    O4 - Exploring the biological properties and regenerative potential of biomaterials using cell culture models

    Life expectancy has improved signifcantly and, along with the declining birthrate, has contributed to the aging of populations, especially in industrialized countries. Alas, aging is intrinsically associated with the incidence of health problems including bone and tooth loss that require suitable solutions to support the quality of life. To meet these demands, signifcant research eforts have been undertaken to develop novel biomaterials, both orthopedic and dental implants. The feld of biomaterials for bone tissue engineering is increasingly evolving. The most recent generations of biomaterials have increasingly more activity and interaction with the biological environment and stimulate the regeneration of functional tissue. Natural polymers and compounds have been combined with each other to improve workability and are strategically integrated with ceramics or bioactive glasses to reinforce the structure of the fnal system, thus producing composites with a better mechanical performance. Our research group has been focused on the biological characterization of diferent added-value materials and composites, namely by evaluating their antimicrobial, biocompatibility, and regenerative properties. Some of our recent work results allowed us to conclude that marine fungal extracts, as well as sol–gel-derived bioactive glass nanoparticles, have inhibitory efects on the growth of C. albicans and E. faecalis (main pathogens in persistent root canal infections). Additionally, we have characterized cuttlefsh bone powders for endodontic applications. We are also committed to developing strategies for monitoring cell response to these biomaterials at the molecular level that could be used to follow infammation and osteoconduction.info:eu-repo/semantics/publishedVersio

    P8 - Marine fungi exhibit antimicrobial activity against human oral pathogens

    The emergence of resistance to antibiotics and antimycotics has become a challenge in the treatment of infectious diseases, including infections of the oral cavity. Marine fungi are a source of novel biologically active compounds, namely in what concerns the development of antimicrobial and anticancer solutions. Our study aimed to test the antimicrobial activity and the cytotoxicity of the extracts of the two recent identified species of marine fungi, Penicillum lusitanum and Aspergillus affinis. Candida spp. and Enterococcus faecalis isolated from oral pathologies were included to evaluate the antimicrobial potential of the marine fungi by the disk diffusion assay. The cytotoxicity of the effective concentrations of the extract was tested using the Vero cell line (ECACC 88020401, African Green Monkey Kidney cells, GMK clone), according to the ISO 10993-5. The extracts of P. lusitanum and A. affinis were active against C. albicans and E. faecalis, respectively. Penicillum lusitanum active extracts are non-cytotoxic, in contrast to A. affinis extracts that showed high cytotoxic effects on Vero cells, for all concentrations tested. The results on the biological characterization of the P. lusitanumextract are promising and support the development of new disinfecting solutions that may be used during root canal therapy cleaning and shaping.info:eu-repo/semantics/publishedVersio

Publisher Copyright: © 2022, The Author(s).Laser-induced graphene (LIG) is as a promising material for flexible microsupercapacitors (MSCs) due to its simple and cost-effective processing. However, LIG-MSC research and production has been centered on non-sustainable polymeric substrates, such as polyimide. In this work, it is presented a cost-effective, reproducible, and robust approach for the preparation of LIG structures via a one-step laser direct writing on chromatography paper. The developed strategy relies on soaking the paper in a 0.1 M sodium tetraborate solution (borax) prior to the laser processing. Borax acts as a fire-retardant agent, thus allowing the laser processing of sensitive substrates that other way would be easily destroyed under the high-energy beam. LIG on paper exhibiting low sheet resistance (30 Ω sq−1) and improved electrode/electrolyte interface was obtained by the proposed method. When used as microsupercapacitor electrodes, this laser-induced graphene resulted in specific capacitances of 4.6 mF cm−2 (0.015 mA cm−2). Furthermore, the devices exhibit excellent cycling stability (> 10,000 cycles at 0.5 mA cm−2) and good mechanical properties. By connecting the devices in series and parallel, it was also possible to control the voltage and energy delivered by the system. Thus, paper-based LIG-MSC can be used as energy storage devices for flexible, low-cost, and portable electronics. Additionally, due to their flexible design and architecture, they can be easily adapted to other circuits and applications with different power requirements.     Monitoring biotechnological processes through quantitative image analysis: application to 2-phenylethanol production by Yarrowia lipolytica

    Available online 20 March 2023Quantitative image analysis (QIA) is a simple and automated tool for process monitoring that, when combined with chemometric techniques, enables the association of changes in microbiota morphology to various operational parameters. To that effect, principal component analysis, multilinear regression, and ordinary least squares methods were applied to the obtained dataset of the biotransformation conditions for Y. lipolytica through the monitor of yeast morphology, substrates (glycerol, L-phenylalanine - L-Phe) consumption and metabolites (2-phenylethanol 2-PE) production was developed. Glycerol and L-Phe were successfully monitored by the proposed approach, though with a lower monitoring ability for 2-PE, and mostly related to yeast and cluster size and proportion, yeasts contents and cluster morphology. The chemometric approach also allowed to identify significant morphological modifications related with the change in the stirring speed in the experiments at 600rpm, 600/400rpm (600rpm for 24h, and 400rpm until the end of the experiment) and in pH from 5.5 to 7.5. This work demonstrated, for the first time, that QIA combined with chemometric analysis can be considered a valuable tool to monitor biotechnological processes, namely the 2-PE production by Y. lipolytica, by analyzing yeast and cluster morphology.info:eu-repo/semantics/publishedVersio

    P9 - Antimicrobial properties and bioactivity potential of smart nanoparticles for dental applications

    Tooth decay is one of the greatest causes of tooth loss in the world. This not only afects the patient’s quality of life but also carries an economic burden associated with the need for multiple reinterventions. Endodontic treatment aims to preserve teeth by cleaning, disinfecting and flling/sealing the root canal. Despite the high success rate of endodontic treatment, failures do occur in a large number of cases. Several new biomaterials for dentistry have been developed, however their bioactivity is often misunderstood. Our work focuses on the biological characterization of novel bioactive glass nanoparticles, including the evaluation of their antimicrobial and biocompatibility properties. Candida albicans (ATCC 11225) and Enterococcus faecalis (ATCC 29212) were included to evaluate the antimicrobial potential by the drop plate method [1]. The cytotoxicity was tested using the MC3T3-E1 cell line, through the resazurin reduction assay. The novel bioactive glass nanoparticles demonstrated antimicrobial activity against C. albicans and E. faecalis, being able to inhibit their growth but also, in some incubation times, decreased the survival of these microorganisms. After 24 h of incubation of MC3T3-E1 osteoblast cells with bioactive glass nanoparticles conditioned medium, around 48% cell viability was achieved.     White spot lesions: diagnosis and treatment - a systematic review

    BACKGROUND: White spot lesions represent the first stage of caries and their prevalence has been increasing in recent years, particularly in patients undergoing orthodontic treatment. DIferential diagnosis and lesion activity are essential to decide on the clinical approaches to treatment. The aim of this study is to understand if the new diagnostic tools such as fluorescence, microradiography and computed microtomography have the potential to change the conventional treatment of white spots". METHODS: A systematic search of available studies in the literature was carried out, using PRISMA guidelines, in Pubmed and Scopus electronic databases and manually to identify relevant articles to answer the PICO question: "Do the new diagnostic tools have the potential to change the conventional treatment of white spots?". This systematic review included randomized controlled trials (RCT), cross-sectional and longitudinal studies complying with the following inclusion criteria: (i) studies in humans, (ii) studies about white spot lesions, (iii) studies published between 2012 and 2023, (iv) studies having both diagnosis and treatment and (v) studies with full text available. In this review we excluded other systematic reviews of clinical trials and in vitro studies. The RoB tool was used to assess the risk of bias. RESULTS: The systematic literature search identified 143 potentially relevant references, which after applying the exclusion criteria, resulted in 20 articles. Regarding diagnostic methods, most articles found were based on conventional methods of visual examination (n:10) or fluorescence (n:7). The least referenced diagnostic techniques were based on the use of clinical photographs (n:2), cross-sectional microradiography (n:1) and computed microtomography (n:1). The use of DIAGNOdent was reported by 3 in vitro studies. With regard to therapies, most studies reported the use of infiltrating resin (n:7) and fluoride-based products (n:5). Other studies have reported the use of self-assembling peptide P11-4 (n:1), home care (n:1), casein phosphopeptide-amorphous calcium phosphate (n:2) and hydrochloric acid (n:1). Combination therapies were also considered. CONCLUSION: Diagnostic tool does not have the potential to change the form of treatment, whether it is a conventional method or a more differentiated one.info:eu-repo/semantics/publishedVersio
