    Transcriptional activators and repressors for the neuron-specific expression of a metabotropic glutamate receptor

    Metabotropic glutamate receptor 1 (mGlu1) has a discrete distribution in the central nervous system restricted to neurons. Its expression undergoes important changes during development and in response to physiological and pathological modifications. Here, we have determined the structure of the mGlu1 gene and demonstrated that mGlu1 transcription takes places at alternative first exons. Moreover, we have identified active promoter regions upstream from the two most expressed first exons by means of luciferase reporter gene assays performed in primary cerebellar granule neurons. Targeted mutations of active elements constituting the core promoter and electrophoretic mobility shift assays demonstrated that the factors thyroid transcription factor-1 and CCAAT/enhancer-binding proteins β act synergistically to promote mGlu1 transcription. We have also elucidated the molecular bases for the neuron-specific expression of mGlu1 identifying a neural restrictive silencing element and a regulatory factor for X box element, which suppressed mGlu1 expression in nonneuronal cells. These results reveal the molecular bases for cell- and context-specific expression of an important glutamate receptor critically involved in synaptogenesis, neuronal differentiation, synaptic transmission, and plasticity. © 2007 by The American Society for Biochemistry and Molecular Biology, Inc

    The psycholinguistic and affective structure of words conveying pain

    Despite the flourishing research on the relationships between affect and language, the characteristics of pain-related words, a specific type of negative words, have never been systematically investigated from a psycholinguistic and emotional perspective, despite their psychological relevance. This study offers psycholinguistic, affective, and pain-related norms for words expressing physical and social pain. This may provide a useful tool for the selection of stimulus materials in future studies on negative emotions and/or pain. We explored the relationships between psycholinguistic, affective, and pain-related properties of 512 Italian words (nouns, adjectives, and verbs) conveying physical and social pain by asking 1020 Italian participants to provide ratings of Familiarity, Age of Acquisition, Imageability, Concreteness, Context Availability, Valence, Arousal, Pain-Relatedness, Intensity, and Unpleasantness. We also collected data concerning Length, Written Frequency (Subtlex-IT), N-Size, Orthographic Levenshtein Distance 20, Neighbor Mean Frequency, and Neighbor Maximum Frequency of each word. Interestingly, the words expressing social pain were rated as more negative, arousing, pain-related, and conveying more intense and unpleasant experiences than the words conveying physical pain

    A new test of action verb naming: normative data from 290 italian adults

    Introduction Verbs and nouns can be selectively impaired, suggesting that they are processed, at least in part, by distinct neural structures. While several tests of object naming are available, tasks involving action verb naming with normative data are lacking. We report the construction and standardization of a new test for the assessment of picture naming of actions. Material and methods The test includes 50 stimuli, strictly controlled for several confounding variables. Normative data on 290 Italian subjects pooled across homogenous subgroups for age, sex, and education are reported. Results Multiple regression analyses revealed that age and education significantly correlated with the subject\u2019s score. In particular, increasing age negatively affected performance, while the performance increased with a higher education. Conclusions In the clinical practice, the availability of equivalent scores will help the comparison with performance in the picture naming of objects. This test allows investigating action naming deficits in aphasic patients, in Parkinson\u2019s disease patients and in further neurodegenerative disorders, in which a specific impairment of action verbs is expected, filling a gap in the clinical neuropsychological assessment

    Mammalian models of extended healthy lifespan

    Over the last two centuries, there has been a significant increase in average lifespan expectancy in the developed world. One unambiguous clinical implication of getting older is the risk of experiencing age-related diseases including various cancers, dementia, type-2 diabetes, cataracts and osteoporosis. Historically, the ageing process and its consequences were thought to be intractable. However, over the last two decades or so, a wealth of empirical data has been generated which demonstrates that longevity in model organisms can be extended through the manipulation of individual genes. In particular, many pathological conditions associated with the ageing process in model organisms, and importantly conserved from nematodes to humans, are attenuated in long-lived genetic mutants. For example, several long-lived genetic mouse models show attenuation in age-related cognitive decline, adiposity, cancer and glucose intolerance. Therefore, these long-lived mice enjoy a longer period without suffering the various sequelae of ageing. The greatest challenge in the biology of ageing is to now identify the mechanisms underlying increased healthy lifespan in these model organisms. Given that the elderly are making up an increasingly greater proportion of society, this focused approach in model organisms should help identify tractable interventions that can ultimately be translated to humans

    Ultrafast photodoping and effective Fermi-Dirac distribution of the Dirac particles in Bi2Se3

    We exploit time- and angle- resolved photoemission spectroscopy to determine the evolution of the out-of-equilibrium electronic structure of the topological insulator Bi2Se. The response of the Fermi-Dirac distribution to ultrashort IR laser pulses has been studied by modelling the dynamics of the hot electrons after optical excitation. We disentangle a large increase of the effective temperature T* from a shift of the chemical potential mu*, which is consequence of the ultrafast photodoping of the conduction band. The relaxation dynamics of T* and mu* are k-independent and these two quantities uniquely define the evolution of the excited charge population. We observe that the energy dependence of the non-equilibrium charge population is solely determined by the analytical form of the effective Fermi-Dirac distribution.Comment: 5 Pages, 3 Figure

    Evidence of reduced surface electron-phonon scattering in the conduction band of Bi_{2}Se_{3} by non-equilibrium ARPES

    The nature of the Dirac quasiparticles in topological insulators calls for a direct investigation of the electron-phonon scattering at the \emph{surface}. By comparing time-resolved ARPES measurements of the TI Bi_{2}Se_{3} with different probing depths we show that the relaxation dynamics of the electronic temperature of the conduction band is much slower at the surface than in the bulk. This observation suggests that surface phonons are less effective in cooling the electron gas in the conduction band.Comment: 5 pages, 3 figure

    Form and Function: A Study on the Distribution of the Inflectional Endings in Italian Nouns and Adjectives

    Inflectional values, such as singular and plural, sustain agreement relations between constituents in sentences, allowing sentence parsing and prediction in online processing. Ideally, these processes would be facilitated by a consistent and transparent correspondence between the inflectional values and their form: for example, the value of plural should always be expressed by the same ending, and that ending should only express plural. Experimental research reports higher processing costs in the presence of a non-transparent relation between forms and values. While this effect was found in several languages, and typological research shows that consistency is far from common in morphological paradigms, it is still somewhat difficult to precisely quantify the transparency degree of the inflected forms. Furthermore, to date, no accounts have quantified the transparency in inflection with regard to the declensional classes and the extent to which it is expressed across different parts of speech, depending on whether these act as controllers of the agreement (e.g., nouns) or as targets (e.g., adjectives). We present a case study on Italian, a language that marks gender and number features in nouns and adjectives. This work provides measures of the distribution of forms in the noun and adjective inflection in Italian, and quantifies the degree of form-value transparency with respect to inflectional endings and declensional classes. In order to obtain these measures, we built Flex It, a dedicated large-scale database of inflectional morphology of Italian, and made it available, in order to sustain further theoretical and empirical research

    Preliminary study on MC1R polymorphism in some cattle breeds raised in Italy

    Ricerche preliminari sul polimorfismo del gene MC1R in alcune razze bovine allevate in Italia – Il gene MC1R è stato analizzato in 193 soggetti appartenenti a 8 razze bovine, tramite PCR, per la presenza di due mutazioni ad effetto fenotipico noto sulla pigmentazione del mantello: la delezione G310 e la sostituzione T296C, associate rispettivamente al fenotipo feomelanico (e) ed eumelanico nero (Ed). Sessanta soggetti di razza Limousine e Pezzata Rossa Italiana presentano genotipo e/e; 27 soggetti di razza Frisona Italiana mostrano genotipo Ed/Ed mentre 2 genotipo Ed/e. Gli 84 soggetti appartenenti alle razze Cabannina, Chianina, Marchigiana e Piemontese non presentano tali mutazioni, analogamente a 18 soggetti di razza Romagnola, nella quale però si sono anche osservati 2 soggetti portatori dell’allele e allo stato eterozigote
