Epistemic and social scripts in computer-supported collaborative learning
Collaborative learning in computer-supported learning environments typically means that learners work on tasks together, discussing their individual perspectives via text-based media or videoconferencing, and consequently acquire knowledge. Collaborative learning, however, is often sub-optimal with respect to how learners work on the concepts that are supposed to be learned and how learners interact with each other. One possibility to improve collaborative learning environments is to conceptualize epistemic scripts, which specify how learners work on a given task, and social scripts, which structure how learners interact with each other. In this contribution, two studies will be reported that investigated the effects of epistemic and social scripts in a text-based computer-supported learning environment and in a videoconferencing learning environment in order to foster the individual acquisition of knowledge. In each study the factors ‘epistemic script’ and ‘social script’ have been independently varied in a 2×2-factorial design. 182 university students of Educational Science participated in these two studies. Results of both studies show that social scripts can be substantially beneficial with respect to the individual acquisition of knowledge, whereas epistemic scripts apparently do not to lead to the expected effects

Single molecule study of a processivity clamp sliding on DNA
Using solution based single molecule spectroscopy, we study the motion of the polIII {beta}-subunit DNA sliding clamp ('{beta}-clamp') on DNA. Present in all cellular (and some viral) forms of life, DNA sliding clamps attach to polymerases and allow rapid, processive replication of DNA. In the absence of other proteins, the DNA sliding clamps are thought to 'freely slide' along the DNA; however, the abundance of positively charged residues along the inner surface may create favorable electrostatic contact with the highly negatively charged DNA. We have performed single-molecule measurements on a fluorescently labeled {beta}-clamp loaded onto freely diffusing plasmids annealed with fluorescently labeled primers of up to 90 bases. We find that the diffusion constant for 1D diffusion of the {beta}-clamp on DNA satisfies D {le} 10{sup -14} cm{sup 2}/s, much slower than the frictionless limit of D = 10{sup -10} cm{sup 2}/s. We find that the {beta} clamp remains at the 3-foot end in the presence of E. coli single-stranded binding protein (SSB), which would allow for a sliding clamp to wait for binding of the DNA polymerase. Replacement of SSB with Human RP-A eliminates this interaction; free movement of sliding clamp and poor binding of clamp loader to the junction allows sliding clamp to accumulate on DNA. This result implies that the clamp not only acts as a tether, but also a placeholder

Mature exotic conifer stands have greater catches of the EU-protected Geomalacus maculosus than adjacent peatland or clear-felled stands—implications for forestry

Context: The discovery of EU-protected Geomalacus maculosus in commercial plantations requires an understanding of the implications of forestry practices for the species within the context of sustainable forest management. Aims: 1.Compare Geomalacus maculosus captures across mature exotic Sitka spruce-dominated stands, previously clear-felled stands and adjacent peatland habitats.2.Assess the suitability, for forest managers, of population estimate models for G. maculosus.3.Assess the implications of felling by comparing relative abundances of G. maculosus directly before and after clear-felling at a mature exotic Sitka spruce-dominated stand. Methods: Geomalacus maculosus catches were compared at four sites across two to three mature (43–45 years old) conifer stands per site, one clear-felled stand per site, and one adjacent peatland per site using refuge traps and hand searching. Capture-mark-recapture studies were undertaken to estimate population sizes. A BACIP (Before-After-Control-Impact-Paired) analysis was undertaken in one forest stand at one forest site to determine impacts of a clear-felling event. Results: Mean catches of Geomalacus maculosus adults in the mature forest stands were over 10 and 11 times greater than mean catches on peatland and clear-fell stands, respectively. The Schnabel model for estimating population size was most suited for mature forest stands but could not be utilised for the other habitats. BACIP analysis showed a significant impact of clear-felling with a 95% reduction in mean G. maculosus catches after a clear-felling event where none of the individuals marked prior to felling were recaptured compared to 21% recapture rates at the control site. Greater tree circumference in mature conifer stands correlated with greater catches. Conclusion: Guidelines are needed to ensure the protection of Geomalacus maculosus in commercial forestry. Interventions could include patch/tall stump retention at final felling and/or translocation of the protected species
Nest Architecture, Colony Productivity, and Duration of Immature Stages in a Social Wasp, Mischocyttarus consimilis
This study examined the nest architecture, colony productivity, and duration of the immature stages of the social wasp Mischocyttarus consimilis Zikáán (Hymenoptera: Vespidae). The study was carried out under field conditions. Nests of M. consimilis consist of a single uncovered comb, which is attached to the substratum by a single petiole. The data for the nest architecture showed a positive and significant correlation between the size of the comb and the diameter of the petiole, and also between the height and diameter of the cells. The nests were constructed on horizontal, vertical, and sloping substrata with no apparent preference for a specific orientation. The colonies produced 72.9 cells and 40.7 adults on average. The mean frequency of productive cells was 33.3%, and 19.4% of the cells were reused. The mean duration of the immature stages combined was 69.7 days and the egg, larval, and pupal stages had mean durations of 14.9, 36.0, and 18.8 days, respectively. The duration of each immature stage was significantly shorter in the warmhumid season, and the larval and pupal stages were shorter during the colony pre-emergence stage
Demographics of sources of HIV-1 transmission in Zambia: a molecular epidemiology analysis in the HPTN 071 PopART study
BACKGROUND: In the last decade, universally available antiretroviral therapy (ART) has led to greatly improved health and survival of people living with HIV in sub-Saharan Africa, but new infections continue to appear. The design of effective prevention strategies requires the demographic characterisation of individuals acting as sources of infection, which is the aim of this study. METHODS: Between 2014 and 2018, the HPTN 071 PopART study was conducted to quantify the public health benefits of ART. Viral samples from 7124 study participants in Zambia were deep-sequenced as part of HPTN 071-02 PopART Phylogenetics, an ancillary study. We used these sequences to identify likely transmission pairs. After demographic weighting of the recipients in these pairs to match the overall HIV-positive population, we analysed the demographic characteristics of the sources to better understand transmission in the general population. FINDINGS: We identified a total of 300 likely transmission pairs. 178 (59·4%) were male to female, with 130 (95% CI 110-150; 43·3%) from males aged 25-40 years. Overall, men transmitted 2·09-fold (2·06-2·29) more infections per capita than women, a ratio peaking at 5·87 (2·78-15·8) in the 35-39 years source age group. 40 (26-57; 13·2%) transmissions linked individuals from different communities in the trial. Of 288 sources with recorded information on drug resistance mutations, 52 (38-69; 18·1%) carried viruses resistant to first-line ART. INTERPRETATION: HIV-1 transmission in the HPTN 071 study communities comes from a wide range of age and sex groups, and there is no outsized contribution to new infections from importation or drug resistance mutations. Men aged 25-39 years, underserved by current treatment and prevention services, should be prioritised for HIV testing and ART. in the two decay mechanisms, and hence future experimental measurements of this
ratio will provide valuable information for substantiating the hybrid nature of
