    Spreading in Social Systems: Reflections

    In this final chapter, we consider the state-of-the-art for spreading in social systems and discuss the future of the field. As part of this reflection, we identify a set of key challenges ahead. The challenges include the following questions: how can we improve the quality, quantity, extent, and accessibility of datasets? How can we extract more information from limited datasets? How can we take individual cognition and decision making processes into account? How can we incorporate other complexity of the real contagion processes? Finally, how can we translate research into positive real-world impact?     Preterm birth is not associated with asymptomatic/mild SARS-CoV-2 infection per se: Pre-pregnancy state is what matters

    Evidence for the real impact of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection on preterm birth is unclear, as available series report composite pregnancy outcomes and/or do not stratify patients according to disease severity. The purpose of the research was to determine the real impact of asymptomatic/mild SARS-CoV-2 infection on preterm birth not due to maternal respiratory failure. This case-control study involved women admitted to Sant Anna Hospital, Turin, for delivery between 20 September 2020 and 9 January 2021. The cumulative incidence of Coronavirus disease-19 was compared between preterm birth (case group, n = 102) and full-term delivery (control group, n = 127). Only women with spontaneous or medically-indicated preterm birth because of placental vascular malperfusion (pregnancy-related hypertension and its complications) were included. Current or past SARS-CoV-2 infection was determined by nasopharyngeal swab testing and detection of IgM/IgG antibodies in blood samples. A significant difference in the cumulative incidence of Coronavirus disease-19 between the case (21/102, 20.5%) and the control group (32/127, 25.1%) (P= 0.50) was not observed, although the case group was burdened by a higher prevalence of three known risk factors (body mass index > 24.9, asthma, chronic hypertension) for severe Coronavirus disease-19. Logistic regression analysis showed that asymptomatic/mild SARS-CoV-2 infection was not an independent predictor of spontaneous and medically-indicated preterm birth due to pregnancy-related hypertension and its complications (0.77; 95% confidence interval, 0.41-1.43). Pregnant patients without comorbidities need to be reassured that asymptomatic/mild SARS-CoV-2 infection does not increase the risk of preterm delivery. Preterm birth and severe Coronavirus disease-19 share common risk factors (i.e., body mass index > 24.9, asthma, chronic hypertension), which may explain the high rate of indicated preterm birth due to maternal conditions reported in the literature

    The influence of climatic factors on downy mildew in two vineyards from France and Romania

    The objective of this study was to evaluate the influence of climate variability from the last two years, namely 2021-2022, on the development of the pathogen Plasmopara viticola in two vineyards renowned for the quality of their wines, Murfatlar (Romania) and Val de Loire (France). The study was carried out on four grapevine cultivars: 'Sauvignon blanc' and 'Cabernet sauvignon', authorized for the production of CDO Murfatlar wines; and Cot and Chenin, authorized for the production of CDO Touraine wines. The results regarding the appearance and development of the disease on leaves and bunches in different grapevine development stages, on the two variants of the experiments (treated and untreated) highlighted different levels of attack depending on the distribution of precipitation and the evolution of average air temperatures during the vegetation period. The excess rainfall regime in 2021 of 339.3 mm in Murfatlar and 351.8 mm in Val de Loire, and the air temperatures of 20-25°C generated optimal conditions for the spread of disease on the leaves in the untreated variant, reaching a maximum of 48.3% in Murfatlar and 9.5% in Val de Loire during veraison. The disease also spread to the bunches, reaching a maximum of 86.2% in Murfatlar, endangering almost completely the production of grapes. In the case of the variant where phytosanitary treatments were applied, the degree of attack was a maximum of 4%. In 2022, against the background of quantitatively reduced precipitation compared to 2021, with 64.15% in Murfatlar and 45.01% in Val de Loire, the first oil spots were identified in the untreated variant during BBCH 55 (the emergence of inflorescences), having an insignificant spread, reaching a maximum of 8.9% in the cluster compaction phenophase (BBCH 77)

    From Uterus to Brain: An Update on Epidemiology, Clinical Features, and Treatment of Brain Metastases From Gestational Trophoblastic Neoplasia

    In this review, we provide the state of the art about brain metastases (BMs) from gestational trophoblastic neoplasia (GTN), a rare condition. Data concerning the epidemiology, clinical presentation, innovations in therapeutic modalities, and outcomes of GTN BMs are comprehensively presented with particular attention to the role of radiotherapy, neurosurgery, and the most recent chemotherapy regimens. Good response rates have been achieved thanks to multi-agent chemotherapy, but brain involvement by GTNs entails significant risks for patients’ health since sudden and extensive intracranial hemorrhages are possible. Moreover, despite the evolution of treatment protocols, a small proportion of these patients ultimately develops a resistant disease. To tackle this unmet clinical need, immunotherapy has been recently proposed. The role of this novel option for this subset of patients as well as the achieved results so far are also discussed

    Is there a place for immune checkpoint inhibitors in vulvar neoplasms? A state of the art review

    Vulvar cancer (VC) is a rare neoplasm, usually arising in postmenopausal women, although human papilloma virus (HPV)-associated VC usually develop in younger women. Incidences of VCs are rising in many countries. Surgery is the cornerstone of early-stage VC management, whereas therapies for advanced VC are multimodal and not standardized, combining chemotherapy and radiotherapy to avoid exenterative surgery. Randomized controlled trials (RCTs) are scarce due to the rarity of the disease and prognosis has not improved. Hence, new therapies are needed to improve the outcomes of these patients. In recent years, improved knowledge regarding the crosstalk between neoplastic and tumor cells has allowed researchers to develop a novel therapeutic approach exploiting these molecular interactions. Both the innate and adaptive immune systems play a key role in anti-tumor immunesurveillance. Immune checkpoint inhibitors (ICIs) have demonstrated efficacy in multiple tumor types, improving survival rates and disease outcomes. In some gynecologic cancers (e.g., cervical cancer), many studies are showing promising results and a growing interest is emerging about the potential use of ICIs in VC. The aim of this manuscript is to summarize the latest developments in the field of VC immunoncology, to present the role of state-of-the-art ICIs in VC management and to discuss new potential immunotherapeutic approaches

    The study regarding the agrobiological and technological properties of some clonal elites for table grapes, under the conditions of the Murfatlar vineyard

    At the Research and Development Station for Viticulture and Vinification Murfatlar, in the period 2019 – 2022, was carried out clonal selection work on two cultivars for table grapes, respectively: 'Victoria' and 'Coarnă neagră'. The research consisted of the comparative study of five clonal elites of each cultivar, regarding their agrobiological and technological qualities, correlated with the specific climatic conditions of the Murfatlar vineyard. The five elites of each cultivar were studied and analyzed compared to the characters of the initial cultivar (the control). In the case of the 'Victoria' cultivar, out of the five elites under study, the one that stands out is the clonal elite ‘Victoria 57/4/7', which has great vigor, good fertility 63-73%, and high productivity. The preservability of grapes on the bush is long, about 30 days. Grape production is high, over 16-18 t/ha, of which 83-85% is commodity production. When are ripe for consumption, the grapes accumulate 140-150 g/l sugars, and the acidity is relatively low 3.8-4 g/l H2SO4. Regarding the cultivar 'Coarnă neagră', of the five elites under study, the one that stands out is the clonal elite 'Coarnă neagră’ 14/5/22', which has great vigor, high fertility, and medium ripening. The grape is large, cylindrical or cylindrical-conical in shape. Grape production is high, 20-25 t/ha, of which 85% is commodity production. The preservability on the bush is high, between 25-30 days. When are ripe for consumption, grapes accumulate 170–180 g/l sugars, and 5 g/l H2SO4 total acidity

    The sovereign debt crisis: the impact on the intermediation model of Italian banks

    The aim of the contribute is to analyze the impact of the financial crisis, in particular since the start of the sovereign debt phase, on Italian banks and their intermediation model. Italian banks\u2019 specific business model explains why they suffered less than those of other countries during the first phase of the crisis, requiring one of the lowest levels of public facilities in the EC as compared to GDP. Most of these same characteristics have changed from positive to negative factors since the sovereign debt crisis, which hit Italy hard, affecting first banks\u2019 liquidity and secondly the cost and volumes of funding and loans. Italian banks are now facing the effects of the double-dip recession, which has significantly weakened businesses and households, their key customer segments, and their borrowing and saving capability, with an increasing rate of non-performing loans. This situation is impairing the sustainability of the \u201ctraditional\u201d intermediation model and means that banks must introduce strategies for significantly modifying the banking business model they adopt

    CD36 deficiency attenuates experimental mycobacterial infection

    <p>Abstract</p> <p>Background</p> <p>Members of the CD36 scavenger receptor family have been implicated as sensors of microbial products that mediate phagocytosis and inflammation in response to a broad range of pathogens. We investigated the role of CD36 in host response to mycobacterial infection.</p> <p>Methods</p> <p>Experimental <it>Mycobacterium bovis </it>Bacillus Calmette-Guérin (BCG) infection in <it>Cd36<sup>+/+ </sup></it>and <it>Cd36<sup>-/- </sup></it>mice, and <it>in vitro </it>co-cultivation of <it>M. tuberculosis</it>, BCG and <it>M. marinum </it>with <it>Cd36<sup>+/+ </sup></it>and <it>Cd36<sup>-/-</sup></it>murine macrophages.</p> <p>Results</p> <p>Using an <it>in vivo </it>model of BCG infection in <it>Cd36<sup>+/+ </sup></it>and <it>Cd36<sup>-/- </sup></it>mice, we found that mycobacterial burden in liver and spleen is reduced (83% lower peak splenic colony forming units, p < 0.001), as well as the density of granulomas, and circulating tumor necrosis factor (TNF) levels in <it>Cd36<sup>-/- </sup></it>animals. Intracellular growth of all three mycobacterial species was reduced in <it>Cd36<sup>-/- </sup></it>relative to wild type <it>Cd36<sup>+/+ </sup></it>macrophages <it>in vitro</it>. This difference was not attributable to alterations in mycobacterial uptake, macrophage viability, rate of macrophage apoptosis, production of reactive oxygen and/or nitrogen species, TNF or interleukin-10. Using an <it>in vitro </it>model designed to recapitulate cellular events implicated in mycobacterial infection and dissemination <it>in vivo </it>(i.e., phagocytosis of apoptotic macrophages containing mycobacteria), we demonstrated reduced recovery of viable mycobacteria within <it>Cd36<sup>-/- </sup></it>macrophages.</p> <p>Conclusions</p> <p>Together, these data indicate that CD36 deficiency confers resistance to mycobacterial infection. This observation is best explained by reduced intracellular survival of mycobacteria in the <it>Cd36<sup>-/- </sup></it>macrophage and a role for CD36 in the cellular events involved in granuloma formation that promote early bacterial expansion and dissemination.</p

    Association of Cholinergic Basal Forebrain Volume and Functional Connectivity with Markers of Inflammatory Response in the Alzheimer's Disease Spectrum

    BACKGROUND: Inflammation has been described as a key pathogenic event in Alzheimer's disease (AD), downstream of amyloid and tau pathology. Preclinical and clinical data suggest that the cholinergic basal forebrain may moderate inflammatory response to different pathologies. OBJECTIVE: To study the association of cholinergic basal forebrain volume and functional connectivity with measures of neuroinflammation in people from the AD spectrum. METHODS: We studied 261 cases from the DELCODE cohort, including people with subjective cognitive decline, mild cognitive impairment, AD dementia, first degree relatives, and healthy controls. Using Bayesian ANCOVA, we tested associations of MRI indices of cholinergic basal forebrain volume and functional connectivity with cerebrospinal fluid (CSF) levels of sTREM2 as a marker of microglia activation, and serum levels of complement C3. Using Bayesian elastic net regression, we determined associations between basal forebrain measures and a large inflammation marker panel from CSF and serum. RESULTS: We found anecdotal to moderate evidence in favor of the absence of an effect of basal forebrain volume and functional connectivity on CSF sTREM2 and serum C3 levels both in Aβ42/ptau-positive and negative cases. Bayesian elastic net regression identified several CSF and serum markers of inflammation that were associated with basal forebrain volume and functional connectivity. The effect sizes were moderate to small. CONCLUSION: Our data-driven analyses generate the hypothesis that cholinergic basal forebrain may be involved in the neuroinflammation response to Aβ42 and phospho-tau pathology in people from the AD spectrum. This hypothesis needs to be tested in independent samples
