    Super-resolution imaging of live sperm reveals dynamic changes of the actin cytoskeleton during acrosomal exocytosis

    Filamentous actin (F-actin) is a key factor in exocytosis in many cell types. In mammalian sperm, acrosomal exocytosis (denoted the acrosome reaction or AR), a special type of controlled secretion, is regulated by multiple signaling pathways and the actin cytoskeleton. However, the dynamic changes of the actin cytoskeleton in live sperm are largely not understood. Here, we used the powerful properties of SiR-actin to examine actin dynamics in live mouse sperm at the onset of the AR. By using a combination of super-resolution microscopy techniques to image sperm loaded with SiR-actin or sperm from transgenic mice containing Lifeact-EGFP, six regions containing F-actin within the sperm head were revealed. The proportion of sperm possessing these structures changed upon capacitation. By performing live-cell imaging experiments, we report that dynamic changes of F-actin during the AR occur in specific regions of the sperm head. While certain F-actin regions undergo depolymerization prior to the initiation of the AR, others remain unaltered or are lost after exocytosis occurs. Our work emphasizes the utility of live-cell nanoscopy, which will undoubtedly impact the search for mechanisms that underlie basic sperm functions.Fil: Romarowski, Ana.     Classification of strengthening exercises of the lower member posterior chain, based on the amplitude of muscular activation in amateur soccer players between 18-25 years

    The hamstring injuries are recurrent in the sports disciplines of teams such as football, with rehabilitation being a key point to prevent future alterations. Within this process, the strengthening paradigms are rather empirical, therefore, criteria based on the exercises used are required. In this context, the objective of the present investigation is to classify the strengthening exercises of the posterior lower limb chain, based on the amplitude of muscular activation, in amateur soccer players of the Andrés Bello University (UNAB), for which a non-experimental, descriptive, cross-sectional design. The study population was 30 university students, who were studying from first to fifth year, during the academic period 2017. The participants were summoned to the Laboratory of Rehabilitation Sciences, where after signing the informed consent they had to perform a phase of heating on a cycle ergometer, then the MCIV (maximum voluntary isometric contraction) of the semimembranous, semitendinous, biceps femoral and gluteus maximus muscles was calculated, finally applying a battery of strengthening exercises, which were classified according to the intensity of muscle activation. In relation to this, it was obtained that 8 of the 13 exercises implemented were classified as low intensity (p <0.05), 3 of medium intensity (p <0.05) and 2 of high intensity (p <0.05), while the peak of amplitude of the semimembranous, semitendinous and biceps femoral muscles moved to the right in temporal terms, to the extent that the exercises classified as low, medium and high intensity are compared.Las lesiones de isquiotibiales son recurrentes en disciplinas deportivas de equipos como el fútbol, siendo la rehabilitación un punto clave para prevenir alteraciones futuras. Dentro de este proceso, los paradigmas de fortalecimiento son más bien empíricos, por lo tanto, criterios fundamentados en torno a los ejercicios utilizados son requeridos. Bajo este contexto, el objetivo de la presente investigación fue clasificar los ejercicios de fortalecimiento de la cadena posterior de miembro inferior, en base a la amplitud de activación muscular, en futbolistas amateur de la Universidad Andrés Bello (UNAB), para lo cual se utilizó un diseño no experimental de tipo descriptivo y de corte transversal. La población de estudio, fueron 30 estudiantes universitarios, que estaban cursando de primero a quinto año, durante el periodo académico 2017. Los participantes fueron citados al Laboratorio de Ciencias de Rehabilitación de la UNAB, en donde luego de firmar el consentimiento informado debieron realizar una fase de calentamiento en cicloergómetro, posteriormente se calculó la MCIV (Máxima contracción isométrica voluntaria) de los músculos semimembranoso, semitendinoso, bíceps femoral y glúteo mayor, para finalmente aplicar una batería de ejercicios de fortalecimientos, los cuales fueron clasificados según la intensidad de activación muscular. En relación con esto, se obtuvo que 8 de los 13 ejercicios implementados fueron clasificados de baja intensidad (p<0.05), 3 de mediana intensidad (p<0.05) y 2 de alta intensidad (p<0.05), mientras que el peak de amplitud de los músculos semimembranoso, semitendinoso y bíceps femoral se desplazó a la derecha en términos temporales, en la medida que se comparan los ejercicios clasificados como de baja, media y alta intensidad

    El cerro de la Judería de Atienza: primera aproximación arqueológica

    The archaeological excavations carried out in the medieval Jewish quarter of Atienza (Guadalajara) in 2022 constitute the first comprehensive approach to the archaeological site of the walled quarter in which this religious minority resided. Although there are numerous reports from written sources of the presence of an important Jewish community in Atienza during the late medieval centuries, until now there had only been a few superficial approaches to its archaeological remains. The planned archaeological excavation summarised here, carried out as part of a wider territorial study project, has documented a 13th and 14th century domestic context in the central sector of the Jewish quarter. The analysis of the exhumed structures and the archaeological materials found in context, especially ceramics and archaeozoology, allows us to establish relevant considerations regarding the processes of occupation and abandonment of the quarter and about the central question of the archaeological identification of the late medieval Jewish communities. The site is also an exceptional space for the collection of archaeobiological samples, whose potential for advancing the study of the relationship between these medieval communities and the natural environment has barely been glimpsed.Las excavaciones arqueológicas realizadas en la judería medieval de Atienza (Guadalajara) en 2022 constituyen la primera aproximación integral al conjunto arqueológico del barrio amurallado en el que residió esa minoría religiosa. A pesar de que son numerosas las noticias procedentes de las fuentes escritas que informan de la presencia de una importante comunidad judía en Atienza durante los siglos bajomedievales, hasta ahora apenas se habían hecho algunas aproximaciones superficiales a sus restos arqueológicos. La excavación arqueológica programada que aquí se sintetiza, realizada en el marco de un proyecto de estudio territorial más amplio, ha documentado un espacio doméstico de los siglos XIII y XIV del sector central de la judería. El análisis de las estructuras exhumadas y de los materiales arqueológicos hallados en contexto, especialmente cerámicos y arqueozoológicos, permite establecer consideraciones relevantes en lo relativo a los procesos de ocupación y abandono del barrio y en lo que respecta a la cuestión central de la identificación arqueológica de las comunidades judías bajomedievales.     Recovery of eutrophized marine ecosystems using the European flat oyster, Ostrea edulis

    The development of tourism and intensification of agriculture has released large amounts of nitrogen and phosphorus into the Mar Menor coastal lagoon in South-east Spain, resulting in a phytoplankton bloom in 2016. This bloom turned the clear and transparent waters turbid and greenish, and killed approximately 85% of benthic macrophytes. Nutrient bioextraction by flat oysters, Ostrea edulis, has been proposed for remediation of these eutrophication events and water quality recovery. This research aims to quantify the clearance rate and investigate the genetic origin of Mar Menor oysters under eutrophized conditions for potential applications to bioremediation projects. Oligotrophic and eutrophic conditions were replicated in the laboratory, and oyster feeding behaviour (i.e. clearance rates, ingestion rates, absorption efficiency and absorption rates) were studied using a flow-through system. The genetic characterization of oysters showed no significant difference between individuals from the Mar Menor and individuals collected from a nearby Mediterranean bed (Tabarca Island). Based on the physiological results observed, oysters were grouped into high-feeder (HF) and low-feeder (LF) categories according to their clearance rate, that was 3-fold higher in the HF group. Different responses in feeding behaviour were observed under eutrophic conditions in both oyster groups. Constraints in the absorption capacity of LF oysters seemed to be related to their reduced filtering activity. Lower body condition of LF oysters was evidenced by their negative scope for growth value. From this work, several conclusions can be drawn for future restoration/bioextraction actions: (i) the recovery of half of the oyster population that existed in the past would act as an effective top-down control on the phytoplankton community; (ii) using clearance rate measurements is recommended to select oysters as broodstock; and (iii) Mediterranean oysters could be used as broodstock in the event that Mar Menor oysters fail to reproduce.We appreciate the work done by Francisco Gomez in the maintenance and feeding of the oyster and support on the physiological measurements.     A comparison of Thellier-type and multispecimen paleointensity determinations on Pleistocene and historical lava flows from Lanzarote (Canary Islands, Spain)

    Sixteen Miocene, Pleistocene, and historic lava flows have been sampled in Lanzarote (Canary Islands) for paleointensity analysis with both the Coe and multispecimen methods. Besides obtaining new data, the main goal of the study was the comparison of paleointensity results determined with two different techniques. Characteristic Remanent Magnetization (ChRM) directions were obtained in 15 flows, and 12 were chosen for paleointensity determination. In Thellier-type experiments, a selection of reliable paleointensity determinations (43 of 78 studied samples) was performed using sets of criteria of different stringency, trying to relate the quality of results to the strictness of the chosen criteria. Uncorrected and fraction and domain-state corrected multispecimen paleointensity results were obtained in all flows. Results with the Coe method on historical flows either agree with the expected values or show moderately lower ones, but multispecimen determinations display a large deviation from the expected result in one case. No relation can be detected between correct or anomalous results and paleointensity determination quality or rock-magnetic properties. However, results on historical flows suggest that agreement between both methods could be a good indicator of correct determinations. Comparison of results obtained with both methods on seven Pleistocene flows yields an excellent agreement in four and disagreements in three cases. Pleistocene determinations were only accepted if either results from both methods agreed or a result was based on a sufficiently large number (n>4) of individual Thellier-type determinations. In most Pleistocene flows, a VADM around 5 31022 Am2 was observed, although two flows displayed higher values around 931022 Am2.project CGL2012-32149 (Ministerio de Econom ıa y Competitividad, Spain), project 320/2011 (Ministerio de Medio Ambiente y Medio Rural y Marino, Spain) and the European Regional Development Fund (ERDF)

    Selected Schizosaccharomyces pombe Strains Have Characteristics That Are Beneficial for Winemaking

    At present, wine is generally produced using Saccharomyces yeast followed by Oenococus bacteria to complete malolactic fermentation. This method has some unsolved problems, such as the management of highly acidic musts and the production of potentially toxic products including biogenic amines and ethyl carbamate. Here we explore the potential of the fission yeast Schizosaccharomyces pombe to solve these problems. We characterise an extensive worldwide collection of S. pombe strains according to classic biochemical parameters of oenological interest. We identify three genetically different S. pombe strains that appear suitable for winemaking. These strains compare favourably to standard Saccharomyces cerevisiae winemaking strains, in that they perform effective malic acid deacidification and significantly reduce levels of biogenic amines and ethyl carbamate precursors without the need for any secondary bacterial malolactic fermentation. These findings indicate that the use of certain S. pombe strains could be advantageous for winemaking in regions where malic acid is problematic, and these strains also show superior performance with respect to food safety

El mérito del libro radica en una triple condición: alimenta la investigación académica ecuatoriana, contribuye a llenar el vacío de producción científica automotriz direccionada a las necesidades del Ecuador y reconoce el esfuerzo de los investigadores que se dedican a la producción académica técnica. La Universidad Politécnica Salesiana —en su sede Guayaquil— realizó en 2018, las Segundas Jornadas Científicas de Ingeniería Automotriz; este texto es el producto final de ese evento académico, cuyas memorias técnicas son constituidas por ocho resultados de investigaciones en Ingeniería Automotriz que aportarán desarrollo sostenible al Ecuador en áreas como: el diseño, el control de contaminación, la eficiencia energética y la movilidad.     Key Factors Associated With Pulmonary Sequelae in the Follow-Up of Critically Ill COVID-19 Patients

    Introduction: Critical COVID-19 survivors have a high risk of respiratory sequelae. Therefore, we aimed to identify key factors associated with altered lung function and CT scan abnormalities at a follow-up visit in a cohort of critical COVID-19 survivors. Methods: Multicenter ambispective observational study in 52 Spanish intensive care units. Up to 1327 PCR-confirmed critical COVID-19 patients had sociodemographic, anthropometric, comorbidity and lifestyle characteristics collected at hospital admission; clinical and biological parameters throughout hospital stay; and, lung function and CT scan at a follow-up visit. Results: The median [p25–p75] time from discharge to follow-up was 3.57 [2.77–4.92] months. Median age was 60 [53–67] years, 27.8% women. The mean (SD) percentage of predicted diffusing lung capacity for carbon monoxide (DLCO) at follow-up was 72.02 (18.33)% predicted, with 66% of patients having DLCO < 80% and 24% having DLCO < 60%. CT scan showed persistent pulmonary infiltrates, fibrotic lesions, and emphysema in 33%, 25% and 6% of patients, respectively. Key variables associated with DLCO < 60% were chronic lung disease (CLD) (OR: 1.86 (1.18–2.92)), duration of invasive mechanical ventilation (IMV) (OR: 1.56 (1.37–1.77)), age (OR [per-1-SD] (95%CI): 1.39 (1.18–1.63)), urea (OR: 1.16 (0.97–1.39)) and estimated glomerular filtration rate at ICU admission (OR: 0.88 (0.73–1.06)). Bacterial pneumonia (1.62 (1.11–2.35)) and duration of ventilation (NIMV (1.23 (1.06–1.42), IMV (1.21 (1.01–1.45)) and prone positioning (1.17 (0.98–1.39)) were associated with fibrotic lesions. Conclusion: Age and CLD, reflecting patients’ baseline vulnerability, and markers of COVID-19 severity, such as duration of IMV and renal failure, were key factors associated with impaired DLCO and CT abnormalities

    CIBERER : Spanish national network for research on rare diseases: A highly productive collaborative initiative

    Altres ajuts: Instituto de Salud Carlos III (ISCIII); Ministerio de Ciencia e Innovación.CIBER (Center for Biomedical Network Research; Centro de Investigación Biomédica En Red) is a public national consortium created in 2006 under the umbrella of the Spanish National Institute of Health Carlos III (ISCIII). This innovative research structure comprises 11 different specific areas dedicated to the main public health priorities in the National Health System. CIBERER, the thematic area of CIBER focused on rare diseases (RDs) currently consists of 75 research groups belonging to universities, research centers, and hospitals of the entire country. CIBERER's mission is to be a center prioritizing and favoring collaboration and cooperation between biomedical and clinical research groups, with special emphasis on the aspects of genetic, molecular, biochemical, and cellular research of RDs. This research is the basis for providing new tools for the diagnosis and therapy of low-prevalence diseases, in line with the International Rare Diseases Research Consortium (IRDiRC) objectives, thus favoring translational research between the scientific environment of the laboratory and the clinical setting of health centers. In this article, we intend to review CIBERER's 15-year journey and summarize the main results obtained in terms of internationalization, scientific production, contributions toward the discovery of new therapies and novel genes associated to diseases, cooperation with patients' associations and many other topics related to RD research

    Naturaleza y cultura en Ámerica Latina

    La concreción del XVIII Foro de Estudiantes Latinoamericanos de Antrología y Arqueología: Cultura y naturaleza en América Latina: escenarios para un modelo de desarrollo no civilizatorio, efectuado en Quito desde el 17 al 23 de julio del 2011, se constituyó en un acontecimiento sumamente significativo para la antropología latinoamericana debido a dos motivos. Primero porque coincidió con la emergencia del movimiento universitario estudiantil latinoamericano que expresaba sus tendencias, propuestas y exigencias de cambios tanto de las prácticas académicas como de los patrones civilizatorios que rigen las relaciones actuales. Segundo, porque se inscribía en un contexto de consolidación de las nuevas democracias de los países andinos, de carácter antineoliberal y basadas en los sujetos de derecho entre los cuales se incluye la naturaleza. Estos contextos determinaron que el Foro no ponga en escena certidumbres teóricas o metodológicas, ni se preste al exhibicionismo estéril de los avances disciplinares. Más bien, la convocatoria de la antropología y la arqueología fue apenas un pretexto para hablar, con su lenguaje, de nosotros mismos, de lo que somos, de lo que pensamos, de lo que aspiramos y sentimos sobre nuestra Latinoamérica. Lo que hemos visto, oído y compartido, en realidad, no han sido solamente ideas o conceptos sino opciones y toma de posiciones respecto a múltiples encrucijadas. Posición ante situaciones que amenazan la vida, la justicia y los derechos de todos, un desafío epistemológico todavía en ciernes y que no termina de cuajar aún en nuestras prácticas académicas
