    Background: The effect of gene polymorphisms and promoter methylation, associated with maladaptive developmental outcomes, vary depending on environmental factors (e.g., parental psychopathology). Most studies have focused on 0- to 5-year-old children, adolescents, or adults, whereas there is dearth of research on school-age youths and pre-adolescents. Methods: In a sample of 21 families recruited at schools, we addressed parents' psychopathological symptoms (through SCL-90-R); offspring emotional-behavioral functioning (through CBCL-6-18); dopamine transporter gene (DAT1) for epigenetic status of the 5'-untranslated region (UTR) and for genotype, i.e., variable number of tandem repeats polymorphism at the 3'-UTR. Possible associations were explored between bio-genetic and psychological characteristics within the same individual and between triplets of children, mothers, and fathers. Results: DAT methylation of CpG at positions M1, M6, and M7 in mothers was correlated with maternal (phobic) anxiety, whereas in fathers' position M6 was related to paternal depression, anxiety, hostility, psychoticism, and higher Global Severity Index (GSI). No significant correlations were found between maternal and offspring DAT methylation. Significant correlations were found between fathers' methylation at CpG M1 and children's methylation at CpG M6. Linear regressions showed that mothers and fathers' GSI predicted children's methylation at CpG sites M2, M3, and M6, whereas fathers' GSI predicted children's methylation at CpG sites, particularly M1, M2, and M6. Moreover, offspring methylation of DAT at CpG M2 predicted somatic complaint, internalizing and attention problems; methylation of DAT at CpG M6 predicted withdraw. Conclusion: This study may have important clinical implication for the prevention and treatment of emotional-behavioral difficulties in children, as it adds to previous knowledge about the role of genetic and environmental factors in predicting psychopathological symptoms within non-clinical population

    Engineering geological mapping of near-surface rock mass quality of folded and thrusted arenaceous flysch units in the Northern Apennines (Italy)

    The regionalized knowledge of the quality of near-surface rock masses is an important tool for land management/planning, as well as for guiding further in-depth studies aimed at landslide and earthquake risk assessment and civil engineering planning. The characterization of heterogeneous rock masses like flysch units represents a relevant challenge to engineering geologists due to the complex structure of these materials, which results from both their depositional context and tectonic history. Flysches are widespread all over the Apennines chain and their mechanical characterization is a difficult task given the occurrence of intercalation of layers with different lithology and strength. Moreover, the complexity of the thrust and fold tectonic framework makes the regional distribution of these characters difficult to predict. The aim of this work is to provide a method to map the near-surface rock masses quality for an arenaceous flysch widely cropping out in the outer Northern Apennines (Torrente Carigiola Formation, Aquitanian; Bettelli et al., 2002). This formation is mapped in both the geological map of the Regione Toscana (Italy) at the scale of 1:10,000 and the geological sheet “252 – Barberino di Mugello” (Bettelli et al., 2002) of the Italian Geological Map at the scale of 1:50,000 (CARG). It is made up by intercalated arenaceous (A) and pelitic (P) layers characterized by variable A/P ratio. The rock mass quality is evaluated by estimating, for a set of representative rock outcrops, the Rock Mass Quality Index (RQI; Disperati et al. 2016; Mammoliti et al. 2018). This index results from the analysis of both systematic Schmidt hammer rebound measurements (R) acquired at the nodes of a regular grid (ca. 20 R measurements for ca. 15-25 nodes) and the determination of the unit weight for representative outcrop rock samples. For the same outcrops, also the A/P ratio and bedding attitude are determined. The results show a positive linear correlation between RQI and the A/P ratio, confirming that the latter parameter is an important feature controlling the rock mass strength. This correlation is used to assess the distribution of both parameters within a set of geological cross sections traced normal to the regional structures trend (main thrusts and km-scale folds). Then, the structural features available from the literature geological maps allow us to extrapolate both the RQI and A/P ratio from the profiles to the map scale. Finally, a further set of the same rock outcrop data acquired after the above-described modelling procedure is used to check the accuracy of the method

Le travail présenté consiste en une étude comparée de la langue utilisée pour la vulgarisation de la médecine en Italie et en France. L’analyse concerne notamment la langue employée dans les notices de médicaments, rédigées selon les directives européennes. Le but de la recherche est d’évaluer les analogies et les différences qui existent entre les stratégies adoptées à des fins de simplification. La notice du Doliprane/Tachipirina, en langue italienne et française, a été analysée. Chaque notice a été analysée sur les plans : formel – syntaxique – morphologique – lexical – pragmatique

    Are public health professionals prepared for public health genomics? A cross-sectional survey in Italy

    Background: Public health genomics is an emerging multidisciplinary approach, which aims to integrate genome-based knowledge in a responsible and effective way into public health. Despite several surveys performed to evaluate knowledge, attitudes and professional behaviors of physicians towards predictive genetic testing, similar surveys have not been carried out for public health practitioners. This study is the first to assess knowledge, attitudes and training needs of public health professionals in the field of predictive genetic testing for chronic diseases. Methods: A self-administered questionnaire was used to carry out a cross-sectional survey of a random sample of Italian public health professionals. Results: A response rate of 67.4% (797 questionnaires) was achieved. Italian public health professionals have the necessary attitudinal background to contribute to the proper use of predictive genetic testing for chronic diseases, but they need additional training to increase their methodological knowledge. Knowledge significantly increases with exposure to predictive genetic testing during postgraduate training (odds ratio (OR) = 1.74, 95% confidence interval (CI) = 1.05-2.88), time dedicated to continuing medical education (OR = 1.53, 95% CI = 1.14-2.04) and level of English language knowledge (OR = 1.36, 95% CI = 1.07-1.72). Adequate knowledge is the strongest predictor of positive attitudes from a public health perspective (OR = 3.98, 95% CI = 2.44-6.50). Physicians show a lower level of knowledge and more public health attitudes than other public health professionals do. About 80% of public health professionals considered their knowledge inadequate and 86.0% believed that it should be improved through specific postgraduate training courses. Conclusions: Specific and targeted training initiatives are needed to develop a skilled public health workforce competent in identifying genomic technology that is ready for use in population health and in modeling public health genomic programs and primary care services that need to be developed, implemented and evaluated

    Methylation dynamics on 5′-UTR of DAT1 gene as a bio-marker to recognize therapy success in ADHD children

    : Attention-deficit/hyperactivity disorder (ADHD), a neuropsychiatric condition characterized by inattention, hyperactivity, and impulsivity, afflicts 5% of children worldwide. Each ADHD patient presents with individual cognitive and motivational peculiarities. Furthermore, choice of appropriate therapy is still up to clinicians, who express somewhat qualitative advice on whether a child is being successfully cured or not: it would be more appropriate to use an objective biomarker to indicate whether a treatment led to benefits or not. The aim of our work is to search for such clinical biomarkers. We recruited 60 ADHD kids; psychopathological scales were administered at recruitment and after six weeks of therapy. Out of such a cohort of ADHD children, we rigorously extracted two specific subgroups; regardless of the initial severity of their disease, we compared those who obtained the largest improvement (ΔCGAS > 5) vs. those who were still characterized by a severe condition (CGAS < 40). After such a therapy, methylation levels of DNA extracted from buccal swabs were measured in the 5’-UTR of the DAT1 gene. CpGs 3 and 5 displayed, in relation to the other CpGs, a particular symmetrical pattern; for “improving” ADHD children, they were methylated together with CpG 2 and CpG 6; instead, for “severe” ADHD children, they accompanied a methylated CpG 1. These specific patterns of methylation could be used as objective molecular biomarkers of successful cures, establishing if a certain therapy is akin to a given patient (personalized medicine). Present data support the use of post-therapy molecular data obtained with non-invasive technique

    People are plugged into an intangible sound universe. But only a tiny part of the sounds we are exposed to have been purposefully designed. Recently, designers are bashfully approaching these intangible products’ quality. Product Sound Design represents, in fact, a promising research field still scarcely explored. The design community is answering this concern through new design methods. An Italian university developed a patented method-and-tool, conceived to collect, analyze, and recreate various sounds to develop a new generation of products with designed mechanical (and, eventually, digital) sounds. Spreading this innovation within the design community is fundamental to stimulate future more focused and aware practices. As well as all new technologies, the new patent didn’t have its own identity from the beginning. Extensive work conducted with the scientific approach has therefore been undertaken to redesign its identity to make its disruptiveness intelligible and understandable

    Domain-domain interactions in Filamin A (16-23) impose a hierarchy of unfolding forces

    The quaternary structure of Filamin A (FLNa) 16-23 was recently shown to exhibit multiple domain-domain interactions that lead to a propeller-like construction. Here we present single molecule force spectroscopy experiments to show a wide variety of mechanical responses of this molecule and compare it with its linear counterpart FLNa 1-8. The compact structure of FLNa 16-23 leads to a broad distribution of rupture forces and end-to-end lengths in the force-extension mode and multiple unraveling timescales in the force-clamp mode. Moreover, a subset of force-extension trajectories reveals a mechanical hierarchy in which the rupture of domain-domain interactions at high forces (200 pN) liberates the unfolding of individual domains at low forces (100 pN). This mechanism may also explain the order of magnitude difference in the rates of the biexponential fits to the distribution of unfolding dwell times under force-clamp. Overall, FLNa 16-23 under a force of 100 pN is more compliant than the linear FLNa 1-8. Since a physiological role of FLNa is to crosslink actin filaments, this range of responses allows it to accommodate a broad spectrum of forces exerted by the cell and its environment

    The new engineering geological map (carta litotecnica) of Tuscany (Italy)

    Municipal administrations in Italy must be provided with thematic maps and documentation which describe the geological, geomorphological, lithological, hydrogeological and hydraulic characters useful to manage spatial planning issues. Among these documents, a “Lithotechnical” (or “Lithological-Technical”) map is drawn up, generally at the scale of 1:10,000, by organizing the geological formations into lithotechnical units according to their lithological and physical-mechanical properties. Often, this map also integrates the results of previous field and borehole investigations. However, this map is characterized by a certain degree of subjectivity because it is supported by few specific quantitative data. We present a new method for the regional scale engineering geological classification of sub-surface rock and soil masses obtained by integrating the geological map at the scale of 1:10,000 as a reference document, with a large set of data obtained through the collection and processing of new lithological and physical-mechanical observations and measurements of the outcropping geological formations. The adopted procedure involves both the extensive in situ use of the Schmidt's hammer and the execution of laboratory tests, such as the Slake Durability Test (Franklin & Chandra, 1972) and the determination of the rock unit weight. These tools and tests allow us to acquire a large set of quantitative in situ and laboratory data with known repeatability to obtain a regional scale GIS database providing the classification of the lithological and physical-mechanical characteristics of a wide range of geological formations. As a first step, each outcrop is classified according to a new engineering geological nomenclature system described by the code XXv[y]_[Z] whose values are obtained by integrating: i) a lithological parameter XXv evaluated from both typical characters of the geological formations under analysis and outcrop observations; ii) an engineering geological parameter [y] obtained by the results of the Slake Durability Test; iii) an engineering geological parameter [Z] (Rockmass Quality Index - RQI) evaluated at the outcrop scale on the basis of a large set of sclerometric measurements. The results of outcrop classification are stored into a point topology GIS dataset and are then processed and spatialized in order to assign the XXv[y]_[Z] code to the geological formations, thus obtaining the new engineering geological map. Within the framework of research agreements among Regione Toscana administration, the Consorzio LaMMA, the CNRIGG and the Department of Earth, Environmental and Physical Sciences of the University of Siena, the latter being the leader for their implementation, more than 300 geological formations were analysed and classified, and the new engineering geological GIS map was realized in Tuscany for the provinces of Arezzo, Florence, Lucca, Massa-Carrara, Pistoia, Prato and Siena (ca. 15,000 km2)
