Urinary Calculi
The chemical composition of 256 urinary calculi on the Witwatersrand has been determined in 3 population groups. 'Calcium stones' comprised 53,1% of the total and most of the remainder were triple phosphate. The distribution of these stones in the different population groups was similar. cis-Regulatory Changes in Kit Ligand Expression and Parallel Evolution of Pigmentation in Sticklebacks and Humans
SummaryDramatic pigmentation changes have evolved within most vertebrate groups, including fish and humans. Here we use genetic crosses in sticklebacks to investigate the parallel origin of pigmentation changes in natural populations. High-resolution mapping and expression experiments show that light gills and light ventrums map to a divergent regulatory allele of the Kit ligand (Kitlg) gene. The divergent allele reduces expression in gill and skin tissue and is shared by multiple derived freshwater populations with reduced pigmentation. In humans, Europeans and East Asians also share derived alleles at the KITLG locus. Strong signatures of selection map to regulatory regions surrounding the gene, and admixture mapping shows that the KITLG genomic region has a significant effect on human skin color. These experiments suggest that regulatory changes in Kitlg contribute to natural variation in vertebrate pigmentation, and that similar genetic mechanisms may underlie rapid evolutionary change in fish and humans
Osteoblastic flare in a patient with advanced gastric cancer after treatment with pemetrexed and oxaliplatin: implications for response assessment with RECIST criteria
<p>Abstract</p> <p>Background</p> <p>The RECIST guidelines are commonly used in phase II and III clinical trials. The correct definition of response can be controversial in some situations, as in the case we describe.</p> <p>Case presentation</p> <p>A 43 year-old man with advanced gastric cancer was enrolled in a phase II trial where he was treated with pemetrexed 500 mg/m<sup>2 </sup>plus oxaliplatin 120 mg/m<sup>2 </sup>every 3 weeks. At baseline, the target lesions were lymph-nodes, and the non-target lesions were small pulmonary nodules. At first re-evaluation, the target lesions showed partial response and the non-target lesions showed complete response, but new diffuse osteoblastic lesions appeared. The investigator decided to continue treatment until the second re-evaluation. CT scan confirmed the response of the target and non-target lesions, while the osteoblastic lesions did not change.</p> <p>Conclusion</p> <p>The appearance of osteoblastic lesions after an active antitumor treatment, a phenomenon known as flare, can complicate the definition of the best overall response using RECIST criteria. This possibility should be considered by oncologists involved in clinical trials.</p

Growth dynamics of deciduous species during their life period: A case study of urban green space in India
It is evident that grass density (GD) and shoot growth rate (SGR) governs the differential settlement of substructure, groundwater recharge, and stability of green infrastructure. GD and SGR are usually assumed to be constant during the entire life period of vegetation. However, spatial and temporal dynamics of GD and SGR in urban green space were rarely explored previously. The main objective of this study is to explore the spatial and temporal dynamics of GD and SGR in urban space vegetated with deciduous species (mix grass i.e., Poaceae and Bauhinia purpurea). Field monitoring was conducted in the urban green space for one year (i.e., life period of selected species). The monitoring period includes the growth period and gradual wilting period. Substantial spatial variation of GD was found during the first six months. GD away from the tree trunk was found to be 1.02–56.3 times higher than that near the tree trunk during the first six months. Thereafter, any spatial variation of GD was not found in the next six months. Unlike the GD, SGR was found to vary during the entire life period of mix grass. In addition, SGR away from the tree trunk was found to be 1.1–4.6 times higher than that near the tree trunk. Any relationship between GD and rainfall depth was not found. Whereas, SGR mainly depends on rainfall depth. The hypothesis of uniformity in GD and SGR during the life period of deciduous species was not found to be true
Normalizing single-cell RNA sequencing data: challenges and opportunities
Single-cell transcriptomics is becoming an important component of the molecular biologist's toolkit. A critical step when analyzing data generated using this technology is normalization. However, normalization is typically performed using methods developed for bulk RNA sequencing or even microarray data, and the suitability of these methods for single-cell transcriptomics has not been assessed. We here discuss commonly used normalization approaches and illustrate how these can produce misleading results. Finally, we present alternative approaches and provide recommendations for single-cell RNA sequencing users
