Cytokinin Acts through the Auxin Influx Carrier AUX1 to Regulate Cell Elongation in the Root
Hormonal interactions are crucial for plant development. In Arabidopsis, cytokinins inhibit root growth through effects on cell proliferation and cell elongation. Here, we define key mechanistic elements in a regulatory network by which cytokinin inhibits root cell elongation in concert with the hormones auxin and ethylene. The auxin importer AUX1 functions as a positive regulator of cytokinin responses in the root; mutation of AUX1 specifically affects the ability of cytokinin to inhibit cell elongation but not cell proliferation. AUX1 is required for cytokinin-dependent changes of auxin activity in the lateral root cap associated with the control of cell elongation. Cytokinin regulates root cell elongation through ethylene-dependent and -independent mechanisms, both hormonal signals converging on AUX1 as a regulatory hub. An autoregulatory circuit is identified involving the control of ARR10 and AUX1 expression by cytokinin and auxin, this circuit potentially functioning as an oscillator to integrate the effects of these two hormones. Taken together, our results uncover several regulatory circuits controlling interactions of cytokinin with auxin and ethylene, and support a model in which cytokinin regulates shootward auxin transport to control cell elongation and root growth
Experiments in quasi-static manipulation of an elastic rod
The purpose of this dissertation is to experimentally validate a new approach to robotic manipulation of deformable objects. As a case study, it will focus on the manipulation of objects that can be modeled as Kirchhoff elastic rods, for example a metal wire that is held at each end by robotic grippers. Any curve traced by this wire when in static equilibrium can be described as the solution to an optimal control problem with boundary conditions that vary with the position and orientation of each gripper. Recent work has shown that the set of all local solutions to this problem over all possible boundary conditions is a smooth manifold of finite dimension that can be parameterized by a single chart, the coordinates for which have a direct interpretation as forces and torques. These coordinates-in principle-allow the problem of manipulation planning to be formulated as finding a path of the wire through its set of equilibrium configurations, something that was previously thought impossible and that has significant advantages. However, this approach has never before been applied to hardware experiments.
We begin by considering a metal wire that is confined to a planar workspace. We derive global coordinates for this wire and characterize the extent to which they accurately describe its shape during robotic manipulation. In particular, we show that differences between predicted and observed manipulation (which can be quite large) derive primarily from small errors in the position and orientation of each robotic gripper. We reduce these differences in two ways. First, we give an algorithm for manipulation planning that locally minimizes sensitivity to errors in gripper placement. Second, we give a feedback control policy (based on force sensor data as well as on position and orientation estimates) that locally minimizes the sum-squared error between planned and observed paths in our global coordinate chart for the wire.
We conclude by showing-again, with hardware experiments-that these results extend directly to enable robotic manipulation of a metal wire in a three-dimensional workspace
Virtualizing the access network via open APIs
Residential broadband consumption is growing rapidly, in-creasing the gap between ISP costs and revenues. Mean-while, proliferation of Internet-enabled devices is congesting access networks, frustrating end-users and content providers. We propose that ISPs virtualize access infrastructure, using open APIs supported through SDN, to enable dynamic and controlled sharing amongst user streams. Content providers can programmatically provision capacity to user devices to ensure quality of experience, users can match the degree of virtualization to their usage pattern, and ISPs can real-ize per-stream revenues by slicing their network resources. Using video streaming and bulk transfers as examples, we develop an architecture that specifies the interfaces between the ISP, content provider, and user. We propose an algo-rithm for optimally allocating network resources, leveraging bulk transfer time elasticity and access path space diver-sity. Simulations using real traces show that virtualization can reduce video degradation by over 50%, for little extra bulk transfer delay. Lastly, we prototype our system and validate it in a test-bed with real video streaming and file transfers. Our proposal is a first step towards the long-term goal of realizing open and agile access network service quality management that is acceptable to users, ISPs and content providers alike
The effect of coating characteristics on implant-bone interface mechanics
Successful osseointegration of press-fit implants depends on the initial stability, often measured by the micromotions between the implant and bone. A good primary stability can be achieved by optimizing the compressive and frictional forces acting at the bone-implant interface. The frictional properties of the implant-bone interface, which depend on the roughness and porosity of the implant surface coating, can affect the primary stability. Several reversible (elastic) and non-reversible (permanent) deformation processes take place during frictional loading of the implant-bone interface. In case of a rough coating, the asperities of the implant surface are compressed into the bone leading to mechanical interlocking. To optimize fixation of orthopaedic implants it is crucial to understand these complex interactions between coating and bone. The objective of the current study was to gain more insight into the reversible and non-reversible processes acting at the implant-bone interface. Tribological experiments were performed with two types of porous coatings against human cadaveric bone. The results indicated that the coefficient of friction depended on the coating roughness (0.86, 0.95, and 0.45 for an Ra roughness of 41.2, 53.0, and a polished surface, respectively). Larger elastic and permanent displacements were found for the rougher coating, resulting in a lower interface stiffness. The experiments furthermore revealed that relative displacements of up to 35 µm can occur without sliding at the interface. These findings have implications for micromotion thresholds that currently are assumed for osseointegration, and suggest that bone ingrowth actually occurs in the absence of relative sliding at the implant-bone interface

Palliative care services in families of males with muscular dystrophy: Data from MD STARnet
Introduction: Information on use of palliative care services among individuals with Duchenne and Becker muscular dystrophy is scant despite the clearly documented need. Methods: We examined associations between uptake of palliative care services by 233 males with Duchenne and Becker muscular dystrophy aged 12 and older for both caregiver and affected male characteristics using the Muscular Dystrophy Surveillance Tracking and Research Network baseline interview. Results: Ninety-one percent of caregivers (213/233) used at least one palliative care service. Case management had the highest frequency of use (59%). Use of palliative care was more frequently associated with the characteristics of affected males, as were some individual palliative care services. Utilization of six individual services differed among Muscular Dystrophy Surveillance Tracking and Research Network sites. While research suggests that pain is a frequent problem in Duchenne and Becker muscular dystrophy, only 12.5% reported use of pain management services. Discussion: Although palliative care use among families of males with Duchenne and Becker muscular dystrophy is high overall, there is much variability in use of individual services. Use of palliative care is driven by disease experience in the affected male.
