    The experience of meaning in circle dance

    Circle dance, which derives from the tradition of folk dances, is practised worldwide. This article explores the meanings participants attribute to it. In-depth interviews with 39 participants, teachers and coordinators of teacher training programmes from the circle dance network in the United Kingdom were undertaken. Applying a constructivist grounded theory approach, major categories, representing respectively the experiences of circle dance participants, teachers and coordinators, were developed. This article specifically focuses on the first major category, termed “I can’t imagine life without it”, which relates to the experience of 22 dancers. From an occupational perspective, the study reveals how participants realise a sense of meaning and satisfaction through engagement in circle dance and the potential contribution of this occupation to well-being

    Chitosan/sulfated locust bean gum nanoparticles: In vitro and in vivo evaluation towards an application in oral immunization

    This work proposes the design of nanoparticles based on locus bean gum (LBG) and chitosan to be used as oral immunoadjuvant for vaccination purposes. LBG-based nanoparticles were prepared by mild polyelectrolyte complexation between chitosan (CS) and a synthesized LBG sulfate derivative (LBGS). Morphological characterization suggested that nanoparticles present a solid and compact structure with spherical-like shape. Sizes around 180-200 nm and a positive surface charge between +9 mV and +14 mV were obtained. CS/LBGS nanoparticles did not affect cell viability of Caco-2 cells after 3 h and 24h of exposure when tested at concentrations up to 1.0 mg/mL. Two model antigens (a particulate acellular extract HE of Salmonella enterica serovar Enteritidis, and ovalbumin as soluble antigen) were associated to CS/LBGS nanoparticles with efficiencies around 26% for ovalbumin and 32% for HE, which resulted in loading capacities up to 12%. The process did not affect the antigenicity of the associated antigens. BALB/c mice were orally immunized with ovalbumin-loaded nanoparticles (100 mu g), and results indicate an adjuvant effect of the CS/LBGS nanoparticles, eliciting a balanced Th1/Th2 immune response. Thus, CS/LBGS nanoparticles are promising as antigen mucosal delivery strategy, with particular interest for oral administration. (C) 2017 Elsevier B.V.     Probing cellulose–solvent interactions with self-diffusion NMR: Onium hydroxide concentration and co-solvent effects

    The molecular self-diffusion coefficients were accessed, for the first time, in solutions of microcrystalline cel-lulose, dissolved in 30 wt% and 55 wt% aqueous tetrabutylammonium hydroxide, TBAH (aq), and in mixtures of 40 wt% TBAH (aq) with an organic co-solvent, dimethylsulfoxide (DMSO), through pulsed field gradient stim-ulated echo NMR measurements. A two-state model was applied to estimate alpha (i.e., average number of ions that "bind" to each anhydroglucose unit) and Pb (i.e., fraction of "bound" molecules of DMSO, TBAH or H2O to cellulose) parameters. The alpha values suggest that TBA+ ions can bind to cellulose within 0.5 TBA+ to 2.3 TBA+/ AGU. On the other hand, the Pb parameter increases when raising cellulose concentration for TBA+, DMSO and water in all solvent systems. Data suggests that TBAH interacts with the ionized OH groups from cellulose forming a sheath of bulky TBA+ counterions which consequently leads to steric hindrance between cellulose chains.info:eu-repo/semantics/publishedVersio

    Sulphated locust bean gum-coated lipid nanocapsules as potential lung delivery carriers

    Drugs pertaining to Biopharmaceutics Classification System (BCS) classes II and IV have limitations in their delivery, including in the lung. Therefore, drug delivery carriers have been proposed to improve the therapeutic effectiveness of such drugs. This work proposes lipid nanocapsules (LNC) as a potential platform for lung drug delivery. Locust bean gum (LBG), which is a galactomannan, was used as polymeric shell, protecting the oily core of the nanocapsules and providing their surface with hydrophilic character. Due to the neutral character of LBG, in order to enable nanocapsule formation, a sulphate derivative (LBGS) was prepared, which was confirmed by Fourier-transformed infrared (FTIR) spectroscopy. The electrostatic interaction between the negatively charged sulphate groups of LBGS and the positively charged groups of the used cationic lipid (1,2-dioleoyloxy-3- trimethylammoniumpropanchloride, DOTAP), allowed the formation of monodisperse nanocapsules, with sizes around 200 nm and strongly negative zeta potentials, between -70 and -85 mV. Envisaging potential lung drug delivery, the LBGS-coated LNC were co-formulated with mannitol using spray-drying, producing microencapsulated nanocapsules. Feret’s diameter was determined to be 2.6 ± 1.8 µm and 3.1 ± 1.9 µm for Man (control) and Man/LNC microparticles, respectively.     In situ synthesis of magnetite nanoparticles in carrageenan gels

    Magnetite nanoparticles have been successfully synthesized in the presence of carrageenan polysaccharides using an in situ coprecipitation method. Iron coordination to the sulfate groups of the polysaccharide was confirmed by FTIR. The polysaccharide type ( , é, or ì) and concentration have been varied and their effects on particle morphology and chemical stability of the resultant nanocomposite investigated. The presence of carrageenan induces the formation of smaller particles, compared to those formed in the absence of polymer, and their average size depends on the nature and concentration of the polysaccharide used. The chemical stability of magnetite nanoparticles toward oxidation was also seen to depend on biopolymer type with magnetite formed in é-carrageenan showing the highest chemical stability. A general tendency toward lower stability is observed as the polysaccharide concentration is increased. It is suggested that magnetite chemical stability in the carrageenan composites is determined by a fine balance between particle size and gel strength, the latter determining oxygen diffusion rates through the medium

    Comparative study of the production of cellulose nanofibers from agro-industrial waste streams of Salicornia ramosissima by acid and enzymatic treatment

    The study of the suitability of two isolation processes to produce cellulose nanofibers (CNFs) from Salicornia ramosissima waste, with potential applicability as a reinforcing agent of polymeric composites was carried out. To separate the cellulose fibrils from the cell wall and obtain CNFs an alkaline treatment was applied followed by a bleaching treat-ment and, the insoluble residue was next hydrolyzed by either an acid treatment (AT) or an enzyme treatment (ET). SEM and TEM images indicated fiber exposure caused by both treatments. The diameter, length, aspect ratio, and polydispersity index, were measured for both CNFs. CNF (ET) showed high zeta potential values suggesting that ET produces more electrically stable and thinner nanofibers. The FTIR spectra revealed that both treatments effectively removed the amorphous components allowing the CNFs isolation, and XRD patterns evidenced the increase in the degree of crystallinity of both CNFs. Nonetheless, CNF(AT) presented a lower mechanical resistance due to its smaller particle size, compared to the CNF(ET). In summary, the (ET) could successfully isolate CNFs from the Salicornia waste, encouraging the use of this treatment, once when compared to (AT), it does not generate toxic residues, presents mild thermal conditions, and produces CNFs with higher-value applications.info:eu-repo/semantics/publishedVersio

    Nanotechnology, energy and nanomedicine: a survey on scientific communications

    Nanotechnology is a broad spectrum of products and processes spanning mainly human health and energy, but also agriculture, the environment and many other industries and classifications. Different attempts are made for measuring activity and investments to help inform strategy and policy decision making on the scientific, economic, health, environmental and social impacts of nanotechnology. For that reason, an overview of the current state of research communications on specific areas in connection to nanotechnology is necessary. In the present work, a survey was performed considering just the published scientific papers using online databases like Web of Science and accessing using different related terms and combinations. Data were collected since the first register from the 1970s until May 2015. The relationship between nanotechnology applied to energy transformations was established. In particular, the relationship between nuclear energy and nanotechnology is quite limited now, but it is expected to increase in the short term. Independently of the term and the combination of terms employed, a tendency is showed: nanotechnology exhibits almost exponential growth and this interdisciplinary technology can be considered as a new industrial revolution

    Genomic and phenotypic characterization of Campylobacter fetus subsp. venerealis strains

    Research Areas: MicrobiologyThe pathogenesis mechanisms of Campylobacter fetus subsp. venerealis (Cfv), the etiologic agent of Bovine Genital Campylobacteriosis remain elusive. This study evaluated the virulence potential and biovar characteristics of Cfv isolates (n = 13) by PCR screening of putative virulencefactor (VF) genes, Multilocus Sequence Typing (MLST) analysis, antimicrobial susceptibility to tetracycline, penicillin, enrofloxacin and streptomycin testing and whole-genome sequencing (WGS; n = 5), also comparing the latter with 26 other whole-genome sequences of Cfv strains. The putative VF genes encoding type IV secretion system of Cfv (virB2-virB11/virD4) were absent in 92% of isolates, including isolates from aborted foetuses, evidencing that these VF genes are not essential for Cfv pathogenicity. The parA gene, used as a Cfv diagnostic molecular target, was detected in only 3 of 13 isolates, invalidating its use for diagnosis purposes. Three novel sequence types were identified by MLST. Although no in vitro antimicrobial resistance was detected, WGS identified antimicrobial resistance-related genes, including those encoding the multidrug efflux pumps CmeABC and YkkCD, indicating that their presence is not enough to provide antimicrobial resistance. The SNP and accessory protein families analysis segregated the Cfv and Cfv biovar intermedius (Cfvi) strains into different clusters. In conclusion, this study evidenced virulence potential and biovar characteristics of Cfv and Cfvi, which are of relevance for the control of Bovine Genital Campylobacteriosis.info:eu-repo/semantics/publishedVersio

    Toxicological Study Employing Repeated Doses of Garcinielliptone FC, a Polyisoprenylated-Benzophenone Isolated from Seed of Platonia Insignis Mart

    The major constituent from the hexane extract of the seeds of P. insignis is GFC (garcinielliptone FC). Doses of 25, 50and 75 mg/kg of GFC were aseptically suspended in 0.05% Tween 80 dissolved in 0.9% saline (vehicle) and orally administered for30, 90 and 120 consecutive days to adult Swiss mice. In this work, the repeated oral administration, in animals of both sexes,demonstrates that this compound is not able to induce mortality and/or behavioral changes in adult mice. In addition, body weightgain, feed intake and disposal of excreta were not altered by the administration of this compound with repeated doses. Furthermore,no differences in weight and macroscopic structure of the brain, liver, kidney, lung, heart and spleen between groups of male andfemale adult mice were observed after treatment. During the periods of treatment, GFC produced no significant changes onhaematological and biochemical parameters in male and female mice treated with all doses used.     Structural characterization of exopolysaccharides obtained from Porphyridium cruentum exhausted culture medium

    Microalgae produce and secrete large quantities of polysaccharides into the culture medium that is discarded when biomass is separated. The main objective of this study was to add value to the exhausted culture medium (ECM) of Porphyridium cruentum through the recovery of extracellular polysaccharides (EPS) by different extracting and purifying methods for the future biotechnological applications. The ECM was submitted to pre-cipitation with cold absolute ethanol, using ultrasound, dialysis, and trichloroacetic acid (TCA) as purification methods. The purification provided a lower yield, with higher car-bohydrate content. The TCA purified sample presented up to twice as much total carbo-hydrates as the non-purified samples, mainly composed of xylose, galactose, and glucose. Scanning Electron Microscopy (SEM) images showed the smooth structure of P. cruentum EPS, in which TCA post-treatment and dialysis allowed obtaining larger and purest par-ticles, being a good candidate for film making. Microalgal polymeric by-products are a sustainable source to recover valuable compounds, and the purification treatment proved to be an important step to valorize this material.(c) 2023 The Authors.
