Streptozotocin-induced mechanical hypernociception is not dependent on hyperglycemia
Since streptozotocin (STZ)-induced diabetes is a widely used model of painful diabetic neuropathy, the aim of the present study was to design a rational protocol to investigate whether the development of mechanical hypernociception induced by STZ depends exclusively on hyperglycemia. Male Wistar rats (180-200 g; N = 6-7 per group) received a single intravenous injection of STZ at three different doses (10, 20, or 40 mg/kg). Only the higher dose (40 mg/kg) induced a significant increase in blood glucose levels, glucose tolerance and deficiency in weight gain. However, all STZ-treated rats (hyperglycemic or not) developed persistent (for at least 20 days) and indistinguishable bilateral mechanical hypernociception that was not prevented by daily insulin treatment (2 IU twice a day, sc). Systemic morphine (2 mg/kg) but not local (intraplantar) morphine treatment (8 µg/paw) significantly inhibited the mechanical hypernociception induced by STZ (10 or 40 mg/kg). In addition, intraplantar injection of STZ at doses that did not cause hyperglycemia (30, 100 or 300 µg/paw) induced ipsilateral mechanical hypernociception for at least 8 h that was inhibited by local and systemic morphine treatment (8 µg/paw or 2 mg/kg, respectively), but not by dexamethasone (1 mg/kg, sc). The results of this study demonstrate that systemic administration of STZ induces mechanical hypernociception that does not depend on hyperglycemia and intraplantar STZ induces mechanical sensitization of primary sensory neurons responsive to local morphine treatment
Perfil e distribuição da síndrome cólica em equinos em três unidades militares do Estado do Rio de Janeiro, Brasil
Este estudo teve como objetivo avaliar o perfil e a distribuição da síndrome cólica em eqüinos de três unidades militares no Estado do Rio de Janeiro, o Regimento Escola de Cavalaria (REsC), a Academia Militar das Agulhas Negras (AMAN) e o Esquadrão Escola de Cavalaria (EEC). No período entre 2003 e 2004, 770 eqüinos foram acompanhados para a ocorrência de casos clínicos de cólica. Realizou-se análise descritiva dos dados, e foram calculadas a taxa de incidência de cólica e a proporção de eqüinos acometidos. O teste do χ2 foi utilizado para avaliar a associação entre síndrome cólica e variáveis relativas às características dos eqüinos e do manejo. A incidência variou entre as unidades militares, 0,12 na AMAN, 0,21 no EEC e 0,95 casos/eqüino-ano no REsC, sendo acometidos 15% dos eqüinos da AMAN, 30% do EEC e 69% do REsC. A maior incidência foi de episódios de origem gástrica, 76,5%. Casos de reincidência foram elevados, sendo 62,5% no REsC, 36,7% na AMAN e 29,0% no EEC. A síndrome cólica estava significativamente associada às variáveis unidade militar, sistema de criação, quantidade de grãos ingerido e suplemento mineral-vitamínico. As altas incidências, reincidências e proporção de animais acometidos observadas ocorreram de forma diferenciada nas três unidades, indicando que, apesar de possuírem como característica comum pertencerem a unidades militares, os eqüinos formam um grupo heterogêneo no que diz respeito à ocorrência de síndrome cólica, provavelmente devido às diferentes condições de manejo.This research aimed to verify the profile and distribution of colic syndrome in horses of three military units in Rio de Janeiro, Brazil, which were Regimento Escola de Cavalaria (REsC), Academia Militar das Agulhas Negras (AMAN), and Esquadrão Escola de Cavalaria (EEC). From 2003 to 2004, 770 horses were followed up for the incidence of colic cases. A descriptive analysis of clinical data, incidence of colic cases and rate of horses affected was established. Chi-squared test was used to evaluate the association among colic syndrome and variables related to horses characteristics and management. Colic syndrome incidence varied among military units as follows: 0.12 cases/horse-year at AMAN, 0.21 at EEC and 0.95 at REsC, affecting 15% of horses in AMAN, 30% in EEC and 69% in REsC. The highest incidence was of gastric episodes (76.5%). Recurrence cases were 62.5% in REsC, 36.7% in AMAN and 29.0% in EEC. Colic syndrome occurred associated with variables: military unit, management, amount of intake grains and supplemented mineral-vitamin. High incidence, recurrence and horses rate with colic were differentiated among the three units. Although the horses of military units had common characteristic , they formed a heterogeneous group in respect to colic syndrome occurrence, probably due to different management conditions
TÉCNICA RESTAURADORA SEMI-DIRETA: REVISÃO DE LITERATURA
Introdução: A resina composta é um material restaurador que encontra-se em constante evolução, o que confere a ela um uso clínico amplo e diversificado. Clinicamente, o cirurgião dentista (CD) tem à sua disposição diferentes tipos de resinas, o que lhe permite realizar procedimentos estéticos e funcionais bem satisfatórios. Contudo, quando as cavidades a serem restauradas são mais amplas os procedimentos diretos tornam-se mais difíceis, posto isso optamos por um procedimento indireto. Este por sua vez possui mais sessões clínicas, além do custo com o protético, acarretando em muitas das vezes a desistência do tratamento. Neste sentido, optamos pela restauração semidireta. Estas após a moldagem, podem ser confeccionadas sobre modelos rígidos (em gesso) ou semirrígidos (silicone de adição) pelo próprio CD utilizando as resinas de uso direto e fotopolimerizadas por LEDs convencionais. A seguir, são levadas ao aparelho de micro-ondas imersas em água destilada por cinco minutos. Esta etapa eleva o grau de conversão das resinas compostas por meio de uma termo polimerização, proporcionando aumento de resistência ao desgaste e da estabilidade dimensional da resina. Objetivos: O objetivo deste trabalho foi relembrar aos acadêmicos de odontologia e aos CD’s formados uma alternativa extremamente viável devido ao baixo custo, estética favorável, melhoria das propriedades físicas e mecânicas, maior controle da contração de polimerização e facilidade de reprodução anatômica. Metodologia: O presente trabalho foi baseado em análises de artigos científicos sobre a temática. Foi pesquisado “restauração semidireta” nos sites : Scielo e BVS, sendo que, não houve nenhum encontrado no Scielo e quatro encontrados no BVS. Foi feito, também, a pesquisa “semi-direct restauration” no site PubMed e foram encontrados dois artigos sobre o assunto. Considerações: Diante do exposto concluímos que a técnica restauradora semidireta consiste em uma alternativa viável para restaurações extensas em dentes posteriores, unitárias ou múltiplas, em sessão única, facilitando e ampliando as opções de tratamento
Gastrin-releasing peptide receptor (GRPR) mediates chemotaxis in neutrophils
Neutrophil migration to inflamed sites is crucial for both the initiation of inflammation and resolution of infection, yet these cells are involved in perpetuation of different chronic inflammatory diseases. Gastrin-releasing peptide (GRP) is a neuropeptide that acts through G protein coupled receptors (GPCRs) involved in signal transmission in both central and peripheral nervous systems. Its receptor, gastrin-releasing peptide receptor (GRPR), is expressed by various cell types, and it is overexpressed in cancer cells. RC-3095 is a selective GRPR antagonist, recently found to have antiinflammatory properties in arthritis and sepsis models. Here we demonstrate that i.p. injection of GRP attracts neutrophils in 4 h, and attraction is blocked by RC-3095. Macrophage depletion or neutralization of TNF abrogates GRP-induced neutrophil recruitment to the peritoneum. In vitro, GRP-induced neutrophil migration was dependent on PLC- β2, PI3K, ERK, p38 and independent of Gαi protein, and neutrophil migration toward synovial fluid of arthritis patients was inhibited by treatment with RC-3095.We propose that GRPR is an alternative chemotactic receptor that may play a role in the pathogenesis of inflammatory disorders
Smoking-induced aggravation of experimental arthritis is dependent of aryl hydrocarbon receptor activation in Th17 cells.
Background:
Epidemiologic studies have highlighted the association of environmental factors with the development and progression of autoimmune and chronic inflammatory diseases. Among the environmental factors, smoking has been associated with increased susceptibility and poor prognosis in rheumatoid arthritis (RA). However, the immune and molecular mechanism of smoking-induced arthritis aggravation remains unclear. The transcription factor aryl hydrocarbon receptor (AHR) regulates the generation of Th17 cells, CD4 T cells linked the development of autoimmune diseases. AHR is activated by organic compounds including polycyclic aromatic hydrocarbons (PAHs), which are environmental pollutants that are also present in cigarette smoke. In this study, we investigated the role of AHR activation in the aggravation of experiment arthritis induced by exposure to cigarette smoke.
Methods:
Mice were exposed to cigarette smoke during the developmental phase of antigen-induced arthritis and collagen-induced arthritis to evaluate the effects of smoking on disease development. Aggravation of articular inflammation was assessed by measuring neutrophil migration to the joints, increase in articular hyperalgesia and changes in the frequencies of Th17 cells. In vitro studies were performed to evaluate the direct effects of cigarette smoke and PAH on Th17 differentiation. We also used mice genetically deficient for AHR (Ahr KO) and IL-17Ra (Il17ra KO) to determine the in vivo mechanism of smoking-induced arthritis aggravation.
Results:
We found that smoking induces arthritis aggravation and increase in the frequencies of Th17 cells. The absence of IL-17 signaling (Il17ra KO) conferred protection to smoking-induced arthritis aggravation. Moreover, in vitro experiments showed that cigarette smoke can directly increase Th17 differentiation of T cells by inducing AHR activation. Indeed, Ahr KO mice were protected from cigarette smoke-induced arthritis aggravation and did not display increase in TH17 frequencies, suggesting that AHR activation is an important mechanism for cigarette smoke effects on arthritis. Finally, we demonstrate that PAHs are also able to induce arthritis aggravation.
Conclusions:
Our data demonstrate that the disease-exacerbating effects of cigarette smoking are AHR dependent and environmental pollutants with AHR agonist activity can induce arthritis aggravation by directly enhancing Th17 cell development
Nutritional supplementation and growth after hospital discharge in very low birthweight newborns: Randomized controlled trial / Suplementação nutricional e crescimento de recém-nascidos de muito baixo peso após alta hospitalar: Ensaio clínico randomizado
Background Evidence is insufficient to show whether fortification has any effect on growth in preterm infants after discharge. Objective to verify whether VLBW preterm infants who are supplemented with multicomponent present greater anthropometric measurements than those not supplemented. Study Design Parallel randomized controlled trial. A computer-generated random number table was used to allocate the participants. Participants Preterm infants discharged from the NICU of a University Hospital from northeast, Brazil, weighing less than 1,500 g exclusively breastfed at discharge and followed up until they reached 6 months corrected gestational age. Intervention intervention group received Nestlé® PreNan® formula, fractionated in 2 g of powder, mixed with the mother's milk twice a day. Control group was exclusively breastfed. Follow-up was conducted until the infants reached 6 months corrected gestational age (CGA). Outcomes Growth of the anthropometrics parameters weight, head circumference (HC) and lenth with 6 months of corrected age. Mixed effects model for longitudinal data was used. Interaction according to sex was detected and ajusted. Results Weight gain was significantly higher in the intervention group. This effect was verified only for males (p = 0.001). No statistically significant association was observed between the intervention and the head circumference or length (p = 0.211; 0.597). The weaning rate at the end of follow-up was similar in both groups. Conclusions Breastmilk supplementation may improve the weight gain of very low birthweight preterm infants up to six months corrected gestational age. This effect differed by sex and was considered significant only for males

Leishmania parasites are transmitted to their vertebrate hosts by infected Phlebotomine sand flies during the blood meal of the flies. Sand fly saliva is known to enhance Leishmania spp. infection, while pre-exposure to saliva protects mice against parasitic infections. We demonstrated that inoculating SGE once (SGE-1X) or three times (SGE-3X), which represented a co-inoculation or a pre-exposure to saliva, respectively, resulted in different cellular infiltrate profiles. Whereas SGE-1X led to the recruitment of all leucocytes subtypes including CD4+ T cells, CD4+CD25+ T cells, dendritic cells, macrophages and neutrophils, the immune cell profile in the SGE-3X group differed dramatically, as CD4+ T cells, CD4+CD25+ T cells, dendritic cells, macrophages and neutrophils were decreased and CD8+ T cells were increased. The SGE-1X group did not show differences in the ear lesion size; however, the SGE-1X group harbored a higher number of parasites. On the other hand, the SGE-3X group demonstrated a protective effect against parasitic disease, as the parasite burden was lower even in the earlier stages of the infection, a period in which the SGE-1X group presented with larger and more severe lesions. These effects were also reflected in the cytokine profiles of both groups. Whereas the SGE-1X group presented with a substantial increase in IL-10 production, the SGE-3X group showed an increase in IFN-γ production in the draining lymph nodes. Analysis of the inflammatory cell populations present within the ear lesions, the SGE-1X group showed an increase in CD4+FOXP3+ cells, whereas the CD4+FOXP3+ population was reduced in the SGE-3X group. Moreover, CD4+ T cells and CD8+ T cells producing IFN-γ were highly detected in the ears of the SGE-3X mice prior to infection. In addition, upon treatment of SGE-3X mice with anti-IFN-γ monoclonal antibody, we observed a decrease in the protective effect of SGE-3X against L. braziliensis infection.\ud

Diversidade de mamíferos fósseis de grande porte do quaternário na porção sudoeste do estado de Mato Grosso do Sul
A porção Sudoeste do Estado de Mato Grosso do Sul tem revelado um potencial paleontológico da megafauna do Quaternário, destacando-se a Serra da Bodoquena e a Bacia Sedimentar do Pantanal. O objetivo deste trabalho foi realizar um levantamento sobre a ocorrência de fósseis da megafauna do Quaternário de Mato do Grosso do Sul e discutir como estas novas descobertas auxiliam na compreensão dos paleoambientes e paleoecologia locais. Apesar de pouco estudadas, a maioria das unidades litoestratigráficas localizadas no Sudoeste do estado têm apresentado registros fossilíferos, principalmente ao longo de canais fluviais e cavernas. Dentre os registros osteológicos fósseis recuperados destacam-se as seguintes Ordens: Xenarthra, Perissodactyla, Cetartiodactyla, Litopterna, Notoungulata, Proboscidea, Carnivora e Crocodilia. Essas ordens conviveram mutuamente em um ambiente submetido às variações paleoclimáticas, especialmente no Pleistoceno de modo a influenciar seu hábito alimentar, comportamental, dispersão e distribuição geográfica.Palavras-Chave: Megafauna do Quaternário, Fósseis, Estado de Mato Grosso do Sul
