o numerach
rs: 1803938 (a/g), 11024 (g/c), 1801463 (a/c) i 1131516 (c/t).
W badanych grupach w analizowanych miejscach polimorficznych
stwierdzono: brak polimorfizmu (a/g rs 18039380),
polimorfizm zgodny z bazą danych (g/c rs 11024 ), a u pozostałych
niezgodny z bazą danych (a/g rs 1801463) i (c/g rs
1131516). Nie stwierdzono istotnych różnic w występowaniu
zbadanych polimorfizmów genu LGALS3BP pomiędzy
analizowanymi grupami.
Wnioski: Wykazane w bazie danych NCBI polimorfizmy
SNP genu LGALS3BP nie są charakterystyczne dla raka brodawkowatego
tarczycy, gruczolaka pęcherzykowego i wola
guzowatego
Influence of PPARγ agonists and polymorphism Pro12 Ala on blood pressure values in patients with type 2 diabetes. Initial coverage
Wstęp Receptor aktywowany przez proliferatory peroksysomów
γ (PPARγ, peroxisome proliferator-activated
receptor-γ) jest czynnikiem transkrypcyjnym
o szerokim spektrum działania. Zaburzenia jego ekspresji,
uwarunkowane wariantami polimorficznymi
genu, są nierzadko odpowiedzialne za otyłość, insulinooporność,
a także nadciśnienie tętnicze.
Celem pracy była ocena wpływu agonisty receptora
PPARγ (roziglitazon) na wartości ciśnienia tętniczego
u chorych na cukrzycę typu 2 w zależności od
polimorfizmu Pro12Ala receptora PPARγ.
Materiał i metody U 45 pacjentów z niewyrównaną
metabolicznie cukrzycą typu 2 i insulinoopornością,
leczonych pochodnymi sulfonylomocznika i metforminą,
dołączono roziglitazon w dawce 4 mg/d.
U wszystkich chorych oznaczono polimorfizm
Pro12Ala [SnaPshot; ABI PRISM310]. Przed i po
22-tygodniowej terapii roziglitazonem dokonano
oceny wartości ciśnienia tętniczego (średnia z 3 pomiarów).
Pozostałe badania wykonano za pomocą
rutynowych metod laboratoryjnych.
Wyniki U 12 osób ustalono genotyp Pro12Ala, a u 33
chorych genotyp Pro12Pro. Po zastosowanej terapii
stwierdzono istotne statystycznie obniżenie ciśnienia
skurczowego (przed leczeniem: 145,35 ± 20,25 mm Hg vs. po leczeniu: 139,40 ± 15,39 mm Hg; p < 0,045).
Składowa rozkurczowa również uległa obniżeniu,
jednak w stopniu nieistotnym statystycznie (przed
leczeniem: 86,37 ± 10,77 vs. po leczeniu: 83,73 ± 10,19 [mm Hg]; p = 0,170). Uwzględniając genotyp
badanych, obniżenie ciśnienia tętniczego obserwowano
jedynie u pacjentów posiadających jeden
zmutowany allel (p < 0,023). U pacjentów z genotypem
Pro12Pro nie wykazano istotnego statystycznie
zmniejszenia wartości ciśnienia tętniczego (p = 0,307).
Wnioski 1. Egzogenna stymulacja receptora PPARγ
przebiega z istotnym spadkiem skurczowych wartości
ciśnienia tętniczego. 2. Efekt hipotensyjny rozyglitazonu
dotyczy przede wszystkim chorych z genotypem
Pro12Ala.Background PPARγ is a transcriptor factor with wide spectrum
activity. Its expression disorders, connected with polymorphic
variants, are responsible for obesity, insulin
resitance and also hypertension.
The aim of this study was to assess the influence of PPARγ
agonist (roziglitazon) and polymorphism Pro12Ala of this
receptor on blood pressure values in type 2 diabetes
Material and methods In 45 patients, who were treated
sulfonylurea and metformin with inadequate metabolic control
and insulin resitance, rosiglitazone 4 mg per day was
administered. Polymorphism Pro12Ala was estimated
[SnaPshot; ABI PRISM310]. In 45 patients with type 2 diabetes
before and following 22 weeks of rosiglitazone therapy
blood pressure valuses were estimated (mean of three
measurments). Other tests were conducted using routine
lab tests.
Results A heterozygous genotyp Pro12Ala was found in 12
patients and and a homozygous genotyp Pro12Pro in 33
patients. Following rosiglitazone therapy a statistically significant
downward tendency of systolic blood pressure
valuses was noted (before: 145.35 ± 20.25 mm Hg vs. after:
139.40 ± 15.39 mm Hg; p < 0.045 ). A downward tendency
of diastolic blood pressure valuses was observed as
well, but it did not achieve significance level (before: 86.37
± 10.77 vs. after: 83.73 ± 10.19 [mm Hg]; p = 0.170).
Blood pressure reduction was noted only in patients with
genotype Pro12Ala PPARγ (p < 0.023). A statistically significant
downward tendency of blood pressure was not
noted in patients with Pro12Pro genotype (p = 0.307).
Conclusions 1. Exogenous stimulation of PPARγ receptor
is connected with statistically significant downward tendency
of systolic blood pressure values. 2. Hypotensive effect
of rosiglitazone appears first in all patients with
Pro12Ala genotype
Serotonin-Related Gene Variants in Patients with Irritable Bowel Syndrome and Depressive or Anxiety Disorders
Aim. To assess the association of six polymorphisms in serotonin-related genes with depressive or anxiety disorders in patients with irritable bowel syndrome (IBS). Methods. The lifetime prevalence of depressive and anxiety disorders was assessed in 95 IBS patients (85% women) using the Munich version of the Composite International Diagnostic Interview (CIDI). IBS was diagnosed according to the Rome III criteria. SCL6A4 HTTLPR polymorphism (rs4795541) was determined using PCR-based method. Single-nucleotide polymorphisms in HTR1A (rs6295), HTR2A (rs6313 and rs6311), HTR2C (rs6318), and TPH1 (rs1800532) were detected by minisequencing method. Results. IBS patients with depressive disorders were characterized by higher frequency of 5-HTTLPR L allele in comparison to IBS patients with anxiety disorders. The lower frequency of 1438 A allele in HTR2A was found in IBS patients with depressive disorders in comparison to IBS patients without mental disorders. The lower G allele frequency in HTR2C rs6318 polymorphism among IBS patients with anxiety disorders was also observed. Conclusions. Our results provide further evidence for the involvement of SLC6A4 rs4795541 and HTR2A rs6311 polymorphisms in the pathophysiology of depressive disorders in IBS patients. The new findings indicate that HTR2C rs6318 polymorphism may be associated with the susceptibility to anxiety disorders in IBS patients
Identification and characterization of novel rapidly mutating Y-chromosomal short tandem repeat markers
Short tandem repeat polymorphisms on the male‐specific part of the human
Y‐chromosome (Y‐STRs) are valuable tools in many areas of human genetics. Although
their paternal inheritance and moderate mutation rate (~10−3 mutations per marker
per meiosis) allow detecting paternal relationships, they typically fail to separate male
relatives. Previously, we identified 13 Y‐STR markers with untypically high mutation
rates (>10−2
), termed rapidly mutating (RM) Y‐STRs, and showed that they improved
male relative differentiation over standard Y‐STRs. By applying a newly developed in
silico search approach to the Y‐chromosome reference sequence, we identified
27 novel RM Y‐STR candidates. Genotyping them in 1,616 DNA‐confirmed father–son
pairs for mutation rate estimation empirically highlighted 12 novel RM Y‐STRs. Their
capacity to differentiate males related by 1, 2, and 3 meioses was 27%, 47%, and 61%,
respectively, while for all 25 currently known RM Y‐STRs, it was 44%, 69%, and 83%.

Comprehensive mutation analysis of 17 Y-chromosomal short tandem repeat polymorphisms included in the AmpFlSTR® Yfiler® PCR amplification kit
The Y-chromosomal short tandem repeat (Y-STR) polymorphisms included in the AmpFlSTR® Yfiler® polymerase chain reaction amplification kit have become widely used for forensic and evolutionary applications where a reliable knowledge on mutation properties is necessary for correct data interpretation. Therefore, we investigated the 17 Yfiler Y-STRs in 1,730–1,764 DNA-confirmed father–son pairs per locus and found 84 sequence-confirmed mutations among the 29,792 meiotic transfers covered. Of the 84 mutations, 83 (98.8%) were single-repeat changes and one (1.2%) was a double-repeat change (ratio, 1:0.01), as well as 43 (51.2%) were repeat gains and 41 (48.8%) repeat losses (ratio, 1:0.95). Medians from Bayesian estimation of locus-specific mutation rates ranged from 0.0003 for DYS448 to 0.0074 for DYS458, with a median rate across all 17 Y-STRs of 0.0025. The mean age (at the time of son’s birth) of fathers with mutations was with 34.40 (±11.63) years higher than that of fathers without ones at 30.32 (±10.22) years, a difference that is highly statistically significant (p < 0.001). A Poisson-based modeling revealed that the Y-STR mutation rate increased with increasing father’s age on a statistically significant level (α = 0.0294, 2.5% quantile = 0.0001). From combining our data with those previously published, considering all together 135,212 meiotic events and 331 mutations, we conclude for the Yfiler Y-STRs that (1) none had a mutation rate of >1%, 12 had mutation rates of >0.1% and four of <0.1%, (2) single-repeat changes were strongly favored over multiple-repeat ones for all loci but 1 and (3) considerable variation existed among loci in the ratio of repeat gains versus losses. Our finding of three Y-STR mutations in one father–son pair (and two pairs with two mutations each) has consequences for determining the threshold of allelic differences to conclude exclusion constellations in future applications of Y-STRs in paternity testing and pedigree analyses
A global analysis of Y-chromosomal haplotype diversity for 23 STR loci
In a worldwide collaborative effort, 19,630 Y-chromosomes were sampled from 129 different populations in 51 countries. These chromosomes were typed for 23 short-tandem repeat (STR) loci (DYS19, DYS389I, DYS389II, DYS390, DYS391, DYS392, DYS393, DYS385ab, DYS437, DYS438, DYS439, DYS448, DYS456, DYS458, DYS635, GATAH4, DYS481, DYS533, DYS549, DYS570, DYS576, and DYS643) and using the PowerPlex Y23 System (PPY23, Promega Corporation, Madison, WI). Locus-specific allelic spectra of these markers were determined and a consistently high level of allelic diversity was observed. A considerable number of null, duplicate and off-ladder alleles were revealed. Standard single-locus and haplotype-based parameters were calculated and compared between subsets of Y-STR markers established for forensic casework. The PPY23 marker set provides substantially stronger discriminatory power than other available kits but at the same time reveals the same general patterns of population structure as other marker sets. A strong correlation was observed between the number of Y-STRs included in a marker set and some of the forensic parameters under study. Interestingly a weak but consistent trend toward smaller genetic distances resulting from larger numbers of markers became apparent.Peer reviewe
Comparison of various decalcificators in preparation of DNA from human rib bone
Various calcium-binding buffers and agents in DNA preparation from fresh human rib bones were compared. We showed that the buffer described by Kuntze et al. [1996] yielded the highest quantaties of DNA, but the obtained DNA was strongly degraded. The best quality DNA was obtained using two buffers: of Hänni et al. [1994] and Ivanov et al. [1995]. Plans for the museum path of the Medical University in Wrocław
People have always been interested in discovering their past and seeking answers to their existential questions. Therefore institutions like museums, which collect exhibits related to the history and development of humans, have been known since antiquity. However, museology today has changed and is no longer confined to establishments related to popular fields like painting, sculpture and archaeology. Nowadays, there are also museums dedicated to medical sciences, which present the history of humanity in the sphere of its biological development. The Medical University in Wrocław can boast of several such establishments, although so far they have been treated as separate institutions. For this reason, the idea was created to combine them all into a single museum route, and to make it available to a broader visitor base. Thanks to this, people can view items from old and modern medicine practice located in many different sites, allowing them to deepen their knowledge and expand their interests. To make it easier for visitors to navigate through such a diverse complex, a special museum path has been created, along with a description of each location
