    Calibration of the Ames Anechoic Facility. Phase 1: Short range plan

    A calibration was made of the acoustic and aerodynamic characteristics of a small, open-jet wind tunnel in an anechoic room. The jet nozzle was 102 mm diameter and was operated subsonically. The anechoic-room dimensions were 7.6 m by 5.5 m by 3.4 m high (wedge tip to wedge tip). Noise contours in the chamber were determined by various jet speeds and exhaust collector positions. The optimum nozzle/collector separation from an acoustic standpoint was 2.1 m. Jet velocity profiles and turbulence levels were measured using pressure probes and hot wires. The jet was found to be symmetric, with no unusual characteristics. The turbulence measurements were hampered by oil mist contamination of the airflow

    Calibrating Type Ia Supernovae using the Planetary Nebula Luminosity Function I. Initial Results

    We report the results of an [O III] lambda 5007 survey for planetary nebulae (PN) in five galaxies that were hosts of well-observed Type Ia supernovae: NGC 524, NGC 1316, NGC 1380, NGC 1448 and NGC 4526. The goals of this survey are to better quantify the zero-point of the maximum magnitude versus decline rate relation for supernovae Type Ia and to validate the insensitivity of Type Ia luminosity to parent stellar population using the host galaxy Hubble type as a surrogate. We detected a total of 45 planetary nebulae candidates in NGC 1316, 44 candidates in NGC 1380, and 94 candidates in NGC 4526. From these data, and the empirical planetary nebula luminosity function (PNLF), we derive distances of 17.9 +0.8/-0.9 Mpc, 16.1 +0.8/-1.1 Mpc, and 13.6 +1.3/-1.2 Mpc respectively. Our derived distance to NGC 4526 has a lower precision due to the likely presence of Virgo intracluster planetary nebulae in the foreground of this galaxy. In NGC 524 and NGC 1448 we detected no planetary nebulae candidates down to the limiting magnitudes of our observations. We present a formalism for setting realistic distance limits in these two cases, and derive robust lower limits of 20.9 Mpc and 15.8 Mpc, respectively. After combining these results with other distances from the PNLF, Cepheid, and Surface Brightness Fluctuations distance indicators, we calibrate the optical and near-infrared relations for supernovae Type Ia and we find that the Hubble constants derived from each of the three methods are broadly consistent, implying that the properties of supernovae Type Ia do not vary drastically as a function of stellar population. We determine a preliminary Hubble constant of H_0 = 77 +/- 3 (random) +/- 5 (systematic) km/s/Mpc for the PNLF, though more nearby galaxies with high-quality observations are clearly needed.Comment: 25 pages, 12 figures.     Toward a New Kind of Asteroseismic Grid Fitting

    Recent developments in instrumentation (e.g., in particular the Kepler and CoRoT satellites) provide a new opportunity to improve the models of stellar pulsations. Surface layers, rotation, and magnetic fields imprint erratic frequency shifts, trends, and other non-random behavior in the frequency spectra. As our observational uncertainties become smaller, these are increasingly important and difficult to deal with using standard fitting techniques. To improve the models, new ways to compare their predictions with observations need to be conceived. In this paper we present a completely probabilistic (Bayesian) approach to asteroseismic model fitting. It allows for varying degrees of prior mode identification, corrections for the discrete nature of the grid, and most importantly implements a treatment of systematic errors, such as the "surface effects." It removes the need to apply semi- empirical corrections to the observations prior to fitting them to the models and results in a consistent set of probabilities with which the model physics can be probed and compared. As an example, we show a detailed asteroseismic analysis of the Sun. We find a most probable solar age, including a 35 +- 5 million year pre-main sequence phase, of 4.591 billion years, and initial element mass fractions of X_0 = 0.72, Y_0 = 0.264, Z_0 = 0.016, consistent with recent asteroseismic and non-asteroseismic studies.Comment: 15 pages, 5 figures, accepted for publication in The Astrophysical Journal; v2 contains minor changes made in the proofs (updated references & corrected typos

    Response of OH airglow emissions to mesospheric gravity waves and comparisons with full-wave model simulation at a low-latitude Indian station

    Quasi-monochromatic gravity-wave-induced oscillations, monitored using the mesospheric OH airglow emission over Kolhapur (16.8° N, 74.2° E), India, during January to April 2010 and January to December 2011, have been characterized using the Krassovsky method. The nocturnal variability reveals prominent wave signatures with periods ranging from 5.2 to 10.8 h as the dominant nocturnal wave with embedded short-period waves having wave periods of 1.5–4.4 h. The results show that the magnitude of the Krassovsky parameter, viz. |η|, ranged from 2.1 to 10.2 h for principal or long nocturnal waves (5.2–10.8 h observed periods), and from 1.5 to 5.4 h for the short waves (1.5–4.4 h observed periods) during the years of 2010 and 2011, respectively. The phase (i.e., Φ) values of the Krassovsky parameters exhibited larger variability and varied from −8.1 to −167°. The deduced mean vertical wavelengths are found to be approximately −60.2 ± 20 and −42.8 ± 35 km for long- and short-period waves for the year 2010. Similarly, for 2011 the mean vertical wavelengths are found to be approximately −77.6 ± 30 and −59.2 ± 30 km for long and short wave periods, respectively, indicating that the observations over Kolhapur were dominated by upward-propagating waves. We use a full-wave model to simulate the response of OH emission to the wave motion and compare the results with observed values

    ON-bipolar cell gene expression during retinal degeneration: Implications for optogenetic visual restoration.

    PURPOSE: Retinal bipolar cells survive even in the later stages of inherited retinal degenerations (IRDs) and so are attractive targets for optogenetic approaches to vision restoration. However, it is not known to what extent the remodelling that these cells undergo during degeneration affects their function. Specifically, it is unclear if they are free from metabolic stress, receptive to adeno-associated viral vectors, suitable for opsin-based optogenetic tools and able to propagate signals by releasing neurotransmitter. METHODS: Fluorescence activated cell sorting (FACS) was performed to isolate labelled bipolar cells from dissociated retinae of litter-mates with or without the IRD mutation Pde6brd1/rd1 selectively expressing an enhanced yellow fluorescent protein (EYFP) as a marker in ON-bipolar cells. Subsequent mRNA extraction allowed Illumina® microarray comparison of gene expression in bipolar cells from degenerate to those of wildtype retinae. Changes in four candidate genes were further investigated at the protein level using retinal immunohistochemistry over the course of degeneration. RESULTS: A total of sixty differentially expressed transcripts reached statistical significance: these did not include any genes directly associated with native primary bipolar cell signalling, nor changes consistent with metabolic stress. Four significantly altered genes (Srm2, Slf2, Anxa7 & Cntn1), implicated in synaptic remodelling, neurotransmitter release and viral vector entry had immunohistochemical staining colocalising with ON-bipolar cell markers and varying over the course of degeneration. CONCLUSION: Our findings suggest relatively few gene expression changes in the context of degeneration: that despite remodelling, bipolar cells are likely to remain viable targets for optogenetic vision restoration. In addition, several genes where changes were seen could provide a basis for investigations to enhance the efficacy of optogenetic therapies

    Associations between Sleep Quality and Heart Rate Variability: Implications for a Biological Model of Stress Detection Using Wearable Technology.

    INTRODUCTION: The autonomic nervous system plays a vital role in the modulation of many vital bodily functions, one of which is sleep and wakefulness. Many studies have investigated the link between autonomic dysfunction and sleep cycles; however, few studies have investigated the links between short-term sleep health, as determined by the Pittsburgh Quality of Sleep Index (PSQI), such as subjective sleep quality, sleep latency, sleep duration, habitual sleep efficiency, sleep disturbances, use of sleeping medication, and daytime dysfunction, and autonomic functioning in healthy individuals. AIM: In this cross-sectional study, the aim was to investigate the links between short-term sleep quality and duration, and heart rate variability in 60 healthy individuals, in order to provide useful information about the effects of stress and sleep on heart rate variability (HRV) indices, which in turn could be integrated into biological models for wearable devices. METHODS: Sleep parameters were collected from participants on commencement of the study, and HRV was derived using an electrocardiogram (ECG) during a resting and stress task (Trier Stress Test). RESULT: Low-frequency to high-frequency (LF:HF) ratio was significantly higher during the stress task than during the baseline resting phase, and very-low-frequency and high-frequency HRV were inversely related to impaired sleep during stress tasks. CONCLUSION: Given the ubiquitous nature of wearable technologies for monitoring health states, in particular HRV, it is important to consider the impacts of sleep states when using these technologies to interpret data. Very-low-frequency HRV during the stress task was found to be inversely related to three negative sleep indices: sleep quality, daytime dysfunction, and global sleep score

    Molecular basis of oocyte-paracrine signalling that promotes granulosa cell proliferation

    Copyright © 2006 Company of BiologistsOocytes regulate follicle growth by secreting paracrine growth factors that act on neighbouring granulosa cells (GCs). Those factors identified to date are mainly members of the transforming growth factor-ß (TGFß) superfamily, but little is known about which specific receptor/signalling system(s) they employ. This study was conducted to determine the requisite pathways utilised by oocytes to promote GC proliferation. We used an established oocyte-secreted mitogen bioassay, where denuded mouse oocytes are co-cultured with mural GCs. Oocytes, growth differentiation factor-9 (GDF9), TGFß1 and activin-A all promoted GC DNA synthesis, but bone-morphogenetic protein 6 (BMP6) did not. Subsequently, we tested the capacity of various TGFß superfamily receptor ectodomains (ECD) to neutralise oocyte- or specific growth factor-stimulated GC proliferation. The BMP type-II receptor (BMPR-II) ECD antagonised oocyte and GDF9 bioactivity dose-dependently, but had no or minimal effect on TGFß1 and activin-A bioactivity, demonstrating its specificity. The TGFßR-II, activinR-IIA and activinR-IIB ECDs all failed to neutralise oocyte- or GDF9-stimulated GC DNA synthesis, whereas they did antagonise the activity of their respective native ligands. An activin receptor-like kinase (ALK) 4/5/7 inhibitor, SB431542, also antagonised both oocyte and GDF9 bioactivity in a dose-dependent manner. Consistent with these findings, oocytes, GDF9 and TGFß1 all activated SMAD2/3 reporter constructs in transfected GC, and led to phosphorylation of SMAD2 proteins in treated cells. Surprisingly, oocytes did not activate the SMAD1/5/8 pathway in transfected GCs although exogenous BMP6 did. This study indicates that oocyte paracrine factors primarily utilise a similar signalling pathway first identified for GDF9 that employs an unusual combination of TGFß superfamily receptors, the BMPR-II and a SMAD2/3 stimulatory ALK (4, 5 or 7), for transmitting their mitogenic actions in GC. This cell-signalling pathway may also have relevance in the hypothalamic-pituitary axis and in germ-somatic cell interactions in the testis.Robert B. Gilchrist, Lesley J.     Stress Watch: The Use of Heart Rate and Heart Rate Variability to Detect Stress: A Pilot Study Using Smart Watch Wearables.

    Stress is an inherent part of the normal human experience. Although, for the most part, this stress response is advantageous, chronic, heightened, or inappropriate stress responses can have deleterious effects on the human body. It has been suggested that individuals who experience repeated or prolonged stress exhibit blunted biological stress responses when compared to the general population. Thus, when assessing whether a ubiquitous stress response exists, it is important to stratify based on resting levels in the absence of stress. Research has shown that stress that causes symptomatic responses requires early intervention in order to mitigate possible associated mental health decline and personal risks. Given this, real-time monitoring of stress may provide immediate biofeedback to the individual and allow for early self-intervention. This study aimed to determine if the change in heart rate variability could predict, in two different cohorts, the quality of response to acute stress when exposed to an acute stressor and, in turn, contribute to the development of a physiological algorithm for stress which could be utilized in future smartwatch technologies. This study also aimed to assess whether baseline stress levels may affect the changes seen in heart rate variability at baseline and following stress tasks. A total of 30 student doctor participants and 30 participants from the general population were recruited for the study. The Trier Stress Test was utilized to induce stress, with resting and stress phase ECGs recorded, as well as inter-second heart rate (recorded using a FitBit). Although the present study failed to identify ubiquitous patterns of HRV and HR changes during stress, it did identify novel changes in these parameters between resting and stress states. This study has shown that the utilization of HRV as a measure of stress should be calculated with consideration of resting (baseline) anxiety and stress states in order to ensure an accurate measure of the effects of additive acute stress

    The British antibiotic and silver-impregnated catheters for ventriculoperitoneal shunts multi-centre randomised controlled trial (the BASICS trial): study protocol

    BACKGROUND: Insertion of a ventriculoperitoneal shunt (VPS) for the treatment of hydrocephalus is one of the most common neurosurgical procedures in the UK, but failures caused by infection occur in approximately 8% of primary cases. VPS infection is associated with considerable morbidity and mortality and its management results in substantial cost to the health service. Antibiotic-impregnated (rifampicin and clindamycin) and silver-impregnated VPS have been developed to reduce infection rates. Whilst there is some evidence showing that such devices may lead to a reduction in VPS infection, there are no randomised controlled trials (RCTs) to support their routine use. METHODS/DESIGN: Overall, 1,200 patients will be recruited from 17 regional neurosurgical units in the UK and Ireland. Patients of any age undergoing insertion of their first VPS are eligible. Patients with previous indwelling VPS, active and on-going cerebrospinal fluid (CSF) or peritoneal infection, multiloculated hydrocephalus requiring multiple VPS or neuroendoscopy, and ventriculoatrial or ventriculopleural shunt planned will be excluded. Patients will be randomised 1:1:1 to either standard silicone (comparator), antibiotic-impregnated, or silver-impregnated VPS. The primary outcome measure is time to VPS infection. Secondary outcome measures include time to VPS failure of any cause, reason for VPS failure (infection, mechanical failure, or patient failure), types of bacterial VPS infection (organism type and antibiotic resistance), and incremental cost per VPS failure averted. DISCUSSION: The British antibiotic and silver-impregnated catheters for ventriculoperitoneal shunts multi-centre randomised controlled trial (the BASICS trial) is the first multi-centre RCT designed to determine whether antibiotic or silver-impregnated VPS reduce early shunt infection compared to standard silicone VPS. The results of this study will be used to inform current neurosurgical practice and may potentially benefit patients undergoing shunt surgery in the future.     Farmers as experts: interpreting the "hidden" messages of participatory video across African contexts

    Recent scholarship has contributed important insights into the political dynamics inherent in the process of making and showing participatory videos (PV). As a research method and an instrument for social change, participatory video has both potential and limitations for overturning the power dynamics embedded within research and in development processes. This paper focuses on experiences of incorporating participatory video in land management projects in four countries in Africa. Along with other participatory methods, the videos represented an effort to include community perspectives and objectives into the research process. Analysis of PV has largely focused on examining the tensions and contradictions involved in the process of making participatory videos. There has been less focus on the content of the videos themselves and what it might suggest for empowerment, voice and representation. This paper attempts to address this gap by examining the implications of the narratives that emerge in five different videos. On the surface, the participants appear to repeat dominant national and global narratives about land degradation. However, the fact that farmers present themselves as experts on these topics and the ways in which they appropriate and reconfigure the dominant narratives, can be seen as an act of empowerment. In this way, they preclude the need for external intervention on how to manage their resources
