    Low Velocity Granular Drag in Reduced Gravity

    We probe the dependence of the low velocity drag force in granular materials on the effective gravitational acceleration (geff) through studies of spherical granular materials saturated within fluids of varying density. We vary geff by a factor of 20, and we find that the granular drag is proportional to geff, i.e., that the granular drag follows the expected relation Fprobe = {\eta} {\rho}grain geff dprobe hprobe^2 for the drag force, Fprobe on a vertical cylinder with depth of insertion, hprobe, diameter dprobe, moving through grains of density {\rho}grain, and where {\eta} is a dimensionless constant. This dimensionless constant shows no systematic variation over four orders of magnitude in effective grain weight, demonstrating that the relation holds over that entire range to within the precision of our data

    Nel 1988, l\u2019Organizzazione mondiale della Sanit\ue0 (OMS) ha approvato il piano di eradicazione mondiale della poliomielite con l\u2019obiettivo di annullare la circolazione del virus a partire dall\u2019anno 2000. Tale target, per motivi economici, organizzativi, culturali, bellici e financo religiosi, \ue8 stato successivamente spostato in avanti negli anni fino a giungere attualmente al 2015. A seguito di questo impegno, negli anni si \ue8 registrata una riduzione dell\u2019incidenza di poliomielite di oltre il 99%, passando dai 350.000 casi annui di polio paralitica documentati nel 1988 in 125 paesi, alle poche centinaia di casi riscontrati nel 2013 sia negli ultimi 3 paesi endemici (Nigeria, Afghanistan, Pakistan) sia in paesi polio-free spesso confinanti con quelli endemici ed interessati da un corollario di epidemie di dimensioni variabili

    Shear-promoted drug encapsulation into red blood cells: a CFD model and μ-PIV analysis

    The present work focuses on the main parameters that influence shear-promoted encapsulation of drugs into erythrocytes. A CFD model was built to investigate the fluid dynamics of a suspension of particles flowing in a commercial micro channel. Micro Particle Image Velocimetry (μ-PIV) allowed to take into account for the real properties of the red blood cell (RBC), thus having a deeper understanding of the process. Coupling these results with an analytical diffusion model, suitable working conditions were defined for different values of haematocrit

    In the industrial panorama, many processes operate under time-varying conditions. Adapt¬ing high-performance diagnostic techniques under these relatively more complex situations is ur¬gently needed to mitigate the risk of false alarms. Attention is being paid to fault anticipation, requiring an in-depth study of prediction techniques. Predicting remaining life before the occurrence of faults allows for a comprehensive maintenance management protocol and facilitates the wear management of the machine, avoiding faults that could permanently compromise the integrity of such machinery. This study focuses on canonical variate analysis for fault detection in processes operating under time-varying conditions and on its contribution to the diagnostic and prognostic analysis, the latter of which was performed with machine learning techniques.     Fault diagnosis of a granulator operating under time-varying conditions using canonical variate analysis

    Granulators play a key role in many pharmaceutical processes because they are involved in the production of tablets and capsule dosage forms. Considering the characteristics of the production processes in which a granulator is involved, proper maintenance of the latter is relevant for plant safety. During the operational phase, there is a high risk of explosion, pollution, and contamination. The nature of this process also requires an in-depth examination of the time-dependence of the process variables. This study proposes the application of canonical variate analysis (CVA) to perform fault detection in a granulation process that operates under time-varying conditions. Beyond this, a different approach to the management of process non-linearities is proposed. The novelty of the study is in the application of CVA in this kind of process, because it is possible to state that the actual literature on the theme shows some limitations of CVA in such processes. The aim was to increase the applicability of CVA in variable contexts, with simple management of non-linearities. The results, considering process data from a pharmaceutical granulator, showed that the proposed approach could detect faults and manage non-linearities, exhibiting future scenarios for more performing and automatic monitoring techniques of time-varying processes

    A novel stepwise micro-TESE approach in non obstructive azoospermia

    Background: The purpose of the study was to investigate whether micro-TESE can improve sperm retrieval rate (SRR) compared to conventional single TESE biopsy on the same testicle or to contralateral multiple TESE, by employing a novel stepwise micro-TESE approach in a population of poor prognosis patients with non-obstructive azoospermia (NOA). Methods: Sixty-four poor prognosis NOA men undergoing surgical testicular sperm retrieval for ICSI, from March 2007 to April 2013, were included in this study. Patients inclusion criteria were a) previous unsuccessful TESE, b) unfavorable histology (SCOS, MA, sclerahyalinosis), c) Klinefelter syndrome. We employed a stepwise micro-TESE consisting three-steps: 1) single conventional TESE biopsy; 2) micro-TESE on the same testis; 3) contralateral multiple TESE. Results: SRR was 28.1 % (18/64). Sperm was obtained in both the initial single conventional TESE and in the following micro-TESE. The positive or negative sperm retrieval was further confirmed by a contralateral multiple TESE, when performed. No significant pre-operative predictors of sperm retrieval, including patients’ age, previous negative TESE or serological markers (LH, FSH, inhibin B), were observed at univariate or multivariate analysis. Micro-TESE (step 2) did not improve sperm retrieval as compared to single TESE biopsy on the same testicle (step 1) or multiple contralateral TESE (step 3). Conclusions: Stepwise micro-TESE could represent an optimal approach for sperm retrieval in NOA men. In our view, it should be offered to NOA patients in order to gradually increase surgical invasiveness, when necessary. Stepwise micro-TESE might also reduce the costs, time and efforts involved in surgery

    Evaluation of the burden of HPV-related hospitalizations as a useful tool to increase awareness: 2007–2017 data from the sicilian hospital discharge records

    In light of the implementation of human papillomavirus (HPV) prevention strategies, epidemiological studies in different geographical areas are required in order to assess the impact of HPV-related diseases. The purpose of the present study was to describe the burden of HPV-related hospitalizations in Sicily. A retrospective observational study estimated 43,531 hospitalizations attributable to HPV from 2007 to 2017. During the observed period, there was a decrease for all HPV-related conditions with a higher reduction, among neoplasms, for cervical cancer (annual percent change (APC) = −9.9%, p < 0.001). The median age for cervical cancer was 45 years old, with an increasing value from 43 to 47 years (p < 0.001). The age classes with greater decreases in hospital admissions for invasive cancers were women aged 35 years or more (APC range from −5.5 to −9.86) and 25–34 years old (APC = −11.87, p < 0.001) for women with cervical carcinoma in situ. After ten years for vaccine introduction and sixteen years for cervical cancer screening availability, a relatively large decrease in hospital admissions for cervical cancer and other HPV-related diseases in Sicily was observed. Some clinical characteristics of hospitalization, such as increasing age, are suggestive clues for the impact of preventive strategies, but further research is needed to confirm this relationship

    Targeted delivery of anti-inflammatory therapy to rheumatoid tissue by fusion proteins containing an IL-4-linked synovial targeting peptide

    We provide first-time evidence that the synovial endothelium-targeting peptide (SyETP) CKSTHDRLC successfully delivers conjugated IL-4 to human rheumatoid synovium transplanted into SCID mice. SyETP, previously isolated by in vivo phage display and shown to preferentially localize to synovial xenografts, was linked by recombinant technology to hIL-4 via an MMP-cleavable sequence. Both IL-4 and the MMP-cleavable sequence were shown to be functional. IL-4-SyETP augmented production of IL-1ra by synoviocytes stimulated with IL-1[beta] in a dose-dependent manner. In vivo imaging confirmed increased retention of SyETP-linked-IL-4 in synovial grafts which was enhanced by increasing number of copies (one to three) in the constructs. Strikingly, SyETP delivered bioactive IL-4 in vivo as demonstrated by increased pSTAT6 in synovial grafts. Thus, this study provides proof of concept for peptide-tissue-specific targeted immunotherapy in rheumatoid arthritis. This technology is potentially applicable to other biological therapies providing enhanced potency to inflammatory sites and reducing systemic toxicity
