Visualization of DNA G-quadruplexes in herpes simplex virus 1-infected cells
We have previously shown that clusters of guanine quadruplex (G4) structures can form in the human herpes simplex-1 (HSV-1) genome. Here we used immunofluorescence and immune-electron microscopy with a G4-specific monoclonal antibody to visualize G4 structures in HSV-1 infected cells. We found that G4 formation and localization within the cells was virus cycle dependent: viral G4s peaked at the time of viral DNA replication in the cell nucleus, moved to the nuclear membrane at the time of virus nuclear egress and were later found in HSV-1 immature virions released from the cell nucleus. Colocalization of G4s with ICP8, a viral DNA processing protein, was observed in viral replication compartments. G4s were lost upon treatment with DNAse and inhibitors of HSV-1 DNA replication. The notable increase in G4s upon HSV-1 infection suggests a key role of these structures in the HSV-1 biology and indicates new targets to control both the lytic and latent infection
Pleomorphic adenoma rehabilitative treatment in growing up patient: a 20-years follow-up
OBJECTIVE: Although tumors of minor salivary glands are rare, the pleomorphic adenoma is the most common pathology among the benign neoplasm and can be found with high prevalence in the junction between hard palate and soft palate. Most of the maxillary tumors are surgically treated through either a total or partial maxillectomy. However, surgical defects lead to both clinical and psychologic disorders for the patient. A postoperative obturator prosthesis is a good option in patients who underwent maxillectomy. It allows to restore both masticatory and speaking functions, as well as aesthetic appearance. When reconstruction of the surgical site is possible, an implant-supported prosthesis can be considered to guarantee a better function and aesthetic's rehabilitation. CASE REPORT: This clinical report presents the prosthetic rehabilitation of a patient who underwent maxillectomy because of a pleomorphic adenoma of hard palate minor salivary glands. The patient was treated with a palatal obturator prosthesis first and with an implant-supported prosthesis after surgical site's reconstruction and complete healing. CONCLUSIONS: The rehabilitation of the patient after maxillectomy through both these devices was an excellent option and provided clinical benefits, improving the patient's quality of life, allowing the patient's reinsertion into societ
Immediate implant loading: a comparison of trabecular metal and tapered screw-vent dental implants
Aims: The aim of the present study is to compare osteointegration and marginal bone loss of immediately loaded Trabecular
Metal® and Tapered Screw-Vent® Dental Implants (Zimmer Dental Inc., Carlsbad, CA, USA).
Methods: Eighty-seven (87) patients were selected and randomly divided into Group A and Group B. Twenty-six (26) patients
were enrolled in Group A, and were rehabilitated using Zimmer Trabecular Metal Dental Implants®. Sixty-one (61) patients were
enrolled in Group B, and were rehabilitated using Zimmer Tapered Screw-Vent Dental Implants®.
Results: The mean value of marginal bone loss after one year was 0.44 ± 0.40 mm for Group A and 0.95 ± 0.62 mm for Group B
(p<.003). Mean marginal bone loss after 18 months was 0.46 ± 0.42 mm for group A and 0.97 ± 0.65 mm for group B (p<.003). No
TM implant was lost (Group A), whereas one TSV implant (Group B) was lost before osseointegration and was not included in the
statistical analysis.
Conclusion: Both Trabecular Metal and Tapered Screw-Vent dental implants showed satisfying levels of osteointegration and
marginal bone loss; however, statistical analysis revealed a value significantly lower of marginal bone loss for TM. Thus, it may be
deduced that when implants are immediately loaded, the average loss of marginal bone around the TM implants is lower than that of
the Tapered Screw-Vent implants

The Drosophila Citrate Lyase Is Required for Cell Division during Spermatogenesis
The Drosophila melanogaster DmATPCL gene encodes for the human ATP Citrate Lyase (ACL) ortholog, a metabolic enzyme that from citrate generates glucose-derived Acetyl-CoA, which fuels central biochemical reactions such as the synthesis of fatty acids, cholesterol and acetylcholine, and the acetylation of proteins and histones. We had previously reported that, although loss of Drosophila ATPCL reduced levels of Acetyl-CoA, unlike its human counterpart, it does not affect global histone acetylation and gene expression, suggesting that its role in histone acetylation is either partially redundant in Drosophila or compensated by alternative pathways. Here, we describe that depletion of DmATPCL affects spindle organization, cytokinesis, and fusome assembly during male meiosis, revealing an unanticipated role for DmATPCL during spermatogenesis. We also show that DmATPCL mutant meiotic phenotype is in part caused by a reduction of fatty acids, but not of triglycerides or cholesterol, indicating that DmATPCL-derived Acetyl-CoA is predominantly devoted to the biosynthesis of fatty acids during spermatogenesis. Collectively, our results unveil for the first time an involvement for DmATPCL in the regulation of meiotic cell division, which is likely conserved in human cells
Since 2015, the first solitary bee hotels in Sicily located in the small island of Isola delle Femmine Nature Reserve, Sicily (Italy)
Natural pollinators, mainly represented by solitary apoidea, play an essential role in ecosystems
and biodiversity. Their population has sharply declined recently, putting both flora and fauna
biodiversity at risk. Many studies report that artificial nesting for reproducing solitary bees,
also known as bee hotels, can support conservation and increase the number of pollinators.
Unipa and LIPU Isola delle Femmine Nature Reserve wanted to create the first bee hotels in
2015 near the coast. In particular, the goal of this project was to protect and increase biodiversity on the islet of Isola delle Femmine, a peculiar and challenging to-balance environment
where there are more than 200 botanical taxa present on the island, some of which are endemic
to Sicilian north-western coast, including pollination by insects. In the current state of knowledge, it is the first case of installing bee hotels in a coastal area so close to the sea, which entailed a significant technical challenge in the construction using recycled material. The hotels
built were wholly colonized in a very short period, and even when maintenance and replacement of materials were carried out in less than a week, the recolonization was rapid. Most individuals belonged to the Megachilidae family, which is characteristic of the Mediterranean.
The bee hotels have also been a source of curiosity and insight for the reserve visitors. In conclusion, the bee hotel proved useful for colonizing pollinators and increasing reserve use
Mouthwash Based on Ozonated Olive Oil in Caries Prevention: A Preliminary In-Vitro Study
(1) Background: Ozone (O3) proved to oxidize organic and inorganic compounds, and its efficacy against bacteria, viruses and fungi plasma membranes was of interest. Ozone vehicle can be a gaseous form, ozonated water or ozonized oil. The aim of this in-vitro study was to evaluate the efficacy of ozonated olive oil against Streptococcus mutans. (2) Methods: Two different commercial mouthwashes were tested: Ialozon Blu (IB) (Gemavip, Cagliari, Italy), with ozonated olive oil, and Ialozon Rose (IR) (Gemavip, Cagliari, Italy), with ozonated olive oil, hyaluronic acid and vitamin E. All formulates were analyzed in a dilution range from 2- to 256-folds in saline solution, as to reproduce the salivary dilution. Streptococcus mutans CIP103220 strain was used for the antimicrobial susceptibility test, and the Kirby-Bauer inhibition method was performed to evaluate the Minimum Inhibitory (MIC), Minimum Bactericidal (MBC), and Minimum Biofilm Inhibitory Concentration (MBIC). (3) Results: Both formulates showed the same antimicrobial activity. MIC, MBC, and MBIC were observed for dilution factors of 1/32, 1/8 and 1/8, respectively. The mean value of inhibition zone diameter was 16.5 mm for IB, and 18 mm for IR. (4) Conclusions: The results suggested that ozonized olive oil formulates were able to inactivate Streptococcus mutans avoiding the salivary dilution effect in the oral cavity
