Asexuality: Classification and characterization

In a small qualitative study (N = 4), individuals who self-identified as asexual were interviewed to help formulate hypotheses for a larger study. The second larger study was an online survey drawn from a convenience sample designed to better characterize asexuality and to test predictors of asexual identity. A convenience sample of 1,146 individuals (N = 41 self-identified asexual) completed online questionnaires assessing sexual history, sexual inhibition and excitation, sexual desire, and an open-response questionnaire concerning asexual identity. Asexuals reported significantly less desire for sex with a partner, lower sexual arousability, and lower sexual excitation but did not differ consistently from non-asexuals in their sexual inhibition scores or their desire to masturbate. Content analyses supported the idea that low sexual desire is the primary feature predicting asexual identity
Diploastrea heliopora Sr/Ca and δ18O records from northeast Luzon, Philippines : an assessment of interspecies coral proxy calibrations and climate controls of sea surface temperature and salinity

The definitive version was published in Paleoceanography 32 (2017): 424–438, doi:10.1002/2017PA003098.The Indo-Pacific coral Diploastrea heliopora reveals regional multidecadal- to centennial- scale climate variability using coral carbonate δ18O (δ18Oc) as a combined proxy for sea surface temperature (SST) and sea surface salinity (SSS). However, to assess the coral's full potential in resolving climatic events, an independent SST proxy would be more advantageous. We examined both Sr/Ca and δ18O of Diploastrea against an adjacent Porites lobata core collected from northeast Luzon, Philippines. Winter Sr/Ca data from Diploastrea show a significant correlation to SST (r = −0.41, p < 0.05, (root-mean-square of the residual) RMSR = 0.81°C) and provide a proxy with similar sensitivity as Porites (r = −0.57, p < 0.05, RMSR = 0.62°C). An interspecies SST record is shown to be robust and used for a reconstruction of the Pacific Decadal Oscillation during boreal winter (r = −0.70, p = 0.02). While we were unable to generate a robust Diploastrea δ18O-SSS calibration at interannual timescale, the freshening trend toward the present, commonly observed in the region, is qualitatively captured in Diploastrea δ18O. Comparison with Porites δ18O and instrumental SSS records shows that the magnitude of freshening is consistent between coral species. Wet and dry season Porites δ18O provide support for the relative influence of El Niño–Southern Oscillation events and local precipitation to SSS variability at our site. The multiproxy, multispecies approach of this study further strengthens the evidence for Diploastrea as an alternate climate archive in the Indo-Pacific region and seals its potential in helping resolve less understood global-scale climate phenomena.National Research Foundation Singapore (NRF) Grant Number: NRF-RF2012-0
Testosterone Is Associated with Erectile Dysfunction: A Cross-Sectional Study in Chinese Men
Testosterone is essential for the regulation of erectile physiology, but the relationship between low testosterone and erectile dysfunction (ED) has not been firmly established.To examine the association between serum total, free and bio-available testosterone and ED in a population-based sample.A consecutive series of 1776 men aged 20–77 participated in the routine physical examination from September 2009 to December 2009 in Guangxi, China. ED was assessed using the five-item International Index of Erectile Function (IIEF-5) questionnaire. Total testosterone (TT), sex hormone binding globulin (SHBG) and other biochemical profiles were measured. Free testosterone (FT) and bio-available testosterone (BT) were calculated based on Vermeulen’s formula. Data were collected with regard to smoking, alcoholic drinking, physical activity and metabolic syndrome.The prevalence of ED (IIEF-5<22) was 47.6%. Men with ED were significantly older, and more prone to smoke cigarettes (≥20 cigarettes/day) or drink alcohol (≥3 drinks/week), and more likely to have elevated blood pressure (P = 0.036) or hyperglycemia (P<0.001) compared with those without ED. The significant increase in SHBG with age was parallel to its increase with increasing severity of ED (P<0.001). The obscure increase in TT across the ED status was detected without significance (P = 0.418), but TT was positively associated with ED after adjustment for age [odds ratio (OR) = 1.02, 95% CI (confidence internal): 1.00–1.04]. FT and BT were inversely associated with ED (OR = 0.14, 95%CI: 0.06–0.33; OR = 0.92 (95%CI: 0.89–0.96, respectively) in the univariate analysis, and this inverse association appeared to be independent of smoking status, alcoholic drinking, physical activity, hyper-triglyceridemia and hyperglycemia.FT and BT are inversely related to worsening ED, whereas the positive association between TT and ED is most likely due to the increase in SHBG
Cloning whole bacterial genomes in yeast
Most microbes have not been cultured, and many of those that are cultivatable are difficult, dangerous or expensive to propagate or are genetically intractable. Routine cloning of large genome fractions or whole genomes from these organisms would significantly enhance their discovery and genetic and functional characterization. Here we report the cloning of whole bacterial genomes in the yeast Saccharomyces cerevisiae as single-DNA molecules. We cloned the genomes of Mycoplasma genitalium (0.6 Mb), M. pneumoniae (0.8 Mb) and M. mycoides subspecies capri (1.1 Mb) as yeast circular centromeric plasmids. These genomes appear to be stably maintained in a host that has efficient, well-established methods for DNA manipulation
Simulating reactive nitrogen, carbon monoxide, and ozone in California during ARCTAS-CARB 2008 with high wildfire activity
Predictions of O_3, CO, total NO_y and individual NO_y species (NO, NO_2, HNO_3, PAN, alkyl nitrates and aerosol nitrate) from a fine resolution regional air quality modeling system for the South Coast Air Basin (SoCAB) and San Joaquin Valley Air Basin (SJVAB) of California are presented and evaluated for the 2008 ARCTAS-CARB campaign. The measurements of the chemical compounds from the fire plumes during the field campaign allow for the evaluation of the model's ability to simulate fire-influenced air masses as well. In general, the model successfully simulated the broad spatial distribution of chemical compounds in both air basins as well as the variation within the basins. Using inventories that reflect 2008 emissions levels, the model performed well in simulating NO_x (NO + NO_2) in SoCAB. Therefore, the under prediction of O_3 over these areas is more likely caused by uncertainties with the VOC emissions, chemistry, or discrepancies in the meteorology. The model did not capture the relatively high levels of O_3, and some reactive nitrogen species that were measured off shore of the SoCAB, indicating potential missing sources or the transport from on shore to off shore was not successfully captured. In SJVAB, the model had good performance in simulating different chemical compounds in the Fresno and Arvin areas. However, enhanced concentrations of O_3, NO_x, HNO_3 and PAN near dairy farms were significantly underestimated in the model. Negative biases also exist for O_3 and HNO_3 near oil fields, suggesting larger uncertainties associated with these emission sources. While the model simulated the total NO_y mixing ratios reasonably well, the prediction for partitioning between individual compounds showed larger uncertainties in the model simulation. Although the fire emissions inventory was updated to include the latest emissions estimates and speciation profiles, our model shows limited improvement in simulating the enhancement of O_3, CO, and PAN under fire impact as compared to a previous version of the modeling system. Further improvements in simulating fire emissions, especially the timing and the plume injection heights, are desired in order to better simulate the impact of fires
The role of ear mites of the genus Raillietia (Acari: Raillietiidae) in otitis of domestic ruminants
This paper presents a hypothesis on the involvement of species of the genus Raillietia Trouessart in the pathology of parasitic otitis in domestic ruminants. The chelicerae and claws of the tarsi are structures that contribute significantly towards producing primary lesions. The movable digits of the chelicerae accidentally injure the epithelium that lines the ear canal, while the claws of the tarsi perforate the epithelium, allowing the mites to affix themselves while feeding. The lesions in the epithelium caused by the chelicerae and the claws favor multiplication of the bacteria that inhabit the ear canal, resulting in typical cases of otitis
Associação entre Mycoplasma spp. e ácaros do conduto auditivo de bovinos
Esse estudo foi realizado com o objetivo de verificar a associação entre micoplasmas e ácaros (Raillietia auris e R. flechtmanni) no conduto auditivo de bovinos. Foram realizadas lavagens no conduto auditivo externo de 60 bovinos abatidos no Estado do Rio de Janeiro, Brasil. Para a lavagem dos condutos auditivos foi utilizada solução salina tamponada (PBS, pH 7.2) em seringas estéreis de 60mL. Para o isolamento de micoplasmas foram utilizados pools de ácaros por animal, lavados sucessivamente em 1mL de meio Hayflick modificado. Os lavados dos ácaros foram diluídos de 10-1 até 10-5 e repicados em meio Hayflick modificado, sólido e líquido e incubados a 37°C por 48-72 horas em microaerofilia. A identificação das espécies de micoplasmas foi realizada pelo teste da imunoperoxidase indireta (IPI). Verificou-se alta prevalência de ácaros do gênero Raillietia spp. 76,7% (46/60). O parasitismo por ácaros e micoplasmas foi verificado em 40 animais (74,1%), sendo esta associação significativa (p<0,001). Dos ácaros processados para isolamento de micoplasmas, 193 foram fêmeas e 25 machos. A frequência de Mycoplasma em Raillietia spp. foi de 81,2% (177/218) (p<0.001). Das fêmeas identificadas, 52,3% (101/193) foram R. auris e 47,7% (92/193) R. flechtmanni. A frequência de Mycoplasma nas fêmeas de R. auris foi de 75,2% (76/101) e na espécie R. flechtmanni foi de 88% (81/92) (P<0.05). As espécies de micoplasmas tipificadas pela IPI nos ácaros Raillietia auris foram: M. alkalescens 6,9%, M. arginini 3,4%, M. bovirhinis 9,2%, M. conjunctivae 18,4%, M. mycoides mycoides LC 8,0%, M. capricolum 5,7%. Em R. flechtmanni as espécies de micoplasmas identificadas foram: M. alkalescens 12,2%, M. arginini 1,0%, M. bovirhinis 18,9%, M. bovis 2,2%, M. conjunctivae 21,0%, M. mycoides mycoides LC 11,0% e M. capricolum 4,4%. As espécies de micoplasmas identificadas no conduto auditivo externo dos bovinos foram as mesmas presentes nos ácaros R. auris e R. flechtmanni. Os resultados confirmam que o conduto auditivo externo de bovinos é um habitat de Mycoplasma spp., incluindo espécies potencialmente patogênicas para os rebanhos, além dos ácaros R. auris e R. flechtmanni estarem associados com esses molicutes carreando-os em seu organismo
