    Vibrational Analysis of Engine Components Using Neural-Net Processing and Electronic Holography

    The use of computational-model trained artificial neural networks to acquire damage specific information from electronic holograms is discussed. A neural network is trained to transform two time-average holograms into a pattern related to the bending-induced-strain distribution of the vibrating component. The bending distribution is very sensitive to component damage unlike the characteristic fringe pattern or the displacement amplitude distribution. The neural network processor is fast for real-time visualization of damage. The two-hologram limit makes the processor more robust to speckle pattern decorrelation. Undamaged and cracked cantilever plates serve as effective objects for testing the combination of electronic holography and neural-net processing. The requirements are discussed for using finite-element-model trained neural networks for field inspections of engine components. The paper specifically discusses neural-network fringe pattern analysis in the presence of the laser speckle effect and the performances of two limiting cases of the neural-net architecture

    Neural Networks Used to Compare Designed and Measured Time-Average Patterns

    Electronic time-average holograms are convenient for comparing the measured vibration modes of fan blades with those calculated by finite-element models. At the NASA Lewis Research Center, neural networks recently were trained to perform what had been a simple visual comparison of the predictions of the design models with the measurements. Finite-element models were used to train neural networks to recognize damage and strain information encoded in subtle changes in the time-average patterns of cantilevers. But the design-grade finite element models were unable to train the neural networks to detect damage in complex blade shapes. The design-model-generated patterns simply did not agree well enough with the measured patterns. Instead, hybrid-training records, with measured time-average patterns as the input and model-generated strain information as the output, were used to effect successful training

    Calcium-Sensing Receptor Genotype and Response to Cinacalcet in Patients Undergoing Hemodialysis

    BACKGROUND AND OBJECTIVES: We tested the hypothesis that single nucleotide polymorphisms (SNPs) in the calcium-sensing receptor (CASR) alter the response to the calcimimetic cinacalcet. DESIGN, SETTING, PARTICIPANTS, & MEASUREMENTS: We analyzed DNA samples in the Evaluation of Cinacalcet HCl Therapy to Lower Cardiovascular Events (EVOLVE) trial, a randomized trial comparing cinacalcet to placebo on a background of usual care. Of the 3883 patients randomized, 1919 (49%) consented to DNA collection, and samples from 1852 participants were genotyped for 18 CASR polymorphisms. The European ancestry (EA; n=1067) and African ancestry (AfAn; n=405) groups were assessed separately. SNPs in CASR were tested for their association with biochemical measures of mineral metabolism at baseline, percent change from baseline to 20 weeks, and risk of clinical fracture as dependent variables. RESULTS: There were modest associations of CASR SNPs with increased baseline serum parathyroid hormone and bone alkaline phosphatase primarily with the minor allele in the EA group (all P≀0.03), but not in the AfAn sample. In contrast, there was a modest association of decreased baseline serum calcium and FGF23 with CASR SNPs (P=0.04) primarily with the minor allele in the AfAn but not in the EA sample. The minor allele of two SNPs was associated with decreased percent reduction in parathyroid hormone from baseline to 20 weeks in the EA population (P<0.04) and this was not altered with cinacalcet. In both EA and AfAn, the same SNP (rs9740) was associated with decreased calcium with cinacalcet treatment (EA and AfAn P≀0.03). Three SNPs in high linkage disequilibrium were associated with a higher risk of clinical fracture that was attenuated by cinacalcet treatment in the EA sample (P<0.04). CONCLUSIONS: These modest associations, if validated, may provide explanations for differences in CKD-mineral bone disorder observed in EA and AfAn populations, and for differential biochemical responses to calcimimetics

    An international cohort comparison of size effects on job growth

    The contribution of different-sized businesses to job creation continues to attract policymakers’ attention; however, it has recently been recognised that conclusions about size were confounded with the effect of age. We probe the role of size, controlling for age, by comparing the cohorts of firms born in 1998 over their first decade of life, using variation across half a dozen northern European countries Austria, Finland, Germany, Norway, Sweden and the UK to pin down size effects. We find that a very small proportion of the smallest firms play a crucial role in accounting for cross-country differences in job growth. A closer analysis reveals that the initial size distribution and survival rates do not seem to explain job growth differences between countries, rather it is a small number of rapidly growing firms that are driving this result

    The James Webb Space Telescope Mission

    Twenty-six years ago a small committee report, building on earlier studies, expounded a compelling and poetic vision for the future of astronomy, calling for an infrared-optimized space telescope with an aperture of at least 4m4m. With the support of their governments in the US, Europe, and Canada, 20,000 people realized that vision as the 6.5m6.5m James Webb Space Telescope. A generation of astronomers will celebrate their accomplishments for the life of the mission, potentially as long as 20 years, and beyond. This report and the scientific discoveries that follow are extended thank-you notes to the 20,000 team members. The telescope is working perfectly, with much better image quality than expected. In this and accompanying papers, we give a brief history, describe the observatory, outline its objectives and current observing program, and discuss the inventions and people who made it possible.     A stepwise approach for the management of poisoning with extracorporeal treatments

    The use of an extracorporeal treatment (ECTR) in a poisoned patient may be life-saving in a limited number of scenarios. The decision-processes surrounding the use of ECTR in poisoning is complex: most nephrologists are not trained to assess a poisoned patient while clinical toxicologists rarely prescribe ECTRs. Deciding on which ECTR is most appropriate for a poison requires a good understanding of the poison's physicochemical and pharmacokinetic properties. Further, a detailed understanding of the capabilities and limitations of the different ECTRs can be useful to select the most appropriate ECTR for a given clinical situation. This manuscript provides a stepwise approach to assess the usefulness of ECTRs in poisoning

    Effect of 1‑Substitution on Tetrahydroisoquinolines as Selective Antagonists for the Orexin‑1 Receptor

    Selective blockade of the orexin-1 receptor (OX<sub>1</sub>) has been suggested as a potential approach to drug addiction therapy because of its role in modulating the brain’s reward system. We have recently reported a series of tetrahydroisoquinoline-based OX<sub>1</sub> selective antagonists. Aimed at elucidating structure–activity relationship requirements in other regions of the molecule and further enhancing OX<sub>1</sub> potency and selectivity, we have designed and synthesized a series of analogues bearing a variety of substituents at the 1-position of the tetrahydroisoquinoline. The results show that an optimally substituted benzyl group is required for activity at the OX<sub>1</sub> receptor. Several compounds with improved potency and/or selectivity have been identified. When combined with structural modifications that were previously found to improve selectivity, we have identified compound <b>73</b> (RTIOX-251) with an apparent dissociation constant (<i>K</i><sub>e</sub>) of 16.1 nM at the OX<sub>1</sub> receptor and >620-fold selectivity over the OX<sub>2</sub> receptor. In vivo, compound <b>73</b> was shown to block the development of locomotor sensitization to cocaine in rats

    Neurotensin receptor type 2 (NTS2) compounds display analgesic activity in animal pain models. We have identified the first high-affinity NTS2-selective antagonist (<b>8</b>) that is active in vivo. This study also revealed that the NTS2 FLIPR assay designation for a compound, agonist, partial agonist, and so forth, did not correlate with its in vivo activity as observed in the thermal tail-flick acute model of pain. This suggests that calcium mobilization is not the signaling pathway involved in NTS2-mediated analgesia as assessed by the thermal tail-flick model. Finally, we found a significant bias between rat and human for compound <b>9</b> in the NTS2 binding assay
